Determination of Terbutaline Enantiomers in Human Urine by Capillary Electrophoresis Using $Hydroxypropyl-{\beta}-cyclodextrin$ as a Chiral Selector

  • A method for the determination of terbutaline enantiomers in human urine by capillary elctrophoresis has been developed. Optimum resolution was achieved using 50 mM phosphate buffer, pH 2.5, containing 15 mM of hydroxypropyl-$\beta$-cyclodextrin as a chiral selector. Urine samples were prepared by solid-phase extraction with Sep-pak silica, followed by CE. The assay was linear between 2-250 ng/mL (R = 0.9998 for (S)-(+)-terbutaline and R = 0.9999 for (R)-(-)-terbutaline) and detection limit was 0.8 ng/mL. The intra-day variation ranged between 6.3 and 14.5% in relation to the measured concentration and the inter-day variation was 8.2-20.1%. It has been applied to the determination of (S)-(+)-terbutaline and (R)-(-)-terbutaline in urine from healthy volunteer dosed with racemic terbutaline sulfate.

  • Chiral separation of aryalcohols such as 1-phenyl-propanol, 1-phenyl-2-proanol, and 2-phenyl-1-propanol by capillary electrophoresis has been optimized using the overlapping resolution mapping (ORM) scheme. Three critical parameters of the electrophoretic media, i.e. phosphate concentration, sulfated ${\beta}$-cyclodextrin (CD) concentration and pH, were chosen for optimization. The working ranges were initially presumed by 7 preexperiments. Further optimization was carried out by another seven experiments within the narrow working ranges. From the final overlapping resolution mapping all peak pairs, the area of maximum separations were located. Using the conditions of a point in this area, we found that the target compounds were a baseline separated within 30 min. The maximum separation conditions of arylalcohols were a chiral selector concentration of 5.4%, a phosphate concentration of 28 mM, and a pH of 5.0.

Capillary Electrophoresis Detection of Hydrogen Peroxide by Using Titanium Ion and 4-(2-thiazolylazo)resorcinol

  • A novel method for the detection of hydrogen peroxide in aqueous solution was developed via reaction between $H_2O_2$, trivalent titanium ion ($Ti^{3+}$) and 4-(2-thiazolylazo) resorcinol (TAR), resulting in a ternary complex with a maximum UV absorbance at 530 nm. The CE detection of $H_2O_2$ was fast, sensitive and cost-effective without pretreatment procedures. $H_2O_2$ was detected within 15 min at 1 to $100{\mu}M$ range with the lowest detection limit at $1.0{\mu}M$. Under the optimized CE conditions, the concentration of $H_2O_2$ in coffee or tea extract was quantitatively determined. Our results show that CE detection of the ternary complex of $H_2O_2-Ti^{3+}$-TAR has potential applications for the detection of $H_2O_2$ in aqueous sources.

Investigation of Enantiomer Separation Using Chiral Crown Ethers as Chiral Selectors

  • A number of chiral selectors have been developed and applied for enantiomer separation of a variety of chiral compounds. Among these chiral selectors are chiral crown ethers, a class of synthetic host polyether molecules that bind protonated chiral primary amines with high selectivity and affinity. In this paper, two important chiral crown ethers as chiral selectors of bis-(1,1'-binaphthyl)-22-crown-6 and (18-crown-6)-2,3,11,12-tetracarboxylic acid (18-C-6-TA) are focused. They have been widely used to resolve the enantiomers of chiral compounds containing a primary amino moiety using chiral stationary phases (CSPs) or chiral selectors by high-performance liquid chromatography (HPLC), capillary electrophoresis (CE) and so on in chirotechnology. Also, it was described that the commercially available covalent type HPLC CSPs derived from (+)- and (-)-18-C-6-TA have been developed and successfully applied for the resolution of various primary amino compounds including amino acids.

Discrimination of Domestic Rice Cultivars by Capillary Electrophoresis (Capillary Electrophoresis를 이용한 국내산 쌀의 품종 판별)

  • Capillary electrophoresis (CE) with rice proteins was used to discriminate 10 domestic rice cultivars in less than 25 min. Most cultivars were differentiated quickly and easily using P-ACN buffer system. CE of rice prolamins allowed classifying ten varieties of Korean rice into three groups. Peak h was characteristic peak for Dongjinbyeo, Gaehwabyeo and Yongnambyeo which were classified into the group of Dongjinbyeo. Chuchungbyeo, Odaebyeo, Mangeumbyeo and Bonggwangbyeo easily differentiated from the group of Dongjinbyeo by the absence of peak h which were classified into the group of Chuchungbyeo. Peak g typical for Illpumbyeo, Hwaseungbyeo and Hwayoungbyeo accounted for 70% of total peak area. They belong to the group of Illpumbyeo. Some cultivars showed specific peak patterns among ten cultivars, Illpumbyeo was differentiated from others by several peaks between peak c and peak f, and the peak d was apparently detected in Odaebyeo not in others. Other minor differences were also found within each group. The result of the study showed that CE has potential for discrimination of rice cultivars. It also possesses the inherent advantages such as low mass requirements, fast seperations, and quantitative analysis through on-capillary UV detection.

Investigation of serum protein electrophoresis fractions analysis in Gyeongju DongGyeong dogs (경주지방의 무미 또는 단미 형태의 개(경주개 동경이)의 혈청 단백질의 분획에 대환 실태조사)

  • Serum proteins of Korean native dogs (Gyeongju DongGyeong dogs) was analyzed by capillary electrophoresis system. Electrophoretic patterns showed six fractions. Total serum protein and ratio of albumin to globulin were $5.99{\pm}0.83$ (g/dl) and $1.41{\pm}0.29$ (g/dl) in DongGyeong dogs. Relative percentages of total serum albumin and $\alpha-1$, $\alpha-2$, $\beta-1$, $\beta-2$, $\gamma$-globulin fraction were $57.94{\pm}5.43$, $3.15{\pm}2.30$, $7.49{\pm}4.09$, $9.43{\pm}3.50$, $7.63{\pm}5.70$, and $14.36{\pm}7.63$, respectively. It was observed that $\beta$-globulin was higher than other fractions. The most striking alternation with age was founded in the $\gamma$-fractions. Also, it was observed that ratios of albumin to globulin in DongGyeong dogs were higher than on other dogs.
