Potential Feeding Value of Deoiled Rice Bran by Japanese Quails. 1. The Metabolisable Energy Content

  • The apparent metabolisable energy (AME) and N-corrected ME (AMEn) of deoiled rice bran (DORB) were determined with adult quails at 6 and 10- week of age. The DORB obtained from two types of extraction process, Batch (DORB-B) and the Continuous (DORB-C), was each included in a practical type of the reference diet at 20 or 40% level. The analysed crude protein, ether extract, total ash, calcium, phosphorus, glucose and starch content of DORB-B and DORB-C were found at 19.0, 0.79, 17.05, 0.11, 1.92, 2.3, 11.22, and 15.02, 1.56, 13.0, 0.40, 2.76, 2.16, 19.0, respectively. The level of inclusion of DORB in diet appeared to exert a significant effect on the AME and AMEn values. When bioassayed at 20% inclusion level the DORB was found to have a significantly (p<0.01) lower value than that obtained at 40% inclusion level. However, no significant effect of age of quails on the AME values of DORB was evident. The ME bioassays with quails gave significantly (p<0.01) higher AME values for DORB-C than DORB-B thereby indicating that the continuous system of solvent extraction of rice bran is superior to the batch system from this point of view. The AME value of DORB predicted from its chemical composition also revealed that the DORB-C contained approximately 15% more energy than that in DORB-B.

  • The implications of incorporating deoiled rice bran (DORB) in diet on the utilization of certain nutrients had been examined through balance studies made with 6- and 10- week-old Japanese quails. The stuffs evaluated in this study had their origin from two different types of processes- a batch extraction (DORB-B) and another continuous process (DORB-C), and each included at 10, 20 or 30% level in an iso-nitrogenous and iso-caloric diet. On chemical analyses, the DORB-B was found to contain crude protein, 19.0; ether extract, 0.79; total ash, 17.05; Ca, 0.11; P, 1.92; glucose, 2.35 and starch 11.22 g/100 g day matter. Corresponding values for DORB-C were 15.02, 1.56, 13.0, 0.40, 2.76, 2.16 and 19.0, respectively. The data on nutrient balances with quails fed diets containing the two-types of DORB suggested no significant (p>0.05) differences in per cent retention of diet dry matter (DM), organic matter (OM), ether extract (EE), gross energy (GE), nitrogen (N), crude fiber (CF), calcium (Ca) or phosphorus (P) content attributable to the extraction process. The age of the birds also appeared to have no significant (p>0.05) effect on nutrient utilization by them. However, the level of inclusion of DORB had a significant (p<0.05) effect on nutrient utilization by quails in that the retention of dietary N, EE, CF, Ca and P was better at lower than at higher levels. It is concluded that the DORB, irrespective of extraction process, can safely be employed upto 20% level in diet of adult Japanese quails without noticeable interference in utilizing the major dietary nutrients.

  • Soybeans contain the phytoestrogens genistein and daidzein, their glucosides genistin and daidzin and coumesterol. These isoflavonoid compounds are capable of producing an estrogenic response in a number of diverse species. This study determined optimum conditions for the extraction of the main isoflavones(daidzin, genistin, daidzein, genistein) in defatted soybean meal using high-performance liquid chromatography. The best optimum extraction was achieved at 75% ethanol, $80^{circ}C$, pH4 and a three hour contact time. In addition, isoflavones with high purity were separated by adding up to 4%(w/v) of calcium chloride dihydrate. Most soybean extracts were composed of $beta$-glucosidic conjugate(daidzin, genistin) which is difficult to adsorb in body. Therefore, $beta$-glucosidase was used to convert as conjugate to aglycone form (daidzein, genistein) which is easy to adsorb. The optimal conditions of enzyme reaction involved to be 8.4 units of enzyme concentration, pH5.0, $40^{circ}C$ and 40 minutes.

  • Of the 200, 000 known species of molds, only 50 or 60 are known to be harmful to humans or livestock. Certain fungi that grow on grains and grasses can produce chemical substances called mycotoxins that adversely affect performance in poultry. There are several methods of preserving feed ingredients. The list includes: drying, antioxidants, mold inhibitors, organic acids, phosphates, cooking or toasting, fat extraction, blending and fermenting. Mold inhibitors are manufactured to inhibit mold growth and prevent the production of toxic substances. They are fungistats and not fungicides, that is, they only stop the growth of molds. Practical and cost-effective methods to detoxify mycotoxin containing feedstuffs are in great demand. 0.5% hydrated sodium calcium aluminosilicate added to the diet protected chicks from the deleterious effects of aflatoxin-contaminated feed. The dietary addition of antioxidants and methionine also significantly diminished the negative effects on body weight in chicks toxicated with 3.0 ppm aflatoxin B1.

