Changes of Chemical Components During Seed Development in Black Soybean (Glycine max L.)

  • Changes in the level of metabolites in leaves and pods were examined with respect to the seed chemical composition in black soybean. There was no further increase in pod length after 42 days after flowering (DAF). Pod weight, however, persistently increase until 73 DAF, thereafter the weight was slightly lowered. The seed storage protein, however, increased drastically as the increasing rate of pod weight was lessened at 61 DAF. The accumulation of seed storage proteins was occurred conspicuously as the increasing rate of pod weight was slowed down. The chlorophyll content both in leaves and pods was drastically decreased after 50 DAF. The beginning of drastic reduction in chlorophyll content was occurred concomitantly with the reduction of soluble protein content in leaves. The sugar content in leaves showed similar tendency with chlorophyll and soluble protein content. The starch level in leaves, however, showed different changing pattern during seed development. The starch content in leaves was increased persistently until 66 DAF, thereafter the content was decreased drastically to about $55\%$ of maximal value at 66 DAF. Total phenolics content in leaves and the anthocyanins content in seeds were stable without noticeable increase until 66 DAF. The contents were increased dramatically after 66 DAF showing the synchronized pattern with the decrease in starch level in leaves. The levels of the selected metabolites in leaf and seed suggested that the accumulation of chemical components of black soybean seed is launched actively at 66 DAF. The profile of storage proteins was nearly completed at 61 DAF because there was no large difference in densitometric intensity among protein subunits after 61 DAF. In soybean, chemical maturation of seed begins around 61 to 66 DAF at which most metabolites in vegetative parts are decreased and remobilized into maturing seeds.

시중에 유통되는 콩의 종류에 따른 품질 특성 (Quality Characteristics of Various Beans in Distribution)

  • 콩의 일반성분 분석에서 조지방은 백태 17.60${\pm}$0.14%, 조단백질은 서목태 42.38${\pm}$0.15%로 가장 높고, 조회분의 경우 비슷한 값을 나타내었다. 색도는 L값 작두콩 64.07${\pm}$0.97, a값 강낭콩 15.64${\pm}$0.48, b값 백태 22.92${\pm}$0.09로 각각 가장 높은값을 나타내었다. 유리당 중 fructose, glucose, maltose는 서목태가 각각 13.92 mg/g, 20.52 mg/g, 2.61 mg/g으로 가장 높은 함유량을 보였으며, sucrose는 서리태가 54.23 mg/g으로 가장 높은 함유량을 보였다. 유기산 중 oxalic acid, malic acid, tartaric acid는 각각 서목태가 23.26 mg/100 g, 18.24 mg/100g, 23.6 mg/100 g으로 가장 높은 함유량을 보였으며, citric acid의 경우 청태가 7.22 mg/100 g으로 가장 높은 함유량을 보였다. 콩의 종류에 따라 각각 함유 성분에 차이가 있었다. 구성아미노산은 glutamic acid가 2.68~6.18 g/100 g으로 가장 많이 함유하고 있으며, 그 중 서리태가 가장 많이 함유하고 있는 것으로 나타났다. 콩의 무기질은 K, Mg이 가장 많이 함유되어 있고, Co, Cu, Zn은 미량 함유되어 있는 것으로 나타났다. 이상의 결과에서 콩의 품종에 따른 차이와 동일한 품종일지라도 재배 및 환경 요인, 건조 방법, 건조 상태에 따라 많은 영향을 받는 것으로 생각된다.

발아와 고압처리가 검정콩 사포닌 추출물의 암세포주 증식억제에 미치는 영향 (Influence of High Hydrostatic Pressure Treatment after Germination on Anti-proliferation Effects of Soyasaponin-rich Fraction in Black Soybean (Glycine max L.))

  • The objective of this study was to determine the effect of high hydrostatic pressure (HHP) treatment on proliferation of human cancer cell lines (MCF-7, HCT-116, PC-3 and AGS) of crude soyasaponin extracts in germinated black soybean. Black soybean was germinated and subjected to HHP, followed by preparation of crude soyasaponin extracts. Cell treatments done with extracts less than $400{\mu}g/mL$ concentrations had no significant effect on 3T3-L1 adipocyte cell viability. The inhibitory effect of crude soyasaponin extracts with germination periods and applied pressure on breast cancer cell (MCF-7), human colon cancer cell (HCT-116), human gastriccancer cell (AGS) and prostate cancer cell (PC-3) growth were investigated using MTT assay. The highest anti-proliferation of human cancer cell line of crude soyasaponin extracts was observed at 150 MPa treatment after germination for 4 days (150 MPa-Day 4). The cell viability on MCF-7, HCT-116, PC-3 and AGS cell lines of crude soyasaponin extract in 150 MPa-Day4 was 48.82%, 57.37%, 39.89% and 23.94% at $400{\mu}g/mL$, respectively. These results suggest that soyasaponin extracts from black soybean subjected to HHP after germination may mediate physiological activity.

