Current Status of the Research and Development of Bispecific Antibodies

  • A bispecific antibody (BsAb) is an artificial protein containing two kinds of specific antigen binding sites. BsAb can connect target cells to functional cells or molecules, and thus stimulate a directed immune response. Last several decades a wide variety of bsAb formats and production technologies have been developed. BsAbs are constructed either chemically or biologically, exploiting techniques like cell fusion and recombinant DNA technologies. There are over 100 different formats of bsAb so far developed, but they could be classified into the two main categories such as Fc-based (with a Fc region) bsAbs and fragment-based (without a Fc region) bsAbs. BsAb has a broad application prospect in tumor immunotherapy and drug delivery. Here, we present a brief introduction to the structure of antibody, pharmacological mechanisms of antibodies and the trend in the production technologies of therapeutic antibodies. In addition, we address a review on the current status of various bsAb format development and their production technologies together with global situation in the clinical studies of bsAb.

Anti-HER-2×anti-CD3 Bi-specific Antibodies Inhibit Growth of HCT-116 Colorectal Carcinoma Cells in Vitro and in Vivo

  • Objective: This study is conducted to evaluate the effects of anti-HER-2${\times}$anti-CD3 bi-specific antibodies(BsAb) on HER-2/neuover-expressing human colorectal carcinoma cells. Methods: Growth was assessed by MTT assays after exposure of HCT-116 cells to Herceptin, anti-CD3 and BsAb antibodies. Immunocytochemistry was applied to test the HER-2 level of HCT-116. In a nude mouse model, HER-2${\times}$CD3 BsAb was combined with effector cells (peripheral blood lymph cells from normal human being) for observations on in Vivo growth of tumors. Results: Compared with the control group, using effector cells combined with anti-CD3 McAb, Herceptin or HER2${\times}$CD3 BsAb, tumor cell growth in vitro and in vivo was significantly inhibited (P<0.05), most remarkably in the HER2${\times}$CD3 BsAb case. The growth of xenografts with HER2${\times}$CD3 BsAb combined with effector cells was also significantly inhibited when compared with the anti-CD3 McAb or Herceptin groups (P<0.05). Conclusion: HER-2/neu might be a useful target for immunotherapy in colorectal carcinoma, anti-HER2${\times}$anti-CD3 BsAb exerting clear anti-tumor effects.

Changes in Quality Characteristics of Gochujang Prepared with Different Ingredients and Meju Starters (고추장 재료와 종균 첨가에 따른 고추장의 품질 변화)

  • Changes in quality characteristics of gochujang prepared with different ingredients (white rice, barley, sorghum and millet, bamboo salt) and meju starters were studied. The gochujang samples were fermented for 6 weeks at $40^{\circ}C$ and analyzed for changes in their physicochemical properties and sensory characteristics. Eight different gochujang samples were prepared: WR-SS-AB, WR-SS-ABL, WR-BS-AB, WR-BS-ABL, MG-SS-AB, MG-SS-ABL, MG-BS-AB, MG-BS-ABL (WR: white rice, MG: mixed grains, SS: solar salt, BS: bamboo salt, AB: Aspergillus oryzae/Bacillus subtilis starter meju, ABL: Aspergillus oryzae/Bacillus subtilis/Lactobacillus plantarum starter meju). There were significant differences between experimental groups in terms of moisture content, pH, acidity, and color value. All gochujang samples showed increased moisture contents and pH as well as increased acidity and amino-type nitrogen during fermentation. MG-BS-ABL showed the highest contents of amino-type nitrogen and free amino acids after 6 weeks of fermentation compared with other experiment groups. In addition, MG-BS-ABL showed the highest sensory analysis score, including appearance, flavor, taste, and overall acceptability after the fermentation of samples. Based on the results, MG-BS-ABL exhibited similar quality characteristics as general gochujang but showed the highest sensory scores and contents of amino-type nitrogen and free amino acids. These results might be due to the high protein contents of mixed grains and high mineral contents of bamboo salt and fermented products from mixed probiotic starters.

  • In the era of immunotherapeutic control of cancers, many advances in biotechnology, especially in Ab engineering, have provided multiple new candidates as therapeutic immuno-oncology modalities. Bispecific Abs (BsAbs) that recognize 2 different antigens in one molecule are promising drug candidates and have inspired an upsurge in research in both academia and the pharmaceutical industry. Among several BsAbs, T cell engaging BsAb (TCEB), a new class of therapeutic agents designed to simultaneously bind to T cells and tumor cells via tumor cell specific antigens in immunotherapy, is the most promising BsAb. Herein, we are providing an overview of the current status of the development of TCEBs. The diverse formats and characteristics of TCEBs, in addition to the functional mechanisms of BsAbs are discussed. Several aspects of a new TCEB-Blinatumomab-are reviewed, including the current clinical data, challenges of patient treatment, drawbacks regarding toxicities, and resistance of TCEB therapy. Development of the next generation of TCEBs is also discussed in addition to the comparison of TCEB with current chimeric antigen receptor-T therapy.

