A Study of Biological Hydrolysis Efficiency for Methane Digestion with Municipal Solid Waste (메탄발효를 위한 도시쓰레기 초고온 가용화 방법의 효율성 검토)

  • The efficiency of biological hydrolysis at $80^{\circ}C$ on municipal solid waste mixed with anaerobic digestion sludge was investigated in 100L batch reactors. The hydrolysis effect was observed within a day, when the hydrolysis reactor used for a pre-treatment reactor for methanogenesis, and the effect was observed during two days, When the reactor used for post-treatment reactor. For both configurations, methane production rate decreased, when hydrolysis was carried out more than a day. Gaseous ammonia in the hydrolysis reactors was successtully removed by the ammonia stripping system. Microbial diversity analysis on the hydrolysis reactors indicated dependency of microbial diversity on the configuration of the hydrolysis reactors. Carbohydrate and lactate degrading microbes dominated in the hydrolysis reactor, when the hydrolysis reactor used for a pre-treatment reactor for methanogenesis, while protein degrading microbes dominated in the post-treatment reactor.

  • This study analyzed the hydrogen conversion rate and microbial community in conjunction with changes in carbohydrate concentration during hydrogen fermentation using food waste, and presented comprehensive research results for the condition 80 g Carbo COD/L, which showed the highest efficiency with a carbohydrate removal rate of 98.1% and a hydrogen conversion rate of 1.76 mol H2/mol. The microbial community analysis found that Clostridium sp., widely known as a hydrogen-producing microorganism, was released in 80 g Carbo COD/L and confirmed that it was a dominant species at 98.1%. Conversely, in 100 g Carbo. Under COD/L conditions, Leuconostoc sp. showed the maximun prevalence, which is believed to hinder hydrogen production.

Development of an anti-obesity dietary supplement inhibiting the digestion of carbohydrate and lipid (비만 개선 효과를 지닌 탄수화물 및 지방 흡수 억제 기능성 식이조성물 개발)

  • In a previous study, we developed a new food additive as an egg yolk antibody (IgY) against carbohydrate digestion enzymer for the regulation of blood glucose level and weight control. The IgY delayed and decreased the increment if blood glucose level after administration of sucrose in human being by 30% in 20∼30 min. We also developed a lipase inhibitor as a water extract of two kinds of herb, Platycodon grandiflorum and Solanum Melongena, Twenty three volunteers were subjected to the intake of the egg yolk IgY Plus the herbal extracts for 50 days. In average, the treated subjects appeared to lose 1.96 kg of body weight and 3.4 kg of body fat mass during the treated period. Furthermore, Panniculus adiposus and breech size were significantly decreased during the experimental period. Above results suggested that the administration of the dietary additives composed of egg yolk IgY and natural herbal extract improve the obesity by the decrement of body weight and body fat mass.

A Feasibility Study on Biogas Production from Anaerobic Digestion of Straw (볏짚의 혐기성소화시 소화가스 생성에 관한 연구)

  • Quantity and composition of biogas from semi-continuous anaerobic digestion of straw were obtained experimentally in laboratory scale digesters fed with 1 liter of 5% straw-water mixture and maintained at 35$^{\circ}C$. Experiments were carried out for hydraulic retention time(HRT) of 8, 10 and 15days, respectively. The amount and composition of biogas produced were measured until steady-state was achieved for each run. The amount of biogas and methane percent go through a maximum and decrease continuously towards the steady-state after three times operation of hydraulic retention time(HRT). Methane gas production rates at steady-state increase with the increasing of HRT. Biogas production of 0.45 liter/day with 25% methane, 0.42 liter/day with 33.7% methane and 0.492 liter/day with 31.7% methane were obtained for 8, 10 and 15days of HRT, respectively. The high proportion of soluble carbohydrates present in straw makes the volatile fatty acids to build up within the digester causing a drop in pH that inhibits digestion. Regular control of pH is therefore necessary by adding alkalinity. Reductions in COD increase with increase in HRT. The stratification of plant material within the digester is different from that of manure, and modifications in design and operation of digesters may be necessary if they are fed with plant matter.

