  • The results of chromosome aberration test in mammalian cells in culture (Chinese hamster lung fibroblast cells) showed no induction of structural and numerical aberrations by antifungal agents of YH1715 series regardless of metabolic activation. While positive control group (mitomycin C and benzo(a)pyrene) showed structural chromosome aberrations of 37% and 23%, respectively. The in vivo induction of micronuclei was measured in polychromatic erythrocytes in bone marrow of male ddY mouse given YH1715R and YH1729R at 1, 0.5, 0.25 g/kg by p.o. once. After 24 hours, animals were sacrificed and evaluated 40 the incidence of micronucleated polychromatic erythrocytes in whole erythrocytes. Although a positive response for induction of micronuclei in animals treated with mitomycin C demonstrated the sensitivity of the test system for detection of a chemical clastogen, YH1715R did not induce micronuclei in bone marrow of ddY male mice but induced cytotoxicity to bone marrow cells at the highest concentration (1 g/kg, p〈0.05), and YH1729R induced micronuclei in bone marrow of ddY male mice dose dependently (p<0.05) but did not induce cytotoxicity to bone marrow cells.

Effect of Molybdenum Induced Copper Deficiency on Peripheral Blood Cells and Bone Marrow in Buffalo Calves

  • Copper deficiency was induced in eight male buffalo calves by adding molybdenum (30 ppm wet basis) to their diet. Copper status was monitored from the liver copper concentration and a level below 30 ppm (DM basis) was considered as deficient. Haemoglobin, haematocrit, total and differential leucocyte numbers were determined. The functions of peripheral neutrophils were assessed by in vitro phagocytosis and killing of Staphylococcus aureus. The effect of molybdenum induced copper deficiency on bone marrow was monitored. The mean total leucocyte count was unaffected whereas a significant fall in neutrophil count coincided with the fall in hepatic copper level to $23.9{\pm}2.69$ ppm. Reduced blood neutrophil numbers was not accompanied by any change in the proportion of different neutrophil precursor cells in bone marrow. It was hypothesised that buffalo calves were more tolerant to dietary molybdenum excess than cattle. It was concluded that neutropenia in molybdenum induced copper deficiency occurred without any effect on their synthesis and maturation process. Bone marrow studies in healthy calves revealed higher percentage of neutrophilic myelocytes and metamyelocytes as compared to cattle.

Combined Genotoxic Effects of Aflatoxin B1, Ochratoxin A and Zearalenone in Rat Bone Marrow and Blood Leukocytes

  • Mycotoxins such as aflatoxin B1 (AFB1), ochratoxin A (OTA) and zearalenone (ZEA) are widespread contaminants of food and feedstuffs. It is very likely, that humans and animals are always exposed to mixtures of mycotoxins rather than to individual compounds. Therefore, risk assessments should consider mixture toxicity data. In the present study the combination of AFB1, OTA and ZEA was tested for genotoxicity in rat bone marrow and blood leukocytes after 15, 30 and 60 days treatment. The level of DNA damage was determined by the comet assay. The tail intensity and Olive tail moment in leukocytes and bone marrow cells were significantly higher than in controls. At the same time, the level of DNA damage in bone marrow cells was higher than in leukocytes. The data suggests that prolonged exposure to mycotoxins combination through food consumption can induce DNA damage contributing to the harmful effects in vivo.

Hematopoietic Effect of Phellinus linteus Polysaccharide in Mouse Splenocytes and Bone Marrow Cells

  • In anti-cancer therapies, radiotherapy and chemotherapy show a superior inhibition effect on cancer cell growth, but those are very toxic to normal tissues and organs. Particularly, drugs for neutropenia, one of chemotherapy agents, which suppress the function of bone marrow, are needed to be controlled in terms of their dosage and therapy period because of their side effect. Phellinus linteus polysaccharide (PL) has been reported to increase the number of splenocytes and bone marrow cells. PL has been shown to decrease the side effects of cyclophosphamide (CYC) treatment to the cancer patients. PL showed no effects in semisolid clonogenic assay, suggesting that PL doesn't contain substantial compounds to substitute for colony stimulating factors (CSFs). On the other hand, PL increased the expression of SCF, IL-3, GM-CSF, TPO genes. These results indicate that PL may promote the growth and proliferation of splenocytes and bone marrow cells through indirect or CSFs-dependent pathway, which may lead to a hematopoiesis.

