대량발효 후 미세여과막과 MWCO 100K, 50K의 한외여과막을 이용한 막분리시스템을 적용하여 MTGase를 분리한 결과 pore size 1.6, $0.7\;{\mu}m$의 cellulose fiber 재질의 예비여과막과 $0.45\;{\mu}m$ cellulose acetate재질의 미세여과막을 이용하여 얻어진 분리액을 MWCO 100K와 50K를이용하여 농축시킨 결과 enzyme의 농도가 1.29 units/ml, 효소 비활성도는 약 0.2 units/mg protein 으로 나타났으며 초기 배양액에 비해 3.7배의 농축효과를 보였다.
회분식 포말분리관에 효모용액을 충전하여 효모분리실험을 수행하며 효모 액면 높이, 질소 공급량, 효모농도, pH와 온도 및 $CaCl_2$ 첨가 등의 변화에 따른 포말 분리관의 분리 및 특성 등을 연구하였다. 액면 높이가 20cm인 경우 최대 효모분리비 70.6을 얻을 수 있었고 포말 생성시간도 6분으로 가장 길었다. 질소 공급량이 120ml/min인 경우 3분 분리운전후 액본체 효모의 90~94%가 제거되어 매우 신속하고 효과적인 분리법이었다. 효모농도가 3.92~8.4g/l 범위가 pH가 5~7 범위에서 분리시간에 따른 효모 제거율은 비슷하였다. 효모용액의 온도가 26~$36^{\circ}C$ 범위에서 온도가 상승할수록 분리시간에 따른 효모 분리비가 증대하였다. 효모의 응집성을 증대하는 $CaCl_2$ 농도는 0.2g/l일 때 효모 분리비가 가장 높았다.
In this study, various pre-treatment methods were evaluated for microalgae separation. These methods aimed to facilitate safe, rapid, and cost-effective online imaging for real-time observation and cell counting. As pre-treatment techniques, heating, chemical hydrolysis, heating combined with chemical hydrolysis, and sonication were employed. The effectiveness of these methods was evaluated in the context of online imaging quality through experimentation on cultivated microalgae (Chlorella vulgaris and Scenedesmus quadricauda). The chemical treatment method was found to be inappropriate for improving image acquisition. The heating pre-treatment method exhibited a drawback of prolonged cell dispersion time. Additionally, the heating combined with chemical hydrolysis method was confirmed to have the lowest dispersion effect for Chlorella vulgaris. Conversely, ultrasonication emerged as a promising technique for microalgae separation in terms of repeatability and reproducibility. This study suggests the potential for selecting optimal pre-treatment methods to effectively operate real-time online monitoring devices, paving the way for future research and applications in microalgae cultivation and imaging.
Cell recovery from high cell density broths of Alcaligenes eutrophus by pretreatment with aluminum-based coagulants such as aluminum sulfate, polyaluminum hydrooxide chloride silicate (PACS), and polyaluminum hydrooxide chloride (Hi-PAX) was carried out. Cells coagulated with coagulants could be successfully recovered above 95-99% by centrifugation or filtration. The optimum initial pH of fermentation broths for cell recovery was in the range of 10 to 12. Optimum coagulants dosage for cell recovery increased with increasing of cell concentrations (21-160 g/L). The optimum coagulant dosages to recover cells with more than 95% cell recovery by centrifugation for the cell concentrations ranged 21-160 g/L were as follows: aluminum sulfate, 416-1708 mg Al/L; PACS, 211-826 mg Al/L; Hi-PAX, 320-960 mg Al/L. At optimum conditions for the coagulation of cells, centrifugal forces for 95% of cell recovery were dependent on the cell concentration. The centrifugal forces at 82 g/L and 160 g/L of cell concentration were only 45${\times}$g and 1600${\times}$g, respectively.