Connection of spectral pattern of carbohydrate molecular structure to alteration of nutritional properties of coffee by-products after fermentation

  • Objective: The objective of this study was to determine internal structure spectral profile of by-products from coffee processing that were affected by added-microorganism fermentation duration in relation to truly absorbed feed nutrient supply in ruminant system. Methods: The by-products from coffee processing were fermented using commercial fermentation product, consisting of various microorganisms: for 0 (control), 7, 14, 21, and 28 days. In this study, carbohydrate-related spectral profiles of coffee by-products were correlated with their chemical and nutritional properties (chemical composition, total digestible nutrient, bioenergy values, carbohydrate sub-fractions and predicted degradation and digestion parameters as well as milk value of feed). The vibrational spectra of coffee by-products samples after fermentation for 0 (control), 7, 14, 21, and 28 days were determined using a JASCO FT/IR-4200 spectroscopy coupled with accessory of attenuated total reflectance (ATR). The molecular spectral analyses with univariate approach were conducted with the OMNIC 7.3 software. Results: Molecular spectral analysis parameters in fermented and non-fermented by-products from coffee processing included structural carbohydrate, cellulosic compounds, non-structural carbohydrates, lignin compound, CH-bending, structural carbohydrate peak1, structural carbohydrate peak2, structural carbohydrate peak3, hemicellulosic compound, non-structural carbohydrate peak1, non-structural carbohydrate peak2, non-structural carbohydrate peak3. The study results show that added-microorganism fermentation induced chemical and nutritional changes of coffee by-products including carbohydrate chemical composition profiles, bioenergy value, feed milk value, carbohydrate subfractions, estimated degradable and undegradable fractions in the rumen, and intestinal digested nutrient supply in ruminant system. Conclusion: In conclusion, carbohydrate nutrition value changes by added-microorganism fermentation duration were in an agreement with the change of their spectral profile in the coffee by-products. The studies show that the vibrational ATR-FT/IR spectroscopic technique could be applied as a rapid analytical tool to evaluate fermented by-products and connect with truly digestible carbohydrate supply in ruminant system.

Isolation of $\alpha$-glucosiadase Inhibitor Producing Actinomycetes from Soil Sample (토양시료로부터 $\alpha$-glucosidase 저해제 생성 방선균의 분리)

  • To find $\alpha$-glucosidase inhibitors produced by Actinomycetes, bacteria belonging to Actinomycetes were isolated from soil sample using Bennett's medium. The inhibitory activity induced by these bacteria on $\alpha$-glucosidase, which is the key enzymes far carbohydrates digestion and the prevention of diabetic complications, was investigated. A strain of these bacteria, PM718 potently inhibited $\alpha$-glucosidase activity in vitro.

Effects of a Hwanggi-tang Ethanol Extract on Glucose Uptake and Metabolism in Murine Myotubes (근육세포주에서 당 흡수 및 대사 조절에 대한 황기탕 에탄올 추출물의 효과)

  • Objectives: The aim of this study was to evaluate the effects of Hwanggi-tang on glucose digestion, uptake, and metabolism in murine C2C12 myotubes. Methods: Hwanggi-tang was prepared according to the Dong-ui-bo-gam (≪東醫寶鑑≫) prescription by 70% ethanol extraction. The effect on glucose digestion was examined by determining the inhibitory effect of Hwanggi-tang on α-glucosidase activity. We also compared and verified the gene and protein expression of genes related to glucose uptake in C2C12 myotubes treated with Hwanggi-tang or insulin. Glucose metabolism was assessed by the expression levels of associated enzymes. Results: Hwanggi-tang caused a dose-dependent inhibition of α-glucosidase activity, induced glucose uptake by activation of the PI3K/Akt/mTOR pathway in the insulin signaling pathway, and promoted glucose oxidation and β-oxidation. Conclusions: Hwanggi-tang exerts an anti-diabetic effect on murine myotubes by inhibiting glucose digestion and inducing glucose uptake and consumption.

