방사선 처리에 의한 톨 페스큐 돌연변이 식물체 선발 (In vitro Technique for Selection of Radiation Induced Mutants of Tall Fescue)

  • In vitro culture and radiation techniques were used for obtaining mutants tin tall fescue. Endophyte free and friendly tall fescue cultivars Kentucky-31 and Jesup were used for induction of genetic variability through in-vitro mutagenesis. Mature seeds was used for callus induction on 6 mg/L 2,4-D. Actively growing and compact callus was treated with three different doses of gamma rays (10 Gy, 30 Gy and 50 Gy). Maximum proliferation and plantlets regeneration growth was observed in control and minimum at 10 Gy. Furthermore, the maximum number of tiller in the irradiated population was observed in 10 Gy. The treatments 30 Gy and 50 Gy exhibited negative impact on the tillering potential of the tall fescue plant. The object of this study was to develop protocols for mutation breeding in tall fescue through radiation techniques.

오이 다형성 마커를 이용한 유전분석 (Genetic Analysis of Polymorphic DNA Markers in Cucumber)

  • DNA 마커는 유전현상 분석이나 품종육성에 널리 사용되고 있다. 본 연구에서는 내냉성 오이 계통인 'NC76'과 냉해 감수성 계통인 'GY14'로부터 내냉성 연관 마커을 목적으로 기존의 총 995개 SSR 마커의 다형성을 평가하였다. Agarose gel 전기영동법으로 'NC76과 'GY14' 간 PCR증폭 산물의 길이 다형성을 보이는 145개 SSR 마커를 개발하였으며, high resolution melting (HRM) 기술을 사용하여 염기서열 다형성을 보이는 30개의 SSR 마커를 확인하였다. 개발된 175개 SSR 마커 중 20개 마커를 선발하여 'NC76'과 'GY14' 간 $F_2$ 분리 집단에 대한 연관지도를 작성하였으며 그 결과 13개의 마커가 예상했던 연관군에 일치하여 위치됨을 확인할 수 있었다. 따라서 본 연구에서 확인된 175개의 SSR 마커는 향 후 냉해 저항성 연관 마커 개발을 위한 오이 유전자 지도 작성 및 이를 통한 품종 육성에 크게 활용될 수 있을 것으로 기대된다.

SLA Homozygous Korean Native Pigs and Their Inbreeding Status Deduced from the Microsatellite Marker Analysis

  • The porcine MHC (Major Histocompatibility Complex), encoding the SLA (Swine Leukocyte Antigen) genes, is one of the most significant regions associated with immune rejection in relation to transplantation. In this study, three SLA class I (SLA-1, SLA-3, SLA-2) loci and three SLA class II (DRB1, DQB1, DQA) loci were investigated in the previously unidentified Korean native pig (KNP) population that was closely inbred in the Livestock Technology Research Station in Cheongyang, Korea. Total thirteen KNPs from four generations were genotyped for the SLA alleles and haplotypes were investigated using PCR-SSP (Sequence-Specific Primer) method. The results showed that all of these KNPs had Lr-56.30/56.30 homozygous haplotype, indicating high level of inbreeding in the SLA genes. The inbreeding status of these animals was also investigated using microsatellite (MS) markers. From the 50 MS markers investigated, 17 MS markers were fixed in all generations and the fixed alleles are increased as 26 loci for the fourth generation. Two MS markers, S0069 and SW173, were heterozygous for all the animals tested. Observed and expected heterozygosities were calculated and the average inbreeding coefficients for each generation were also calculated. In the fourth generation, the average inbreeding coefficients was 0.732 and this may increase with further inbreeding process. Analysis of the SLA haplotypes and MS alleles can give important information for breeding the pigs for xenotransplantation studies.

