Heritabilities, Genotypic and Phenotypic Correlations, and Selection Indices in Aromatic Tobacco Varieties(Nicotiana tabacum L. (향끽미종 잎담배 유전에 관한 연구)

  • To obtain genetic information for breeding aromatic tobacco, broad sense heritabilities, phenotypic, and genotypic correlations were obtained for the following characters : days to frowert, plant height, number of leaves, leaf shape and nicotine. Six pure line varieties and the first and second generations of 5 crosses among them were employed in this study. The heritabilities for number of leaves and nicotine were very high in pure line varieties. However, the heritabilities for days to flower and yield were low in six varieties as well as in F2. Genotypic correlations had higher values than phenotypic did. Heritabilities calculated from segregating generations were lower than those from pure varieties, regardless of crosses involved. Positive phenotypic correlations between yield and plant height were observed in all crosses. Same phenomenon was observed with correlations between days to flower and number of leaves. The genotypic and phenotypic variances and covariances entering into the computation of correlations were used to construct the seletion indices for yield. The use and limitation of selection index was also discussed.

  • A high seed yield '232' was evaluated for morphological characteristics and tissue culture re-sponse. The effect of mowing and urea application on coverage in '232' was also studied and the results were as follows ;1.'232' produced high seed yield than any other lines, but its other characteristics such as coverage, establishment rate, stolon length, stolon number and colour change were not good. 2.Callus induction of '232'(52.9%) was relatively high, and higher than that of Z. sinica(49.0%). However, embryogenic callus formation of '232' was not as good as that of Z. japonica and Z. sinica. 3.Coverage of '232' was very much enhanced by mowing on July 18, but not at all by mowing on May 29 and September 4. 4.Urea application did not increase the coverage in '232',Therefore, it can be used as a breeding material rather than direct use.

  • Pectobacterium carotovorum subsp. carotovorum (Pcc) causes soft rot disease in various plants. Although many studies about Pcc have been going on, little is known yet about the defense genes from plants. To identify defense associated genes in response to Pcc, we screened about 20 thousand Arabidopsis T-DNA knock out lines by inoculation with Pcc. We obtained a line (Atspt1) showing more susceptible symptom compared to WT (Col-0) on 1 day after the inoculation of Pcc on leaves of Arabidopsis with toothpicks. In this study, we optimized the system to select resistant and susceptible lines to Pcc from T-DNA inserted pool of Arabidopsis and expect the system and Atspt1 might be used for molecular breeding to produce resistant vegetables against Pcc.

A New Early-Heading, High-Yielding Triticale Cultivar for Forage, 'Shinseong' (숙기가 빠르고 종실 수량이 많은 트리티케일 신품종 '신성')

  • 'Shinseong', a winter forage triticale cultivar (X Triticosecale Wittmack), was developed at the Department of Rice and Winter Cereal Crop, NICS, RDA in 2015. The cultivar 'Shinseong' was selected from the cross RONDO/2$^*$ERIZO_11//KISSA_4/3/ASNO/ARDI_3//ERIZO_7 by CIMMYT (Mexico) in 1998. Subsequent generations were handled in pedigree selection programs at Mexico from 1999 to 2004, and a line 'CTSS98Y00019S-0MXI-B-3-3-5' was selected for earliness and good agronomic characteristics. After preliminary and advance yield testing in Korea for 3 years, the line was designated 'Iksan47'. The line was subsequently evaluated for earliness and forage yield in seven locations, Jeju, Iksan, Cheongwon, Yesan, Gangjin, Daegu, and Jinju from 2013 to 2015 and was finally named 'Shinseong'. Cultivar 'Shinseong' has the characteristics of light green leaves, yellow culm and spike, and a medium grain of brown color. The heading date of cultivar 'Shinseong' was April 24 which was 3 days earlier than that of check cultivar 'Shinyoung'. The tolerance or resistance to lodging, wet injury, powdery mildew, and leaf rust of 'Shinseong' were similar to those of the check cultivar. The average forage dry matter yield of cultivar 'Shinseong' at milk-ripe stages was $15MT\;ha^{-1}$, which was 3% lower than that ($15.5MT\;ha^{-1}$) of the check cultivar 'Shinyoung'. The silage quality of 'Shinseong' (6.7%) was higher than that of the check cultivar 'Shinyoung' (5.9%) in crude protein content, while was similar to the check cultivar 'Shinyoung' in acid detergent fiber (34.6%), neutral detergent fiber (58.6%), and total digestible nutrients (61.6%). It showed grain yield of $7.2MT\;ha^{-1}$ which was 25% higher than that of the check cultivar 'Shinyoung' ($5.8MT\;ha^{-1}$). This cultivar is recommended for fall sowing forage crops in areas in which average daily minimum mean temperatures in January are higher than $-10^{\circ}C$.

