Occurrence of Fusarium wilt on Cyclamen Casued by Fusarium oxysporum f. sp. cyclaminis and Selection of Resistant Cultivars (Fusarium oxysporum f. sp. cyclaminis에 의한 시클라멘 시들음병 발생 및 저항성 품종 선발)

  • A wilt disease of commercial cyclamen (Cyclamen persicum) which grown in greenhouse was found in Gyeo-nggi province of Korea during the period from August, 2001 to July, 2002. The disease incidence was up to 42.7% in Kimpo, Gyeonggi province. The disease was more severe in ebb and flow irrigation system than con-ventional overhead flooding\'s. The wilted cyclamen plants showed the chlorosis of leaves and followed by the death. The vascular tissues of the infected basal stem and bulb were discolored with black streaks. The casual fungus was identified to be Fusarium oxysporum f. sp. cyclaminis on the basis of mycological characteristics. Effect of infected soil showed 100% infection rate when the cyclamen plants were grown in potting with infested soil. Examine resistant cultivar showed ‘Matis red’, ‘Schubert’, ‘Victoria’ and ‘Chopin’ were the resistant cultivars but most of cultivars were susceptible to Fusarium wilt. The control effect of chemicals for cyclamen wilt was not much efficient to curative effects even though. Benomyl WP and Fludioxonil SC on the Fusarium wilt had only preventive effects by soil drenching in the pot before inoculation of F. oxysporum. This is the first report on the fusarium wilt of cyclamen caused by F. oxysporum f. sp. cyclaminis in Korea.

Biological Control of White Rot in Garlic Using Burkholderia pyrrocinia CAB08106-4 (Burkholderia pyrrocinia CAB08106-4 균주를 이용한 마늘 흑색썩음균핵병의 생물학적 방제)

  • White rot caused by Sclerotium cepivorum was reported to be severe soil-born disease on garlic. Disease progress of white rot of garlic (Allium sativum L.) was investigated during the growing season of 2009 to 2011 at Taean and Seosan areas. The white rot disease on bulb began to occur from late April and peaked in late May. The antifungal bacteria, Burkholderia pyrrocinia CAB08106-4 was tested in field bioassay for suppression of white rot disease. As a result of the nucleotide sequence of the gene 16S rRNA, CAB008106-4 strain used in this study has been identified as B. pyrrocinia. B. pyrrocinia CAB080106-4 isolate suppressed the white rot with 69.6% control efficacy in field test. These results suggested that B. pyrrocinia CAB08106-4 isolate could be an effective biological control agent against white rot of garlic.

Effects of Aeration Rate and Sparger Type on Growth and Ginsenoside Accumulation in Bioreactor Cultures of Ginseng Adventitious Root(Panax ginseng C.A. Meyer) (생물반응기내의 공기주입량 및 Sparger 형태가 인삼 (Panax ginseng C.A. Meyer) 부정근의 생장과 Ginsenoside 함량에 미치는 영향)

  • The two different ways to supply air inside the bioreactor were examined in the adventitious root cultures of Panax ginseng C.A. Meyer. First, the aeration rate varied at 0.05, 0.1, 0.2 and 0.3 vvm, respectively which were supplied during the whole culture period. Second, the amount of air supply was increased from 0.05 to 0.3 vvm at 10-day intervals in proportion to the root growth. Both the root growth and the ginsenoside accumulation were maximized to 175.8 g dry wt. of root growth and 4.3 mg/g dry wt. of ginsenoside accumulation when the aeration rate was increased gradually. The effect of the sparger pore size (15, 30 and $60\;{\mu}m$) in the bioreactor was also investigated, which suggested the greatest root growth (175.9 g dry wt.) in the $15{\mu}m$-sized sparger and the highest ginsenoside content (4.3 mg/g dry wt.) in the $60\;{\mu}m$ size. Finally, the diameter of a sparger ($15\;{\mu}m$-sized) varied at 1.5, 3.0, 5.0 and 8.0 cm, respectively. The highest root growth (191.9 g dry wt.) and the ginsenoside content (4.9 mg/g dry wt.) were obtained in the sparger diameter of 8.0 cm.

