New established cell lines from undifferentiated pleomorphic sarcoma for in vivo study

  • As a high-grade soft-tissue sarcoma (STS), undifferentiated pleomorphic sarcoma (UPS) is highly recurrent and malignant. UPS is categorized as a tumor of uncertain differentiation and has few options for treatment due to its lack of targetable genetic alterations. There are also few cell lines that provide a representative model for UPS, leading to a dearth of experimental research. Here, we established and characterized new cell lines derived from two recurrent UPS tissues. Cells were obtained from UPS tissues by mincing, followed by extraction or dissociation using enzymes and culture in a standard culture environment. Cells were maintained for several months without artificial treatment, and some cell clones were found to be tumorigenic in an immunodeficient mouse model. Interestingly, some cells formed tumors in vivo when injected after aggregation in a non-adherent culture system for 24 h. The tissues from in vivo study and tissues from patients shared common histological characteristics. Pathways related to the cell cycle, such as DNA replication, were enriched in both cell clones. Pathways related to cell-cell adhesion and cell-cell signaling were also enriched, suggesting a role of the mesenchymal-to-epithelial transition for tumorigenicity in vivo. These new UPS cell lines may facilitate research to identify therapeutic strategies for UPS.

Clinicopathological studies on the subclinical fascioliasis in the Korean native cows in Chonnam area (순수번식단지(純粹繁殖團地) 한우(韓牛)의 간질증(肝蛭症)에 관한 임상적(臨床的) 연구(硏究))

  • Fecal samples were taken from 402 cows in Posung, Chonnam where was designated as a place for Korean native cattle breeding. Prevalence of internal parasitisms were determined by the fecal examinations using the floatation and sedimentation procedures. 62.9% of the cows were found as positive cases with excretion of the eggs of Fasciola hepatica in the fecal specimens. Of those infected with F hepatica 97 cows free of other pathogenic intestinal parasites were chosen for albendazole treatment. Albendazole tablets(10mg/kg) were administered to the cows twice at the interval of 4 weeks. Blood samples were collected via jugular vein prior to the first treatment, four weeks after the first treatment and four weeks after the second treatment, respectively. At the same time fecal samples were collected for parasitological examinations by sedimentation methods. The mean pretreatment count was 44 fluke eggs per gram of feces, which compared with 27 epg and 17 epg four weeks after the first and second treatment, respectively. Most of the hematological and biochemical values fluctuated within the normal ranges during the experiment. Eosinophil counts were high initially, decreased after the first treatment and thereafter remained steady. The opposite was the case with aspartate and alanine aminotransferases.

Comparison of in Vitro Cytotoxicity and Apoptogenic Activity of Magnesium Chloride and Cisplatin as Conventional Chemotherapeutic Agents in the MCF-7 Cell Line

  • Breast cancer is the most common malignancy and also the second leading cause of cancer death among women and also in women that have a high mortality. Previous studies showed that magnesium (Mg) has cytotoxic effects on malignant cell lines. However, the anti-cancer effects of Mg on MCF-7 breast cancer cells are uncertain. This study was aimed at the comparison of the cytotoxic effect of Mg salt (MgCl2) and cisplatin on MCF-7 cells and fibroblasts (as normal cells). After treatment with various concentrations of MgCl2, and cisplatin as a positive control for 24 and 48 hours (h), cytotoxicity activity was measured by MTT assay. In addition, apoptosis was determined by annexin V/propidium iide assay. Both cisplatin and the MgCl2 exhibited dose-dependent cytotoxic effects in the MCF-7 cell line, although the LD50 of the Mg was significantly higher when compared to cispaltin ($40{\mu}g/ml$ vs. $20{\mu}g/ml$). Regarding annexin V/propidium results, treatment of MCF-7 cells with LD50 concentrations of cisplatin and Mg showed 59% and 44% apoptosis at 24h, respectively. Finally, the results indicated that Mg has cytotoxic effects on MCF-7 cells, but less than cisplatin as a conventional chemotherapeutic agent. However, regarding the side effects of chemotherapy drugs, it seems that Mg can be considered as a supplement for the treatment of breast cancer.