  • Limestone is an important commodity in Philippines. Limestone has numerous uses that range from agricultural applications to building materials to medicines. Many limestone products require rock with specific physical and chemical characteristics. Most limestone is biochemical in origin meaning the calcium carbonate in the stone originated from shelled oceanic creatures. In this paper, we reported the natural sources of limestone, geological formation of limestone and the oyster shell waste in Cebu, Bantayan, Philippines were reported. Due to the mining or quarrying in Cebu, Bantayan, in a limestone area poses the threat of groundwater pollution (since limestone is a porous geologic formation with a high transmissivity). The other environmental issue is oyster shell waste. The oyster shell waste is the major source of limestone. We developed and applied appropriate technologies for the extraction of limestone from oyster shell waste and utilizes as high value added material.

Applicability Test of Various Stabilizers for Heavy Metals Contaminated Soil from Smelter Area (제련소 주변 오염토양의 중금속 안정화를 위한 다양한 안정화제의 적용성 연구)

  • There are several remediation technologies for heavy metal contaminated soils but increasing cost limits the application of the technology if the contaminated area is large. Therefore, stabilization, which blocks the release of heavy metals or makes slow the release, is one of the applicable technology for the heavy metal contaminated soil. Current study is an applicability test for a smelter area with various stabilizer such as magnetite, hematite, zeolite-A, zeolite-X, zeolite-Y, zinc oxide, calcium oxide, carbon trioxide, manganese oxide, manganese dioxide, fish bone, sodium phosphate. The soil contaminated with arsenic, lead, copper, nickel, and zinc could not be stabilized only one stabilizer which is known to have stability for certain metal. Many of the stabilizer works for a few metal but not all of the heavy metal. In several cases, stabilizers increase the release of the other metals while they stabilize some metals. In general, the stabilizing efficiency was increased with time. For Ni, Pb, calcium oxide, carbon trioxide, manganese oxide had good stabilizing effect in water extractable portion. For Cu, manganese oxide, zeolite showed good results especially in the exchangeable portion of the sequential extraction. For As, magnetite had good ability but most of the metal oxide which showed good result for other heavy metals increased with the release of As. Current study suggest that multiple stabilizers are needed for the contaminated soil and dose of the stabilizer and stabilizing time should be carefully considered for the soil contaminated with various metals.

Mineral extraction from by-products of brown rice using electrodialysis and production of mineral salt containing lower sodium (전기투석을 이용한 현미부산물로부터의 미네랄성분 추출 및 나트륨감량형 미네랄 소금 제조)

  • The purpose of this study was to develop a supplemental healthy food that can help prevent high blood pressure-related diseases caused due to the excessive consumption of sodium in salt. This was achieved by using ion-displacement techniques to produce mineral salt with lower sodium content by using fermented brown rice by-products rich in minerals. Mineral salt containing 2019.2 mg/100 g of potassium, 678.5 mg/100 g of magnesium, 48.7 mg/100 g of calcium, and 19.5 mg/100 g of sodium was obtained by fermenting brown rice by-products to create a culture medium for the mineral salt. Mineral salt containing 1769.7 mg/100 g of potassium, 573.6 mg/100 g of magnesium, 35.3 mg/100 g of calcium, and 19.5 mg/100 g of sodium was obtained by filtering and refining the by-product extract of fermented brown rice. The results showed that when the stream velocity of the instrument used for electrolysis was 200 mL/min and the current and the concentration of the reactive liquid in the purified water chamber were higher, the effect of electrolysis was greater. Ion hot water extraction of the fermented brown rice by-products improved by up to 95% and was collected as purified water within 90 min of the reaction time. Chloride ions with pH 7.4 were produced by mixing sodium hydroxide in a purified saline water chamber with electro-analyzed water. The salt produced in this study contained low sodium, 5.7~30%, as compared to 40% sodium content of the normal salt.

