Six Sigma Analysis of Vitamin D Measurement Using External Quality Assessment Program (외부정도관리 프로그램을 이용한 비타민 D 검사의 6 시그마 분석)

  • Standardization of vitamin analysis continues around the world, and much effort has been made to improve the accuracy of the results. This study analyzed the sigma metrics of the vitamin D test using the external quality assessment (EQA) program. Sigma metrics is used for quantitative tests performed in the laboratory, and the test results can be objectively visualized in terms of quality. This analysis was performed based on the accuracy of the College of American Pathologists (CAP) using the results of the 2019 accuracy-based vitamin D (ABVD) survey, and about 300 laboratories participated in the survey. Reference values were obtained by the Center for Disease Control and Prevention (CDC) reference laboratory. At six different concentrations, the sigma metrics were analyzed to be 1.00, 1.85, 2.42, 1.01, 1.54 and 0.78, respectively. An average of 1.43 sigma metrics was determined. In particular, only positive biases for ABVD-16 and 17 were shown in the liquid chromatography tandem-mass spectrometry (LC-MS/MS), which is the standard method for vitamin D determination when compared to the reference values. The causes of the difference can be explained by cross reactivity to various vitamin D metabolites. Laboratories need to improve their overall performance.

Antibacterial Properties of Pit and Fissure Sealant Containing S-PRG filler on Streptococcus mutans (S-PRG filler를 포함한 치면열구전색제의 Streptococcus mutans에 대한 항미생물 특성에 관한 연구)

  • The purpose of this study was to evaluate the antibacterial properties of a sealant containing S-PRG filler compared to those of two contemporary commercial sealants to determine the inhibition of bacterial growth in broth culture and biofilm formation using the CDC Biofilm Reactor. The BeautiSealant containing S-PRG filler, the fluoride releasing Clinpro$^{TM}$ sealant, which are known to have higher antibacterial effects, and the non-fluoride releasing Concise$^{TM}$ sealant were selected for this study. A Streptococcus mutans culture in BHI broth without sealant served as a negative control in the planktonic growth inhibition test. As a result, bacterial growth was inhibited in all three sealant groups compared to that in the control. The Clinpro$^{TM}$ sealant showed a significantly reduced number of CFUs compared to those of the BeautiSealant and Concise$^{TM}$ sealants. However, no significant difference was detected between the BeautiSealant and Concise$^{TM}$ sealants. The Clinpro$^{TM}$ sealant significantly decreased biofilm formation compared to that by the BeautiSealant and Concise$^{TM}$ sealants. No significant difference was observed between the BeautiSealant and Concise$^{TM}$ sealants. In conclusion, the sealant containing S-PRG filler had a less potent anti-bacterial property and increased biofilm formation capacity compared to those of the fluoride releasing Clinpro$^{TM}$ sealant.

Analysis of the effect of trichloroacetic acid and epidermal growth factor release on cytoskeleton gene expression using the nano-controlled releasing system (나노방출제어시스템을 이용한 trichloroacetic acid와 epidermal growth factor 방출이 세포골격형성 유전자 발현에 미치는 영향 분석)

  • Purpose: Here, we verified that the actin cytoskeletal gene expression of human gingival fibroblasts was altered by the administration of trichloroacetic acid (TCA) and epidermal growth factor (EGF) using the nano-controlled releasing system. Materials and methods: The control and experimental groups were divided into 3 groups: the group with the TCA-only nano-controlled releasing system (EXP1), the group with the TCA- and EGF nano-controlled releasing system (EXP2), and the control group (CON) with 48-h incubation. Expression of 26 genes involved in the regulation of actin cytoskeleton were analyzed by real-time PCR followed by the determination of correlations and influential factors using the Pearson correlation analysis and multiple regression analysis. Results: Among 23 genes upregulated in EXP1 and EXP2, expression of 14 genes were significantly increased in EXP2 compared to EXP1. On the other hand, LPAR1 was downregulated only in EXP1, GNA13 was upregulated only in EXP2, and F2R was downregulated only in EXP2. Three Rac1-related genes and CDC42 were identified as the influential factors of the actin gene upregulation. Conclusion: The actin cytoskeleton genes in human gingival fibroblast were upregulated by the administration of TCA and EGF using HGC-based nano-controlled releasing system.

