Effect of xylo-oligosaccharides on reproduction, lipid metabolism, and adipokines of hens during the late egg-laying period

  • Objective: The present study aimed to investigate the effect of xylo-oligosaccharides (XOS) administration on egg production, reproductive hormones, serum lipids and adipokines of hens at the late cycle of reproduction. Methods: Four treatments included control (basal diet) and XOS addition at 2.0 (XOS-2), 4.0 (XOS-4), or 6.0 (XOS-6) g/kg of diet using 288 commercial Hy-Line brown hens from 73 to 84 wk of age. Egg production, body fat deposition, reproductive tract and hormones, lipid metabolism and adipokines were determined. Results: At 84 wk, compared to the control, XOS supplementation at the three doses increased (p<0.001) egg-laying rates by 13.2% averagely, which led to a higher egg mass by 131 g/hen throughout the whole trial period. Abdominal fat and skinfold of XOS treatments were decreased (p<0.001) by 26.1% and 18.6%, respectively; large follicles and ovary weight were increased (p<0.001) by 0.73 follicle/hen and 18.6%, respectively. For serum parameters, cholesterol and triglyceride were decreased (p<0.001) by 17.5% and 29.2%, respectively; luteinizing hormone, follicle-stimulating hormone, and progesterone were increased (p≤0.001) by 16%, 31%, 29%, respectively; adiponectin and visfatin were increased (p<0.001) by 34% and 44%, respectively; but chemerin and leptin were decreased (p≤0.001) by 22% and 14%, respectively. With the increased XOS doses, linear decreases (p<0.05) were found on abdominal skinfold and serum triglyceride. Conclusion: The obtained data indicate that XOS can be used as an additive to improve fecundity by beneficially modulating fat deposition, lipid metabolism, reproductive hormones, and adipokines of hens at the late cycle of reproduction.

Combined Treatment of Silymarin and Jakyakgamcho-tang Suppresses Hepatic Lipid Accumulation and Inflammation in C57BL/6 Mice (Silymarin과 작약감초탕 병용투여의 C57BL/6 마우스 간조직 지질축적 및 염증 억제효과)

  • Objective : The aim of the present study is to examine hepatic lipid-lowering and anti-inflammatory effects of silymarin combined with Jakyakgamcho-tang on non-alcoholic fatty liver disease in a high fat diet-induced obese mice model. Methods : C57BL/6 mice were divided into four dietary groups: (1) Normal, (2) Control (60% high-fat diet), (3) Control + silymarin 50 mg/kg/day (Silymarin), (4) Control + Silymarin 50 mg/kg/day + Jakyakgamcho-tang 100 mg/kg/day (SPG). After 12 weeks administration, mice were sacrificed and lipids and inflammation-related biomarkers were analyzed liver and plasma. Results : Silymarin and SPG treatments significantly lowered body and liver weights compared to the Control. Serumlipids (triglyceride (TG), total cholesterol) and pro-inflammatory cytokines (tumor necrosis factor alpha, interleukin 1𝛽, and IL-6) concentrations were significantly lowered in the Silymarin and SPG groups than the Control group. Silymarin and SPG treatments suppressed hepatic TG level and hepatic lipid droplets compared to the Control. Theses two treatments significantly increased hepatic kinase B1 and AMP-activated protein kinase protein levels, and significantly decreased hepatic key lipogenic enzymes (acetyl-CoA carboxylase, fatty acid synthase and stearyl coenzyme A desaturase 1) protein levels than the Control. SPG also significantly increased hepatic fatty acid oxidation-related protein (peroxisome proliferator-activated receptor alpha and uncoupling protein 2) levels than the Control. Conclusions: Silymarin and SPG suppressed hepatic lipid accumulation by regulating hepatic protein expression, and lowered blood pro-inflammatory cytokines concentrations though the synergic effect of silymarin and Jakyakgamchotang was not clear.