Medicago truncatula in Interaction with Fusarium and Rhizoctonia Phytopathogenic Fungi: Fungal Aggressiveness, Plant Response Biodiversity and Character Heritability Indices

  • Fusarium and Rhizoctonia genera are important pathogens of many field crops worldwide. They are constantly evolving and expanding their host range. Selecting resistant cultivars is an effective strategy to break their infection cycles. To this end, we screened a collection of Medicago truncatula accessions against Fusarium oxysporum, Fusarium solani, and Rhizoctonia solani strains isolated from different plant species. Despite the small collection, a biodiversity in the disease response of M. truncatula accessions ranging from resistant phenotypes to highly susceptible ones was observed. A17 showed relative resistance to all fungal strains with the lowest disease incidence and ratings while TN1.11 was among the susceptible accessions. As an initiation of the characterization of resistance mechanisms, the antioxidant enzymes' activities, at the early stages of infections, were compared between these contrasting accessions. Our results showed an increment of the antioxidant activities within A17 plants in leaves and roots. We also analyzed the responses of a population of recombinant inbred lines derived from the crossing of A17 and TN1.11 to the infection with the same fungal strains. The broad-sense heritability of measured traits ranged from 0.87 to 0.95, from 0.72 to 0.96, and from 0.14 to 0.85 under control, F. oxysporum, and R. solani conditions, respectively. This high estimated heritability underlines the importance of further molecular analysis of the observed resistance to identify selection markers that could be incorporated into a breeding program and thus improving soil-borne pathogens resistance in crops.

Genetic diversity analysis of tea (Camellia sinensis (L.) O. Kuntze) germplasm in Korea genebank

  • Tea plant (Camellia sinensis (L.) O. Kuntze) has been cultivated widely in many developing Asian, African, and South American countries, where it is the most widely consumed beverage in the world next to water. It has critical importance to understand the genetic diversity and population sturcutre for effective collection, conservation, and utilization of tea germplasm. In this study, 410 tea accessions collected from South Korea were analyzed using 21 SSR markers. Among 410 tea accessions, 85.4% (310 accessions) accessions were collected from Jeollanam-do. A total of 286 alleles were observed, and the genetic diversity and evenness were estimated to be averagely 0.79 and 0.61, respectively, across all the tested samples. Using discriminant analysis of principal components, the four clusters were detected in 410 tea accessions. Among them, cluster 1 showed higher frequency of rare alleles (less than 1%) than other clusters. Using calculation of the index of association and rbaD value, each cluster showed a clonal mode of reproduction. The result of AMOVA showed that most of the variation observed was within populations (99%) rather than among populations (1%). Our results might contribute to provide data about genetic diversity for the conservation of tea germplasm and for future breeding programs.

Microsatellite Markers를 이용한 따오기의 유전적 특성 분석 (Genetic analysis of endangered species Crested Ibis (Nipponia nippon) microsatellite markers)

  • 세계적으로 멸종위기종인 따오기 (Nipponia Nippon)는 한국에서도 멸종위기종으로 구분되어 있으며, 이를 복원하기 위해 2008년 10월에 중국에서 따오기 1쌍을 도입하여 한국 최초로 인공번식에 성공하였다. 이 후 우포늪 따오기 종 복원을 통해 2017년까지 개체수가 200마리 이상으로 늘어났다. 본 연구에서는 우포늪 따오기 228 개체를 대상으로 성별을 결정하고, microsatellite 마커를 이용하여 유전적 유연관계를 분석하였다. 그 결과, 115마리의 암컷과 113마리의 수컷으로 판별되었으며, 2016년보다 2017년에 서식하고 있는 개체의 이형접합도와 다형성 정보지수가 모두 감소한 것으로 나타났다. 이는 작은 집단으로부터 개체 수를 늘려나가다 보니 2017년에 근친율이 증가한 것으로 사료된다. 향 후 본 연구는 한국 따오기의 번식사업 및 복원사업을 위한 기초자료로 유용하게 활용될 것으로 기대된다.

Impact of Confinement and Population Size on the Instrumentally Inseminated Queen's Performance of Apis cerana Species in South Korea

  • Instrumental insemination of honey bee is an attractive alternative to natural mating in breeding program as it allows mating crosses between desirable queen and specific drone. However, nursery condition that the queen is kept before and after insemination is major factor affected to the performance of instrumentally inseminated queen. In this study, we evaluated the influences of three different nursery-conditions of push-in cages, mini nuclei and normal colonies on number of spermatozoa stored in the spermatheca, body weight, onset of ovipositon and performance of instrumentally inseminated Apis cerana queen. Our results demonstrated that instrumentally inseminated queens kept in mini nuclei and in normal hives showed no significant difference in queen's weight (159.8 and 166.2mg, respectively), number of spermatozoa in spermatheca ($2.02{\times}10^6$ and $2.76{\times}10^6$, respectively), proportion of queen supersedure (33.3 and 66.7% queen survival at 11 months after oviposition, respectively) and brood production, compared to naturally mated queens. In contrast, instrumentally inseminated queens kept in push-in cages showed significant difference of those above data in comparison to queens mated naturally. Our results suggested that instrumentally inseminated queens could be kept in mini nuclei containing about 1.000 attendant bees to have desirable performance of queen whereas the push-in method should be practiced for the purpose of using queen in the length of time less than 7 months.

