Immunostimulatory and Bone Health-Promoting Activities of Litsea japonica Fruit Extract (까마귀쪽나무 열매 추출물의 면역증가 및 뼈 건강 효과)

  • This study was performed with hot water extract (LJF-W) and 70% ethanol extract (LJF-70E) of Litsea japonica fruit to investigate the immunostimulatory activity and bone health-promoting effect of L. japonica fruit. The production of pro-inflammatory mediator (nitric oxide) and pro-inflammatory cytokines (TNF-α, IL-6 and IL-1β) in murine macrophage RAW264.7 cells were estimated to examine the immunostimulatory activity of the fruit extracts. The immunostimulatory activity of LJF-W was higher than that of positive control (geinsenosides). However, there was no effect in LJF-70E. Futhermore, both LJF-W and LJF-70E appeared to stimulate the proliferation of MC3T3-E1 cells, proving the effect as a bone health agent. From this result it could be presumed that L. japonica fruit extracts not only stimulate the immune system, but also the ability to promote bone health.

Effects of Sea Tangle Extract on Formation of Collagen and Collagen Cross-link in Ovariectomized Rats (다시마 추출물이 갱년기 유도 흰쥐의 collagen 및 collagen 가교물질의 형성에 미치는 영향)

  • The purpose of this study was to investigate the effects of sea tangle (ST) extracts on formation of collagen and collagen cross-link in ovariectomized rats. From day 3 until 42 after the ovariectomy, Sprague-Dawley female rats were randomly assigned to the following groups: sham-operated rats(Sham), ovariectomized control rats (OVX-control), ovariectomized rats supplemented with ST at 50 mg/kg bw/day (OVX-ST50), 200 mg/kg bw/day (OVX-ST200). The ethanol extraxcr of ST was orally administrated at 1 ml/day. The change of collagen content was investigated in bone, cartilage and skin of ovariectomized rats. Effects of ST extract on the amount of collagen was examined by measuring the hydroxyproline, which is a specific amino acid existing in collagen. The ovariectomy resulted in a decrease in the levels of collagen content in bone and cartilage tissues. However, the supplementation with the sea tangle extract prevented the decrease in the collagen level in bone and cartilage tissues. Pyridinoline is pyridinium cross-link formed in the mature form of collagen from lysine and hydroxylysine residues. Although the pyridinoline content in bone collagen declined after ovariectomy, it was recovered to a normal level of Sham group by the supplementation with the ST extracts. In addition, the deoxypyridinoline content in bone collagen, which was reduced after ovariectomy was enhanced to normal level by the supplementation with the ST extract. These results was consistent with the conclusions based on estrogenic activities of ST.

Investigation of anodized titanium implants coated with triterpenoids extracted from black cohosh: an animal study

  • PURPOSE. The purpose of this study was to evaluate bone response to anodized titanium implants coated with the extract of black cohosh, Asarum Sieboldii, and pharbitis semen. MATERIALS AND METHODS. Forty anodized titanium implants were prepared as follows: group 1 was for control; group 2 were implants soaked in a solution containing triterpenoids extracted from black cohosh for 24 hours; group 3 were implants soaked in a solution containing extracts of black cohosh and Asarum Sieboldii for 24 hours; group 4 were implants soaked in a solution containing extracts of pharbitis semen for 24 hours. The implants from these groups were randomly and surgically implanted into the tibiae of ten rabbits. After 1, 2, and 4 weeks of healing, the nondecalcified ground sections were subjected to histological observation, and the percentage of bone-to-implant contact (BIC%) was calculated. RESULTS. All groups exhibited good bone healing with the bone tissue in direct contact with the surface of the implant. Group 2 ($52.44{\pm}10.98$, $25.54{\pm}5.56$) showed a significantly greater BIC% compared to that of group 3 ($45.34{\pm}5.00$, $22.24{\pm}2.20$) with respect to the four consecutive threads and total length, respectively. The BIC% of group 1 ($25.22{\pm}6.00$) was significantly greater than that of group 3 ($22.24{\pm}2.20$) only for total length. CONCLUSION. This study did not show any remarkable effects of the extract of black coshosh and the other natural products on osseointegration of anodized titanium implants as coating agents. Further studies about the application method of the natural products on to the surface of implants are required.

