전자선 조사로 개질된 PLA의 유변학적 물성 (Rheological Properties of Poly(lactic acid) Modified by Electron Beam Irradiation)

  • 본 연구에서는 폴리락트산(PLA)의 용융강도를 높이기 위하여 PLA에 글리시딜 메타크릴레이트(GMA)를 PLA에 첨가한 후 전자선을 조사하여 PLA를 개질하였다. GMA는 5 phr(parts per hundred parts of resin)로 고정하고 전자선 조사량을 변화시키며 개질 PLA을 제조하였으며, 이후 열적, 유변학적 특성 및 젤화도를 조사하였다. 주파수 0.1 rad/s에서 조사량 300 kGy로 개질된 PLA와 순수 PLA의 유변학적 물성을 서로 비교해보면 복합점도는 약 210배, 탄성률은 약 14500배 상승하는 예외적으로 아주 큰 변화를 보였다. 젤화도의 결과로 조사량 200 kGy 이하로 조사된 시료에서 일어난 분자반응은 가교화 반응보다는 가지화 반응이 우세하다는 것을 알 수 있었다.

  • 10%의 $\alpha-(1\rightarrow3)$가지 결합을 갖고 $\alpha-(1\rightarrow6)$으로 연결된 댁스트란을 합성하는 텍스트란수크라제를 coding하는 유전자 (dsCB)를 Leuconostoc mesenteroides B-742CB로부터 분리하여 염기서열과 아미노산 서열을 결정하였다. dsCB가 포항된 6 6.lkb띄 DNA fragment는 4,536 bp의 염기로 구성된 하나의 open reading 잔ame(ORF)를 가지고 있었다. 추정된 아미노산 서열은 ORF의 698벤째 nucleotide 위치에 있는 start codon (ATG)으로부터 5,223번째 위치에 있는 slop codon(TAA)까지 였다. 구조 유전자의 아마노산은 1,5087H로 구성되고 분자량의 계산값은 168.6 kDa었고 non-denatured SDS-PAGE를 이용하여 활성 band툴 분석한 결과 170 kDa이었다. pDSCB를 까지고 있는 재조합 E. coli는 2 % sucrose배치에서 세포외로 댁스트란수크라제를 생산하였으며, soluble과 insoluble 텍스트 란을 생산하였다. 텍스트란수크라체의 효소 촉매작용에 관여 하는 것으로 얄려져있는 conserved region의 아미노산 중 Asp-492를 Asparagine으로 바꾸고자 point mutation을 시도하였고, 결과로 얻어친 D492N은 돌연변이 이전의 균에 비하여 활성이 1.6배 감소함을 확인하였다.

High-Level Production of Low-Branched Levan from Pseudomonas aurantiaca S-4380 for the Production of $di-\beta-D-Fructofuranose$ Dianhydride IV

  • The IscA gene, encoding a levansucrase of 424 amino acids (aa) residues, was cloned from the genomic DNA of Pseudomonas aurantiaca S-4380, and overexpressed in Escherichia coli. The recombinant levansucrase overexpressed in E. coli was then used to produce levan from sucrose. Levan crystals with 98% purity could be obtained from the reaction mixture with $62\%$ yield using an alcohol precipitation method. The molecular weight of the levan was $7\times10^5$ daltons. Methylation studies showed that the levan was branched: main linkage C-2,6; branched linkage C-2,1; and degree of branching $6\%$. Three bacterial levans from different strains were incubated with levan fructotransferase (LFTase) from Arthrobacter ureafaciens K2032, which produced $di-\beta-D-fructofuranose$ dianhydride IV (DFA IV); final conversion yields from the levans to DFA IV were $39\%$ in Zymomonas mobilis, $53\%$ in Serratia levanicum, and $59\%$ in P. aurantiaca S-4380 levansucrase. The levan from P. aurantiaca S-4380 levansucrase gave the highest conversion yield of levan to DFAIV so far reported.