Evaluating productive performance, meat quality and oxidation products of Italian White breed rabbits under free-range and cage rearing system

  • Objective: Free-range systems have been increasingly available to the consumer due to increased demand for more sustainable meat-products. In the current study, the effect of free-range (FR) and cage system (CS) was explored on growth performance, meat quality and oxidation products in Italian White breed rabbits during the growing-fattening phase (5 to 13 weeks of age). Methods: Forty rabbits were randomly allotted to two treatment groups according to the rearing system, and each treatment group was replicated five times with four subjects in each replicate (20 rabbits per treatment-group). All rabbits fed the same diet as pelleted, and under FR system, no additional feeds were available to animals. Results: Rearing system had significant effect on rabbit growth performance, where CS group resulted in higher final body weight (p<0.045) and gain (p<0.029) and better feed efficiency (p<0.025) compared to FR rabbits. Most carcass traits were not affected by rearing system; however, a reduction of abdominal fat content (p<0.015) and meat lipids (p<0.034) was observed in FR rabbits. Rearing system had no effects on meat fatty acid profile, whereas meat from FR rabbits resulted less susceptible to lipid and protein oxidation compared to caged animals. Conclusion: In overall, FR system could be suggested as a substitute for conventional caged system because of FR system preserved rabbit meat from oxidation.

A Review on Phytochemistry and Pharmacology of Sparassis crispa (꽃송이버섯의 구성성분과 약리효과에 대한 리뷰)

  • Objective : Sparassis crispa has been recognized for its therapeutic value since the late 20th century because of its high β-glucan content. Since then, researches have been conducted on the pharmacological effects but most of these are individual studies on the effects of β-glucan from S. crispa and the comprehensive reviews are lacking. The purpose of this study was to review the compounds composition and pharmacological effects of S. crispa. Methods : This review analyzes the papers about chemical and nutritional composition and pharmacological effects of S. crispa. The data in this review is based on selected papers after reviewing all studies containing the keyword "sparassis crispa" for PubMed, NDSL, and J-Stage published before February 2019. Results : S. crispa is composed of protein, lipids, and carbohydrates. Most of the compounds are carbohydrates and the highest content is β-glucan. More than 40% of the dried fruiting body of S. crispa is composed of β-glucan. In addition, it contains polyphenols, flavonoids, terpenoids and phthalide-based compounds. Broad spectrum of its pharmacological actions have been established which include immunomodulatory, anticancer, antiinflammatory, antioxidant, hypoglycemic, antiobesic and neuroprotective effects. Conclusion : The most studied fields have been shown to have immunomodulatory and anticancer effects by inhibiting the proliferation of cancer cells and angiogenesis and increasing hematopoitic responses. Unique structure and characteristic of high molecular weight β-glucan are considered to have high immunomodulatory effects of S. crispa. And low molecular fractions or phthalides of S. crispa also have antioxidant, immunomodulatory and anticancer effects.

  • The biogenesis and biological roles of extracellular vesicles (EVs) in the progression of liver diseases have attracted considerable attention in recent years. EVs are membrane-bound nanosized vesicles found in different types of body fluids and contain various bioactive materials, including proteins, lipids, nucleic acids, and mitochondrial DNA. Based on their origin and biogenesis, EVs can be classified as apoptotic bodies, microvesicles, and exosomes. Among these, exosomes are the smallest EVs (30-150 nm in diameter), which play a significant role in cell-to-cell communication and epigenetic regulation. Moreover, exosomal content analysis can reveal the functional state of the parental cell. Therefore, exosomes can be applied to various purposes, including disease diagnosis and treatment, drug delivery, cell-free vaccines, and regenerative medicine. However, exosome-related research faces two major limitations: isolation of exosomes with high yield and purity and distinction of exosomes from other EVs (especially microvesicles). No standardized exosome isolation method has been established to date; however, various exosome isolation strategies have been proposed to investigate their biological roles. Exosome-mediated intercellular communications are known to be involved in alcoholic liver disease and nonalcoholic fatty liver disease development. Damaged hepatocytes or nonparenchymal cells release large numbers of exosomes that promote the progression of inflammation and fibrogenesis through interactions with neighboring cells. Exosomes are expected to provide insight on the progression of liver disease. Here, we review the biogenesis of exosomes, exosome isolation techniques, and biological roles of exosomes in alcoholic liver disease and nonalcoholic fatty liver disease.

