Prevalence of canine scabies in the Korean stray dogs (국내 유기견의 개 옴 감염 상황)

  • Recently, the number of stray dogs is proportionate to the increase of the number of the companion dogs. Sarcoptic scabiei var canis, causing scabies, is one of the most important canine zoonotic arthropods in Korea and around the world. Thus, we have tried to know the prevalence of canine scabies in the stray dogs in Korea. A total of 565 stray dogs were collected from the rescue centers all over the country from Jan. 2006 to Dec. 2006. They were grouped with euthanasia or natural death and examined for the gender and age estimated by dental formula. To identify the lesions, the whole body was grossly examined and tested pathologically. Thirty two (5.66%) of 565 dogs were diagnosed as canine scabies. Dogs from urban areas had fewer scabies (0.62%) than those of rural areas (12.5%). Prevalence of scabies in male and female dogs was no difference as 5.96% and 5.25%. Euthanasia group showed higher prevalence (6.48%) than natural death group (2.44%) in scabies. Old dogs over five years showed lower infestration (1.82%) in scabies. In histopathological findings, there were mites in the burrows formed in the subcorneal space. Acanthosis, hyperkeratosis with crust, and vascular dilatation were main findings. One hundred thirteen (20%) of 565 stray dogs were diagnosed to have skin disease. Among them, canine scabies is the most prominent ectoparasite as 5.66 %. With previous reports on human infection in Korea, canine scabies must be regarded as the important zoonotic canine skin disease. Accordingly, for the human and canine hygiene it is imperative that stray dogs with skin problems are segregated and tested for the parasites to treat properly as soon as arriving at rescue shelter.

Canine hemangiopericytoma in a Golden Retriever: A case report

  • A 7-year-old intact female Golden Retriever was presented for examination. The dog had large irregular subcutaneous masses in the abdomen which were ruptured or encapsulated. Those were removed surgically. Histopathologically, the masses consisted of spindle cells that often formed distinct whorls around a central capillary. Immuno-histochemical analysis revealed that the neoplastic cells were strong diffuse cyto-plasmic immunolabelling for vimentin and focal immunoreactivity for smooth muscle actin, whereas not immunoreactive for cytokeratin, desmin, von Willebrand factor, glial fibrillary acidic protein, or S-100. The neoplastic cells ultrastructurally had processes attached by desmosome-like structures, swollen mitochondria and dilated rough endoplasmic reticulum. Based on the above results, this case was diagnosed as a canine hemangiopericytoma in the abdominal subcutis of a Golden Retriever.

Cytotoxicity of natural killer cells on canine mammary carcinoma cells (개 유선종양세포에 대한 자연살해세포 독성)

  • Natural killer (NK) cells play have a crucial role in the early phase of immune responses against various pathogens. We compared characteristics of canine NK cells against two canine mammary carcinoma cell lines, REM134 and CF41.Mg. REM134 showed higher expression of progesterone receptor, proliferative cell nuclear antigen, Ki67, multiple drug resistance, Bmi-1, c-myc, E-cadherin, and human epidermal growth factor receptor type-2 than that of CF41.Mg. For specific expansion and activation of NK cells, we isolated CD5 negative cells from canine peripheral blood mononuclear cells and co-cultured K562 cells in the presence of interleukin (IL)-2, IL-15, and IL-21 for 21 days. As a result, we found that expression markers of activated NK cells such as NKp30, NKp44, NKp46, NKG2D, CD244, perforin, granzyme B, and tumor necrosis factor alpha were highly upregulated. In addition, we found there was upregulated production of interferon gamma of activated NK cells against target cells such as REM134 and CF41.Mg. Specifically, we observed that cytotoxicity of NK cells against target cells was more sensitively reacted to CF41.Mg than REM134. Based on the results of this study, we recommend the development of an experimental application of CF41Mg, which has not been reported in canine mammary carcinoma research.