완도납석으로부터 제조한 $\beta$-Sialon에 대한 SiC의 복합화 효과 (Effect of SiC Dispersion of $\beta$-Sialon Prepared from Wando Pyrophylite)

  • 완도납석과 carbon black 의 혼합물을 135$0^{\circ}C$에서 질소분위기를 사용하여 탄소환원질화법으로 $\beta$-Sialon 분물에 $\beta$-SiC를 제2상으로 넣어서 $Y_2O_3$$ZrO_2$를 각 소결조제로 사용하여 175$0^{\circ}C$에서 2시간 동안 상압소결하였다. 10wt %의 $Y_2O_3$를 소결조제로 사용하였을 때에 파괴인성은 3.8MN/m^{3/2}$, 3점꺽임 강도는 470MN/$m^2$ 그리고 경도는 13.7MN/$m^2$로서 좋은 값을 나타내었다. 이와 같은 파괴인성과 꺽임강도의 증가는 균열편향과 균열나눠짐에 의한 것으로 생각된다.

수소/이산화탄소/알곤 혼합 연료의 비예혼합 대향류 화염에서 NO 배출 특성 연구 (A Study of NO Fmission Characteristics in a Non-premixed Counterflow Flame with $H_2/CO_2/Ar$ Blended-fuel)

  • The detailed chemistry with reaction mechanism of GRI 2.11, which consists of 49 species and 279 elementary reactions, have been numerically conducted to investigate the flame structure and NO emission characteristics in a non-premixed counterflow flame of blended fuel of $H_2/CO_2/Ar$. The combination of $H_2,\;CO_2$, and Ar as fuel is selected to clearly display the contribution of hydrocarbon products to flame structure and NO emission characteristics due to the breakdown of $CO_2$. Radiative heat loss term is involved to correctly describe the flame dynamics especially at low strain rates. All mechanisms including thermal, $NO_2,\;N_2O$, and Fenimore are also taken into account to separately evaluate the effects of $CO_2$ addition on NO emission characteristics. The increase of added $CO_2$ quantity causes flame temperature to fall since at high strain rates diluent effect is prevailing and at low strain rates the breakdown of $CO_2$ produces relatively populous hydrocarbon products and thus the existence of hydrocarbon products inhibits chain branching. It is also found that the ratio of the contribution by Fenimore mechanism to that by thermal mechanism in the total mole production rate becomes much larger with increase in the $CO_2$ quantity and strain rate, even though the absolute quantity of NO production is deceased. Consequently, as strain rate and $CO_2$ quantity increase, NO production by Fenimore mechanism is remarkably augmented.

아세트산비닐의 삼차부틸알코올계 저온 중합 및 비누화에 의한 고분자량 폴리비닐알코올의 합성 (Synthesis of High Molecular Weight Poly(vinyl alcohol) by Low Temperature Polymerization of Vinyl Acetate in Tertiary Butyl Alcohol and the Following Saponification)

  • 아세트산비닐(VAc)을 아조비스디메틸발레로니트릴(ADMVN) 및 삼차부틸알코올 (TBA)을 각각 개시제 및 용매로 하여 30, 40 및 5$0^{\circ}C$에서 용액중합하였다. 합성된 폴리아세트산비닐 (PVAc)을 비누화함으로써 고분자량 혼성배열 폴리비닐알코올(PVA)을 제조하였다. 중합조건들이 전환률, 가지화도 및 PVAc와 PVA의 분자량에 미치는 영향을 고찰하였다. TBA에서의 VAc의 중합 속도는 ADMVN 농도의 0.49승에 비례하였고, 이는 이론치 0.5와 잘 일치하였다. 저온에서 개시가 가능한 ADMVN 및 낮은 사슬이동상수를 갖는 TBA를 사용함으로써 고분자량 및 고수율의 PVA가 얻어졌다. PVAc의 평균 중합도는 전환률 약 35%부터 70%의 범위에서 10000~13000이었고, 이를 비누화하여 얻은 PVA의 평균 중합도는 2400~6100이었다. 교대배열 다이애드기 함량은 중합온도를 낮춤에 따라 조금씩 증가하였고, 중합시 TBA의 입체장애 효과 때문에 TBA의 양을 증가시킴에 따라서도 증가하였다.