Vitamin D Deficiency in Patients Hospitalized for Heart Failure Living in the Tropics

  • Background and Objectives: Vitamin D, as a steroid hormone, has multiple effects on human body and its deficiency has been associated with an increased risk of heart failure (HF) and unfavorable outcomes. The present study investigated the prevalence of vitamin D deficiency (VDD) and its relationship with cardiometabolic parameters in patients hospitalized for HF living in the city of Recife (latitude 8° South). Methods: Analytical cross-sectional study, with men and women aged 40-64 years. The HF group was recruited during hospitalization due to decompensation. A matched control group was recruited from the general endocrine clinics. Vitamin D status was assessed by measuring serum 25-hydroxyvitamin D (25OHD), considering deficiency when 25OHD <20 ng/mL (<50 nmol/L). Results: A total of 243 patients were evaluated (HF group: 161, control group: 82). Lower serum 25OHD levels were observed in the HF group (25.2±9.4 vs. 30.0±7.7ng/mL; p<0.001), as well as a higher prevalence of VDD (27.3% vs. 9.8%; prevalence ratio, 2.80; 95% confidence interval, 1.38-5.67; p=0.002). In patients with HF, VDD was associated with diabetes mellitus (65.9% vs. 41.0%; p=0.005) and female sex (65.9% vs. 44.4%; p=0.015). In the subgroup with VDD, higher values of hemoglobin A1c (7.9% [6.0-8.9] vs. 6.2% [5.7-7.9]; p=0.006) and dyslipidemia were also observed. Conclusions: We found higher rates of VDD in patients hospitalized for HF and this was associated with deleterious laboratory metabolic parameters.

The Beneficial Effects of Pectin on Obesity In vitro and In vivo (In vitro 및 In vivo에서 펙틴의 비만 억제 효과)

  • The effects of pectin on obesity was studied using 3T3-L1 pre-adipocytes and rats fed 20% high fat diets. The concentration of leptin released from 3T3-L1 adipocytes in the presence of pectin was significantly decreased by 85% compared to that of the control (p<0.05), however, glycerol concentration was not changed. These data indicate that pectin seems to inhibit lipids accumulation in the adipocytes rather than enhance the lipolytic activity. Forty Sprague Dawley rats were fed 20% high fat diet for 8 weeks to induce obesity and then divided equally into four groups. Experimental groups were normal diet group (ND), high fat diet group (HFD), HDF with 10% pectin group (HFP10), and HDF with 20% pectin group (HFP20). Diet for the each group was prepared to be iso-caloric following AIN-76 guideline. After obesity was induced, rats were placed on an restricted diet for 9 weeks. The body weight of HFD increased 50% (p<0.05) compared to the ND, while it was decreased by 12% and 16% for HFP10 and HFP20, respectively (p<0.05). The relative amount of visceral fats for HFDl0 and HFD20 were decreased by 45% and 59% compared to that of HDF (130%), respectively (p<0.05). Pectin seems to have a greater effect on reducing visceral fats accumulation than weight reduction. Significantly increased level of triglyceride, total cholesterol or LDL-cholesterol in the plasma of HFD was returned to the normal or even below the normal by pectin diet, while the level of HDL-cholesterol increased. Lipid lowering effect was also observed in the liver and heart. These effects of pectin were dosedependent. In conclusion, the beneficial effect of pectin on the obesity was observed from cell culture experiment and animal study in terms of inhibiting the accumulation of lipids in the adipocytes.

