Streptozotocin으로 유발한 당뇨동물 모델에서 아프리칸 망고 추출물의 혈당 조절 효과 (Effect of African Mango (Irvingia gabonesis, IGOB 131TM) Extract on Glucose Regulation in STZ-Induced Diabetes)

  • 본 연구는 당뇨유발모델 쥐에서 아프리칸 망고(Irvingia gabonesis, IGOB $131^{TM}$) 추출물의 혈당 조절 효과를 알아보기 위해 SD계 수컷 흰쥐에 IGOB $131^{TM}$ 1%, IGOB $131^{TM}$ 2%(IGOB $131^{TM}$ 353 mg/kg body weight, IGOB $131^{TM}$ 706 mg/kg body weight)와 pinitol을 5주간 매일 강제 경구 투여하였다. 그리고 streptozotocin을 복강 주사하여 당뇨를 유발시킨 후 경구 당부하 검사(OGTT)를 실시하였다. 정상군에 비해 당뇨 유발군(DM, pinitol, IGOB $131^{TM}$ 1%, IGOB $131^{TM}$ 2%)의 체중 증가는 유의적으로 감소하였고, 당뇨 유발군 간의 유의적인 차이는 나타나지 않았다. 경구 당부하 검사 결과에서 DM군이 정상군에 비해 유의적으로 높았고, pinitol군, IGOB $131^{TM}$ 1%, IGOB $131^{TM}$ 2%군은 DM에 비해 각각 39.70%, 33.00%, 29.67% 유의적으로 감소하였다. 혈청 glucose 농도는 정상군에 비해 당뇨 유발군에서 유의적으로 높게 나타났고, pinitol, IGOB $131^{TM}$ 1%, IGOB $131^{TM}$ 2%군은 DM에 비해 유의적으로 감소하였다. 또한 혈청 insulin 농도는 정상군에 비해 당뇨 유발군에서 유의적으로 낮게 나타났고, pinitol, IGOB $131^{TM}$ 1%, IGOB $131^{TM}$ 2%군은 DM에 비해 유의적으로 증가한 것으로 나타났다. 따라서 아프리칸 망고 추출물이 STZ 유도 당뇨쥐의 혈청 insulin 함량을 개선시켜 혈청 glucose를 감소하는 효과가 있는 것으로 사료된다.

고지방·고단순당 식이 섭취 마우스에서 토종보리수 열매의 인슐린 저항성 및 고혈당 개선 효과 (Effects of autumn olive berry on insulin resistance and hyperglycemia in mice fed a high-fat, high-sucrose diet)

  • 토종보리수 열매의 항당뇨 효과를 규명하기 위하여, 마우스를 네 군으로 나누어, 기본 식이, 고지방·고단순당 식이, 토종보리수 열매 추출물을 저농도 (0.5%) 및 고농도 (1.0%)로 첨가한 고지방·고단순당 식이를 12주간 제공하였다. 고지방·고단순당 식이를 섭취한 군은 대조군에 비해 체중 및 체중 증가량, 부고환 지방무게가 유의적으로 증가하였으나, 고농도 보리수 열매 추출물의 급여는 부고환 지방무게를 유의적으로 감소시켰다. 고지방·고단순당군은 대조군에 비해 혈당, 인슐린, HOMA-IR값이 유의적으로 증가하였으나, 고농도 보리수 열매 추출물은 인슐린 농도를 감소시켰고, 저농도 및 고농도 추출물은 혈당 및 HOMA-IR값을 감소시켰다. 고지방·고단순당군은 대조군에 비해 IRS-2 및 AMPK 단백질 발현도가 유의적으로 감소하였으나, 고농도 보리수 열매 추출물의 급여는 IRS-2 발현도를 증가시켰고, 저농도 및 고농도 보리수 열매 추출물의 급여는 AMPK 발현도를 증가시켰다. 따라서, 토종보리수 열매는 제2형 당뇨병 동물에서 인슐린 저항성을 개선시켜 고혈당 개선효과를 나타낸 것으로 나타났다. 고지방·고단순당 식이를 섭취한 마우스에서 저농도 및 고농도 토종보리수 열매 추출물의 급여는 혈청 중성지방 농도 및 간조직의 총 지질과 중성지방 함량을 감소시켰고, 고농도추출물의 급여는 혈청 콜레스테롤 농도를 감소시켜, 토종보리수 열매는 지방간과 이상지질혈증 개선효과를 나타내었다.

