강도별 유산소운동이 중년여성의 레닌-알도스테론, 신경전달물질 및 인지기능, 작업기억의 변화에 관한 연구 (The Study of Change in Renin-Aldosterone, Neurotransmitters, Cognitive Function and Working Memory in Middle Aged Women by Intensities Aerobic Exercise)

  • 본 연구는 건강한 중년여성 28명을 대상으로 강도별 유산소운동을 12주간 주 3회로 실시하여 혈압, 레닌-알도스테론계, 신경전달물질 및 인지기능, 작업기억의 변화와 처치 후 인지기능에 영향을 미치는 요인을 분석하고자 실시하였다. 모집된 대상자를 통제집단 9명, 중강도 유산소운동집단 10명(50%V02max), 고강도 유산소운동집단 9명(70%V02max)으로 구분하여 처치 전과 후 반복측정 분산분석과 다중회귀분석을 실시한 결과 다음과 같은 결과가 나타났다. 중강도 유산소운동은 레닌, BDNF, 인지기능과 작업기억을 증가시키고, 알도스테론, 안지오텐신II, 알도스테론-레닌 비율을 감소시켰다. 고강도 유산소운동은 BDNF, 인지기능, 작업기억을 증가시키고 수축기혈압을 감소시켰다. 또한 강도별 유산소운동 후 인지기능에 미치는 요인을 회귀분석한 결과 중강도 유산소운동은 이완기혈압, 레닌-알도스테론 비율의 감소와 작업기억, BDNF의 증가가 인지기능에 영향을 미쳤고, 고강도 유산소운동은 작업기억 BDNF, 세로토닌의 증가가 인지기능에 영향을 미쳤다. 따라서 중강도 이상의 유산소운동은 중년여성의 인지기능 및 작업기억을 향상시키는 것을 알 수 있었다. 또한 인지기능 향상에 영향을 미치는 대사적 요인으로 중강도 유산소운동은 레닌-알도스테론 조절과 작업기억이었고 고강도 유산소운동은 신경전달물질과 작업기억으로 나타났다.

해인탕 추출물이 흰쥐 좌골신경 손상 모델에서 기능회복과 뇌의 c-Fos 발현에 미치는 영향 (Effects of Haein-tang(Hairen-tang) Extract on Functional Recovery in Sciatic Nerve and c-Fos Expression in the Brain after Crushed Sciatic Nerve Injury in Rats)

  • Objectives : Peripheral nerve injuries are commonly encountered clinical problems and often result in severe functional deficits. The purpose of this study was to evaluate the effects of Haein-tang(Hairen-tang) extract on functional recovery and pain release in the sciatic nerve after crushed sciatic nerve injury in rats. Methods : 1. Sciatic functional index(SFI) were performed on functional recovery. 2. c-Fos immunohistochemistry were performed on c-Fos expressions in the paraventricular nucleus(PVN) and ventrolateral periaqueductal gray(vIPAG). 3. Neurofilament immunohistochemistry were performed on neurofilament regeneration. 4. Western blot were performed on brain-derived neurotrophic factor(BDNF) and nerve growth factor(NGF) expression. Results : 1. Haein-tang(Hairen-tang) extract significantly enhanced the SFI value in the sciatic nerve injury and 100 mg/kg, 200 mg/kg Haein-tang(Hairen-tang)-treated group. 2. Haein-tang(Hairen-tang) extract significantly suppressed the sciatic nerve injury-induced increment of c-Fos expressions in the PVN and vIPAG in the sciatic nerve injury and 100 mg/kg, 200 mg/kg Haein-tang(Hairen-tang)-treated group. 3. Haein-tang(Hairen-tang) extract significantly increased neurofilament expression in the sciatic nerve injury and 50 mg/kg, 100 mg/kg, 200 mg/kg Haein-tang(Hairen-tang)-treated group. 4. Haein-tang(Hairen-tang) extract significantly controled the sciatic nerve injury-induced increment of BDNF and NGF expressions in the sciatic nerve injury and 100 mg/kg, 200 mg/kg Haein-tang(Hairen-tang)-treated group. Conclusions : These results suggest that Haein-tang(Hairen-tang) treatment after sciatic nerve injury is effective for the functional recovery by enhancing of axonal regeneration and suppressing of pain.

Effects of systemic administration of ibuprofen on stress response in a rat model of post-traumatic stress disorder

  • Pro-inflammatory cytokine and brain-derived neurotrophic factor (BDNF) are modulated in post-traumatic stress disorder (PTSD). This study investigated the effects of ibuprofen (IBU) on enhanced anxiety in a rat model of PTSD induced by a single prolonged stress (SPS) procedure. The effects of IBU on inflammation and BDNF modulation in the hippocampus and the mechanisms underlying for anxiolytic action of IBU were also investigated. Male Sprague-Dawley rats were given IBU (20 or 40 mg/kg, i.p., once daily) for 14 days. Daily IBU (40 mg/kg) administration significantly increased the number and duration of open arm visits in the elevated plus maze (EPM) test, reduced the anxiety index in the EPM test, and increased the time spent in the center of an open field after SPS. IBU administration significantly decreased the expression of pro-inflammatory mediators, such as tumor necrosis $factor-{\alpha}$, $interleukin-1{\beta}$, and BDNF, in the hippocampus, as assessed by reverse transcription-polymerase chain reaction analysis and immunohistochemistry. These findings suggest that IBU exerts a therapeutic effect on PTSD that might be at least partially mediated by alleviation of anxiety symptoms due to its anti-inflammatory activity and BDNF expression in the rat brain.