Extraction of Antioxidants from Lonicera japonica and Sophora japonica L.: Optimization Using Central Composite Design Model (금은화와 회화나무꽃으로부터 항산화성분의 추출 : 중심합성계획모델을 이용한 최적화)

  • In this study, an antioxidant was extracted from Lonicera japonica and Sophora japonica L, which was optimized by using the central composite design (CDD) model of response surface methodology (RSM). The response value of CDC model establishes the extraction yield and the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity. The extraction time, volume ratio of ethanol/ultrapure water, and extraction temperature were selected as quantitative factors. According to the result of CDC, optimal extraction conditions of Lonicera japonica were as follows; the extraction time of 2.08 h, volume ratio of ethanol/ultrapure water of 41.53 vol.%, and extraction temperature of $55.08^{\circ}C$. At these conditions the expected results indicated that the yield and DPPH radical scavenging activity were estimated as 37.88 wt% and 40.37%, respectively. On the other hand, optimal extraction conditions of Sophora japonica L. could be found as the extraction time of 2.13 h, volume ratio of ethanol/ultrapure water of 62.89 vol.%, and temperature of $50.42^{\circ}C$. Under the conditions, the (possible) maximum values of yield and DPPH radical scavenging activity were found as 35.43 wt% and 55.7%, respectively.

Effects of Salviae miltiorrhizae Radix Extract on Gene Expression of Dendritic cells. (단삼이 수지상 세포의 유전자 발현에 미치는 영향)

  • Objectives and Methods : Salviae miltiorrhizae Radix (SMR) promotes blood circulation to remove blood stasis, cools the blood to relieve carbuncle, clears away heat from the heart and tranquilizes the mind. This study was designed to investigate the effects of SMR on immuno-potentiative action in terms of changes in the genetic profile of dendritic cells (DC) using by microarray analysis. Results and Conclusion: In this experiment, treatments with more than 250 ${\mu}g/ml$ upto 1000 ${\mu}g/ml$ of SMR elevated the proliferation rates of DC. Microscopic observations confirmed the tendency on proliferation rates. Expression levels of genes related with cellular methabolic process, cell communication, and macromolecule metabolic process were elevated by treatment with SMR in comparison of functional distribution in a Biological Process. In molecular functions, expression levels of genes related with receptor activation, nucleotide binding and nucleic acid binding were elevated. In cellular components, expression levels of genes related to cellular membrane-bound organelles were elevated. In addition, expression levels of genes related to Wnt signalling pathways and the glycerophospholipid metabolism were elevated through analysis using pathway analysis between up-and down-regulated genes in cells treated with SMR. Finally, genes related to JAK2, GRB2, CDC42, SMAD4, B2M, FOS and ESRI located the center of Protein interaction network of genes through treatment with SMR.

A Survey of the Health Risk Behaviors of Middle School Students in Kyungju (경주지역 일부 중학생의 건강관련 위험행동)

  • The purpose of this study was to collect basic data to develop school health policy and health promotion program. This survey measured the levels of risk behaviors in six categories in a middle school students in Kyungju(n=490). The data was collected from November to December 2000. It was done using a 1999 version of the Youth Risk Behavior Survey of CDC, which was translated and make a partial modification. The major results were as follows. Many middle school students engaged in behaviors that will increase the likelihood of death or illness. 1) A high percentage of middle school students engaged in behaviors that contribute to unintentional injuries and school-related violence (42.1 -78%) 2) Students who perceived their school life are happy experienced more depression(p=.000) and Students who perceived their school and family life are so-and-so seriously considered attempting suicide(p = .000) than other subgroup. 3) In spite of they were normal weight, 14.5% of students considered themselves overweight. 4) There was significantly difference or correlation between health risk behaviors and grade, parents educational level perception of school life and performance, perception of family life and religion. Based on this results of study, it is necessary to develop school health program and school health policy to prevent health risk behaviors and improve health promotion especially considering characteristics of middle school students.

Activation of MAP Kinase during Maturation in Porcine Ooctyes (돼지 미성숙란의 체외배양시 MAP Kinase의 활성)

  • In an attempt to evaluate the function of MAP kinase of porcine oocytes and to develop a method of assessment for kinase activity, we used MBP as a substrate to detect the MAP kinase activity of porcine oocytes matured in in vitro. The MAP kinase which had lower activity during the first 20 hours of culture started to show an increased amount of activity at 25 hours at which a collapse in nuclear membrane was induced. Significant (P<0.05) a, pp.ared at 30 hours of being cultured. The gel phosphorylation method, MBP which has been known to be a substrate for kinase such as cdc2 kinase, was phosphorylated at two positions corresponding to ERK 1 (44kDa) and ERK2 (42 kDa) which are known as mammalian MAP kinase. The existence of MARKK and MAP kinase were identified with western blotting at 0 hour culture of immature GV oocytes. The amount of those proteins did not increase during 40 hours of culture, which suggest that the increase of MAP kinase activity was caused by phosphorylaton rather than due to change in protein amount. MAPKK and MAP kinase were shown to be dephosporylated with deactivated at M 1 stage by inhibition of protein synthesis with cycloheximide added at the strat following the cultrue. We have reulsts that indicate the existedence of MAP kinase cascade which was activated simultaneously with start of porcine oocyte maturation (GVBD).

