Aflatoxin-mediated Sperm and Blood Cell Abnormalities in Mice Fed with Contaminated Corn

  • The effect of aflatoxin-contaminated corn on albino mice was investigated using the sperm morphology assay. Blood parameter levels including; total white blood cells (WBC), total red blood cells (RBC), packed cell volume (PCV), serum bilirubin (SB) and fasting blood sugar (FBS) were also determined in the tested mice. Test mice were exposed to aflatoxin-contaminated corn (contamination level of 100 ppb) for $1{\sim}4$ weeks while aflatoxin-free corn and cyclophosphamide were used as negative and positive controls, respectively. Sperm cells showed varieties of morphological abnormality when assessed after 5 weeks. The percentage frequencies of the negative and positive controls were 18.8% and 48.87%, respectively, while the percentage abnormalities for the 1, 2, 3 and 4 weeks exposures were 41.38%, 48.17%, 57.13% and 61.67%, respectively. PCV, WBC, total bilirubin and glucose level values of mice in all concentrations were higher and statistically significant as compared to the negative control values using Dunnett's test. Therefore, abnormal sperm cell induction is concentrationdependent such that continuous consumption of aflatoxin-contaminated corn is capable of negatively affecting spermatogenesis by inducing or increasing the frequency of morphologically abnormal sperm cells produced.

Association of GNB3 Polymorphism with Obesity Index, Blood Pressure, Glucose and Lipid Level (비만한 한국인에서 GNB3 유전자다형성과 비만 지표, 혈압, 혈당, 지질 대사의 관계)

  • Objectives: The aim of this study is to establish the association of GNB3 with obesity-index, blood pressure, glucose and lipid level. Methods: A total of 209 Korean obese subjects participated in the study. Body weight, body mass index(BMI), waist circumference, body fat, body fat mass, resting metabolic rate, blood pressure, total cholesterol, high density lipoprotein, triglyceride and fasting blood sugar test were measured by using standardized protocols. The GNB3 was genotyped by using polymerase chain reaction with restriction fragment length polymorphism. Results: The frequency of CC, CT, TT genotype were, respectively, 18,2%, 56.9%, 24,9%, and the frequency of T allele was 53.3% in Korean obese population. There were no association of GNB3 with obesity index, blood pressure, glucose and lipid level in Korean obese population. Conclusions: Obese Korea population have a higher frequency of T alle than general Korean population. There are no association of GNB3 with obesity index, blood pressure, glucose and lipid level in Korean obese population.

Effect of Case Management Program on Self-Efficacy, Self-Care Behaviors and Glycometabolic Control in Patient with Diabetes (당뇨병 환자의 자기효능감, 자가간호행위 및 혈당조절에 미치는 사례관리 프로그램의 효과)

  • - Abstract - Objectives: We examined the effects of diabetes mellitus (DM) case management program developed by National Health Insurance Corporation in Korea. Methods: One trained nurse performed the case management for 30 DM patients recruited through the reimbursement information of National Health Insurance in a rural community. The case manager had four home visits and two telephone calls for 12 weeks. We evaluated the effectiveness of the program in terms of self-efficacy, self-care behaviors and glyco-metabolic control. Results: The self-efficacy score was significantly increased from 70.0 of pre-intervention to 80.5 of post-intervention (p<0.001). The self-care score was also significantly increased from 2.86 to 3.60 (p<0.001). The scores of its components such as nutritional care, self-test of sugar level, foot care, oral care, and aerobic exercise were significantly improved (p<0.001). However, the improvement of score in medication management was statistically marginal (p=0.071). The fasting blood sugar level and HbA1c level was significantly decreased from 128.6 mg/dl to 114.5 mg/dl (p<0.001) and from 6.78 % to 6.54 % (p<0.001) respectively. Conclusions: The case management program was effective to improve the self-efficacy and self-care behaviors, and to control blood sugar level in DM patients.

Influncences of Litium and Ethanol on the Convulsion and Hyperglycemia Induced with Pircotoxin (Picrotoxin의 경련(痙攣) 및 血糖增加作用(혈당증가작용)에 미치는 Lithium과 Ethanol의 영향(影響))

