Determination of Semen Quality and Antibacterial Susceptibility Pattern of Bacteria Isolated from Semen of Iraqi Subjects

  • Infertility is a key issue affecting mood and behavior in men. Microorganisms are one of the primary etiological agents that may be associated with infertility. The objective of the present study was to identify bacterial causative agents from the semen of infertile subjects and determine the effect of bacterial infection on sperm quality, as well as determine the susceptibility of these bacteria to drugs. Forty semen samples from 30 infertile patients and 10 fertile individuals were collected. The pH, volume, motility, and concentration of semen were analyzed. The samples were processed and identified by biochemical testing using API identification kits. The antibiotic susceptibility pattern was determined using the disc diffusion method. Abnormal sperm quality was observed. The mean age of the individual and their sperm morphology, concentration, progressive motility, pH level, and pus cell content were 31.9 years, 2.7%, 10.4 million/ml, 27.3%, 8.3, and 5.7, respectively. Among the tested samples, oligoasthenozoospermia was found to show the highest occurrence, at 27/30 samples, followed by teratozoospermia, at 25/30 samples, and asthenozoospermia, at 22/30 samples. Of the tested infertile patients' sperm, 19, 6, and 5 isolates were identified as Escherichia coli, Klebsiella pneumonia, and Staphylococcus epidermidis, respectively. The results also revealed multi-drug resistance in the bacteria. Compared to that shown by the other tested antibiotics, amikacin showed higher activity against all isolated bacteria. However, the bacteria exhibited maximum resistance against gentamicin, cefotaxime, levofloxacin, and ampicillin. In conclusion, leukocytospermia and bacterial infections are possibly responsible for sperm abnormalities. Multi-drug resistant bacteria were detected. Gentamicin, cefotaxime, levofloxacin and ampicillin were shown the highest resistance, while amikacin was the most effective antimicrobial agent against the isolated bacteria.

Prevalence of Multi-Antibiotic Resistant Bacteria Isolated from Children with Urinary Tract Infection from Baghdad, Iraq

  • Urinary tract infections (UTIs) are one of the most common infections in different age groups, including children. Bacteria are the main etiological agents of UTIs. The aim of the present study was to isolate, identify, and determine the antibiotic susceptibility of bacteria isolated from children with UTIs from Baghdad, Iraq. Three hundred and two urine samples were collected from children aged 6 months to 12 years. The samples were cultured on blood agar and MacConkey agar. The selected colonies were subjected to biochemical tests and antibiotic susceptibility analysis using the Vitek® 2 Compact automated microbial identification system. In this sample, 299 bacteria were identified, of which, 267 were gram-negative bacteria, and 32 were gram-positive bacteria. Escherichia coli (56%) was the most commonly isolated gram-negative bacteria, followed by Pseudomonas aeruginosa (14%), Enterobacter spp. (10.48%), Klebsiella pneumoniae (9.36%), Proteus spp. (7.8%), Acinetobacter baumannii (1.5%), and Morganella morganii (0.37%). Enterococcus faecalis (62.5%) was the most commonly detected gram-positive bacteria, followed by Staphylococcus aureus (37.5%). E. coli and P. aeruginosa were the most antibiotic-resistant bacteria. Among the tested antibiotics, meropenem showed 100% sensitivity, followed by imipenem (97.4%), amikacin (91.8%), and tobramycin (83.5%). In contrast, the high frequencies of resistance were observed with cefixime (93.2%), cefotaxime (78.7%), and ceftriaxone/cefotaxime (71.2%). In conclusion, carbapenems and aminoglycosides are highly recommended for the empirical treatment of UTIs, while, Quinolones, penicillins, and cephalosporins are not suggested. Frequent antibiotics susceptibility testing are warranted to determine the resistance pattern of UTI bacteria.