  • 불소의 적절한 활용을 통하여 미국인들의 구강건강이 확실하게 증진되었으며 수돗물의 인공적인 불소첨가 사업은 지난 2000년 동안에 건강과학이 이룩한 가장 획기적인 건강관리 예방사업으로 인정되었다. 음료수에 불소를 적정량 첨가하여 치아우식증을 퇴치하기 위하여 적극적 예방책을 마련함으로써 치과의학의 과학적인 연구의 기틀을 확고히 하여 치과의학이 현대건강과학의 지도적 위치를 확보하는 과정에 중요한 임무를 담당했다. 그러므로 미국의 중앙건강관리 및 예방 연구소는 정기적으로 불소사용과 관련된 연구 업적들에 대하여 관련 분야의 세계적인 학자들로 하여금 조리 정연하게 주기적으로 분석 검토하여 구강건강을 향상시킬 수 있는 방안을 제시하기 위하여 불소사용법에 대한 안내서를 마련하고 이다. 미국 중앙 질병 관리 및 예방연구소는 평생을 불소에 관한 연구를 하고 있는 11명의 전문가들과 긴밀한 연락을 취하면서 22명의 과학자들과 함께 그 동안에 발표된 관계문헌 270개를 분석 검토하여 2년 동안에 걸쳐 작성한 최근의 불소 사용 안내서를 지난 2001년 8월 17일 발표했다. 그 내용이 중요하고 한국의 치과의사들에게 큰 도움이 되리라고 확신하기 때문에 CDC의 허락을 받고 한글판을 작성하게 되었다. 한국의 관심 있는 독자들의 이해를 돕기 위하여 더욱 자세한 내용을 알고자 하는 분들이 실무진에 참여한 연구자들 및 과학자들과 직접 접촉할 수 있도록 그들의 직책과 현재의 소속을 밝혔으며 이 안내서에 사용된 문허 270개 참고문헌 전체를 소개한다.

故障許容電算體系의 設計와 信賴度

  • 전산기의 신뢰도(reliability)라 함은 사용자가 제출한 입력에 대하여 전산 기가 제공하는 결과의 신빙성의 척도라할 수 있는데, 이것은 주어진 전산기의 부 분품 하나하나가, 그리고 프로그램의 하나하나의 instruction이 설계당시에 목적한 성능을 얼마나 잘 유지하고 있는가를 측정하는 척도라고 볼 수 있습니다. 이 신 뢰도는 전산기의 수명, 필요할 때 전산기가 가동할 확율, 또는 전산기의 성능으로 나타낼 수 있습니다. 제2세대 이전의 전산기들에서는 전자공업과 전산기 기술의 불충분한 발전으로 인하여 비용과 기계의 크기의 한정 때문에 신뢰도 향상을 위 한 대책이 거의 없었습니다. 따라서 현재 볼 수 있는 American Air Line의 SABRE(Semi Automatic Business Research Environment), Bell 전화 연구소의 ESS-I, II, III(Electronic Switching System), IBM의 FMS(Future Manufacturing System)과 같은 real-time 씨스템으로서의 응용분야의 개발은 상 당히 어려운 문제였습니다. 그러나 전자공업의 비약적인 발전에 힘입어 금세대의 범용전산기의 설계가 가능하게 되었고, 오퍼레이팅 씨스템의 발전으로 인하여 multiprogramming, time-sharing, real-time 씨스템 등의 응용분야의 개발이 활발 하게 되었습니다. 이러한 응용분야의 활발한 개발과, 대규모 집적회로 (LSI)의 개 발로 ROM(Read Only Memory)의 가격화, 그리고 microprogram의 보급 등으로 특수 목적의 time sharing operation을 위한 소형 전산기가 발전하게 되었으며 종 래의 범용 전산기 대신에 CDC의 string unit과 pipeline을 이용한 STAR 100과 일리노이 대학의 256processor와 Burrough의 B6500로 구성된 ILLIAC-IV와 같은 초대형 전산기가 등장하게 되었습니다.

Knockdown of Pyruvate Kinase M Inhibits Cell Growth and Migration by Reducing NF-κB Activity in Triple-Negative Breast Cancer Cells

  • Altered genetic features in cancer cells lead to a high rate of aerobic glycolysis and metabolic reprogramming that is essential for increased cancer cell viability and rapid proliferation. Pyruvate kinase muscle (PKM) is a rate-limiting enzyme in the final step of glycolysis. Herein, we report that PKM is a potential therapeutic target in triple-negative breast cancer (TNBC) cells. We found that PKM1 or PKM2 is highly expressed in TNBC tissues or cells. Knockdown of PKM significantly suppressed cell proliferation and migration, and strongly reduced S phase and induced G2 phase cell cycle arrest by reducing phosphorylation of the CDC2 protein in TNBC cells. Additionally, knockdown of PKM significantly suppressed $NF-{\kappa}B$ (nuclear factor kappa-light-chain-enhancer of activated B cells) activity by reducing the phosphorylation of p65 at serine 536, and also decreased the expression of $NF-{\kappa}B$ target genes. Taken together, PKM is a potential target that may have therapeutic implications for TNBC cells.
