Effect of deproteinized bovine bone mineral on cell proliferation in the procedure of guided bone regeneration (골유도재생술시 탈단백 우골이 세포증식에 미치는 영향)

  • One of the bone substitutes now in routine use, deproteinized bovine bone mineral(DBBM), is regarded as resorbable and osteoconductive, but some studies refute this. The present study was performed to evaluate the effects of DBBM on guided bone regeneration using titanium membrane on the calvaria of rabbit. At 2 weeks, 4 weeks, 8 weeks, and 12 weeks after surgery, the animal was scrificed. Non-decalcified specimens were produced for histologic analysis. The results of this study were as follows : 1. Titanium membrane was biocompatible and capable of space-maintaining, but there was ingrowth of soft tissue through the pore of titanium membrane. 2. There was no resorption or reduction of DBBM with time. 3. Some of the DBBM particles were combined with newly formed bone. But, apart from host bone, a great part of the particles were surrounded by connective tissue. 4. The bone formation was slight vertically and restricted to superficial area of host bone. Whithin the above results, DBBM dose not appear to contribute to bone formation. DBBM may disturb the migration and proliferation of mesenchymal cell derived from host bone and increase the growth of connective tissue. Therefore, careful caution is needed on selection of bone graft material and surgical protocol at guided bone regeneration for implant placement.

The Effect of Achyranthis Radix Extract on Hard Tissure Regeneration (우슬 추출물의 경조직 재생촉진효과)

  • This study was performed to investigate therapeutic effects of Achyranthis Radix extract and chitosan on the growth and differentiation of rat calvarial cells. it was found that treatment of methanol extract of Achyranthis Radix for 2 days caused 2.4-fold increase in the growth of rat calvarial cells. However, chitosan treatment caused only 1.9-fold increase in the cell growth. Treatment of methanol extract of Achyranthis Radix for 14 days caused 2-fold increase in the growth of rat calvarial cells. Alkaline phosphatase activity, one of the markers for bone cell differentiation, was increased approximately by 1.7-fold and 2.9-fold by the treatment of methanol extract of Achyranthis Radix for 2 days and 14 days, respectively. These results suggest that Achyranthis Radix extract could be beneficial for bone regeneration.

A Study on the Calcium Bioavailability of Eggshell Powder in the Growing Rats (난각 칼슘의 생체 이용성에 관한 연구)

  • This study has investigated the bioavailability of calcium in eggshell powder (ESP) for the purpose of reutilizing eggshells as the calcium source. The experiment was designed 2 ${\times}$ 2 factorial method with two sources, CaCO$_3$ and ESP, and two levels, 0.2% and 0.4% calcium. Weanling SD rats were assigned randomly to one of 4 groups and provided by one of the isocaloric, 20% casein based 4 different experimental diets for 4weeks. Deionized water was given and environment was kept from any contamination of minerals. The body weight, diet intake, feed efficiency ratio (FER), bone growth, Ca contents of bones, and apparent absorption were measured. FER (0.38 - 0.40) and kidney weight were not different among groups and the weight and length, Ca content, strength of two bones Tibia and Femur were not affected by Ca sources except Femur Ca content. Ca content of Femur was greater in ESP groups than that of CaCO$_3$ groups. The body weight gain, bone growth, the Ca contents and strength of bones were significantly greater in 0.4% calcium groups suggesting 0.2% calcium is not sufficient for the optimum growth in the growing rats. These results indicate that ESP be a proper Ca source comparable or superior to CaCO$_3$. However the apparent absorption rate of final 3 days of feeding did not support the observed results showing lower rate in ESP than CaCO$_3$ groups. Further study be needed in the absorption aspect.

  • Antlers from deer species are alternative animal by-products. Due to the oriental trade, the velvet antler industry is rapidly emerging in North America. The unique biological property of antler wish a deciduous natural phenomenon offers the valuable model of biomedical research. Growing antlers showed different structures according to cell populations consisting of mesenchymes, chondroblasts, chondrocytes and osseous tissues from distal to proximal portions of main beam. Their structures were different from two tissues, cartilage and bone, in growing antlers. Zone of maturing ants calcifying chondrocytes referred as upper section was invaded by osteoblasts indicating the occurrence of endochondral ossification. The cartilaginous tissues were gradually replaced by osseous tissues downward. The bony tissues referred as the middle and base sections in this thesis contained spongy bone and cortical bone structure in the difference of the degree of mineralization antral the thickness of cortical bony in adjacent to outer velvet layer. In addition, the endocrinological regulators such as testosterone, prolactin, growth hormones and other growth factors are actively involved in the unique deciduous nature shown in the growth and development of antler.