  • It is well known that picrotoxin, an amaroid substance of Anamirta cocculus, is a classic stimulant on the central nervous system accompanying convulsive activity, and it liberates catecholameine from the adrenal mdulla through its central action to increase blood sugar level. Schou reported that lithium and alcohol have the similar inhibitory property on the $Na^+,\;K^+$-ATPase activity, and recently, the therapeutic efficacies of lithium on the alcohol withdrawal syndrome and the chronic alcoholics have been studied. Many studies about the hypoglycemic effect of lithium and alcohol were reported but the interaction between those hypoglycemic action is little known. Therefore, in this paper, the hypoglycemic effect of lithium and ethanol on the hyperglycemia induced with picrotoxin, and the interaction of them in those hypoglycemic action were investigated with reference to the anticonvulsive action of them. The results were obtained as follows: 1. The convulsive dose (: $CD__{50}$) of picrotoxin in mice was slightly increased by the pretreatment of lithium or ethanol. 2. The blood sugar level was markedly increased by picrotoxin but the level was sugar level was significantly decreased by lithium, ethanol or both. 3. The hyperglycemic effect of picrotoxin was significantly potentiated by the lithium pretreatment, but the potentiation effect of lithium was markedly suppressed by the additional injection of ethanol after lithium injection and more markedly suppressed by the premedication of ethanol before lithium injection 4. The hyperglycemic effect of picrotoxin was markedly inhibited by the ethanol pretreatment, and the inhibitory effect of ethanol was significantly strenthened by the additional injection of lithium after ethanol injection, but on the contrary, the inhibitory effect was completely disappeared by the premedication of lithium before ethanol injection.

Effects of xylooligosaccharide-sugar mixture on glycemic index (GI) and blood glucose response in healthy adults (자일로올리고당을 함유한 설탕이 건강한 성인의 Glycemic Index (GI)와 혈당에 미치는 효과)

  • Purpose: The objective of this study was to investigate the effects of xyloologosaccharide (XOS)-sugar mixture on glycemic index (GI) and blood glucose in human subjects. Methods: Randomized double-blind cross-over studies were conducted to examine the effect of sucrose with 14% xyloologosaccharide powder (Xylo 14) and sucrose with 20% xylooligosaccharide powder (Xylo 20) on GI and postprandial glucose response at 15, 30, 45, 60, 90, and 120 min. Results: GIs of Xylo 14 and Xylo 20 were $60.0{\pm}23.5$ classified within medium GI range, and $54.3{\pm}17.7$ within low GI range, respectively. Xylo 14 and Xylo 20 showed significantly lower area under the glucose curve (AUC) for 0-15 min (p = 0.0113), 0-30 min (p = 0.0004), 0-45 min (p < 0.0001), 0-60 min (p < 0.0001), 0-90 min (p < 0.0001), and 0-120 min (p = 0.0001). In particular, compared with glucose, the blood glucose levels of Xylo 14 and Xylo 20 were significantly lower at every time point between 15 and 120 min. Conclusion: The results of this study suggested that Xylo 14 and Xylo 20 had an acute suppressive effect on GI and the postprandial glucose surge.

ᴅ-Xylose as a sugar complement regulates blood glucose levels by suppressing phosphoenolpyruvate carboxylase (PEPCK) in streptozotocin-nicotinamide-induced diabetic rats and by enhancing glucose uptake in vitro

  • BACKGROUND/OBJECTIVES: Type 2 diabetes (T2D) is more frequently diagnosed and is characterized by hyperglycemia and insulin resistance. $\small{D}$-xylose, a sucrase inhibitor, may be useful as a functional sugar complement to inhibit increases in blood glucose levels. The objective of this study was to investigate the anti-diabetic effects of $\small{D}$-xylose both in vitro and stretpozotocin (STZ)-nicotinamide (NA)-induced models in vivo. MATERIALS/METHODS: Wistar rats were divided into the following groups: (i) normal control; (ii) diabetic control; (iii) diabetic rats supplemented with a diet where 5% of the total sucrose content in the diet was replaced with $\small{D}$-xylose; and (iv) diabetic rats supplemented with a diet where 10% of the total sucrose content in the diet was replaced with $\small{D}$-xylose. These groups were maintained for two weeks. The effects of $\small{D}$-xylose on blood glucose levels were examined using oral glucose tolerance test, insulin secretion assays, histology of liver and pancreas tissues, and analysis of phosphoenolpyruvate carboxylase (PEPCK) expression in liver tissues of a STZ-NA-induced experimental rat model. Levels of glucose uptake and insulin secretion by differentiated C2C12 muscle cells and INS-1 pancreatic ${\beta}$-cells were analyzed. RESULTS: In vivo, $\small{D}$-xylose supplementation significantly reduced fasting serum glucose levels (P < 0.05), it slightly reduced the area under the glucose curve, and increased insulin levels compared to the diabetic controls. $\small{D}$-xylose supplementation enhanced the regeneration of pancreas tissue and improved the arrangement of hepatocytes compared to the diabetic controls. Lower levels of PEPCK were detected in the liver tissues of $\small{D}$-xylose-supplemented rats (P < 0.05). In vitro, both 2-NBDG uptake by C2C12 cells and insulin secretion by INS-1 cells were increased with $\small{D}$-xylose supplementation in a dose-dependent manner compared to treatment with glucose alone. CONCLUSIONS: In this study, $\small{D}$-xylose exerted anti-diabetic effects in vivo by regulating blood glucose levels via regeneration of damaged pancreas and liver tissues and regulation of PEPCK, a key rate-limiting enzyme in the process of gluconeogenesis. In vitro, $\small{D}$-xylose induced the uptake of glucose by muscle cells and the secretion of insulin cells by ${\beta}$-cells. These mechanistic insights will facilitate the development of highly effective strategy for T2D.

