Cysteamine에 의해 유도된 십이지장 궤양 흰쥐의 체내질소대사에 대한 단백질과 단백질 가수분해물의 섭취효과 (Effects of Protein and Protein Hydrolysate on Nitrogen Metabolism in Rats with Cysteamine-induced Duodenal Ulcer)

  • This study aimed to verify the nutritional and curative effects of protein hydrolysate in rats with cysteamine-induced duodenal uncer. Duodenal ulcer rat model was established by intraperitoneal injections of cysteamine. Sprague-Dawley, female rats weighing approximately 200g were intraperitoneally injected twice cysteamine(13mg/100g BW) at intervals of 3h per day. This procedure was repeated 3$\times$at intervals of 3d. Animals fed on 10% casein diet for infection periods. After last injection, 4 kinds of diets(10% casein, 20% casein, 10% casein hydrolysate, 20% casein hydrolysate) were given. Gastric montility, trypsin activity in gastrointestinal content, retention rate of nitrogen, plasma total protein, albumin, amino-N, urinary urea nitrogen, creatinine and hydroxyproline were analyzed for nutritional effects of dietary nitrogen levels(10%, 20%) and sources(casein, casein hydrolysate). In duodenal ulcer rat model, there was no differences between 20% casein diet and 20% casein hydrolysate in the view of severeness of ulcer, gastric emptying rate, serum total protein, serum albumin, plasma $\alpha$-amino-N, UUN, creatinine excretion, GFR, nitrogen retention. On the other hand, rats on 10% casein hydrolysate diet group had more curative effect of the ulcer, higher plasma albumin concentration and nitrogen retention than 10% casein diet group. The casein hydrolysate diet group was lower trypsin activity in small intestinal content than the casein diet group, at both nitrogen levels(10%, 20%). The results suggest that protein hydrolysate be applied in diet therapy for the patients with gastrointestinal ulcer.

  • This study aimed to verify the nutritional and curative effects of protein hydroysate in rats model with gastric ulcer induced by restraint and water-immersion stress. Sprague-Dawley, famale rats weighing approximtely 200g were forced in 5$\times$5$\times$15cm plexiglas cage. The restraint and water immersion stress was carried at 20$\pm$2$^{\circ}C$ for 8-hours. After stress 4 kinds of diets(10% casein, 20% casein, 10% casein hydrolysate, 20% casein hydrolysate) were given for 5 days. In the gastric ulcer rats model, the growth, gastric emptying rate, trypsin activity in gastrointestinal content, plasma total protein, albumin, $\alpha$-amino-N, UUN, creatinine and hydroxyproline of the urine and nitrogen retention were analyzed for nutritional effects of dietary nitrogen levels(10%, 20%) and sources (casein, casein hydrolysate). The results were as follows ; In gastric ulcer rats model, severeness of ulcer, plasma protein, gastric emptying rate, nitrogen retention rate were not different between 20% casein-fed group and 20% casein hydrolysatefed group. But 10% casein hydrolysate-fed group had more curative group. The casein hydrolysate diet-fed group was lower trysin activity in small intestianl content than the casein-fed group, at both casein level(10%, 20%). Finally at 20% levels, there was no difference between casein and casein hydrolysate diet, but 10% level, casein hydrolysate diet was more curative of ulcer than casein diet in gastric ulcer rat model. The results of this study provide useful information concerning diet therapy for the patients with gastrointestinal diseases and the field of enteral diet materials.

  • 초피(椒皮)나무(Zanthoxylum piperitum DC.)의 embryogenic callus로부터 부정배발생(不定胚發生)에 있어 $NH_4NO_3,\;KNO_3$와 casein hydrolysate의 첨가(添加) 효과(效果)를 알아 보기위하여 본(本) 실험(實驗)을 실시(實施)하였던 바 그 결과(結果)는 다음과 같다. 1. 2,4-D 0.1mg/1가 첨가(添加)된 MT배지(培地)에 $NH_4NO_3$를 3300mg/1 첨가(添加)시킨 배지(培地)에서 부정배발생(不定胚發生)이 가장 양호(良好)하였다. 2. $KNO_3$를 3800mg/1 첨가(添加) 시킨 경우(境遇)에는 MT 기본배지(基本培地)와 2,4-D 0.1mg/1를 첨가(添加)시킨 배지(培地)에서 비교적(比較的) 양호(良好)한 부정배(不定胚)를 발생(發生)시키었다. 3. 2,4-D 0.1mg/1가 첨가(添加)된 MT 배지(培地)에 casein hydrolysate를 700mg/1 첨가(添加)시킨 경우(境遇)에 부정배(不定培) 발생(發生)이 양호(良好)하였으며 2,4-D가 첨가(添加)되지 않는 배지(培地)에서는 부정배(不定培) 발생(發生)이 저조(低調)하였다.