A Study on the Body Fat Content and Serum Lipids in Collegs Students (대학생의 체지방량과 혈청지질에 관한 연구)

  • 본 조사는 경기도 용인에 위치한 명지대학교의 남녀 대학생 202명의 대상으로 일반 환경과 혈철지질, 혈압, 체지방량 측정 검사를 중심으로 이루어졌으며, 조사 분석한 결과를 요약하면 다음과 같다. 1. 조사 대상자의 신체발달 상황을 보면 남자의 평균 신장과 체중 및 비체중치는 173.1cm, 65.9kg, 38.03이였고, 여자는 16.7cm, 52.4kg, 32.6로 한국인 성인의 표준치와 매우 유사한 수치였다. 체지방량 비율은 남자 16.4, 여자 24.0으로 나타 설별에 따른 차이가 뚜렸하였다. 2, 조사 대상자들의 수축기와 이완기의 혈압은 평균 $125\pm123mmHg,$ $77.2\pm11.1mmHg이고,$ 남자와 여자의 수축기 혈압은 각각 $132.3\pm1.9mmHg,$ $118.1\pm1.6mmhg로$ 정상 범위였다.확장기 혈압은 남녀 각각 $80.5\pm11.0mmHgm$ $73.9\pm102.m,Hg였으며$ 역시 정상범위에 포함되었다. 3. 혈청 지질의 검사 결과는 Tc는 남자 175.2mg/dl, 여자 183.6mg/dl로 나타났고 TG는 남자 108.4mg/dl, 여자 72.0mg/dl로 나타나 통RP적으로 유의한 차이가 있었다. (p<0.05). 또한 HDL-C는 남자 59.6mg/dl , 여자 64.3mg/dl 로 성별에 다른 유의적인 차이를 보였고 (p<0.05), LDL-C도 역시 남자 93.9mg/dl, 여자 104.8mg/dl로 나타나 통계적으로 성별에 다른 차이가 뚜렷하였다. (p<0.01). 조사결과 다른 연령층의 성인과는 달리 TG를 제외한 TC, H이-C, L이-C에 있어서 남자가 여자 보다 더 낮은 수치를 보였다. 4. 혈압에 따라 세 그룹으로 분류한 후 혈청 지질과의 관계를 보면, 수축기 혈아이 증가함에 따라 TC, TG, LDL-C은 증가하는 경향을 보였으며, HDL-C은 감소되었으나 통계적으인 유의성은 없었다. 이완기혈압의 겨우 89mmHg 이하 보다 90~94mmHg의 경우에 유의적으로 증가되었으며 (p<0.05) HDL-C은 감소하였으나 유의성은 없었다. 5. 혈정 지질과 비만도의 관계는 남녀 모두 양의 상관관계를 나타냈으나 여자보다 남자에서 뚜fut이 나타났다. 남자의 겨우 TC의 수치가 KI에 따라 110% 미만일 때 $169.7\pm31.5mg/dl,$ 110~120%일 때 $193.8\pm36.8mg/dl,$ 120% 이상 일 때 $202.2\pm29.3mg/dl로$ 비만도에 따른 유의적인 증가를 (p<0.05)보여주었으나 여자의 경우는 유의적인 차이가 없었다. TG의 수치는 남자의 경우 KI가 110%미만인 그룹은 $102.9\pm108.2mg/dl,$ 120% 이상인 그룹은 $112.2\pm40.0mg/dl로$ 비만도가 증가함에 따라 TG가 증가되었으나 유의적인 차이는 없었으며, 여자의 경우 유의적인 차이가 없었다. 남자의 경우 HDL-C은 비만도가 증가함에 따라 유의적으로 감소하는 경향을 보였으나(p<0.05) 여자의 경우는 유의적인 차이가 없었다. 6. 조사 대상자의 체지방량 측정결과 체기방률은 남자의 겨우 16.4% 여자의 겨우 24.0%로 나타나 여자의 체지방률이 남자보다 높았다. 7. 신체계측치, 혈압, 혈청 지질간의 상관관계를 살펴보면 TC, TG는 KI, BMI와 유의적인 양의 상관관계를 보였고 (p<0.01), HCL-C은 비체중, BMI, LBM, TBM와 유의적인 음의 상관관계를 보였으며, (p<0.01), KI, SBP와도 음의 상관관계를 보였다. (p<0.05), LCL-C는 KI와 유의적이인 양의 상관관계를 나타내었으며 (p<0.01) BF%, TBF, BMI와도 양의 상관관계를 나타냈었다. (p<0.05). 또한 LPH, Al는 모두 나이, 비체중, BMI, KI와 유의적인 양의 상관관계를 나타내었다. (p<0.01). 이상을 종합해 보면 본 조사 대상자들은 비만도가 높아질수록 혈청 지질 수준이 놓았으며, 이완기 혈압도 형철 지질과 상관관계를 나타났다. 대학생의 혈청 지질과 체지방에 대한 자료가 부족함을 고려할 때, 본 조사자료가 학생의 혈청지질의 체지방에 대한 기초 자료로서 이용될 것을 기대하며 보다 다각적인 연구가 이루어지기를 바라는 바이다.