Aloe vera투여가 Cobalt-60 감마선 조사를 받은 마우스의 생존율과 조혈간세포에 미치는 영향 (Modification of Survival and Blood-forming Stem Cells in Cobalt-60 Gamma Irradiated Mice by Aloe vera)

  • The present study was carried out to investigate whether the aloe had a radioprotective effect in mice exposed to cobalt-60 gamma radiation or not. The survival ratio of mice for 30 days, hematopoiesis of blood-forming stem cells by spleen colony assay, chromosomal aberration frequency of bone marrow cells and histopathological findings of bone marrow were investigated. The survival ratios of aloe administered groups with concentration of 250, 500, 1,000 and 1,500mg for 3 days before irradiation and control group in cobalt-60 gamma irradiated mice(700rads whole body irradiation, dose rate of 50rads/min.) were 77.4, 79.3, 80.6, 90.0 and 53.1%, respectively. The survival ratios of pre-irradiation aloe administered groups were superior to those of post-irradiation aloe groups and control group. In spleen colony assay, Aloe vera administration before irradiation enhanced the recoveries of numbers of blood-forming stem cells of bone marrow of irradiated mice. There were decreased chromosomal aberrations of bone marrow cells at the first day after irradiation in aloe administered groups compared to that of control group. Histopathological findings in the bone marrow of irradiated mice were hypocellularity due to the depletion of myelocytes, abundant of fat vacuoles and these changes were weakened in aloe administered groups compared to that of control group.

  • Gastric cancer is a major cause of cancer-related mortality. At the time of diagnosis, majority of the patients usually have unresectable or metastatic disease. The most common sites of metastases are the liver and the peritoneum, but in the advanced stages, there may be metastases to any region of the body. Bone marrow is an important metastatic site for solid tumors, and the prognosis in such cases is poor. In gastric cancer cases, bone marrow metastasis is usually observed in younger patients and in those with poorly differentiated tumors. Prognosis is worsened owing to the poor histomorphology as well as the occurrence of pancytopenia. The effect of standard chemotherapy is unknown, as survival is limited to a few weeks. This report aimed to evaluate 5 gastric cancer patients with bone marrow metastases to emphasize the importance of this condition.

  • Human bone marrow mesenchymal stem cells are thought to be multipotent cells, which are present in adult marrow, that can replicate as undifferentiated cells and that have the potential to differentiate to lineages of mesenchymal tissues, including bone, cartilage, fat, tenden, muscle, and marrow stroma. Cells that have the characteristics of human mesenchymal stem cells could be isolated from marrow aspirates of human and animals. This study was designed to identify and characterize genes specifically expressed by osteogenic supplements -treated cells by suppression subtractive hybridization(SSH) method. The results were as follows: 1. 2 genes were upregulated genes in osteogenic diffeentiation of hMSCs, which is further proved by Northern blot analysis. 2. IGFBP-2 has been identified playing an important role in bone formation. 3. HF1 was also upregulated during osteogenic differentiation, but its role in bone formation is not clear yet.

어성초 추출물 부탄올 분획이 Bone Marrow Stromal Cell 분열능 및 골다공증 랫드에 미치는 영향 (Effects of Houyttnia cordata on Bone Marrow Stromal Cell and Osteoporetic Rat)

  • This study was performed to evaluate the effect of HCB on the bone mass and its related factors in estrogen-deficient animal model. The model rats of osteoporosis showed a significant decrease in bone density, bone ash density, calcium content of femur bone. At the 14th day after ovariectomy-surgery, rats were administered with HCB, extract of Houyttnia cordata, per orally, and continued for 10 weeks. And osteoporosis related parameters were determined to investigate the effect of HCB. Osteoporotic rats showed lower serum estrogen level, higher body weight than normal rats, and showed atrophy of uterine horns.

Qi therapy as a complementary therapy in chronic myeloid leukemia

  • We describe the successful treatment of a case of chronic myeloid leukemia with Qi therapy. The patient's disease was managed with conventional medical treatment and Qi therapy as a complementary therapy. Before Qi therapy, 95% of the patient's bone marrow showed evidence of disease. A second bone marrow sample five months after Qi therapy revealed that 38% of the bone marrow was normal; one year after Qi therapy the bone marrow was no longer producing any cancer cells. Although these results were obtained for a single case only and may not constitute conclusive evidence, the data suggest that Qi therapy given as a complementary therapy during conventional medical treatment may have beneficial effects on chronic myeloid leukemia.

  • In this study, we showed that neurons could be generated from adult canine bone marrow stem cells by culturing with $DMSO/BHA/FeCl_2$. These neurons differentiated from the bone marrow stem cells formed neurites, expressed neuron-specific markers. This differentiation was enhanced by $FeCl_2$. These results suggest that iron can effectively initiate differentiation of adult bone marrow stem cells into neurons.