커들란은 용액의 pH 7.0에서 salt 상태로 존재하기 때문에 커들란만이 액상에 녹아 있는 경우에는 분리공정은 원심분리를 함으로써 분리가 가능하다. 그러나, 세포외 다당류로 생산된 커들란읜 세포, 염 의 무기물이 섞여 있기 때문에 이러한 물질을 제거하는 것이 커들란 분리의 중요한 변수가 되었다. 커들란의 생산을 위한 분리에 관한 보고는 원심분리 방법을 이용한 것으로 알려져 있음 세포 및 불순물 등을 제거하여 순수한 커들란을 분리하기 위해 pH swing separation (pH가 높은 경우 커들란이 용액에 녹는 성질을 이용)을 이용하였으나 원심분리를 수 차례 반복해야 되기 때문에 소요되는 동력이 많이 필요하여 에너지 절감형 및 오염절감형의 새로운 공정의 개발이 필요하게 되었다. 온 연구에서는 pH swing separation 에서 동력비가 많은 드는 단점을 감소시키기위하여 커들란의 물리 화학적인 특성을 고려한 분리공정을 개발하였다. 향후 공정 개선을 위한 지속적인 연구가 필요하며 완성시 경제적이 공정을 개발하는 것이 가능할 것이다.
Single-cell research has provided a breakthrough in biology to understand heterogeneous cell groups, such as tissues and organs, in development and disease. Molecular barcoding and subsequent sequencing technology insert a single-cell barcode into isolated single cells, allowing separation cell by cell. Given that multimodal information from a cell defines precise cellular states, recent technical advances in methods focus on simultaneously extracting multimodal data recorded in different biological materials (DNA, RNA, protein, etc.). This review summarizes recently developed single-cell multiomics approaches regarding genome, epigenome, and protein profiles with the transcriptome. In particular, we focus on how to anchor or tag molecules from a cell, improve throughputs with sample multiplexing, and record lineages, and we further discuss the future developments of the technology.

Tissue regeneration is one of the ultimate goals of maxillofacial surgery and various types of tissue engineering technologies have been utilized in clinics. Healthy resources of host cells and growth factors are essential for the tissue engineering, therefore autologous blood-derived cell therapy was introduced. In this article, clinical applications of the autologous platelet concentrates and stem cell separation therapy will be summarized and evaluated for their efficacy and feasibility in the current maxillofacial clinics.
The production of recombinant proteins in Escherichia coli often leads to the formation of an intracellular inclusion body. Key process steps that can determine the economics of large-scale protein production from inclusion bodies are fermentation, inclusion body recovery, and protein refolding. Compared with protein refolding and fermentation, inclusion body recovery has received scant research attention. Nevertheless, it can control the final product yield and hence process cost for some products. Optimal separation of inclusion bodies and cell debris can also aid subsequent operations by removing contaminant particulates that foul chromatographic resins and contain antigenic pyrogens. In this review, the properties of inclusion bodies and cellular debris are therefore examined. Attempts to optimise the centrifugal separation of inclusion bodies and debris are also discussed.
Methodologies to improve photovoltaic performance of dye-sensitized solar cell (DSSC) are reviewed. DSSC is usually composed of a dye-adsorbed $TiO_2$ photoanode, a tri-iodide/iodide redox electrolyte and a Pt counter electrode. Among the photovoltaic parameters of short-circuit photocurrent density, open-circuit voltage and fill factor, short-circuit photocurrent density is the collective measure of light harvesting, charge separation and charge collection efficiencies. Internal quantum efficiency is known to reach almost 100%, which indicates that charge separation occurs without loss by recombination. Thus, light harvesting efficiency plays an important role in improvement of photocurrent. In this paper, technologies to improve light harvesting efficiency, including surface area improvement by nano-dispersion, size-dependent light scattering efficiency, bi-functional nano material, panchromatic absorption by selective positioning of three different dyes and transparent conductive oxide (TCO)-less DSSC, are introduced.
This paper presents a set of candidate regulatory requirements for TV band devices (TVBDs) in the Rep. of Korea. To guarantee the protection of incumbent services, especially digital TV (DTV) and wireless microphones, in TV frequency bands, we suggest minimum separation distances of TVBDs from the noise-limited contour according to incumbent users and TVBD types. This paper also deals with multiple sets of separation distances of a co-channel TVBD network from a DTV protected contour on the basis of the radio propagation characteristics of different geographic areas to make good use of TV white space (TVWS) and safely protect the DTV service. We present a low-power transmission mode of TVBDs and the relevant separation distances for small-cell deployment. The service coverage reduction ratio of a TVBD network is investigated in the presence of DTV interference in four geographic areas. The TVWS field verification results, conducted on the island of Jeju (Rep. of Korea), show that incumbent services operate well without harmful interference from neighboring TVBDs with the proposed separation distances.