An Experimental Study on the Early Feeding of Infants (신생아의 조기수유에 관한 실험적 연구)

  • The newborn human is the only mammalian whose mother does not have a food supply ready for it's offspring at birth. From two to four days usually elapse before the mother's supply If milk appears, and during this period, some kind of artificial feeding should be supplied to the infants. Because of this factor, there has been continued debate fog the past hundreds of years as to when the first feeding should be started Accordingly, many experiments were carried out by scholars and because of these, Pre-lateral feedings were believed to be necessary. Many types of pre-lateral feedings were tried and the conclusion was reached that glucose water was the best food for the first infants'feedings. Traditionally, This has been started 12 hours after birth. The causes for the 12 hours delay were thought to (1) provide rest for the infants: (2) prevent regurgitation ana vomiting which tended to be prevalent during this tine: (3) in cases of low weight infants, prevention of aspiration pneumonia. From recent studies of newborn physiology and as pediatric medicine has been rapidly advancing, many studies hare been carried out concerning the improvement of infant nutrition and the early feeding of infants has been emphasized. This author believes it would be very beneficial to try two different kinds of feedings for the infant. (1) experimental feedings ana (2) comparative feeding, and during this period to investigate and compare the infants blood sugar level, hematocrit, gamma globulin level weight changes and to observe the infant reaction ill order to search for a more desirable feeding program. This study was conducted from January to March 1974 with data related to 40 healthy newborn infants (male 21, female 19: weight, 2.79∼4.20㎏ : gestation, 39∼40 weeks) born at Ewha Womens University Hospital and the results obtained were as follows : 1. At time of birth the blood sugar level from the cord sample averaged 88.99㎎/100㎖, but the blood sugar level rapidly dropped after 2 to 3 hours and reached the lowest point after 10 to 11 hours (54.48㎎/100㎖) and rose again by the 24 hour time period (76.80㎎/100㎖). Changes in the blood sugar level of the experiments: groups and the compare-five group was not significantly different until the 6 to 7 hour period, but by the 10 to 11 hour period the blood sugar levels of the experimental group (49,10㎎/100㎖) and the comparative group (49.70㎎/100㎖) were lower than the remainder of the experimental groups. 9. There ware no significant weight changes between the two groups. Average weight at birth was 3.35㎏, but at the 24 hours period birth weight averaged 3.29㎏. (1.8% reduction of birth weight). It continually lowered until at 48 hours, average weight was 3.26㎏ (2.7% reduction from birth weight.) 3. Hematocrit readings showed no significant difference between the groups. Hematocrit, the average value at birth, was 28.07% and abruptly elevated to average 64.35% at the 2 to 3 hour period, then slowly lowered to an average of 59.67% at the 6 to 7 hour period, 55.10% at the 10 to 11 hour period, ana 53.70% at the 24 hour period. 4. At birth, average gamma globulin value averaged 1,39㎎/100㎖. and at the 24 hour period averaged 1,52㎎/100㎖ revealing no significant difference between the two feeding groups. 5. Such factors as voiding, passing of meconium, regurgitation and vomiting showed no significance between the two feeding groups. However, the number of infants voiding and passing meconium in the experimental groups during the first 12 hours was slightly greater. In general there was an increased tendency for regurgitation and. vomiting among a small group of the infants during the first 24 hours which thereafter decreased. 6. Fluid intake averaged 24.38cc at the first feeding and increased to average 30.48cc at the third feeding and further increased to 73. 00cc at the fifteenth feeding. Finally it was suggested that the most reasonable method of early feeding is to give less than 25cc of 5% glucose water and/or 8% powdered milk at 8 to 9 flours after birth in order to prevent hypoglycemia and dehydration.

