Beneficial effect of Combination with Korean Red Ginseng, Gastrodia Rhizoma and Polygoni Multiflori on Cholesterol and Erectile Dysfunction in Hyperlipidemia rats (홍삼, 천마, 적하수오 병용투여에 의한 고지혈증 랫드에서의 콜레스테롤 및 발기부전 개선효과)

  • Objectives : This study was designed to investigate effects of the combination with Korean Red Ginseng (Panax ginseng C.A. Meyer), Gastrodia Rhizoma (Gastrodia elata Blume) and Polygoni Multiflori Radix (Polygonum multiflorum Thunberg) on metabolic disorders including cholesterol and erectile dysfunction in hyperlipidemia rats.Methods : Animals were divided into six groups; Control with normal diet, high fat/cholesterol-diet (HFCD), fluvastatin, Korean Red Ginseng treated (KRG), and the combination treated (Korean Red Ginseng, Gastrodia Rhizoma and Polygoni Multiflori Radix; 1:1:1 for KGP1 and 2:1:1 for KGP2). The experimental groups initially received HFCD for 10 weeks and then treated orally with fluvastatin, KRG, KGP1 and KGP2 during the final 6 weeks. Erectile function was determined by the measurements of intracavernosal pressure (ICP) and maximal arterial pressure (MAP) after electrical stimulation of the cavernosal nerve.Results : KGP2 decreased the level of total cholesterol and LDL cholesterol in the sera of HFCD rats without no changes of body weights. KRG, KGP1 and KGP2 decreased the level of C-reactive protein (CRP) levels except of fluvastatin, synthetic HMG-CoA reductase inhibitor. KRG, KGP1 and KGP2 significantly increased the ICP, ICP/MAP ratio, area under the curve (AUC) compared with those of normal rat. Morphometric analyses showed that KRG, KGP1 and KGP2 increased the volume of smooth muscle and the regular arrangement of collagen fibers in corpus cavernosum of HFCD rats. The penile expression of eNOS was increased by KRG, KGP1 and KGP2.Conclusions : Based on these results, we suggest that the combination with Korean Red Ginseng, Gastrodia Rhizoma and Polygoni Multiflori may improve hyperlipidemia through regulating the lipid profiles and erectile dysfunction in rats.

Hypolipidemic Effects of Glycoprotein Isolated from Ficus Carica Linnoeus in Mice (무화과 당단백질의 혈중지질 저하 효과)

  • Glycoprotein (60 kDa) isolated from Ficus Carica Linnoeus (FCL glycoprotein) was examined by evaluating its hypolipidemic effects on plasma cholesterol levels and hepatic detoxicant enzyme activities in ICR mice. FCL glycoprotein $(100{\mu}g/mL)$ had strong scavenging activities (38%) against lipid peroxyl radicals. When mice were treated with Triton WR-1339 (400 mg/kg), levels of total cholesterol (TC) and low-density lipoprotein (LDL)-cholesterol in plasma significantly increased by 53.9 and 47.5 mg/dL, respectively, compared to the control, whereas, when pretreated with FCL glycoprotein $(100{\mu}g/mL)$, decreased remarkably by 55.4, and 47,0 mg/dL, compared to Triton WR-1339 treatment alone. Interestingly, high-density lipoprotein (HDL)-cholesterol level did not change. Body and liver weights did not change significantly after Triton WR-1339 treatment in presence of FCL glycoprotein. FCL glycoprotein $(100{\mu}g/mL)$ stimulated activities of antioxidative detoxicant enzymes such as superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx), whereas GPx activity significantly increased compared to the control. These results suggest FCL glycoprotein has abilities to scavenge lipid peroxyl radicals, lower plasma lipid levels, and stimulate detoxicant enzyme activity in mouse liver.

Effects of Pleurotus eryngii on Lipid Levels and Enzyme Activity in Male Rats Fed High Cholesterol Diets (콜레스테롤 식이를 섭취한 흰쥐에 새송이버섯이 지질대사 및 효소활성에 미치는 영향)

  • Effects of dietary supplementation of Pleurotus eryngii(PE) to a diet containig the cholesterol on lipid levels in the liver and serum, and enzyme activities of rats were studied by feeding 8-weeks old male rats with the four diets for 4 weeks, respectively: normal diet, control diet (10% fat + 0.5% cholesterol), 3% and 5% PE diets (control diet + 3% and 5% PE dry powder). The body weight gains and food efficiency ratios of the 3% and 5% PE diet groups were significantly lower than those of the control group. The liver and epididymal fat pad weights, and hepatic triglyceride Levels of the 5% PE diet group were more significantly decreased than those of the control group. The concentrations in serum triglyceride, total cholesterol, LDL+VLDL-cholesterol, and atherogenic index ratios were significantly lower in the 3% or 5% PE groups compared to those of the control group. Ratio of HDL-cholesterol to total-cholesterol was significantly higher in the 3% and 5% PE groups compared to that of the control group. The fecal total lipid excretion of the 5% PE diet group was more significantly decreased than that of the control group. There was no significant difference in the hepatic cholesterol, and the serum HDL-cholesterol concentrations, and the GOT, GPT and ${\gamma}-GTP$ activities among the experimental groups. These results showed that the Pleurotus eryngii powder feeding decreased the total cholesterol, LDL-cholesterol and atherogenic index, and increased the ratio of HDL-cholesterol to total-cholesterol in serum of the rats.