Airway analysis in unilateral cleft lip and palate patients (편측성 순$\cdot$구개열자의 기도 분석)

  • This study was designed to analysis the airway which affects to breathing, speech and facial growth pattern in unilateral cleft lip and palate patients. Upper airway and the position of hyoid bone pattern were analyzed on the lateral cephalometric radiographs of the 78 subjects of complete unilateral cleft lip and palate group and each group was divided two sub-groups by circumpubertal growth peak and gender. These data were statistically analyzed to examine the difference between pre-circumpubertal growth peak group and post-circumpubertal growth peak one, and between male and female group. The results of this study were as follows: 1. After circumpubertal growth peak stage, the position of hyoid bone was lower than before in both male and female group. 2. After circumpubertal growth peak stage, the measurement were increased in CV3ia-APH, PNS-ad which related to the volume of pharyngeal space. This was due to the decrease of adenoid and anteroiferior growth of mandible and affected to breathing and speech after circumpubertal growth peak. 3. During circumpubertal growth peak stage, all measurements increased more in male than female group, which due to the different amount of growth in different gender. 4. The position of hyoid bone was lower in male than female group in all age group. 5. After circumpubertal growth peak stage, CV3ia-APH increased more in male. This was due to the more growth in madible of male which resulted in the activation of digastric muscle.

  • This study was performed to evaluate bone formation in the calvaria of rabbit by the concept of guided bone regeneration with titanium mesh membrane and demineralized freeze-dried bone. The animal was sacrificed at 2 weeks, 4 weeks, 8 weeks, and 12 weeks after the surgery. Non-decalcified specimens were processed for histologic analysis. 1. The titanium mesh but the biocompatibility was excellent the cell-occlusiveness was feeble. 2. The cell-occlusiveness was feeble and also the soft tissue growth of the upper part of the newly-formed bone after operating was excellent in early stage. 3. The maintenance ability of the space for the GBR very was excellent. 4. The titanium mesh the tissue-integration was superior the wound fixation ability excellent. 5. The demineralized freeze-dried bone did not promote the bone regeneration. 6. With the lapse of time, formation quantity of the bone some it increased, it increased quantity very it was feeble. Within the above results, the titanium mesh for the guided bone regeneration was excellent, the dεmineralized freeze-dried bone confirmed does not promote bone regeneration.

  • A Giant cell tumor of bone has unusual characteristics of pulmonary metastasis as well as local aggressiveness. Clinical courses of pulmonary metastasis of benign giant cell tumor vary including rapid growth, continuously slow growth or spontaneous regression. We report a case of extensive pulmonary metastasis of giant cell tumor of bone, which regressed spontaneouly.

  • In anti-cancer therapies, radiotherapy and chemotherapy show a superior inhibition effect on cancer cell growth, but those are very toxic to normal tissues and organs. Particularly, drugs for neutropenia, one of chemotherapy agents, which suppress the function of bone marrow, are needed to be controlled in terms of their dosage and therapy period because of their side effect. Phellinus linteus polysaccharide (PL) has been reported to increase the number of splenocytes and bone marrow cells. PL has been shown to decrease the side effects of cyclophosphamide (CYC) treatment to the cancer patients. PL showed no effects in semisolid clonogenic assay, suggesting that PL doesn't contain substantial compounds to substitute for colony stimulating factors (CSFs). On the other hand, PL increased the expression of SCF, IL-3, GM-CSF, TPO genes. These results indicate that PL may promote the growth and proliferation of splenocytes and bone marrow cells through indirect or CSFs-dependent pathway, which may lead to a hematopoiesis.

  • For further improvement of osseo-integration of bone crystal with a dental implant, a design optimization study is carried out for various holes inside its body to deliver bioactive materials and the effect of bioactive material injection on the bone crystal growing. When bioactive material is absorbed, the bone crystal can grow into holes, which would increase the strength of implant bonding as well as a surface integration. The stress concentrations near the uppermost outlet holes were reduced with increasing the number of outlet holes. A design improvement in the uppermost outlet was shown to be effective in reducing the stress concentration. For design parameters under consideration in this study, total area of outlet of 6.38 $mm^2$ and maximum stress of 1.114 MPa, which corresponds to type 6-C. It is due to the minimization of maximum stress and total area of outlet. The design of the outlet facing down was more effective in reducing the maximum stress value compared with a horizontal symmetry.

  • Bone cells produce multiple growth factors and cytokines that have effects on bone metabolism and can be incorporated into the bone matrix. The present study was designed to extend these observations by examining the interactions between transforming growth factor-$\beta$(TGF-$\beta$) or interleukin-$1\beta$(rhIL-$1\beta$) and bone cells in a rat long bone culture model. IL-$1\beta$ regulates several activities of the osteoblast cells derived from rat long bone explants in vitro. IL-$1\beta$ stimulated cellular proliferation as well as the synthesis of prostaglandin $E_2$ and Plasminogen activator activity in the cultured cells in a dose-dependent manner. TGF-$\beta$ is present in the bone matrix and potentially released during bone resorption. TGF-$\beta$ reduced basal bone resorption and inhibited vitamin $D_3[1,25(OH)_2D_3]$-induced bone resorption in rat long bone cells. These results support the role of IL-$1\beta$ in the pathological modulation of bone cell metabolism, with regard to implication in the Pathogenesis of osteoporosis by IL-$1\beta$, and that TGF-$\beta$ positively inhibits the bone resorption.