Effect of Ginsenoside Re on Depression- and Anxiety-Like Behaviors and Cognition Memory Deficit Induced by Repeated Immobilization in Rats

  • In this study, we assessed the effects of ginsenoside Re (GRe) administration on repeated immobilization stress-induced behavioral alterations using the forced swimming test (FST), the elevated plus maze (EPM), and the active avoidance conditioning test (AAT). Additionally, we examined the effect of GRe on the central adrenergic system by observing changes in neuronal tyrosine hydroxylase (TH) immunoreactivity and brain-derived neurotrophic factor (BDNF) mRNA expression in the rat brain. Male rats received 10, 20, or 50 mg/kg GRe (i.p.) 30 min before daily exposures to repeated immobilization stress (2 h/day) for 10 days. Activation of the hypothalamic-pituitary-adrenal (HPA) axis in response to repeated immobilization was confirmed by measuring serum levels of corticosterone (CORT) and the expression of corticotrophin-releasing factor (CRF) in the hypothalamus. Repeated immobilization stress increased immobility in the FST and reduced open-arm exploration in the EPM test. It also increased the probability of escape failures in the AAT test, indicating a reduced avoidance response. Daily administration of GRe during the repeated immobilization stress period significantly inhibited the stress-induced behavioral deficits in these behavioral tests. Administration of GRe also significantly blocked the increase in TH expression in the locus coeruleus (LC) and the decrease in BDNF mRNA expression in the hippocampus. Taken together, these findings indicate that administration of GRe prior to immobilization stress significantly improved helpless behaviors and cognitive impairment, possibly through modulating the central noradrenergic system in rats. These findings suggest that GRe may be a useful agent for treating complex symptoms of depression, anxiety, and cognitive impairment.

노르아드레날린성 신경세포에서의 BDNF 생산 증진 물질 탐색 (Screening of Potential Compounds Promoting BDNF Production in Noradrenergic Locus Coeruleus Neurons)

  • Locus coeruleus (LC)에는 전체 노르아드레날린성 뉴런의 절반 가량이 모여 있는데, 여기서 노르아드레날린성 뉴런이 뇌의 거의 모든 부위로 신경자극을 보내게 된다. LC는 알츠하이머병, 파킨슨병, 헌팅턴병 같은 여러 가지 신경퇴행성 질환에서 공통적으로 타격을 받는 주요 부위이다. 뇌 유래 신경영양인자, BDNF가 LC 노르아드레날린성 뉴런을 포함한 중추신경계 뉴런들의 분화와 신경세포 생존에 중요한 조절자로 작용한다. 본 연구에서는 LC 노르아드레날린성 신경세포에서 여러 가지 작은 분자들과 성장인자들이 BDNF 생산을 촉진할 수 있는지를 조사하였다. 실험에 사용한 분자들로는 neuropeptides, cytokines, 성장인자, 신경전달물질들과 세포내 신호전달물질들이 포함되었다. 여러 가지 작은 분자들과 성장인자들 중에서 FGF8b, BMP-4, forskolin 그리고 dibutyrl cGMP가 LC 노르아드레날린성 뉴런에서 BDNF 분비를 뚜렷하게 증대시킨 것으로 판명되었다. 특히, BMP-4는 BDNF 생산을 2.5배 이상 증가시켰다. LC 노르아드레날린성 뉴런에서 BDNF를 증가시킨 물질들은 여러 가지 신경퇴행성 질환에서 신경세포가 손실되는 것을 막거나 지연시킬 수 있을 것이므로, 치료제나 증상완화제로서의 가능성이 높다.

운동 트레이닝이 비만 어린이의 neurotrophins, HGF (hepatocyte growth factor)와 산화스트레스에 미치는 영향 (The Effects of Exercise on Neurotrophins, Hepatocyte Growth Factor (HGF), and Oxidative Stress in Obese Children)

  • 본 연구는 운동 트레이닝이 비만 어린이의 산화적 스트레스, 신경성장 및 간 세포성장 인자에 미치는 영향을 알아보기 위하여 12주간 유산소 운동을 실시한 후 트레이닝 전과 후의 농도 수준을 비교하였다. 연구결과, NGF와 BDNF는 트레이닝 전과 후 모두 OT군이 NT군 보다 낮았으며, NGF는 트레이닝에 따라 두 그룹 모두 증가되었다. HGF는 트레이닝 전 OT군이 NT군 보다 높게 나타났지만, 트레이닝 후 차이는 없었다. MDA, ox-LDL, 8-OHdG 모두 OT군이 NT군 보다 높은 수치를 나타냈고, 트레이닝에 따른 차이는 ox-LDL에서만 발견되었다. 이상의 결과, 비만은 성장기 어린이에게 산화적 스트레스를 유발하고, 신경과 간 성장인자의 분비 이상을 초래한다는 것을 알 수 있었고, 12주 유산소 운동은 이들 인자들의 개선에 긍정적인 영향을 주고 인지기능 향상에도 도움을 줄 수 있을 것으로 본다.