한국산 겨우살이 추출물(KME)의 2형 당뇨 억제 및 근육세포 미토콘드리아 생성 증가 효과 (Inhibition of Type II Diabetes in ob/ob Mice and Enhancement of Mitochodrial Biogenesis in C2C12 Myotubes by Korean Mistletoe Extract)

  • 본 연구에서는 C57BL/6J ob/ob 마우스를 이용하여 한국산 겨우살이 냉수 추출물(KME)의 항당뇨 활성을 조사하였다. 50 혹은 100 mg/kg의 KME를 1일 1회씩 경구투여 한 결과 KME 투여 개시 5일 후부터 ob/ob 마우스의 혈당이 유의하게 억제되었으며, 10일 후부터 안정된 억제 효과를 나타내고 대조군에 비해 20% 이상의 혈당강하 효과를 나타내었다. 경구 당부하 실험(OGTT)에서는 KME 경구투여 마우스에서 유효한 당부하 억제 활성이 관찰되었다. 또한 KME 경구 투여는 ob/ob 당뇨 마우스의 혈액 내 총 콜레스테롤과 중성 지질의 농도를 억제하는 것으로 나타났다. 한편 C2C12 근육세포를 이용한 in vitro 실험에서 KME를 처리함으로써 glucose uptake가 현저히 증가하였다. 한편 매우 흥미롭게도 KME를 처리한 C2C12 근육세포에 있어서 미토콘드리아 생성과 산화대사 조절물질인 peroxisome proliferator-activated receptor gamma coactivator 1-${\alpha}(PGC-1{\alpha})$를 비롯하여 glucose transporter type 4(GLUT4), estrogen-related receptor-${\alpha}(ERR-{\alpha})$, nuclear respiratory factor-1(NRF-1) 그리고 mitochondrial transcription factor A(TmfA)와 같은 $PGC-1{\alpha}$ 관련 유전자들의 발현이 증가하는 것으로 확인되었다. 이 결과는 KME가 2형 당뇨에 대한 치료물질로서의 작용을 지니며 이러한 KME의 항당뇨 활성은 미토콘드리아 생성의 조절과 관련 있는 것으로 추정된다.

야관청혈탕이 고지방식이로 유도된 흰쥐의 비만 억제에 미치는 영향 (Effect of Yagwan-cheunghyeoltang on Obesity in Rats Induced by High Fat Diet)

  • Background : Obesity, the syndrome caused by a high fat diet, is a disease. At the same time, obesity causes diabetes mellitus, hyperlipidemia, and cardiovascular disease. Recently, its prevalence rate is increasing. Yagwan-cheunghyeoltang (YCT) used in this experiment is the prescription of Yagwanmoon added to Cheunghyeol-tang which is reported to be very effective in weight loss controlling and serum cholesterol. It is also reported that Yagwanmoon has significant antioxidant effects and YCT has a significant effect on blood glucose control. Objectives : This study was conducted to experimentally evaluate the effects of YCT on obesity in rats induced by high fat diet. Methods : The experiment was conducted with 4-week-old male rat s divided into 5 groups. They were a normal diet group, a high fat diet group, a positive drug control group, a 1% YCT group, and a YCT 3% group, and were tested for eight weeks. After four weeks of inducing obesity by a high fat diet, rats were allowed to lose weight by following the normal diet group, approximately 30% compared with 10 rats in each group were determined as still obese. Changes in body weight and organ weight and serum cholesterol, triglyceride, glucose-density, low density lipoprotein cholesterol, antioxidant activity were checked. Results : In the experimental groups, we observed weight loss and visceral fat reduction, improvement of liver function, reduction of serum glucose, activation of HMG-CoA reductase inhibitor, reduction of concentrations of leptin and it showed a significant effect on antioxidants and lipid peroxidation. Conclusions : YCT has significant effects on the regulation of hyperlipidemia and lipid peroxidation associated with obesity and has significant effects on, antioxidants and lipid peroxidation, too. Additional clinical studies are needed.