분지 덱스트린 첨가가 냉동 소프트롤 반죽 및 빵의 저장 중 품질 특성에 미치는 영향 (Effects of Branched Dextrin on the Quality Characteristics of Frozen Soft Roll Dough and its Bread during Storage)

  • In this study, samples of wheat flour and dough were prepared by adding of 1, 3, or 5% branched dextrin, which is produced from the amylopectin of waxy corn starch using a cyclization reaction with a branching enzyme. The samples were then evaluated qualitatively in terms of farinogram, viscogram, and extensogram characteristics. The fermentation power of dough expansion, extensogram characteristics, specific volume, baking loss, external/internal surface appearance, and sensory qualities were also examined after 4 weeks of storage at -20$^{\circ}C$ to determine the effect on freeze-thaw stability and quality improvement of branched dextrins in the soft roll bread formulation. Furthermore, the samples along with a control were compared regarding their quality characteristics, including changes in moisture content, water activity, color, and textural characteristics during a storage period of 4 days at 20$^{\circ}C$ to determine the effect on preventing retrogradation of the branched dextrin. As the branched dextrin content increased, area and extensibility increased, whereas water absorption, fermentation power of dough expansion, resistance/extensibility ratio, baking loss, and brownness of the crust decreased. However, the control group presented significantly higher peak viscosity, resistance, specific volume, taste, overall acceptability, moisture content, water activity, springiness, cohesiveness, and resilience values than those of the branched dextrin samples, whereas lightness, hardness, and chewiness showed the reverse effect. As the storage period increased, lightness, hardness, and chewiness increased, whereas cohesiveness decreased. In conclusion, the results indicate that adding 1~3% branched dextrin into a soft roll bread formulation from frozen dough had no positive effect on freeze-thaw stability or preventing retrogradation but may provide good nutritional properties.

Molecular identification and characterization of Lumpy skin disease virus emergence from cattle in the northeastern part of Thailand

  • Background: Lumpy skin disease (LSD), a disease transmitted by direct and indirect contact with infected cattle, is caused by the Lumpy skin disease virus (LSDV). The disease affects cattle herds in Africa, Europe, and Asia. The clinical signs of LSD range from mild to the appearance of nodules and lesions in the skin leading to severe symptoms that are sometimes fatal with significant livestock economic losses. Objectives: This study aimed to characterize LSDV strains in the blood of infected cattle in Thailand based on the GPCR gene and determine the phylogenetic relationship of LSDV Thailand isolates with published sequences available in the database. Methods: In total, the blood samples of 120 cattle were collected from different farms in four provinces in the northeastern part of Thailand, and the occurrence of LSDV was examined by PCR based on the P32 antigen gene. The genetic diversity of LSDV based on the GPCR gene was analyzed. Results: Polymerase chain reaction assays based on the P32 antigen gene showed that 4.17% (5/120) were positive for LSDV. All positive blood samples were amplified successfully for the GPCR gene. Phylogenetic analysis showed that LSDV Thailand isolates clustered together with LSDVs from China and Russia. Conclusions: The LSD outbreak in Thailand was confirmed, and a phylogenetic tree was constructed to infer the branching pattern of the GPCR gene from the presence of LSDV in Thailand. This is the first report on the molecular characterization of LSDV in cattle in Thailand.

상아질 손상 후 흰쥐 대구치 치수의 calcitonin gene-related peptide(CGRP) 함유 신경섬유 분포에 관한 연구 (A STUDY ON THE DISTRIBUTION OF CALCITONIN GENE-RELATED PEPTIDE CONTAINING NERVE FIBERS IN RAT PULP FOLLOWING DENTINAL INJURY)