Effect of a Mixture of Extracts from Residues of Onion Left after Onion Harvesting and Purslane (Portulaca oleracea) on Productivity and Quality Characteristics of Organic Onions (양파 수확 후 잔재물과 쇠비름 추출물이 유기농 양파의 수확량 및 품질 특성에 미치는 영향)

  • This study assessed the effects of treatment with sap extract from onion residues postharvest and purslane on the quality and quantity of organic onions. At the bending stage, onions treated with the sap extract showed vigorous growth, with higher plant heights, more leaves, and longer sheath lengths than untreated onions. The onion yield was significantly increased when the plant was treated with extracted sap as compared with that of untreated plants (p<0.05). The bulb weight distribution of onions in the mixed onion and purslane treatment was also significantly increased (~300 g) as compared with that of the other treatment (p<0.05). Except for CaO and S, the mineral content of the onions produced from plants treated with the onion and purslane extract mixture was higher than those of onions in the other treatment. The hardness of onions produced from plants treated with the onion and purslane extract was significantly increased (8% and 20%, respectively) as compared with that of onions produced from plants treated with the onion extract only or no treatment (p<0.05). However, there was no significant difference in the sugar contents of the onions produced from extract-treated and nonextract-treated plants. Postharvest, the content of inorganic components (phosphate, calcium, sulfuric acid, and manganese) was higher in soil treated with the onion extract than in soil treated with the onion and purslane extract and non-treated soil. It can be concluded that residues left after onion harvests and purslane extract can be used as natural and environmentally friendly materials for the cultivation of organic onions.

In Vitro Propagation of Lilium Asiatic Hybrid 'Hae Hwa' via the Formation of Shoot Clusters (신초 Cluster 형성에 의한 Lilium Asiatic Hybrid 'Hae Hwa'의 기내번식)

  • This experiment was conducted to micropropagate bulblets via shoot cluster formation and massproduce normal bulblets from the sections of proliferated shoot clusters in Lilium asiatic hybrid 'Hae Hwa'. The induction of shoot clusters from the culture of bulblet sections was more effective than that of bulb scales on MS medium with 1.0 mg/L BA and 0.5 mg/L IAA. Proliferation of shoot clusters from the formed shoot cluster sections was the most favorable on medium containing 5.0 mg/L BA and 0.5 mg/L IAA. The formation and the growth of bulblets from shoot cluster sections were achieved effectively on medium with 60∼90 g/L sucrose. The leaves derived from shoot clusters grew vigorously but the bulblets from shoot clusters grew very poor in 5L air-lift bioreactor culture. By the addition of 30 mL fresh liquid medium containing doulble strength MS salts, 250 g/L sucrose and 5 g/L activated charcoal after 8 weeks in the shoot cluster culture on MS medium with 5.0 mg/L BA and 0.5 mg/L IAA, the number of bulblets was increased in light condition, but the growth of bulblets was not affected by light. Bulblet production was possible with the bulblet product at 53 to 68 mg in fresh weight by liquid medium addition after the proliferation of shoot cluster.

  • Onion has been reported to contain various organosulfur compounds which have antibiotic and anticarcinogenic properties and flavonoid like quercetin which is a valuable natural source of antioxidants. Carbohydrates in onion constitute about 80% of dry matter, and the major non-structural carbohydrate of onion bulb is fructo-oligosaccharides, well known as fructan, followed by glucose, fructose, and sucrose. The sugar concentration is associated with dormancy and storage life of onion, occurring as decrease in glucose, fructose and fructan, particularly towards the end of storage. Forced air pre-drying for 15-20 days at room temperature is an essential procedure to reduce freezing injury and sprouting, then onion bulbs can be stored at $0^{\circ}C$ for 6 months to control sprouting and decay. Bacterial soft rot caused by $Erwinia$ and $Pseudomonas$ is the main postharvest disease when the bulbs are infected with the bacteria and stored at room temperature. Browning in sliced onion is due to oxidation of phenolic compounds by polyphenol oxidase and it can be inhibited by citric acid treatment, packing with nitrogen gas, and polyethylene film.