Modeling of transmission pathways on canine heartworm dynamics

  • Canine heartworm disease is a vector-borne disease that is transmitted from dog to dog by mosquitoes. It causes epidemics that disrupt the health environments of dogs and are burdensome for many dog owners. Recent trends of changing temperatures and weather conditions in South Korea may have an impact on the population of mosquitoes, and it affects the population of dogs at risk of heartworm infection. Mathematical modeling has become an important measure for analyzing the epidemiological characteristics of infectious diseases. However, canine heartworm infection transmission has not been reported yet through mathematical modeling. We develop a mathematical model of canine heartworm infection to predict the population of infected dogs depending on the vector (mosquito) population using a susceptible, exposed, infected, and recovered model. Simulation results show that after 1 year, 3,289 dogs out of 73,602 (about 4.5%) are exposed and 134 (about 0.2%) are infected. Only 0.2% of susceptible dogs become infected after 1 year. However, if all exposed dogs are maintained in the same circumstances without any treatment, then the number of infected subjects will increase over time. This may increase the possibility of other dogs, especially dogs that live outside, being infected.

Development of indirect ELISA for the detection of canine adenovirus type 2 antibodies in dog sera

  • Background: Canine adenovirus type 2 (CAV-2) induces infectious laryngotracheitis in members of the family Canidae, including dogs. To date, no ELISA kits specific for CAV-2 antibody have been commercialized for dogs in Korea. Objectives: We aimed to develop new indirect enzyme-linked immunosorbent assay (I-ELISA) to perform rapid, accurate serological surveys of CAV-2 in dog serum samples. Methods: In total, 165 serum samples were collected from dogs residing in Chungbuk and Gyeongbuk provinces between 2016 and 2018. The Korean CAV-2, named the APQA1701-40P strain, was propagated in Madin-Darby canine kidney cells and purified in an anion-exchange chromatography column for use as an antigen for I-ELISA. The virus-neutralizing antibody titers of CAV-2 in the dog sera were measured by virus neutralization (VN) test. Results: We compared the results obtained between the VN and new I-ELISA tests. The sensitivity, specificity, and accuracy of new I-ELISA were 98.6%, 86.4% and 97.0% compared with VN test, respectively. New I-ELISA was significantly correlated with VN (r = 0.91). Conclusions: These results indicate that new I-ELISA is useful for sero-surveillance of CAV-2 in dog serum.

Animals and diseases prevalence of the rescued dogs in an animal shelter in Incheon (인천지역 유기동물 현황 및 개 질병 조사)

  • Animals and disease frequency of the rescued dogs were investigated in Incheon Veterinary Medical Association Animal Shelter from January in 2012 to December in 2013. Three zoonoses (rabies, brucellosis, and dirofilariosis) and three infectious diseases (canine distemper, canine parvoviral enteritis, and canine influenza) were examined for stray dogs. Among 5,603 heads, 647 (11.5%) went back to their owner and 969 (17.3%) were adopted to new families. Prevalence of dirofilariosis, canine distemper and canine parvoviral enteritis were 2.2% (16/718), 6.0% (24/399) and 6.1% (24/396), respectively. Positive antibody rates against rabies, B. canis and canine influenza virus were 20.5% (41/200), 0.1% (1/718) and 2.0% (4/200), respectively. Protective antibody for canine distemper virus and canine parvovirus were shown in 47.0% (94/200). The data indicate that control measures including facility standards and disease control program are one of the important aspects of the shelter management because stray dogs are exposed to various infectious agents.