Isolation and Identification from Korean Olive Flounders (Paralichthys olivaceus) Showing Abnormal Swimming Behavior, and Sstudy of Antibiotic Susceptibility (이상유영 증상 국내산 양식 넙치에서의 슈도알테로모나스 분리·동정 및 항생제 감수성 연구)

  • Olive flounders which were cultured in commercial fish farm showed abnormal swimming behavior in November 2020. Then, gradual mortality was observed in the fish farm. During the diagnosis, bacterial strain KNCFKW-PN1 was isolated from the kidney of the dead fish. Based on the sequence of gyrase B subunit gene, KNCFKW-PN1 was proved to be Pseudoalteromonas nigrifaciens, showing 99.59% nucleotide identity with that of P. nigrifaciens LMG 2227T. According to the result of antibiotic susceptibility test, P. nigrifaciens KNCFKW-PN1 showed intermediate resistance to ciprofloxacin, and was resistant to ampicillin, cefepime, cefotaxime, ceftazidime, and amikacin. This is the first report of the isolation of multiple-antibiotic-resistant P. nigrifaciens from olive flounder.

Causative Agents and Antimicrobial Sensitivity of Neonatal Sepsis : Ten-year Experience in One Neonatal Intensive Care Unit (단일 신생아중환자실에서 경험한 10년간의 신생아 패혈증의 원인균 및 항생제 감수성 변화)

  • Purpose: To identify trends in causative bacterial organisms for neonatal sepsis and antimicrobial susceptibilities over 10 years in one neonatal intensive care unit. Methods: We retrospectively reviewed the cases of culture-proven neonatal sepsis between January 1998 and December 2007. The 10-year period was divided into two phases (phase I, 1998-2002; phase II, 2003-2007) to distinguish the differences during the entire period. Results: Total 350 episodes of neonatal sepsis were identified in 315 neonates. The common pathogens of early-onset sepsis were S. epidermidis, S. aureus, P. aeruginosa, and E. cloacae in phase I, and S. epidermidis and E. cloacae in phase II. In cases of late-onset sepsis, coagulase negative Staphylococcus, S. aureus, and K. pneumoniae were isolated frequently in both phases. The incidence of sepsis caused by multi-drug resistant organisms decreased with strict infection control. Gram positive organisms showed 0-20% susceptibility to penicillin, ampicillin, and cefotaxime in both phases. Sensitivity to amikacin for Enterobacter spp. increased, whereas P. aeruginosa showed decreased sensitivity in phase II. Between 50% and 60% of other gram negative bacteria, except P. aeruginosa, were susceptible to cefotaxime in phase II in contrast to phase I. Greater than 80% of gram negative bacteria were sensitive to imipenem except P. aeruginosa and ciprofloxacin in both phases. Conclusion: The trend in causative microorganisms and antimicrobial susceptibilities can be used as a guideline for selection of appropriate antibiotics. A particular attention should be paid to infection control, especially to reduce sepsis caused by multi-drug resistant organisms.

In vitro Antibacterial Activity of DWC-751, n New Parenteral Cephalosporin (신규 세파로스포린 항생제 DWC-751의 in vitro 항균작용)

  • We compared in vitro antibacterial activity of DWC-751, a new parenteral cephalosporin antibiotic, with those of cefpirome (CPR), cefotaxime (CTX) and ceftazidime (CAZ). DWC-751 showed a broad antimicrobial spectrum against Gram-positive and negative bacteria. The antibacterial activity of DWC-751 against Stapylococcus aureus was equal to that of CPR and superior to those of CTX and CAZ. The activity of it against Excherichia coli was more potent than those of CPR, CTX and CAZ. Against Pseudomonas aeruginosa, DWC-751 was slightly inferior to that of CAZ and superior to those of CPR and CTX. The antibacterial activity of DWC-751 was superior to those of CPR, CTX and CAZ against clinical isolates and ofloxacin resistant strains. DWC-751 showed bactericidal action against Escherichia coli at concentrations close to the MIC and induced the formation of filament and burge and lysis of Escherichia coli in a microscopic examination.