  • Ink-jet printing technology, which utilizes a digitalized design to print fine ink directly on a substrate, has been of interest in various industries due to its high efficiency and adaptability to various materials. Recently, active attempts have been made to apply ceramic materials having excellent heat resistance, light resistance, and chemical resistance to the ink-jet printing process. In this study, ceramic ink was synthesized by combining ceramic pigments with UV curable polymer. 3D printability at various contact angles between ceramic ink and substrate was analyzed in detail. Rheological properties of the synthesized ceramic ink were optimized to meet the requirements of the ink-jet printing process, and the contact angle of UV curable ceramic ink was controlled through surface treatment of the substrate. The potential for additive manufacturing of ceramic material using ink-jet printing was investigated by analyzing the effect of contact angle control on ceramic ink droplets and their 3D printability.

Nomogram Estimating the Probability of Intraabdominal Abscesses after Gastrectomy in Patients with Gastric Cancer

  • Purpose: Intraabdominal abscess is one of the most common reasons for re-hospitalization after gastrectomy. This study aimed to develop a model for estimating the probability of intraabdominal abscesses that can be used during the postoperative period. Materials and Methods: We retrospectively reviewed the clinicopathological data of 1,564 patients who underwent gastrectomy for gastric cancer between 2010 and 2012. Twenty-six related markers were analyzed, and multivariate logistic regression analysis was used to develop the probability estimation model for intraabdominal abscess. Internal validation using a bootstrap approach was employed to correct for bias, and the model was then validated using an independent dataset comprising of patients who underwent gastrectomy between January 2008 and March 2010. Discrimination and calibration abilities were checked in both datasets. Results: The incidence of intraabdominal abscess in the development set was 7.80% (122/1,564). The surgical approach, operating time, pathologic N classification, body temperature, white blood cell count, C-reactive protein level, glucose level, and change in the hemoglobin level were significant predictors of intraabdominal abscess in the multivariate analysis. The probability estimation model that was developed on the basis of these results showed good discrimination and calibration abilities (concordance index=0.828, Hosmer-Lemeshow chi-statistic P=0.274). Finally, we combined both datasets to produce a nomogram that estimates the probability of intraabdominal abscess. Conclusions: This nomogram can be useful for identifying patients at a high risk of intraabdominal abscess. Patients at a high risk may benefit from further evaluation or treatment before discharge.

Alleviative effects of activated charcoal and Houttuynia cordata Thunb. in broiler chickens during aflatoxicosis (Aflatoxin에 노출된 닭에서 활성탄과 어성초의 독성완화 효과)

  • This study was conducted to evaluate the alleviative effects of activated charcoal (AC) and Houttuynia cordata (HC) singly or in combination in broiler chickens during aflatoxicosis. Activated charcoal (1% or 0.5%) and H. cordata (1% or 0.5%) were mixed into the diets for the ability to reduced the deleterious effects of 2.4mg total aflatoxin $(AFB_1)kg^{-1}$ diet on growing broiler chickens from 1 to 21 days of age. A total of 160 1-day-old (Hyline Variety Brown) broiler chicks were housed in eight treatment groups [Control, $AFB_1$, AC 1%, HC 1%, $AFB_1$ plus AC 1% plus HC 1%, $AFB_1$ plus AC 1% plus HC 0.5%, $AFB_1$ plus AC 0.5% plus HC 1%, $AFB_1$ plus AC 0.5% plus HC 0.5%] each consisting of 20 chicks. Compared to control, 2.4mg $AFB_1$ alone treatment group significantly decreased body weight gains of chickens. The addition of mixed AC 1% and HC 1% including 6, 7 groups to the 2.4mg $AFB_1$-containing diet moderately reduced the adverse effects of $AFB_1$ on performances of chickens. The chickens consuming 2.4mg $AFB_1$ plus AC 0.5% and HC 0.5%-containing diet showed very slightly reduced the adverse effects on investigated parameters compared to the $AFB_1$ only treated group. Also, the single addition of AC or HC to the $AFB_1$-free diet had no adverse effects in chickens. These results suggest that AC and HC mixed can reduced the aflatoxicosis in broilers and may be contribute to a solution of the aflatoxicosis problem in poultry production.