Association Between Trp64arg Polymorphism of the β3 adrenoreceptor Gene and Female Sex in Obese Turkish Children and Adolescents

  • Purpose: The ${\beta}3-adrenergic$ receptor (ADRB3) is expressed in visceral adipose tissue and has been speculated to contribute to lipolysis, energy metabolism, and regulation of the metabolic rate. In this study, we aimed to investigate the association of polymorphism of the ADRB3 gene with the sex of children with obesity and related pathologies. Methods: ADRB3 gene trp64arg genotyping was conducted in 441 children aged 6-18 years. Among these subjects, 264 were obese (103 boys; 161 girls) and 179 were of normal weight (81 boys; 98 girls). In the obese group, fasting lipids, glucose and insulin levels, and blood pressure were measured. Metabolic syndrome (MS) was defined according to the modified World Health Organization criteria adapted for children. Results: The frequency of trp64arg genotype was similar in obese and normal weight children. In obese children, serum lipid, glucose, and insulin levels; homeostasis model assessment of insulin resistance (HOMA-IR) scores; and MS were not different between arg allele carriers (trp64arg) and noncarriers (trp64trp). In 264 obese children, genetic analysis results revealed that the arg allele carriers were significantly higher in girls than in boys (p=0.001). In the normal weight group, no statistically significant difference was found between genotypes of boys and girls (p=0.771). Conclusion: Trp64arg polymorphism of the ADRB3 gene was not associated with obesity and MS in Turkish children and adolescents. Although no relationships were observed between the genotypes and lipids, glucose/insulin levels, or HOMA-IR, the presence of trp64arg variant was frequent in obese girls, which can lead to weight gain as well as difficulty in losing weight in women.

창출·지모·육계 복합추출물의 고지방식이 유도 당뇨병 마우스에서의 항당뇨 효능 및 C2C12 골격근세포에서의 조절기전 연구 (Anti-diabetic effects of the extract from Atractylodes lancea, Anemarrhena asphodeloides and Cinnamomum Cassia mixture in high fat diet-induced diabetic mice and regulation of the function in C2C12 mouse skeletal muscle cells)

  • Objective : This study investigated the anti-diabetic effects of DM1, a herbal mixture with Atractylodis Rhizoma, Anemarrhenae Rhizoma, and Cinnamomi Cortex in high fat diet (HFD)-induced diabetic mice and the mechanism in C2C12 mouse skeletal muscle cells. Methods : The C57B/6 mice were fed high fat for 12 weeks, and then administrated DM1 extract (500 mg/kg, p.o.) for 4 weeks. The changes of body weight, calorie and water intakes, fasting blood glucose levels and the serum levels of glucose, insulin, triglyceride, HDL-cholesterol, AST and ALT were measured in mice. The histological changes of liver and pancreas tissues were also observed by H&E stain. C2C12 myoblasts were differentiated into myotubes and then treated with DM1 extract (0.5, 1, and 2 mg/㎖) for 24 hr. The expression of myosin heavy chain (MHC), PGC1α, Sirt1 and NRF1, and the AMPK phosphorylation were determined in the myotubes by western blot, respectively. Results : The DM1 extract administration significantly decreased the calorie and water intakes, glucose, triglyceride, AST and ALT levels and increased insulin and HDL-cholesterol in HFD-induced diabetic mice. DM1 extract inhibited lipid accumulation in liver tissue and improved glucose tolerance. In C2C12 myotubes, DM1 treatment increased the expression of MHC, PGC1α, Sirt-1, NRF-1 and the AMPK phosphorylation. Conclusion : In our results indicate that DM1 can improve diabetic symptoms by decreasing the obesity, glucose tolerance and fatty liver in HFD-induced diabetic mice, and responsible mechanism is might be related with energy enhancement.

Epigallocatechin Gallate Prevents Autoimmune Diabetes Induced by Multiple Low Doses of Streptozotocin in Mice

  • Cytokines produced by immune cells infiltrating pancreatic islets have been incriminated as important mediators of $\beta$-cell destruction in insulin-dependent diabetes mellitus. In non insulin-dependent diabetes, cytokines are also associated with impaired $\beta$-cell function in high glucose condition. By the screening of various natural products blocking $\beta$-cell destruction, we have recently found that epigallocatechin gallate (EGCG) can prevent the in vitro destruction of RINm5F cell, an insulinoma cell line, that is induced by cytokines. In that study we suggested that EGCG could prevent cytokine-induced $\beta$-cell destruction by down-regulation of nitric oxide synthase (NOS) through inhibition of NF-kB activation. Here, to verify the in vivo antidiabetogenic effect of EGCG, we examined the possibility that EGCG could also prevent the experimental autoimmune diabetes induced by the treatment of multiple low doses of streptozotocin (MLD-STZ), which is recognized as an inducer of type I autoimmune diabetes. Administration of EGCG (100 mg/day/kg for 10 days) during the MLD-STZ induction of diabetes reduced the increase of blood glucose levels caused by MLD-STZ. Ex vivo analysis of $\beta$-islets showed that EGCG downregulates the MLD-STZ-induced expression of inducible NOS (iNOS). In addition, morphological examination showed that EGCG treatment ameliorated the decrease of islet mass induced by MLD-STZ. In combination these results suggest that EGCG could prevent the onset of MLD-STZ-induced diabetes by protecting pancreatic islets. Our results therefore revealed the possible therapeutic value of EGCG for the prevention of diabetes mellitus progression.