Recurrent parent genome (RPG) recovery analysis in a marker-assisted backcross breeding based on the genotyping-by-sequencing in tomato (Solanum lycopersicum L.) (토마토 MABC 육종에서 GBS(genotyping-by-sequencing)에 의한 RPG(recurrent parent genome) 회복률 분석)

  • Marker-assisted backcrossing (MABC) is useful for selecting an offspring with a highly recovered genetic background for a recurrent parent at early generation to various crops. Moreover, marker-assisted backcrossing (MABC) along with marker-assisted selection (MAS) contributes immensely to overcome the main limitation of the conventional breeding and it accelerates recurrent parent genome (RPG) recovery. In this study, we were employed to incorporate rin gene(s) from the donor parent T13-1084, into the genetic background of HK13-1151, a popular high-yielding tomato elite inbred line that is a pink color fruit, in order to develop a rin HK13-1084 improved line. The recurrent parent genome recovery was analyzed in early generations of backcrossing using SNP markers obtained from genotyping-by-sequencing analysis. From the $BC_1F_1$ and $BC_2F_1$ plants, 3,086 and 4868 polymorphic SNP markers were obtained via GBS analysis, respectively. These markers were present in all twelve chromosomes. The background analysis revealed that the extent of RPG recovery ranged from 56.7% to 84.5% and from 87.8% to 97.8% in $BC_1F_1$ and $BC_2F_1$ generations, respectively. In this study, No 5-1 with 97.8% RPG recovery rate among $BC_2F_1$ plants was similar to HK13-1151 strain in the fruit shape. Therefore, the selected plants were fixed in $BC_2F_2$ generation through selfing. MAS allowed identification of the plants that are more similar to the recurrent parent for the loci evaluated in the backcross generations. MABC can greatly reduce breeding time as compared to the conventional backcross breeding. For instance, MABC approach greatly shortened breeding time in tomato.

Development of Near-Isogenic Line of japonica Rice Cultivar Saenuri without Lipoxygenase-3 (새누리 벼 품종 배경 lipoxygenase-3 결핍 자포니카 근동질계통 개발)

  • It is reported that the absence of lipoxygenase-3 (LOX-3) may contribute to a reduction in stale flavor after the storage of rice. To improve the quality of stored rice of the Korean japonica rice cultivar, we conducted a breeding program to develop near-isogenic rice without LOX-3 in the genetic background of Saenuri, a mega variety of Korea. In the first step of the breeding program, we used a donor parent of LOX-3 null, Daw Dam, and a recurrent japonica parent, Sindongjin, to develop HR27873-AC12 by backcross (BC1), color test for introgression of lox-3, and anther culture for rapid fixation. In the second step, we used the donor parent, HR27873-AC12, and the recurrent parent, Saenuri, to develop HR28896-31-3-1-1 by backcross (BC1), marker-assisted selection (MAS) for lox-3, and phenotypic selection (PS) for agronomic traits. Finally, in the third step, we developed HR30960-186-2-1-2-1 (Jeonju624), derived from a cross between Saenuri and HR28896-31-3-1-1, by MAS for lox-3 and PS with high selection pressure for agronomic characteristics. Jeonju624 was confirmed with the introgression of lox-3 by molecular marker. Jeonju624 was a mid-late maturing rice with similar agronomic characteristics to Saenuri, lodging tolerance with short culm, erect plant architecture, and resistance to bacterial blight and rice stripe virus. The yield components of Jeonju624 were mostly similar to Saenuri, except for the 1,000-grain weight of brown rice. The appearance of the grain of Jeonju624 was better than that of Saenuri, and the characteristics of cooked rice were similar to those of Saenuri. In the genetic background analysis using 406 KASP (Kompetitive Allele-Specific PCR) markers, Jeonju624 was confirmed to be the near-isogenic line (NIL) of Saenuri with a 95.8% recovery rate. Jeonju624 is the NIL of Saenuri without LOX-3, and overcomes the linkage drag of Daw Dam with similar agronomic characteristics and genetic background to Saenuri. Jeonju624 can be utilized as a practical cultivar to improve the quality of stored rice, breeding material for the introgression of lox-3, and genetic material to elucidate the effect of introgressed genes.

Combining Ability for Morphological and Biochemical Characters in Mulberry (Morns spp.) under Salinity Stress

  • A line x tester analysis was carried out in mulberry (Morns spp.) under different salinity levels to determine the changes in the genetic interaction of various morpho-biochemical characters. Five mulberry genotypes, 3 females and 2 males, differing in salt tolerance were selected for the study. Clones of these parents along with clones of the F1 hybrids were planted in earthen pots and subjected to different levels of salinity (0.0%, 0.25%, 0.50%, 0.75% and 1.00% NaCl). Data on morphological and biochemical characters were subjected to line x tester analysis. The result revealed significant variation among the parents studied. The prominence of non-additive gene effect under control condition suggests the need for well chalked out breeding program to exploit the non-fixable variance of components for improvement of plant height, leaf size and leaf yield, chlorophyll and photosynthesis in mulberry. However, under salinity stress a shift from non-additive gene effect to additive gene effect for the above said character further suggests the need for a change in breeding strategy. The general combining ability (GCA) analysis has identified English black as the best combiner among the parents and the specific combining ability analysis (SCA) found crosses of English black X C776 and Rotndiloba x Mandalaya were good for Plant height and leaf size and English black X C776 and Rotundiloba x C776 were good for biochemical proline and chlorophyll. From the performance of parents and their crosses under different salinity levels and also under normal cultural conditions it is concluded that in mulberry different approaches are required to develop varieties for the irrigated and saline conditions.