  • Micelle-Enhanced Ultrafiltration (MEUF) is a membrane separation processes that improving ultrafiltration process with the formation of micelles of the surface active agents. Surface active agents are widely used to improve membrane processes due to the ability to trap organic compounds and metals in the treatment of industrial waste water. In this study, surface active agents are used to improve micelle-enhanced ultrafiltration (MEUF) to reduce chemical oxygen demand (COD), total dissolved solid (TDS), turbidity and clogging the membrane in dairy wastewater treatment. Three important operational factors (anionic surface active agent concentration, pressure and pH) and these interactions were investigated by using response surface methodology (RSM) and Box-Behnken design. Results show that due to the concentration polarization layer and increase the number of Micelles; the anionic surface active agent concentration has a negative effect on the flux and has a positive effect on the elimination of contamination indices. pH, and the pressure have the greatest effect on flux. On the other hand, it could be stated that these percentages of separation are in the percentages range of Nano-filtration (NF). While MEUF process has higher flux than NF process. The results have been achieved at lower pressure while NF process needs high pressure, thus making MEUF is the replacement for the NF process.

Physicochemical Properties of Enzymatically Modified Maize Starch Using 4-${\alpha}$-Glucanotransferase

  • Granular maize starch was treated with Thermus scotoductus 4-${\alpha}$-glucanotransferase (${\alpha}$-GTase), and its physicochemical properties were determined. The gelatinization and pasting temperatures of ${\alpha}$-GTase-modified starch were decreased by higher enzyme concentrations. ${\alpha}$-GTase treatment lowered the peak, setback, and [mal viscosity of the starch. At a higher level of enzyme treatment, the melting peak of the amylose-lipid complex was undetectable on the DSC thermogram. Also, ${\alpha}$-GTase-modified starch showed a slower retrogradation rate. The enzyme treatment changed the dynamic rheological properties of the starch, leading to decreases in its elastic (G') and viscous (G") moduli. ${\alpha}$-GTase-modified starch showed more liquid-like characteristics, whereas normal maize starch was more elastic and solid-like. Gel permeation chromatography of modified starch showed that amylose was degraded, and a low molecular-weight fraction with $M_w$ of $1.1{\times}10^5$ was produced. Branch chain-length (BCL) distribution of modified starch showed increases in BCL (DP>20), which could result from the glucans degraded from amylose molecules transferred to the branch chains of amylopectin by inter-/intra-molecular transglycosylation of ${\alpha}$-GTase. These new physicochemical functionalities of the modified starch produced by ${\alpha}$-GTase treatment are applicable to starch-based products in various industries.

Removal of Cd(II) from water using carbon, boron nitride and silicon carbide nanotubes

  • Molecular dynamics simulations were used to study the removal of Cd(II) as a heavy metal from wastewater using armchair carbon nanotube, boron nitride nanotube and silicon carbide nanotubes under applied electric field. The system contains an aqueous solution of $CdCl_2$ as a heavy metal and a (7,7) nanotube as a nanostructured membrane, embedded in a silicon nitride membrane. An external electric field was applied to the considered system for the removal of $Cd^{2+}$ through nanotubes. The simulation results show that in the same conditions, considered armchair nanotubes were capable to remove $Cd^{2+}$ from wastewater with different ratios. Our results reveal that the removal of heavy metals ions through armchair carbon, boron nitride and silicon carbide nanotubes was attributed to the applied electric field. The selective removal phenomenon is explained with the calculation of potential of mean force. Therefore, the investigated systems can be recommended as a model for the water treatment.