소화성 궤양 흰쥐에서 체내 질소이용율 증진을 위한 체내 질소원에 관한 연구 - 단백질과 단백질 가수분해물의 비율을 중심으로 - (A Study on the Nitrogen Sources for the Enhancement of the Nitrogen Bioavailability in Rats with Peptic Ulcer -The Ratio of Casein and Casein Hydrolysate-)

  • This study aimed to verify the nutritional and curative effects of protein hydroysate and optimal ratio between protein and protein hydroysate as nitrogen source in rats with cysteamine-induced duodenal ulcer. Duodenal ulcer rat model was established by intraperitoneal injections of cysteamine. Sprague-Dawley, female rats weighing approximately 200g were intrapertionealy injected twice cysteamine(13mg/100g BW) at intervals of 3hours per day. This procedure was repeated 3 times at intervals of 3 days. Animals fed on 10% casein diet for injection periods. After last injection, 5 kinds of kiets (the ratio of casein and casein hydrolysate was 100 : 0(C100), 75 : 25(CH 25), 50 : 50(CH 50), 25 : 75(CH 75), 0 : 100(CH 100)) were given. The rate were sacrificed after feeding diet, 1, 3, 5 days. Ulcer index, hexosamine content of stomach and duodeum, gastric motility, trypsin activity, blood glutathione, plasma total protein, albumin, amino-N, urinalry urea nitrogen, creatinine, hydroxyproline and retention rate of nitrogen were analyzed for nutritional effects of diet treatments. There were no differences among diet groups in the view of the growth and diet treatments. There difference of ulcer curation by diet was appeared after 3 days. The ulcer indexes of C100 and CH 25 of 3, 5 days were significantly higher than those of CH 50, CH 75 and CH 100. This result was the same as hexosamine content of stomach, plasma protein, albumin concentration and nitrogen retention rate. The more casein hydrolysate diet had, the lower trypsin activity was. The more casein gydroysate diet had, the higher excretion of hydroxyproline was. These results show that protein hydrolysate can be applied in diet therapy for the patients with gastronitestinal ulcer. It suggests that it has curative effect of diet when nitogen sources include at least over than 50% of protein hydrolysate.

Effects of Casein Hydrolysate on the Systolic Blood Pressure and Serum Lipid Profiles in Spontaneously Hypertensive Rats

  • The effects of casein hydrolysate on blood pressure (BP) and serum lipid profiles were investigated in spontaneously hypertensive rats (SHR). Twenty-four 6-wk old male SHRs were assigned to 3 groups in a completely randomized design. Experimental groups were as follow: control, market milk group (MLG) and casein hydrolysate group (CHG). Reference blood pressure (RBP) showed average $198.94{\pm}1.46mmHg$. Blood pressure (BP) of CHG reduced 24 mmHg after 2 wk of treatment, but these increased after 3 wk. BP of CHG was significantly lower than BP of Control in experimental time (p<0.05). Serum lipid profiles were not differ significantly among groups (p<0.05). These data suggest that casein hydrolysate may beneficially improvement of blood pressure level in SHR.

Agrobacterium spp. 에 의하여 형질전환된 감자조직의 생장특성 (Growth Characteristics of Transgenic Potato Using Wild-type Agrobacterium spp.)

  • This study was carried out to obtain the information for growth characteristics of crown gall tumor and hairy root transformed by Agrobacterium spp,. on the media with phytohormones, casein hydrolysate and activated charoal. Crown gall tumors and hairly roots were formed respectively on potato tuber discs infected by tumerfaciens A ch 5 and A.rhizogenes ATCC15834. These tumors and roots could be grown on the phytohormone free media. PCR analysis of Rol C and Vir C gene fragments confirmed that crown gall root was prompted on the medium containing 2,4-D 2mg/l with casein hydrolysate lg/l. The survival ration of crown gall tumor callus derived from potato increased on medium containing the activated charcoal 0.5∼0.2mg/l because of the prevention, on the other hand, hairly roots were necrosis on the same medium. Callus derived from hairly root were excellently grown for a short time by suspension culture on liquid medium containing 2.4-d 2mg/L and casein hydrolysate lg/l.