Effect of Hen Egg Supplementation on Blood Lipid Profile and Fecal Bile Acid of C57BL/6 Mouse Fed Normal and High Cholesterol Diet (계란 급여가 일반식이와 고콜레스테롤 식이를 급여한 C57BL/6 마우스의 혈중지질과 분변 담즙산에 미치는 영향)

  • This study was conducted to evaluate the effect of whole egg supplementation on the blood lipid profiles and cholesterol levels of C57BL/6 mice. Sixty-six mice were divided into two groups: normal-diet supplemented and high-cholesterol diet supplemented. Lyophilized whole egg powder was mixed with the two diets at 2 and 10%: normal diet only, normal diet with 2 and 10% whole egg powder, high cholesterol diet only, high cholesterol diet with 2 and 10% whole egg powder. The mixed diets were fed for 5 wk and feeding condition (body weight change, feed intake, and feed efficiency ratio (FER)), blood lipid profiles (total cholesterol (TC), triglyceride (TG), high density lipoprotein-cholesterol (HDL-C), low density lipoprotein cholesterol, hepatic and fecal lipids (TG, TC)), and fecal bile acids were determined. No significant differences were found in body weight gain or FER after whole egg supplementation in both the normal and high-cholesterol diet fed groups. In the normal-diet fed mice, HDL-C increased significantly in the 2 and 10% whole-egg powder groups. In the high-cholesterol diet fed mice, administering 10% egg powder increased the atherogenic index compared to the control. Furthermore, administration of whole egg powder increased fecal bile acids dose dependently (p<0.05). These results indicate that administering 2% hen whole egg powder did not affect blood lipid profiles and was more beneficial for health by increasing HDL-C and aiding in the excretion of cholesterol by fecal bile acids than those in the control.

Effects of Green Tea or Coffee Consumption on Serum Lipid Profiles (녹차 및 커피 음용과 혈중 지질 성상과의 관계)

  • This study was conducted to investigate the effects of green tea or coffee consumption on the serum lipids profiles. This study analyzed data of 1,343 participants (753 males and 590 females) among the people who underwent medical check-up. The subjects were divided by three groups as pattern of green tea or coffee consumption. Anthropometric measurements, biochemical analysis and nutrient intake using a food frequency questionnaire were performed. There were no significant differences among groups in height, % body fat, waist circumference, and body mass index. There was no significant difference among groups in energy intake. Fat intake was significantly higher in green tea and coffee group than the control group in men subjects. The intakes of dietary Fe, vitamin A, $\beta$-carotene, and folate was the lowest in coffee group (p<0.05). The intakes of dietary fat was the highest in coffee group in male subjects (p<0.05). Serum concentrations of total cholesterol and LDL-cholesterol were green tea< control< coffee group in order by multivariate analysis after adjusting for confounding factors. The serum concentrations of total cholesterol and LDL-cholesterol were significantly lower in green tea group than coffee group by multivariate analysis after adjusting for confounding factors (p<0.05). However, there was no significant difference in the serum concentrations of total cholesterol and LDL-cholesterol between green tea and control group. These results suggest that green tea consumption didn't show positive effects in preventing hyperlipidemia but coffee consumption showed negative effects in hyperlipidemia.