A philological study on poisoning of herbal medicines used to activate blood flow and remove blood stasis(活血祛瘀藥) (활혈거어약(活血祛瘀藥)의 독성(毒性)에 관한 문헌적 고찰)

  • Objectives & Method : We investigated toxicity, poisoning symptoms, poisoning treatment and prevention against poisoning of herbal medicines used to activate blood flow and remove blood stasis(活血祛瘀藥) in order to use herbal medicines accurately. Result : Cnidii Rhizoma(川芎), Olibanum(乳香), Myrrha(沒藥), Corydalis Tuber(玄胡索), Zedoariae Rhizoma(莪朮), Salviae Miltiorrhizae Radix(丹參), Polygoni Cuspidati Radix(虎杖根), Leonuri Herba(益母草), Persicae Semen(桃仁), Carthami Flos(紅花), Manitis Squama(穿山甲), Eupolyphaga(蟅蟲), Hirudo(水蛭), Vaccariae Semen(王不留行), Sappan Lignum(蘇木), Lacca Sinica Exsiccata(乾漆), Draconis Resina(血竭) and Leonuri Semen(茺蔚子) may give rise to some side effects or toxic symptoms in herbal medicines used to activate blood flow and remove blood stasis(活血祛瘀藥). The representative methods of poisoning treatment in western medicines are washing out the stomach, promotion of vomiting, causing diarrhea, supplies of grape sugar and symptomatic treatment, etc. The representative methods of poisoning treatment in oriental medicine take advantage of herbs. And Oriental medical doctor should meet symptoms as patients call for attention. In order to prevent against poisoning of herbal medicines used to activate blood flow and remove blood stasis (活血祛瘀藥), the patients should keep usage, dosage and notes and oriental medical doctors should do processing drugs. Conclusion : We should pay attention to clinical using of Cnidii Rhizoma(川芎), Olibanum(乳香), Myrrha(沒藥), Corydalis Tuber(玄胡索), Zedoariae Rhizoma(莪朮), Salviae Miltiorrhizae Radix(丹參), Polygoni Cuspidati Radix(虎杖根), Leonuri Herba(益母草), Persicae Semen(桃仁), Carthami Flos(紅花), Manitis Squama(穿山甲), Eupolyphaga(蟅蟲), Hirudo(水蛭), Vaccariae Semen(王不留行), Sappan Lignum(蘇木), Lacca Sinica Exsiccata(乾漆), Draconis Resina(血竭) and Leonuri Semen(茺蔚子) in herbal medicines used to activate blood flow and remove blood stasis(活血祛瘀藥).

Nutritional Education for the Diabetic Children (A case study) (당뇨병역에 있는 아동의 영양 교육 사례)

  • Since diet is an essential component of the treatment for diabetic mellitus, a progressive educational plan was designed to educate diabetic children for the proper selection of their foods. Seven suspicious children were chosen according to the previous oral glucose tolerance test and present blood glucose levels (fasting and postprandial 2 hours). The education program includes the basic nutrition study, learning of the five basic food groups. familiarization with the food exchange lists. calorie calculation and menu planning, and follow-up evaluation by checking every day -food intake. The duration of the education was five weeks. The levels of fasting blood glucose and postprandial -2 hours blood sugar of the seven children were significantly reduced after the 5-weeks education:FBS:from 92-125mg% to 67-80mg%, pp-2hours BS: from 130-169mg% to 69-90mg%. The children have felt much better with profound self-confidence after to program . Their oral glucose tolerance test levels were also significantly decreased after the 5-weeks education in comparison with those of one year ago.

Factors associated with Obesity among Korean Adolescents based on the Seventh Korea National Health and Nutrition Examination Survey (2016)

  • Purpose: This study was conducted to examine factors associated with obesity among Korean adolescents. Methods: The participants were 436 Korean adolescents aged 12~18 years who took part in the seventh Korea National Health and Nutrition Examination Survey (2016). The data were analyzed by descriptive statistics, the $x^2$ test, one-way analysis of variance, and multinomial logistic regression using complex sample analysis. Results: Among the participants, 9.7% were classified as being obese, and 11.9% as being overweight. Several health parameters, including systolic blood pressure, diastolic blood pressure, waist circumference, and triglyceride, fasting blood sugar, and alanine aminotransferase levels, varied according to obesity. Distorted perceptions of body shape showed a significant negative association with overweight, and carbohydrate intake was negatively associated with obesity. Conclusion: These findings indicate that obesity in Korean adolescents was associated with lifestyle and health parameters. Therefore, intervention programs for obese or overweight adolescents should focus on preventing them from developing health problems.