Antitumor and Immunologic Activity of Chitosan Extracted from Shell of Shrimp (새우 껍질에서 추출한 키토산의 항암 및 면역활성)

  • This study was designed to investigate the antitumor effect and immunological activities of chitosan extracted from Solenocera prominetis toward on mire bearing sarcoma-180. The growth inhibition ratio of the chitosan toward sarcoma-180 showed at the highest level of 63.84% when chitosan were administrated at the concentration of 40mg/kg. The direct cytotoxic effect of chitosan was not observed in the mice bearing sar-roma-180 in vitro. In the effect of immunological activities, dose-dependent responses indicated by the increase of leucocyte, peritoneal exudate cell than that of control group when chitosan administered to the mice in the concentation of 30mg/kg and 40mg/kg. Also dose-dependent responses showed also by the increase of immunoorgans weights such as body weight, liver, spleen or thymus in the same concentration of 30mg/kg and 40mg/kg. Food pad swelling having the relationship with arthus reaction of antibody-mediated hypersensitivity and delayed type hypersensitivity was recovered the almost normal level. In the efforts of macrophge on phagocytes, there were not substantial differences in phagorytic and corrected phagocytic index. In the number of plaque forming cell(PFC), PFC on the 10$^{7}$ spleen cells were increased the levels of 18.88% and 31.83% when chitosan were adminstersd at the concentration of 30mg/kg and 40mg/kg.

Repeated-Dose Oral Toxicity Study of Lactobacillus Plantarum AF1 Isolated from Kimchi in Rats (김치에서 분리한 Lactobacillus plantarum AF1의 흰쥐에 대한 반복투여독성)

  • This study was performed to investigate the 4-week repeated-dose toxicity of $Lactobacillus$ $plantarum$ AF1 ($Lb.$ $plantarum$ AF1), a lactic acid bacteria isolated from kimchi, in male and female rats. Sprague-Dawley male and female rats were divided into four groups, with 10 animals in each group. The test article was administered once daily by gavage to rats at dosage levels of 0, 0.5, 1.0, and 2.0 g/kg/day for 4-weeks. There were no test articlerelated deaths or abnormal clinical signs in either gender of rat during the observation period. Furthermore, no differences were found between the control and treatment groups in terms of body weight changes, food intake, and water consumptions. Hematological parameters, serum biochemical analysis, and any other findings also showed no significant or dose-dependent alterations. There were no alterations in organ weights upon administration of $Lb.$ $plantarum$ AF1. These results suggest that there were no adverse effects of oral application of $Lb.$ $plantarum$ AF1 up to a dosage level of 2.0 g/kg in both male and female rats.

Detection of Novel Mutations in the FABP3 Promoter Region and Association Analysis with Intramuscular Fat Content in Pigs (돼지 FABP3 Promoter 부위 내 신규 돌연변이 탐색과 근내지방도와의 연관성 분석)

  • Intramuscular fat content(lMF) is considered as one of major economic traits in the pig breeding and industry. In general, high IMF results in better meat quality. Several approaches to detect quantitative trait 10ci( QTL) for IMF indicated a strong possibility of the existence of a QTL related to IMF between the microsatellite marker SW71 and SW1881 on SSC6q. Porcine FABP3 has been considered as a candidate gene affecting IMF due to its physiological roles and position on the pig genome. Two novel mutations, g.-114T> C and g.-158T>G were detected by duplicate sequencing of the porcine FABP3 promoter region. These two mutations were identified as absolute linkage disequilibrium. The g.-158T> G mutation was used for investigating relationships with growth and fat deposition traits. The GG genotype of the g.-158T> G polymorphism showed highly negative effects(P< 0.01) on body weights at 3 and 12 weeks of age, and a positive effect(P< 0.05) on IMF. However, backfat thickness(BF) and carcass fat(CF) content were not significantly associated with the genotype. The result indicates that the novel mutations, identified in this study, could be utilized as possible genetic markers to improve IMF, independent with BF.