Characteristics of Potato Virus Y (PVY) Mutant Isolated from PVY Resistance Breeding Line in Korea (국내 감자바이러스 Y (PVY) 저항성 육성 계통에서 분리한 PVY Mutant의 특성)

  • A mutant of Potato vims Y (PVY) was occurred in PVY resistance flue-cured tobacco breeding line KF0402 $(TC1146{\times}KF117)$ showing vein necrosis at Suwon in Korea. This isolate, PVY-SWM, was differentiated from other PVY based on biological properties and nucleotide sequence analyses of coat protein gene. PVY-SWM caused typical symptoms on 21 indicator plants as compared to the PVY-TOJC37. Remarkably, the PVY-SWM induced distinctly different symptom of systemic vein necrosis on tobacco cultivars V.SCR, PBD6, TN86, TN90, Virgin A Mutant (VAM), Wislica, NC744, KB108 and KB111, which were reported to have the recessive potyvirus resistance gene va. In RT-PCR assays with specific primers for detection of PVY, a single band of about 800bp in length was produced. The amplified DNA was cloned and the nucleotide sequence was determined. The coat protein gene of PVY-SWM showed 88.4%-99.0% and 92.5%-98.5% identities to the 12 different PVY isolates of Genbank Database at the nucleotide and amino acidi respectively. Multiple alignments as well as cluster dendrograms of PVY-SWM isolate revealed close phylogenetic relationship to the $PVY^{NTN}$ subgroup.

QTL Mapping of Agronomic Traits Using an Introgression Line Population Derived from an Intersubspecific Cross in Rice

  • The objectives of this study were to identify QTLs for agronomic traits using introgression lines from a cross between a japonica weedy rice and a Tongil-type rice. A total of 75 introgression lines developed in the Tongil-type rice were characterized. A total of 368 introgressed segments including 285 homozygous and 83 heterozygous loci were detected on 12 chromosomes based on the genotypes of 136 SSR markers. Each of 75 introgression lines contained 0-9 homozygous and 0-8 heterozygous introgressed segments with an average of 5.8 segments per line. A total of 31 quantitative and 2 qualitative loci were identified for 14 agronomic traits and each QTL explained 4.1% to 76.6% of the phenotypic variance. Some QTLs were clustered in a few chromosomal regions. A first cluster was located near RM315 and RM472 on chromosome 1 with QTLs for 1,000 grain weight, culm length, grain width and thickness. Another cluster was detected with four QTLs for 1,000 grain weight, grain length, grain width and grain length/width ratio near the SSR marker RM249 on chromosome 5. Among the 31 QTLs, 9 (28.1%) Hapcheonaengmi3 alleles were beneficial in the Milyang23 background. ILs would be useful to confirm QTLs putatively detected in a primary mapping population for complex traits and serve as a starting point for map-based cloning of the QTLs. Additional backcrosses are being made to purify nearly isogenic lines (NILs) harboring a few favorable Hapcheonaengmi3 alleles in Milyang23 background.

Genomics Approach to Identify the Cause of the Missing Omega-5 Gliadin Protein in O-Free Wheat

  • A previous work developed and identified a new omega-5 gliadin deficient wheat line named O-free by crossing Keumkang and Olgeuru, which is nutritionally quite meaningful in that omega-5 gliadin is one of the known wheat allergens. To verify the characteristics of the O-free, we performed RNA sequencing (RNAseq) analysis of the O-free and the two parent lines (Keumkang and Olgeuru). The results of the similarity analysis with the ESTs for gliadins and glutenins showed that the O-free ESTs had no similarity with the omega-5 gliadin sequences but had similarity to other gliadins and glutenins. Furthermore, mapping results between the raw RNAseq data from the O-free and the omega-5 gliadin sequence showed a clear deletion of the N-terminal sequences which are an important signature of omega-5 gliadin. We also designed specific PCR primers that could identify omega-5 gliadin in the genomic DNA. The results showed that no omega-5 gliadin fragments were detected in the O-free. According to these results, we confirmed that the deficiency of omega-5 gliadin in the O-free is not caused by post-transcriptional or post-translational regulations such as epigenetic phenomena but by a simple deletion in the chromosome. Furthermore, we showed that the low-molecular weight glutenin subunit (LMW-GS) gene in the O-free had a single nucleotide polymorphism (SNP) causing a premature stop codon, resulting in a truncated polypeptide. We expect that the O-free line may serve as an excellent source of wheat that could prevail in the hypo-allergen wheat market, which has recently gained interest world-wide.