  • There are two kinds of calla belonging to the Zantedeschia spp. depending on the growing condition. Between them, various colored calla with flower shape and flower color are very popular as cut flowers. However, colored calla is very low in natural reproduction rate, and it takes a long time to obtain a flowering bulb. So it is urgent to establish a large breeding system of good varieties. In this study, various plant growth hormone treatments were carried out to improve the growth of the plants in calla. Root formation was the highest at 81.5 % in basal MS medium, and the formation of shoot and multiple shoots were the highest at 100.0 % and 36.4 % in the combined treatment of BA 2.0 mg/L and IBA 1.0 mg/L, respectively. In fresh weight, the highest growth rate was observed with 11.2 times increase in BA 2.0 mg/L single treatment. Auxin and cytokinin mixed treatment were widely used in previous studies and positive effects on the growth of calla plants will help to establish the micro-propagation system.

Morphological and Agronomic Traits of Allium longicuspis Regel Collected in Central Asia (중앙아시아에서 수집한 꽃피는 마늘의 형태 및 농업적 특성)

  • Garlic (Allium sativum L.) has been used as both food and medicine in many cultures for thousands of years. Garlic cultivars are completely sterile and propagated through vegetative method. Collection of a large number of fertile accessions of these genus is needed to explore genetic variability. In order to investigate genetic variation among Allium species and its possibilities for direct cultivation in Korea, we characterized 12 accessions of A. longicuspis, flowering wild garlic which had collected from Central Asia, the main center of garlic diversity. Most of A. longicuspis accessions showed higher over-wintering and bolting rate, longer scape length and more number of bulbils than Korean landraces cultivar, Danyang and Euiseong, but A. longicuspis accessions exhibited smaller size of bulbs and bulbils. Most accessions of A. longicuspis had more number of cloves per bulb, except K229596 and K248824 than Korean landraces. All the accessions of A. longicuspis from Central Asia had complete bolters having many flowers and topsets in umbel. Further studies of A. longicuspis should focus on securing true seeds through removal of topsets and crosses among accessions to create the genetic variability.

Physiological Characteristics and Yield of Onion Affected by Rapid Temperature Changes (급변온도 변이에 따른 양파의 생리적 특성 및 수량 변화)

  • BACKGROUND: To evaluate the impact of rapid temperature change at spring and the early summer seasons in climate change, we have investigated the physiological response and yield of onion in a greenhouse with thermostat control system. METHODS AND RESULTS: Seedlings of onion(cv. Sunshine) were planted on October 30, 2012 and harvested on May 30, 2013. The used treatments(March-April-May) for a rapid temperature change were T0(control): $6.0-10.4-17.2^{\circ}C$, T1: $6.0-5.4(-5)-17.2^{\circ}C$, T2: $6.0-10.4-22.2(+5)^{\circ}C$ and T3: $6.0-5.4(-5)-22.2(+5)^{\circ}C$. Total yields of bulb within the temperature change as high temperature treatment T2 and control treatment T0 were increased significantly(p<0.05), as compared to the low temperature treatment T1. Low temperature conditions significantly (p<0.05) reduced plant height, SPAD reading, crude protein and fiber etc., as compared to the TO and T2. CONCLUSION: The rapid temperature changes were highly affected by low temperature than high temperature. These results suggest that rapid climate change of future could need systematic standard model for physiological characteristics and yields of onion.

Inhibitory effect of Allium macrostemon extracts on adipogenesis of 3T3-L1 preadipocytes (산달래 추출물의 3T3-L1 지방전구세포 분화 억제 효능)

  • The aim of this study was to compare the biological activities of whole-plant (WAE), bulb (BAE), and leaf (LAE) extracts of Allium macrostemon. The antioxidant activities, total polyphenol contents, and anti-adipogenic activities of WAE and LAE were superior to those of BAE, whereas the biological effects of WAE and LAE were similar. Therefore, the effect of LAE on adipogenesis was further investigated. Treatment of preadipocytes with LAE at 100 g/mL resulted in the inhibition of intracellular lipid accumulation by 49.64%. Consistent with this result, quantitative reverse transcription-PCR showed that LAE treatment decreased the gene expressions of CCAAT/enhancer-binding protein beta (C/EBPβ), peroxisome proliferator-activated receptor gamma (PPARγ), C/EBPα and stearoyl-CoA desaturase 1 (SCD1). Thus, LAE attenuates the adipogenesis of preadipocytes by suppressing the expression of adipogenic and lipogenic genes. These results suggest that LAE can be potentially useful as a functional ingredient to prevent obesity in the food industry.