The Effect of Dietary Phytase on Nitrogen and Phosphorus Excretion of Gestating and Lactating Sows (Phytase 첨가가 임신돈과 포유돈의 분뇨 배설량 및 질소와 인 배설량에 미치는 영향)

  • This study was carried out to investigate the effect of dietary phytase on nitrogen and phosphorus excretion of gestating and lactating sows (Yorkshire ${\times}$ Landrace ${\times}$ Duroc). Twelve gestating sows and twelve lactating sows were used in this study and were divided into 2 groups (1 control group and 1 treatment group, 6 replications/group) the control group was without phytase and the treatment group was fed with phytase (750 FTU/kg) in the diet, respectively. Body weights of gestating and lactating sows were $208.9{\pm}13.8$ kg and $190.5{\pm}22.9$ kg, respectively. In gestating sows, feed intake was greater in phytase fed group than the control (P<0.05), but water intake and total excretion were not different between the groups. In lactating sows, feed and water intakes and total excretion were not different between the groups. The N intake of lactating sows was higher in phytase fed group than control (P<0.05) but N excretion ratio was not different. In lactating sows, N intake and excretion and N excretion ratio were not significantly different between the groups. P excretion and excretion ratio in gestating sows decreased by phytase treatment (P<0.05) as compared to control. In lactating sows, N intake and excretion was not significantly different by added phytase (P>0.05). Finally, dietary addition of phytase decreased P excretion in feces of gestating and lactating sows.

  • The strains selected in the previous paper were identified according to the manual of Aspergilli by Thom and Raper, and the preparation of the crude enzymes with the strains were investigated. The fermented fodders were made with wheat bran and chaffs containing 20% wheat bran by the strains, and the feeding experiments concerned was conducted. The results obtained are as follows: 1. The selected strains were identified as below; MC-9 was a similar strain of Aspergillus niger van Tieghem, MC-10 was a similar strain of Aspergillus flavus Link, MC-53 was a similar strain of Aspergillus penicilloides Spegazzini and MC-61 was a similar strain of Aspergillus niger mut Schiemanni n. comb. 2. Percentages of crude enzymes obtained from the dried matters of the wheat bran cultures were MC-9: 2.9%, MC-10: 3.9%, MC-53: 6.4% and MC-61:2.6%. And the cellulase activity of MC-9 was the most active among the crude enzymes as the same as the filtrates of wheat bran cultures. 3. As the results of the feeding experiments, changes of body weights showed no trends to be significant as compared with the control group, and the groups of MC-9 and MC-61 showed the increasing significance with P<0.05 in egg-laying rate.

Efficacies of the Modified Ultrafiltration and Peritoneal Dialysis in Removing Inflammatory Mediators After Pediatric Cardiac Surgery (소아 개심술 후 변형 초여과법과 복막투석의 염증 매개체 (inflammatory mediator)의 감소에 대한 효과)

  • capillary leak syndrome and organ dysfunction in infants. Removing harmful cytokines and complement anaphylatoxins after cardiopulmonary bypass may attenuate this response. This study was conducted to see if the modified ultrafiltration and postoperative peritoneal dialysis can reduce plasma inflammatory mediators in pediatric cardiac surgery. Material and Method: 30 infants (age 1.1 to 12.6 months) who underwent closures of ventricular septal defect using cardiopulmonary bypass (CPB) were enrolled in this study. These patients were divided into three groups; 10 patients selected randomly underwent modified ultrafiltration (Group U), 10 with small body weights ($\leq$5 kg) received postoperative peritoneal dialysis (Group P), and 10 patients did not undergo modified ultrafiltration nor receivcd peritoneal dialysis (Group C). Serum samples were obtained before and after CPB, and after peritoneal dialysis. Effluents sample were also obtained after modified ultrafiltration or peritoneal dialysis. C3a and interleukin-6 (IL-6) were measured by radioimmunoassay and enzyme-linked immunosorbent assay respectively. Result: There was no differences in CPB time, aortic cross-clamping title, and lowest temperature during CPB. The effluents of peritoneal dialysis contained significant amount of C3a and IL-6, but there was no definitive decrease of serum concentration of C3a and IL-6. The effluents of modified ultrafiltration had some amount of C3a and negligible IL-6, and there was no decrease of serum concentration of these (actors. Conclusion: The effluents of peritoneal dialysis contained significant amount of proinflammatory cytokine, IL-6 and complement, C3a. However this study failed to elucidate the decrease in serum levels of these factors. The modified ultrafiltration also was not able to reduce the serum levels of C3a or IL-6 in our study as well.

The Norwood-Rastelli Procedure for Left Ventricular Outflow Tarct Obstruction with a Ventricular Septal Defect - Three case report - (심실중격결손이 동반된 좌심실유출로협착 환아에서의 Norwood-Rastelli Procedure -3예 보고 -)

  • Between 2001 and 2006, 3 neonates that had multilevel left ventricular outflow tract obstruction and a ventricular septal defect underwent the Norwood-Rastelli procedure. The body weights ranged from 2.9 to 3.1 kg. The patients had a near normal sized mitral valve and left ventricle. We simultaneously performed a modified Norwood procedure with native tissues-to-tissue anastomosis without circulatory arrest, and a Rastelli type procedure using a non-valved conduit from the right ventricle to the pulmonary artery and intracardiac patch baffling from the left ventricle to the pulmonary valve via the ventricular septal defect. The postoperative courses were uneventful. During follow-up, there was one late mortality caused by a cardiac catheterization related complication at 7 months after surgery, One patient required a Rastelli conduit change. Two patients are doing well during a follow-up period of 1 and 5 years, respectively.