산야유 $\beta-Casein$의 효소 가수분해 특성과 가수분해물의 Angiotensin Converting Enzyme 저해 효과 (Hydrolysis Characteristics of Goat Milk $\beta-Casein$ by Enzyme and Angiotensin Converting Enzyme Inhibition Effects of Hydrolysate)

  • 산양유 $\beta-casein$의 효소에 의한 가수분해 특성과 가수분해물의 ACE 저해 효과를 측정하고자 산양유의 $\beta-casein$을 양이온 교환 컬럼인 Mono S HR 5/5를 이용하여 분리하였으며 분리된 $\beta-casein$을 동물성 분해효소인 trypsin으로 처리하여 가수분해 특성을 확인하였고 가수분해물의 ACE 저해활성을 측정하였다. Mono S HR 5/5 양이온 교환 컬럼을 이용한 산양유 산 케이신으로 부터 순수한 $\beta-casein$의 분리는 SDS-PAGE를 이용하여 확인한 결과 순수한 $\beta-casein$의 분리가 이루어졌음을 확인할 수 있었다. $\beta-casein$$37^{\circ}C4$에서 trypsin으로 처리하여 전기영동으로 확인한 결과 가수분해 직후부터 $\beta-casein$ 위치의 band가 희미해지기 시작하고 저분자량의 band가 나타나기 시작하였으나 120분이 지난 후에는 모든 band가 가수분해되어 사라졌고 산양유에서 분리된 $\beta-casein$을 trypsin으로 처리하여 120분 경과 후 그 가수분해물을 이용하여 ACE 저해효과를 측정한 결과 가수분해하지 않은 $\beta-casein$$1.80\pm1.21\%$의 ACE 저해활성을 보였으나 trypsin으로 가수분해하여 ACE 저해 활성을 측정하였을 때 $25.36\pm0.79\%$의 저해 활성을 나타내었으며, trypsin에 의한 $\beta-casein$가수분해물의 $IC_{50}$을 측정한 결과 $308.7\pm2.77({\mu}g/mL)$로 나타났다.

고구마 정단분열조직으로부터 체세포배발생 및 식물체 재분화에 미치는 casein의 영향 (Effect of Casein on Somatic Embryogenesis and Plant Regeneration in Shoot Apical Meristem Explants of Sweetpotato (Ipomoea batatas L.))

  • 고구마의 정단배양에 의한 배발생 캘러스로부터 대량증식 체계가 개발되어져 왔다. 고구마 정단분열조직은 1mg/L 2,4-D가 첨가된 MS배지에서 배양 4주 경에 최적의 배발생 캘러스가 형성되었다. 또한 2,4-D가 첨가된 배지에 casein을 첨가함으로써 고구마 신천미 품종의 배발생 효율을 최고 90%이상으로 2.4-D단독처리보다 현저하게 증가시켰다. 배발생 캘러스로부터 체세포배의 유도는 식물생장조절제가 제거된 MS 기본배지에서 효과적으로 형성되었으며 300∼500mg/L casein을 첨가한 배지에서는 더 높은 형성 빈도와 녹색의 단단한 체세포배가 발달하였다. 한편, 2mm이하의 체세포배로부터 이차 배발생 캘러스 형성 및 체세포배의 발달이 100∼300mg/L casein의 첨가에 의해 증가하였다 배발생 캘러스에서 얻어진 체세포배는 직접 MS기본배지에서 쉽게 각 기관이 형성되었으며, 발근과 shoot를 발달시켜 정상적인 식물체로 하여 토양에 성공적으로 옮겨 심을 수 있었다.

Casein 효소 가수분해물의 항균 활성과 그 응용 (Application and Antimicrobial Activities of Casein Hydrolysates Treated with Asp.oryzae Protease)

  • 단백 효소 가수분해물의 천연 항균제로서의 응용성을 검토하기 위하여 casein에 5종의 단백질 가수분해 효소를 작용시켜 얻어진 가수 분해물의 항균활성을 측정하고 활성이 가장 높은 가수분해물을 부분 정제하여 그 응용성을 검토하였다. Casein에 5종 단백질 분해효소를 작용시켜 얻은 가수분해물의 항균활성은 Aspergillus oryzae protease에 의한 것이 가장 높았다. 효소처리에 의하여 얻어진 가수분해 물을 30,000, 10,000, 3,000 membrane filter로 한외여과 하였을 때 항균활성은 3,000이하 분획물에 대부분 함유되어 있었으며 공시균주에 대한 최소저해농도는 $1.0\~1.5\;mg/mL$이었다. Aspergillus oryzae pretense로 작용시킨 casein 가수분해물은 $121^{\circ}C$에서 10분간 가열했을 때도 그 항균 활성을 유지하는 것으로 보아 열에 안정하였다. 가수분해물을 HPLC로 220 nm 280 nm에서 검출된 peak 별로 수집하여 항균활성을 측정 한 결과 retention time 12.6, 13.2 분에서 분취된 peptide가 활성이 있었다. 가수분해 동결건조물을 된장에 첨가하였을 때 미생물의 생육이 저해되었으며 실용성이 있었다.