Study on Utilization of Heterosis in Layer Chicken (산란계종의 잡잡강세리용에 관한 연구)

  • This study was carried out to estimate combining abilitie or economic traits in layer chickens. The data used in this study were the record of 10 single crosses produced by half diallel cross of 5 lines of Single Comb White Leghorns, such as A, B, C, K and S lines. Total 720 progenies of the crosses were reared at the Poultry Breeding Farm, College of Agriculture, Seoul National University from Feb. 1979 to August, 1980. Combining abilities were estimated by Grilling's mathematical model for the traits; age the first egg, total egg number, egg weight and body weight. The results attained from the studies were summarized as follows; In estimate of combining ability, an age at first egg of BS cross was largely due to significiantly higher general combining ability (G. C. A.) effect of B and S strains than Cand K strains in G. C. A. effect, and to specific combining ability (S. C. A) effect of B and S-trains. AB and BS crosses showed the highest egg Production. AB cross performance was result from high G.C.A. effect of A ana B strains. BS cross performance was result from high G. C. A. effect of B and high S. C. A. effect of BS cross. Specific combining ability effect in egg. weight was not statiscally significiant, but S strain showed high G. C. A. effect. A and B strains in body weight showed significantly low C. C. A. effect. From the above results, BS cross in an age at first egg, AB and BS crosses in egg Production, S strain in egg weight and AB cross in body weight were superior to other strains or crosses.

A Study of the usage of Corrugated Fiberboard Cartons for Domestic Fresh Produce and Their Physical Properties: Focused on a Gyeongsan City, North Gyeongsan Province (국내 신선 농산물 포장용 골판지 상자의 이용 실태 및 물리적 특성 연구: 경북 경산 소재 지역 중심으로)

  • In this study, forty corrugated fiberboard (CF) boxes for fresh produce were collected from farms located in Gyeongsan city and evaluated its characterizations and physical properties according to Korean Standard (KS). The basis weight, flute, and paperboard combination were determined for the characterizations of CF boxes. The measured basis weights of all samples were within the tolerance limit referred to KS and the flutes were B/F, EB/F, and AB/F flute, respectively. The bursting strength (BS), edgewise crush resistance (ECR), and compression strength (CS) were tested to evaluate the physical properties. There are four single-walled (SW) CF boxes showed BS and ECR higher than 638 kPa and 3.30 kN/m, respectively. One SW CF boxes showed BS and ECR higher than 785 kPa and 3.43 kN/m, respectively. In case of double-walled (DW) CF boxes, sixteen CF boxes showed BS and ECR higher than 785 kPa, 4.96 kN/m (AB flute), or 4.90 (EB flute), respectively. The other nineteen CF boxes showed BS and ECR higher than 981 kPa, 5.29 kN/m (AB flute), or 5.20 (EB flute), respectively. All the CF boxes except two samples showed the acceptable compression strength in reference to the guideline of KS.

COMMON LOCAL SPECTRAL PROPERTIES OF INTERTWINING LINEAR OPERATORS

Iron sulfate and molasses treated anthocyanin-rich black cane silage improves growth performance, rumen fermentation, antioxidant status, and meat tenderness in goats

  • Objective: This study investigated the effects of feeding anthocyanin-rich black cane treated with ferrous sulfate and molasses on animal performance, rumen fermentation, microbial composition, blood biochemical indices, and carcass characteristics in meat goats. Methods: Thirty-two Thai-native×Anglo-Nubian crossbred male goats (14.47±2.3 kg) were divided equally into two groups (n = 16) to investigate the effect of feeding diet containing 50% untreated anthocyanin-rich black cane silage (BS) vs diet containing anthocyaninrich black cane silage treated with 0.03% ferrous sulfate and 4% molasses (TBS) on average daily gain (ADG) and dry matter intake (DMI). At the end of 90 d feeding trial, the goats were slaughtered to determine blood biochemical indices, rumen fermentation, microbial composition, and carcass characteristics differences between the two dietary groups. Results: Goats fed the TBS diet had greater ADG and ADG to DMI ratio (p<0.05). TBS diet did not affect rumen fluid pH; however, goats in the TBS group had lower rumen ammonia N levels (p<0.05) and higher total volatile fatty acid concentrations (p<0.05). Goats in the TBS group had a higher (p<0.05) concentration of Ruminococcus albus but a lower (p<0.05) concentration of methanogenic bacteria. The TBS diet also resulted in lower (p<0.05) thiobarbituric acid-reactive substances concentration but higher (p<0.05) total antioxidant capacity, superoxide dismutase, catalase, glutathione peroxidase, and glutathione reductase concentrations in blood plasma, while having no effect on plasma protein, glucose, lipid, immunoglobin G, alanine transaminase, and aspartate aminotransferase. Meat from goats fed the TBS diet contained more intramuscular fat (p<0.05) and was more tender (p<0.05). Conclusion: In comparison to goats fed a diet containing 50% untreated anthocyanin-rich black cane silage, feeding a diet containing 50% anthocyanin-rich black cane silage treated with 0.03% ferrous sulfate and 4% molasses improved rumen fermentation and reduced oxidative stress, resulting in higher growth and more tender meat.