A Study of Safflower Seed Extracts on Bone Formation in Vitro (홍화인 추출물이 골 형성에 미치는 영향에 관한 실험실적 연구)

  • The ultimate goal of periodontal therapy is the regeneration of periodontal tissue and the repair of function. For more than a decade there have been many efforts to develop materials and methods of treatment to promote periodontal tissue regeneration. Recently many efforts are concentrated on the regeneration potential of material used in traditional medicine. Safflower(Carthamus tinctorius L.) seed extract(SSE) have long clinically used in Korea to promote bone formation and prevent osteoporosis. The purpose of this study was to examine the effects of SSE on bone formation in human osteoblastic cell line. Human fetal osteoblastic cell line(hFOB 1.19) was cultured with DMEM and SSE($1{\mu}g/ml$, $10{\mu}g/ml$, $100{\mu}g/ml$, $1mg/ml$) at $34^{\cdot}C$ with 5% $CO_2$ in 100% humidity. The proliferation, differentiation of the cell was evaluated by several experiments. Cell proliferation was significantly increased at $10{\mu}g/ml$, $100{\mu}g/ml$, 1mg/ml of SSE after 3 and 7 days incubation(p<0.05). Cell spreading assay was significantly increased at $100{\mu}g/ml$ of SSE after 3 days and $1{\mu}g/ml$, $10{\mu}g/ml$, $100{\mu}g/ml$, 1mg/ml of SSE after 7 days(p<0.05). Alkaline Phosphatase(ALP) level was significantly increased in $10{\mu}g/ml$, $100{\mu}g/ml$, 1mg/ml of SSE(p<0.05). Collagen synthesis was significantly increased at $10{\mu}g/ml$, $100{\mu}g/ml$, 1mg/ml of SSE(p<0.05). A quantified calcium accumulation was significantly increased at $10{\mu}g/ml$, $100{\mu}g/ml$ of SSE(p<0.05). ALP and osteocalcin mRNA was expressed in $100{\mu}g/ml$ of SSE by RT-PCR. These results indicate that SSE are capable of increasing osteoblasts mineralization and may play an important role in bone formation.

Nrf-2 Mediated Antioxidative Effect of Korean and Chinese Safflower Seeds (한국산 중국산 홍화자의 Nrf-2 매개 항산화 효과)

  • Safflower (Carthamus tinctorius L.) seeds have been used in Korea and China for promoting bone formation and protection. This study was designed to examine the Nrf-2 mediated anti-oxidative effects of Korean and Chinese safflower seeds. Water and ethanol extracts of safflower seeds were treated to RAW 264.7 cells. Nrf-2 transcriptional activity was measured by reporter gene assay and western blot analysis. Semi-quantitive RT-PCR analysis was adopted to measure Nrf-2 dependent gene expressions. Water extracts of safflower seeds have strongly induced the activation of Nrf-2 transcription than ethanol extracts. Especially, water extracts of Korean safflower seeds has more strongly increased the expression of nuclear Nrf-2. Water extracts of Korea and China safflower seeds have also increased the expression of Nrf-2-dependent genes such as GCLC, NQO-1 and HO-1 in RAW 264.7 cells. However, all kinds of safflower seeds extracts did not increase intracellular ROS production. These results demonstrate that the antioxidant effects of safflower seeds are not related with ROS production, rather it is mediated by the direct activation of Nrf-2.

Effects of Mulberry Fruits on Collagen Content of Connective Tissues in Ovariectomized Rats (오디가 난소를 절제한 흰쥐 결함조직 중의 collagen 함량 변화에 미치는 영향)

  • The aim of this study was to evaluate effects of mulberry cultivars extracts on the collagen content of the connective tissues in ovariectomized rats. From day 2 until day 37 after ovariectomy, Sprague-Dawley female rats were randomly assigned to the following groups : sham-operated rats (Sham), ovariectomized control rats (OVX-control), and ovariectomized rats supplemented with $80\%$ ethyl alcohol extracts from various kinds of Tajikistan mulberry, Korea mulberry and China mulberry at 200 mg/kg bw/day, respectively. The mulberry extracts were orally administrated at 1 mL per day. The OVX rats were significantly heavier than the sham-operated rats were at all time points, but supplementation with the mulberry extracts tended to gain weight less than OVX-control did. The ovariectomy caused a decreasing in the levels of collagen content in bone and cartilage tissues. However, supplementation with mulberry extracts prevented the decrease in the collagen level in bone and cartilage tissues. Therefore, it may be used to possibly improve the quality of life in menopausal women.

Effects of the Rehmanniae Radix Preparat on Ovariectomized Rats (숙지황(熟地黃)이 난소적출(卵巢摘出)로 폐경(閉經)이 유발(誘發)된 흰쥐에 미치는 영향(影響))

  • Objectives : Obese and osteoporosis are diseases characterized by bone loss and morbidity with osteoporotic fracture is increasing in elderly. The present study was carried out to determine the effects of Rehmanniae Radix Preparat(RRP) on the postmenopausal obese. Methods : The variance of body weight, femoral bone weight, serum level of cholesterol and microscopic view of tibia were investigated. Animals were observed for 8 weeks, and vehicle and extracts of prescriptions were administered ad libitum. Results : Body weight increased significantly in 6 weeks after ovariectomy, and the weight of RRP administered group decreased significantly. RRP decreased the serum level of cholesterol. Conclusions : According to above results, RRP can be used for prevention and curing the postmenopausal obese.