  • Seven cases of canine babesiasis were observed by clinically, hematologically and immunologically in the Kyungbook and Pusan area from May 1961 to October 1963. And a survey was conducted on the rising pups for their babesia, infection in the broken out districts of the disease. The results obtained were as follows: 1. Severe cases of canine babesiasis due to the same agent have been encountered in various breed, of dogs in widely separated location in the Kyungpook and Pusan area. 2. The principal symptoms oserved in infected dogs were severe anemia, weakness, inappetence, moderate fever or subnormal body temperature, increased respiration and pulse, palpitation, icterus, hemogrobinuria and redish yellow defecation. 3. Of hematological findings, decrease in erythrocytes count, hemoglobin content was evident and severe anemic changes occured which were associated with macrocytic anisocytosis. The differential leucocytes counts showed a tendency to increased monocytes and decreased monocytes and decreased basophil. 4. The principal anatomic chabges observed in two cases of infected dogs were severe anemic and icteric changes, markedly enlarged spleen, and enlarged liver with distended gall bladder. 5. Haemaphysalis bispinosa was suspected as being the principal vector of the infection. 6. It was suspected that chronic canine babesiasis would prevailed widely in Kyungpook and Pusan area, and that dogs are raised in such infected environment usually suffer only from the mild, chronic form, which may be practically symptomless, while imported dogs usually suffer from the acute form of the disease.

Isolation and identification of canine adenovirus type 2 from a naturally infected dog in Korea

  • Canine adenovirus type 2 (CAV-2) infection results in significant respiratory illness in dogs. Isolating and culturing CAV-2 allows for investigations into its pathogenesis and the development of vaccines and diagnostic assays. In this study, we successfully isolated a virus from a naturally infected dog in Gyeonggi-do, Korea. The virus was propagated in Madin-Darby canine kidney (MDCK) and Vero cells and showed a specific cytopathic morphology that appeared similar to a bunch of grapes. The virus was first confirmed as CAV-2 based on these cytopathic effects, an immunofluorescence assay, hemagglutination assay, and electron microscopy. The viral titer of the isolate designated APQA1601 reached $10^{6.5}$ 50% tissue culture infections dose per mL in MDCK cells and exhibited no hemagglutination units with erythrocytes from guinea pig. The virus was also confirmed by polymerase chain reaction and next-generation sequencing. The APQA1601 strain had the highest similarity (~99.9%) with the Toronto A26/61 strain, which was isolated in Canada in 1976 when the nucleotide sequences of the full genome of the APQA1601 strain were compared with those of other CAV strains. Isolating CAV-2 will help elucidate the biological properties of CAV-2 circulating in Korean dogs.

Prediction survey on the viral diseases of companion animals in Gwangju area, Korea (광주지역 반려동물 바이러스 질병 예측 조사)

  • For the monitoring of six viral disease (CIV: canine influenzavirus, CPIV: canine parainfluenzavirus, CHV: canine herpesvirus, CPV2: canine parvovirus type 2, CCoV: canine coronavirus, CNV: canine norovirus) inspections, a total of 300 samples were collected nasal or feces from the companion dogs of animal hospital (n=98) and the abandoned dogs of animal shelters (n=202) in Gwangju, Korea. Using PCR and RT-PCR, CPV2, CPIV and CHV were detected in 55 (18.3%), 11 (3.7%), 1 (0.3%), respectively. CPV2 was highly detected in May, October and November. and CPIV was highly detected in November. But those agents were not detected the virus in March and July. Based on the results of the investigation continuous monitoring for companion and abandoned dogs will be required.

  • 교정경험, 보철물장착 경험, 교합조정, 상하악골절, 및 전신적 질환이 없는 Angle class I 교합을 갖는 100명(名)의 환자에 있어 1000개의 치아를 임상적으로 평가하여, 그 disclusion pattern과 periodontal disease index와의 상호관계를 조사해 본 결과, 14%가canine protection을 하고, 16%가 progressive disclusion을 하며, 46%가 group function을 하고 나머지 24%가 각각 바른 disclusion pattern을 띤다는 것을 알았다. 여기서 C.P.하는 mouth의 치아가 다른 3group보다도 더 낮은 periodontal disease index를 갖는다는 사실이 유의성 있는 통계자료로써 나타났다.