The Analysis for Calcium and Fructooligosaccharides Contents in Nutrients Fortified Dairy Products (유가공품 중 칼슘 및 프락토올리고당 영양강화 함량 분석)

  • Nutrients fortified dairy products declare their contents on the label for nutrition claim and marketing. However, there are few monitoring studies about relations between actual quantities of fortified nutrients and the described ones on the label. This study was carried out for comparing actual fortified nutrient contents with labeled ones. Forty calcium fortified dairy products and twenty four fructooligosaccharides (FOS) fortified dairy products were sampled at supermarkets located in Anyang, Korea from March to November in 2010. Calcium contents were analyzed by using inductively coupled plasma optical emission spectrometry followed by microwave sample digestion, and FOS contents were analyzed by HPLC-ELSD followed by solvent extraction. In fresh milk, calcium contents ranged from 1.0 to 2.4 mg/mL, and those values were 87~127% of their labeled contents. In fermented milk products and cheeses, calcium contents ranged from 0.3 to 1.6 mg/g (89~131% of their labeled contents), 4.2 to 23.0 mg/g (83~127% of their labeled contents), respectively. FOS contents ranged from 9.09 to 18.89 mg/g in FOS contents labeled products and showed 83~154% compared to their labeled quantity, and ranged from 1.3~30.8 mg/g in products without quantity labeling. In conclusion, the amounts of calcium and FOS in dairy products were above 80% compared to their labeled ones and conformed to the Korean official livestock products labeling standard.

Optimization of Radiostrontium Separation Process Using Sr Resin (Sr resin을 이용한 방사성 스트론튬 분리의 최적화)

  • For the analysis of 90Sr, which is a pure beta emitter, radiochemical separation from the main interfering elements such as Ca, Ba and Ra is required due to their similarity in chemical behavior to strontium. This study describes a radioanalytical procedure using extraction chromatography for separating Sr from interfering elements. The maximum capacity of the resin for Sr was approximately 6 mg per 1.5 mL of bed volume (BV). The recovery of Sr on the resin 1.5 mL (BV) was quantitative for the calcium level of 200 mg at the flow rate of 1 mL min-1. However the chemical yield declined by increasing the flow rate by up to 5 mL min-1 even at the calcium level of 200 mg. When using the same BV of Sr resin, the performance of the resin was enhanced as the cross-sectional area of the Sr resin column is small.

Extracellular Matrix of Fresh and Cryopreserved Porcine Aortic Tissues

  • The effect of cryopreservation on extracellular matrix was studied with the ultimate objective of permiting a prediction of the tendency of aorta conduit tissue to calcify following transplantation. Cryopreserved and fresh porcine aorta conduit tissues were extracted using guanidine-hydrochloride (Gdn-HCl) followed by sequential digestion of the tissues with collagenase, elastase, and papain. Glycosaminoglycans (GAGs) of the proteoglycans (PGs) were isolated and quantitated. Gdn-HCl extracted about 61% and 62% of the total GAG (proteoqlycan) material from cryopreserved and fresh tissues, respectively. Collagenasesolubilized proteoglycans from Gdn-HCl extracted tissue represented 20% and 13%, respectively, of the total GAGs present in cryopreserved and fresh tissues. Subsequent elastase hydrolysis of collagenase-digested tissue released about 11% of total GAGs from cryopreserved tissue and 16% from fresh tissue. The remaining 8%, from cryopreserved tissue, and 9%, from fresh tissue, of the total GAGs were obtained after using a papain hydrolysis. There was essentially no difference between fresh and cryopreserved tissues in the relative distribution of proteoglycans in the extracts and digestions except in the initial digestion step where more proteoglycans were obtained from collagenase solubilization of cryopreserved tissue than fresh tissue (p<0.05). The histologic status of the fresh and cryopreserved porcine aortic conduit did not differ markedly. The normal tissue architecture was not affected markedly by the cryopreservation procedure as neither alteration of elastic structure, fibrous proteins nor alteration of nuclear distribution or smooth muscle cell morphology was detected. Quantitative tissue mineral studies revealed that the mean calcium content of the cryopreserved aorta conduit tissue $(165{\pm}3\;{\mu}g/g\;wet\;tissue)$ was higher than that of the fresh tissue $(105{\pm}4\;{\mu}g/g\;wet\;tissue)$ $(p<0.05)$. The mean phosphorus content was $703{\pm}35\;{\mu}g$ wet tissue from cryopreserved tissue and $720{\pm}26\;{\mu}g$ wet tissue from fresh tissue. The study indicates that there is no significant alteration in the distribution of PGs in properly cryopreserved tissue, but the total calcium level appears to be increased in tissue cryopreserved by the cryopreservation process used in this study.