Effect of Vaccination and Mass Medication for the Prevention of Shipping Fever in Korean Native Cow (한우에서 수송열의 예방을 위한 백신접종과 예방치료의 효과)

  • The objective of this study was to compare occurrence of the shipping fever associated with vaccination and mass medication in Korean Native Cow. Lack of vaccination (Pasteurella haemolytica, infectious bovine rhinotracheitis, bovine viral diarrhea, parainfluenza 3, and bovine respiratory syncytial virus) and mass medication before and after transportation, 67% of gram-negative cocci were detected. However, with vaccination and mass medication, only 33% of Gram-negative cocci were detected. When antimicrobial susceptibility for the detected bacterium was tested, apramycin, ampicillin, amilacin, clindamycin, gentamycin, kanamycin, penicillin, and streptomycin were resistant, whereas tylocin, amoxicillin, linocospetin, nalidixic acid, norfloxacin, oxalinic acid, and cefotaxime were susceptible. Morbidity and average therapeutic effect were 33% and 1.0time with vaccination and mass medication and were 78% and 5.3 times without vaccination and mass medication, respectively. Therefore, the results suggest that shipping fever would be considerably decreased with vaccination including mass medication before and after transportation.

Stability of Paclitaxel with Cephalosporines in $0.9\%$ Sodium Chloride Injection and $5\%$ Dextrose Injection During Simulated Y-Site Administration (Y-Site 동시투여 동안 $0.9\%$ 생리식염수와 $5\%$ 포도당 용액에서의 Paclitaxel과 Cephalosporines의 안전성 연구)

  • Paclitaxel과 cephalosporines(제 1세대인 ceftezole sodium과 cephradine, 제2세대인 cefamandole sodium과 cefmetazole sodium, 제3세대인 cefoperazone sodium과 cefotaxime sodium 그러고 제4세대인 cefepime hydrochloride)을 $5\%$포도당주사액 그리고 $0.9\%$ 염화나트륨주사액과 함께 Y-Site 장치를 써서 환자에게 주입할 때 paclitaxel의 안정성에 관하여 연구하였다. Paclitaxel 0.3 mg/ml 및 1.2 mg/ml과 cephalosporines 20 mg/m을 각각 1 : 1로 혼합한 후 0, 1, 2, 4, 12시간 시점에서 paclitaxel의 농도를 HPLC로 분석하였다. 방해물질에 의한 분석오차를 줄이기 위해 분석법을 여러 상태에서 확인하였으며, 각 농도에서 3차례씩 실험하였고 각 샘플은 2차례 반복하여 HPLC로 분석하였다. 분석전에 각 시료의 투명도, 색의 변화, 침전상태 및 pH를 검사하였다. Paclitaxel 0.3 mg/ml 및 1.2 mg/ml와 cephalosporines 20 mg/ml를 각각 혼합하였을 때 12시간 동안 안정하였으며, 주사액의 혼탁이나 색의 변화 및 침전은 나타나지 않았으며 pH도 변하지 않았다.

  • 새로 합성된 6-exomethylene penamsulfone derivatives 7종에 대해 In Vitro antibacterial activities를 Broth Microdilution법으로 Sulbactam-Na(SAM), Clavulanic acid-K(CLA)와 Cefotaxime-Na(CFT), Ampicillin Trihydrates(AMP), Amoxicillin-Na(AMX), Cefoperazone(CFZ)을 병용투여하여 $\beta$-lactamase 생성 균주(30 여종)에 실시, MIC를 구하였다. 균주에 따라 SAM, CLA에 비해 CH1145, CH1155 등이 우수한 효과를, CH1140, CH2150등이 유사한 효과를 보였다.

Antibacterial and Synergistic Activity of Isocryptomerin Isolated from Selaginella tamariscina

  • We investigated novel antibacterial and synergistic activities of isocryptomerin isolated from Selaginella tamariscina. Isocryptomerin showed potent antibacterial activity against Gram-positive and Gram-negative bacterial strains including clinical isolates of antibiotic-resistant species such as methicillin-resistant Staphylococcus aureus(MRSA). Additionally, we further investigated the synergistic activity of isocryptomerin with a conventional antibiotic against MRSA. The result indicated that isocryptomerin had considerable synergistic activity in combination with cefotaxime. In summary, the present study suggests that isocryptomerin may have potential as a novel therapeutic agent for treatment of infectious diseases by not only human pathogenic bacteria but also multidrug-resistant bacteria.