Protective Effects of Silibinin and Its Possible Mechanism of Action in Mice Exposed to Chronic Unpredictable Mild Stress

  • Silibinin, a natural flavonoid antioxidant isolated from extracts of the milk thistle herb, has recently been identified as having anti-hepatotoxic and anticancer properties. In this paper, we investigated the effects of silibinin on behavior and neuroplasticity in mice subjected to chronic unpredictable mild stress (CUMS). After 5 consecutive weeks of CUMS, the mice were treated with silibinin (100 mg/kg, 200 mg/kg and 400 mg/kg by oral gavage) for 3 consecutive weeks. The results showed that silibinin administration significantly alleviated the CUMS-induced depressive-like behavior, including the total number of squares crossed and the frequency of rearing in the open field test, the immobility time in the tail suspension test and the forced swimming test. Furthermore, silibinin treatment increased the levels of brain-derived neurotrophic factor (BDNF), serotonin (5-HT) and norepinephrine (NE) in the prefrontal cortex and hippocampus. Our study provides new insight into the protective effects of silibinin on the depressive status of CUMS mice, specifically by improving neuroplasticity and neurotransmission.

Wogonin Attenuates Hippocampal Neuronal Loss and Cognitive Dysfunction in Trimethyltin-Intoxicated Rats

  • We examined whether wogonin (WO) improved hippocampal neuronal activity, behavioral alterations and cognitive impairment, in rats induced by administration of trimethyltin (TMT), an organotin compound that is neurotoxic to these animals. The ability of WO to improve cognitive efficacy in the TMT-induced neurodegenerative rats was investigated using a passive avoidance test, and the Morris water maze test, and using immunohistochemistry to detect components of the acetylcholinergic system, brain-derived neurotrophic factor (BDNF), and cAMP-response element-binding protein (CREB) expression. Rats injected with TMT showed impairments in learning and memory and daily administration of WO improved memory function, and reduced aggressive behavior. Administration of WO significantly alleviated the TMT-induced loss of cholinergic immunoreactivity and restored the hippocampal expression levels of BDNF and CREB proteins and their encoding mRNAs to normal levels. These findings suggest that WO might be useful as a new therapy for treatment of various neurodegenerative diseases.

Effects of Gentianae Macrophyllae Radix on the functional recovery and expression of BDNF and c-Fos after sciatic crushed nerve injury in rats

  • Background : Peripheral nerve injuries are a commonly encountered clinical problem and often result in a chronic pain and severe functional deficits. Objective : The aim of this study was to evaluate the effects of Gentianae Macrophyllae Radix (G. M. Radix) on the pain control and the recovery of the locomotor function that results from the sciatic crushed nerve injury in rats. Method : Using rats, we crushed their sciatic nerve, and then orally administered the aqueous extract of G. M. Radix. The effects of G. M. Radix on the recovery locomotor function were investigated by walking track analysis. The effects of G. M. Radix on pain control were investigated by brain-derived neurotrophic factor (BDNF) expression in the sciatic nerve, and c-Fos expression in the paraventricular nucleus (PVN) of the hypothalamus and in the ventrolateral periaqueductal gray (vlPAG). Result : G. M. RADIX facilitates motor function from the locomotor deficit, and thereby increased BDNF expression and suppressed painful stimuli in the PVN and vlPAG after sciatic crushed nerve injury. Conclusion : It is suggested that G. M. Radix might aid recovery locomotor function and control pain after sciatic crushed nerve injury. Further studies on identifying specific the component in G.M. Radix associated with enhanced neural activity in the peripheral nerve injury may be helpful to develop therapeutic strategies for the treatment of peripheral nerve injury.

Administration of Phytoceramide Enhances Memory and Up-regulates the Expression of pCREB and BDNF in Hippocampus of Mice

  • This study was aimed at investigating the possible effects of phytoceramide (Pcer) on learning and memory and their underlying mechanisms. Phytoceramide was orally administered to ICR mice for 7 days. Memory performances were assessed using the passive avoidance test and Y-maze task. The expressions of phosphorylated cAMP response element binding protein (pCREB), brain-derived neurotrophic factor (BDNF) were measured with immunoblot. The incorporation of 5-bromo-2-deoxyuridine (BrdU) in hippocampal regions was investigated by using immunohistochemical methods. Treatment of Pcer enhanced cognitive performances in the passive avoidance test and Y-maze task. Immunoblotting studies revealed that the phosphorylated CREB and BDNF were significantly increased on hippocampus in the Pcer-treated mice. Immunohistochemical studies showed that the number of immunopositive cells to BrdU was significantly increased in the hippocampal dentate gyrus regions after Pcer-treatment for 7 days. These results suggest that Pcer contribute to enhancing memory and BDNF expression and it could be secondary to the elevation of neurogenesis.