Enzymatic N-glycan analysis of 31 kDa molecule in plerocercoid of Spirometra mansoni (sparganum) and its antigenicity after chemical oxidation

  • A highly specific antigenic protein of 31 kDa from plerocercoid of Spirometra mansoni (sparganum) was obtained by gelatin affinity and Mono Q anion-exchange column chromatography. The purified 31 kDa protein was subjected to N-glycan enzymatic digestion for structural analysis. The relative electrophoretic mobility was analyzed by SDS-PAGE, before and after digestion. On SDS-PAGE after enzymatic digestion, the 31 kDa protein showed a molecular shift of approximately 2 kDa, which indicated the possession of complex N-linked oligosaccharides (N-glycosidase F sensitive) but not of high-mannose oligosaccharides (endo-beta-N-acetylglucosaminidase H, non-sensitive). Chemically periodated 31 kDa protein showed statistically non-significant changes with human sparganosis sera by enzyme linked immunosorbent assay (ELISA). Therefore, the dominant epitopes of the 31 kDa molecule in human sparganosis were found to be mainly polypeptide, while N-glycans of the antigenic molecule in sparganum was minimal in anti-carbohydrate antibody production.

Effects of Amylose Contents and Degree of Gelatinization of Rice Flour on In Vitro Starch Digestibility, Physical Characteristics, and Morphological Properties

  • The relationship of in vitro starch digestibility and gel strength was investigated at various concentrations (10-30%) of rice cultivars with different amylose contents (27.9, 17.9, and 5.2%). As the rice flour concentration increased, predicted glycemic index decreased, but gel strength increased regardless of amylose contents. Gel strength correlated strongly with amylose content, whereas in vitro starch digestibility was more highly affected by rice flour concentration than by amylose contents. Moreover, the impact of degree of gelatinization on in vitro starch digestibility of high amylose rice was also examined in terms of structural features and rheological properties. The digestion rate of fully gelatinized flour was 1.7 times higher than that of native flour, while the disrupted structure with a different gelatinization degree during starch digestion was visually demonstrated through the X-ray diffraction and molecular distribution analysis. The rice flour changed from an A-type to a V-type pattern and showed difference in crystalline melting. The low molecular weight distribution increased with increasing degree of gelatinization during starch digestion. The apparent viscosity also increased with degree of gelatinization. These results demonstrated that the starch digestibility of rice was more affected by concentration than by amylose content, as well as by the degree of gelatinization due to structural difference.

Blood Glucose Lowering Effects of Mulberry Leaves and Silkworm Extracts on Mice Fed with High-Carbohydrate Diet (고탄수화물 식이 섭취 마우스에서 상엽 및 누에 추출물의 혈당강하 효과)

  • Mulberry leaves(Mori folium) and silkworm(Bombyx mori) are potnet inhibiters of intestinal $\alpha$-glycosidase, and inhibit the digestion of starch and sucrose in the small intestine. They are able to prevent postprandial hyperglycemia and decrease blood insulin levels. In this study , a high-carbohydrate diet(CHO ; 67.5%, protein ; 20.8%, fat : 11.7%) was received by the control group. In contrast, the experimental groups received a high-carbohydrate diet with extracts of mulberry leaves and silkwork(50mg.100g diet), and acarbose(6.7mg/100g diet). after a 10 week study period , the experimental groups had lower blood glucose and triglyceride levels. The experimental groups tended to have lwer Hb Alc levels. Also, blood insulin levels were lower than the control groups in accordance with blood glucose levels. The activities of intestinal $\alpha$-glucosidase in the middle and distal parts of small intestine were induced by the extracts of mulberry leaves and silkworm in the experimental groups. However, the activities of liver lysosomal glucosidase and the contents of glycogen in the liver were not affected by the mulberry leave and silkworm extracts nor by acarbose. Mulberry leaves and silkworm were able to prevent sudden postprandial peaks in blood glucose as a result of $\alpha$-glycosidase, inhibition, there by decreasing unnecessary insulin secretion.