Exploring effects of different male parent crossings on sheep muscles and related regulatory genes using mRNA-Seq

  • Objective: With improvements in living standards and increase in global population, the demand for meat products has been increasing; improved meat production from livestock could effectively meet this demand. In this study, we examined the differences in the muscle traits of different male crossbred sheep and attempted to identify key genes that regulate these traits. Methods: Dubo sheep×small-tailed Han sheep (DP×STH) and Suffolk×small-tailed Han sheep (SFK×STH) were selected to determine meat quality and production performance by Masson staining. Transcriptome sequencing and bioinformatic analysis were performed to identify differentially expressed genes (DEGs) related to meat quality. The presence of DEGs was confirmed by real-time polymerase chain reaction. Results: The production performance of SFK×STH sheep was better than that of DP×STH sheep, but the meat quality of DP×STH sheep was better than that of SFK×STH sheep. The muscle fiber diameter of DP×STH sheep was smaller than that of SFK×STH sheep. Twenty-two DEGs were identified. Among them, four gene ontology terms were related to muscle traits, and three DEGs were related to muscle or muscle fibers. There were no significant differences in the number of single nucleotide mutations and mutation sites in the different male parent cross combinations. Conclusion: This study provides genetic resources for future sheep muscle development and cross-breeding research.

Genetic parameter analysis of reproductive traits in Large White pigs

  • Objective: The primary objective of this study was to determine the genetic parameters for reproductive traits among Large White pigs, including the following traits: total number born (TNB), number born alive (NBA), litter birth weight (LBW), average birth weight (ABW), gestation length (GL), age at first service (AFS) and age at first farrowing (AFF). Methods: The dataset consisted of 19,036 reproductive records from 4,986 sows, and a multi-trait animal model was used to estimate genetic variance components of seven reproductive traits. Results: The heritability estimates for these reproductive traits ranged from 0.09 to 0.26, with the highest heritability for GL and AFF, and the lowest heritability for NBA. The repeatabilities for TNB, NBA, LWB, ABW, and GL were ranged from 0.16 to 0.34. Genetic and phenotypic correlations ranged from -0.41 to 0.99, and -0.34 to 0.98, respectively. In particular, the correlations between TNB, NBA and LBW, between AFS and AFF, exhibited a strong positive correlation. Furthermore, for TNB, NBA, LBW, ABW, and GL, genetic correlations of the same trait between different parities were moderately to strongly correlated (0.32 to 0.97), and the correlations of adjacent parities were higher than those of nonadjacent parities. Conclusion: All the results in the present study can be used as a basis for the genetic assessment of the target population. In the formulation of dam line selection index, AFS or AFF can be considered to combine with TNB in a multiple trait swine breeding value estimation system. Moreover, breeders are encouraged to increase the proportion of sows at parity 3-5 and reinforce the management of sows at parity 1 and parity ≥8.

QTL Mapping for Protein Content Derived from a Cross between oryza sativa and Weedy Rice

  • Protein is a major nutrient of food and has long been studied for nutritional and utility value. Among them, rice protein is attracting attention because of its hypoallergenic characteristics and nutritional value. Mutant DM225 with increased protein content was selected by EMS treatment on the weed rice Dharial. QTL analysis of Protein content was carried out using BC2F2 populations derived from a cross between "Hanareum2" as a recurrent parent and "DM225" as a donor parent. The protein content of populations was between 5~11%, with an average of 7.7%. To identify QTLs related to Protein content, 117 KASP markers(polymorphic ratio: 15%) showing polymorphisms between the parents were genotyped for the BC2F2 population. One QTL was detected between markers SK07 06 and SK07_10 on chromosome 7(LOD: 28.1). This QTL explained 71.4% of the phenotypic variance for Protein content. This QTL will be useful for protein-related rice breeding program.