Diabetes Mellitus as a Risk Factor for High Grade Renal Cell Carcinoma

  • Background: Diabetes is a chronic disease characterized by impaired fasting blood glucose that leads to disturbances in various organs. In this study, we evaluated relationships between tumor size and grade in a population of diabetic and non-diabetic patients with renal cell carcinoma. Materials and Methods: Between 2007-2013, in our clinic radical nephrectomy performed to 310 patients for renal tumors and pathology reported renal cell carcinoma cases were enrolled in the study. Patients with and without a history of diabetes regarding fasting glucose and HgA1c levels were evaluated during surgery for tumor size and Fuhrman grade. Results: Diabetes was found in 95 patients. The mean age of the patients with and without diabetes mellitus was 64.3 (40-79) and 58.4 (31-87) years, respectively. In the diabetes group 51% of patients had a tumor size over 7 cm and 54% a tumor grade over Fuhrman 3. The respective figures in the non-diabetes group were 35% and 30% (p<0.05 in both cases). Conclusions: Renal cancer appears more aggressive in patients with diabetes. In this study lifestyle and risk factors with diabetes regulation were observed to be important for renal cancer patients. Multicenter studies are needed in larger series for more accurate results.

Anti-diabetic Effect and Mechanism of Korean Red Ginseng in C57BL/KsJ db/db Mice

  • The present study was designed to investigate the anti-diabetic effect and mechanism of Korean red ginseng in C57BL/KsJ db/db mice. The db/db mice were divided into three groups: diabetic control group (DC), Korean red ginseng group (KRG, 100 mg/kg) and metformin group (MET, 300 mg/kg), and treated with drugs once per day for 10 weeks. Compared to the DC group, fasting blood glucose levels were decreased by 19.8% in KRG-, 67.7% in MET-treated group. With decreased plasma glucose and insulin levels, the insulin resistance index of the KRG-treated group was reduced by 27.6% compared to the DC group. The HbA1c levels in KRG and MET-treated groups were also decreased by 11.0% and 18.9% compared to that of DC group, respectively. Plasma triglyceride and non-esterified fatty acid levels were decreased by 18.8% and 16.8%, respectively, and plasma adiponectin and leptin levels were increased by 20.6% and 12.1%, respectively, in the KRG-treated group compared to those in DC group. Histological analyses of the liver and fat tissue of mice treated with KRG revealed significantly decreased number of lipid droplets and decreased size of adipocytes compared to the DC group. From the pancreatic islet double-immunofluorescence staining, we observed KRG has increased insulin contents, but decreased glucagon production. To elucidate action mechanism of KRG, effects on AMP-activated protein kinase (AMPK) and its downstream target proteins responsible for fatty acid oxidation and gluconeogenesis were explored in the liver. KRG activated AMPK and acetyl-coA carboxylase (ACC) phosphorylations, resulting in stimulation of fatty acid oxidation. KRG also caused to down regulation of SREBP1a and its target gene expressions such as FAS, SCD1 and GPAT. In summary, our results suggest that KRG exerted the anti-diabetic effect through AMPK activation in the liver of db/db mice.

키토산과 세리신 및 콜라겐 펩타이드 추출 혼합물이 이상지질혈증의 지질대사 개선에 미치는 영향 (Improvements Caused by Chitosan, Sericin and Collagen Peptide Extract Complexes on Lipid Metabolism in Dyslipidemia)

  • The objective of this study was to assess improvements caused by chitosan, sericin and collagen peptide extract complexes (1:1:1, w/w/w, CSC-F-005) in lipid concentrations in the sera of dyslipidemic rats (SD strain) fed on experimental diets for 5 weeks. Serum concentrations of total cholesterol, HDL-cholesterol, ratio of HDL-cholesterol concentration to total cholesterol, atherosclerotic index, LDL-cholesterol, free cholesterol, cholesteryl ester, triglyceride, phospholipid and blood glucose were effective on the metabolic regulation of dyslipidemic rats. The activities of alkaline phosphatase, aspartate aminotransferase and alanine aminotransferase in serum were remarkably lower in the extract complexes (CSC-F-005) than in the dyslipidemic model. From the above results shows that CSC-F-005 extract complexes were effective on the improvement of the lipid metabolism in sera of dyslipidemic rats.