The Effect of DanSamSaMul-Tang on Hematopoiesis (단삼사물탕이 조혈작용에 미치는 영향)

  • Objectives : Korean traditional medicinal herbs and some formulas have been used to improve the function of the hematopoietic system. The present research is to examine the effect of DanSamSaMul- Tang(DSSMT) as a biological response modifier related to hemato-potentiating function using bone marrow cell as a testing system. Methods : Mice with leukopenia and thrombocytopenia induced by CTX were treated with DSSMT Extracts. Then we observed cytokines mRNA expression level, the number of CFU-GEMM and BFU-E, and the expression of TPO and SCF in bone marrow cells of mice. Results : In the DSSMT group, the gene expressions of hematopoietic cytokines were increased significantly and the number of CFU-GEMM and BFU-E of this group was significantly increased compared to the group treated only with EPO and IL-3. The expressions of TPO and SCF in the group treated with DSSMT were higher than those of the control group. Conclusions : We showed that DSSMT has a good effect on the hematopoietic system. This could also be used as important research data for Korean oriental medicine about hemato-potentiating.

Inhibitory Effect of Water Extracts of Aconiti Lateralis Preparata Radix and Acanthopanacis Cortex on Differentiation of Bone Marrow-Derived Adipocytes and Osteoclasts (부자(附子)와 오가피(五加皮) 물 추출물의 골수유래 지방세포와 파골세포 분화 억제 효과)

  • Objectives : The aim of this study was to evaluate the efficacy of Aconiti Lateralis Preparata Radix (AP) and Acanthopanacis Cortex (AT) extracts in bone-derived adipocyte OP9 cell, osteoclast and osteoblast-like MG63 cells. Methods : MTT assay was used to evaluate the cytotoxicity of AP and AT extracts on OP9, osteoclast and MG63 cells. OP9 cells were treated with AP and AT, and the alterations in fat storage in the cells were determined by the Oil red O. To explain effects of RANKL-induced osteoclast differentiation in bone marrow macrophages, we performed the TRAP staining. The protein level of CAAAT/enhancer binding protein alpha ($C/EBP{\alpha}$) and peroxisome proliferator-activated receptor ${\gamma}$ ($PPAR{\gamma}$) as a adipocyte differentiation marker, and adiponectin was examined using western blot in differentiated OP9 cells. Effects of related genes were confirmed by luciferase assay using reporter assay. Results : AP and AT was not toxic on OP9 and MG63 cells, but AT was a little cytotoxic to osteoclast at the dose of $100{\mu}g/m{\ell}$. They could inhibit differentiation of OP9 cells and osteoclast with results of oil red O staining and TRAP staining. By western blot, AP and AT decreased the expression of $PPAR{\gamma}$ and $C/EBP{\alpha}$ which is the key transcription factor in adipogenesis and adiponectin secretion. AT also inhibited the BMP-4 activity in luciferase assay. AP also inhibited BMP-4 and Wnt3a activity, stimulated ER-${\beta}$ activity but inhibited androgen receptor activity. Conclusions : These results show AP and AT can be useful in osteoporosis and obesity via inhibition of osteoclast and adipocyte differentiation.

Comparative Analysis of the Constituents of the Leaves and Roots of Rumex crispus and their Effects on the Differentiation of Human Osteoblast-like MG-63 Cells (소리쟁이 잎과 뿌리 성분 분석 및 사람 조골 유사 MG-63 세포 분화에 미치는 효과 비교)

  • Rumex crispus (curled dock), which is a perennial wild plant, has long been used as a laxative, astringent, and medicine to treat blood and skin diseases. We recently reported that the roots of R. crispus are an effective nutraceutical for bone. This study prepared ethanol extracts of the leaves and roots of R. crispus, and analyzed the major constituents using liquid chromatography and mass spectrometry. In addition, their effects on the proliferation and differentiation of human osteoblast-like MG-63 cells, such as cell viability, alkaline phosphatase (ALP) activity, collagen content, and mineralization, were compared. The chromatograms of the chemical constituents of the two extracts exhibited quite different profiles: quercetin and quercitrin were identified as major peaks in the leaf extract, whereas cinnamtannin B1 and procyanidin isomers were the major peaks for the root extract. Neither extract was cytotoxic at concentrations of < $25{\mu}g/ml$. ALP activity and collagen synthesis-which are markers of the early stage of osteogenesis-in MG-63 cells were significantly increased upon the addition of the root extract compared with the addition of the leaf extract. In contrast, the leaf extract had a more stimulatory effect on mineralization-which is marker of the late stage of osteogenesis-in MG-63 cells than did the root extract. In conclusion, extracts of both leaves and roots of R. crispus stimulated the bone-forming activity of osteoblasts; in particular, the root extract was more effective in the early stage of osteoblast differentiation, while the leaf extract was more effective in the late stage. This difference in anabolic activity may be due to differences in the constituents of the leaves and roots. The leaves and roots of R. crispus appear to complement each other as stimulators of bone formation.