  • The purpose of this study was to investigate the distribution of calcitonin gene-related peptide containing nerve fibers in rat pulp after dentinl injury by means of immunohistochemistry and confocal laser scanning microscope. The Spague-Dawley rats weighing about 250-300gm were used. The animals were devided into normal control and experimental groups. Experimental animals were sacrified 1, 2, 4, 7, 10, 21days after dentinal injury (dentin cutting, and then acid etching with 35% phosphoric acid) on the maxillary molar teeth. The maxillary teeth and alveolar bone were removed and immersed in the 4% paraformaldehyde in 0.1M phosphate buffer (pH 7.4), then were decalcified with 15% formic acid for 10 days. Serial frozen $50{\mu}m$ thick sections were cut on a cryostat. The rabbit CGRP antibody was used as a primary antibody with a dilution of 1:2000 in 0.01M PB. The sections were incubated for 48 hours at $4^{\circ}C$, and placed into biotinylated antirabbit Ig G as a secondary anti body with dilution of 1:200 in 0.01M PB and incubated in ABC(avidin-biotin complex). The peroxidase reaction was visualized by incubating the sections in 0.05% 3,3 diaminobenzidine tetrahydrochloride containing 0.02% $H_2O_2$. For the confocal laser scanning microscopic examination, Primary antibody reaction was same as immunoperoxidase stainning, but fluorescein isothiocyanate(FITC)-conjugate antirabbit IgG as a secondary antibody was used. The confocal laser scanning microscope was used for the examination. A series of images of optical sections was collected with a 20x objective at $3{\mu}m$ intervals throughout the depth of specimen. FITC fluerescence was registrated through a 488nm and 568nm excitation filter, and images were saved on optical disk. The stereoscopic images and three dimentionnal images were reconstructed by computer software, and then were analyzed. The results were as follows : 1. In normal control group, CGRP containing nerve fibers were coursed through the root with very little branching, and then formed a dense network of terminals in coronal pulp. 2. A slight increase in CGRP containing nerve fibers at 1 and 2day postinjury was noted subjacent to the injury site. In the 4day group, there were an extensive increase in the number of reactive fibers, followed by a partial return toward normal levels at 7~10 day postinjury, and return by 21days. 3. The sprouting of the CGRP containing nerve fibers was evident within 2day after dentinal injury, and by 4days there was a maximal increased, but was decreased at 7days and returned to normal 10~21 day postinjury. 4. In confocal laser scanning microscopic exammination, the distinct distribution pattern and sprouting reaction of CGRP containing nerve fibers were observed in stereoscopic images and three dimentional images. These results suggest that CGRP containing nerve fiber can be important role in the response to dental injury and pain regulation.

F-T 공정으로 합성된 바이오항공유의 화학적 조성에 따른 점화특성 분석 (Analysis on Ignition Characteristics According to the Chemical Composition of Bio Jet Fuel Synthesized by F-T Process)

  • 본 연구에서는 F-T 공정을 통해 합성하여 제조한 바이오항공유(Bio-7629, Bio-5172)와 기존에 사용 중인 석유계항공유(Jet A-1)의 점화특성을 비교하여 분석하였다. Combustion research unit (CRU) 장비를 활용하여 각 항공유의 점화지연시간을 측정하였고, 그 결과를 연료의 물성 및 구성 화합물에 대한 분석을 통해 해석하고자 하였다. 점화지연시간은 Bio-5172가 가장 짧게 측정되었으며 Jet A-1이 가장 길게 측정되었다. 이는 물리적 점화지연시간에 영향을 줄 수 있는 연료의 물성 측면에서 Jet A-1이 가장 큰 표면장력을 가지며 Bio-5172가 가장 낮은 점도를 갖기 때문인 것으로 해석된다. 또한, 각 연료를 구성하는 화합물의 종류 및 비율에 대하여 분석한 결과, 실험 대상 바이오항공유에 없는 방향족화합물이 Jet A-1에는 약 22.8%의 비율로 존재함을 확인하였다. 이는 산화 과정 시에 비교적 반응성이 낮은 benzyl radical을 생성하여 점화지연시간이 길게 측정되는 데에 영향을 주는 것으로 판단된다. Bio-7629와 Bio-5172는 paraffin으로만 구성되어 있으며, n-/iso-의 값은 각각 0.06, 0.80으로 큰 차이를 보였다. 가지화 된 정도가 낮은 paraffin일수록 산화 시에 생성되는 peroxy radical의 이성질화가 빠르게 진행되어 점화의 전파속도 또한 빨라진다. 따라서 n-paraffin의 함량이 비교적 높은 Bio-5172의 경우에 점화지연시간 또한 짧게 측정된 것으로 해석된다.