Variation of Major Isoflavone Contents in Soybeans (Glycine max (L.) Merill.) of Collection Area, Diversity, and Seed Coat Color

  • Soybean seeds are important sources of non-nutritive health promoting metabolites. The contents of these metabolites are affected by both genetic and environmental factors. In the present study, the contents of five common isoflavones including daidzin, genistin, glycitin, malonyldaidzin, and malonylgenistin were analyzed in 72 soybeans of different seed coat colors, diversity, and of different origins including China (22), Japan (9), USA (12), India (4), and Korea (25). The average total isoflavone content (TIC) was maximum in Indian soybeans (3302.36 ㎍/g) and minimum in Chinese landraces (1214.95 ㎍/g). The Korean landraces had higher average TIC (2148.05 ㎍/g) than the USA genetic materials (1580.23 ㎍/g) and Japanese landraces (1485.99 ㎍/g). The content of malonylgenistin was in the range of 54.31 - 2385.68 ㎍/g in the entire population, and was the most abundant isoflavone irrespective of origin although there was content variation among individual soybeans. Besides, glycitin was the least concentrated isoflavone, and its content ranged from 0.00 to 79.79 ㎍/g. With respect to seed coat color, green soybeans from all countries displayed the maximum malonylgenistin and TIC contents. Exceptions were those of Korean and Indian origins where black and yellowish-green soybeans presented the highest malonylgenistin and TIC contents, respectively. In multivariate analysis, 92.72% of the variance was explained by the first two principal components, and the soybeans were grouped in to three clusters based on isoflavone contents. Overall, our findings signify the importance of seed coat color and origin as discriminant parameters, and provide wide spectrum of routes for breeding soybean cultivars.

중북부지역에서 재배한 소립 검정콩의 품질 및 이화학 특성 (Quality and Physicochemical Characteristics of Small Black Soybean Cultivar Cultivated in the North-central Region)

  • Proximate compositions, quality and physicochemical characteristics of small black soybean cultivar, cultivated in the north-central region of South Korea with different seeding periods, were evaluated. Proximate compositions, chromaticity, water binding capacity, water solubility index, swelling power, and antioxidant properties were significantly different among cultivars and different seeding periods. Moisture, crude ash, fat, protein, and carbohydrate contents of small black soybean cultivar were 5.53~6.69, 5.47~6.54, 15.38~19.14, 34.17~40.26, and 32.04~36.85 g/100 g, respectively. Lightness, redness and yellowness were 35.60~38.61, -0.02~0.07 and -0.56~-0.13, and water binding capacity, water solubility index and swelling power were 84.48~148.31, 46.65~54.89 and 29.87~35.12%, respectively. Total polyphenol contents of first, second, and third seedings on small black soybean cultivar were 10.40~15.48, 9.86~14.85 and 8.61~15.39 mg GAE/g; total flavonoid contents were 5.81~7.25, 5.81~7.34 and 5.52~7.64 mg CE/g, respectively. DPPH radical scavenging activity was 4.55~7.86, 3.99~8.79, and 3.74~9.43 mg TE/g, and ABTS radical scavenging activity was 9.32~12.90, 8.64~13.39, and 8.51~14.35 mg TE/g, respectively. Phenol compound of Tawonkong and Socheong cultivars decreased with delay of seeding periods. Radical scavenging activity of Socheong and Jununi cultivars decreased with delay of seeding periods, but Socheong 2 and Socheongja cultivars increased. In the study, phenol compound and radical scavenging activity of small black soybean cultivar were different, depending on cultivars and seeding periods.

검정콩 껍질 유래 안토시아닌의 RBL-2H3 세포에서 탈과립화와 사이토카인 생성 저해 효과 (Inhibitory Effects of Anthocyanins Isolated from Black Soybean (Glycine max L.) Seed Coat on Degranulation and Cytokine Generation in RBL-2H3 Cells)

  • 안토시아닌(anthocyanins)은 플라보노이드에 속하고 항산화 활성들을 포함한 그들의 다양한 건강 유익성에 대해 알려져 있다. 그들 중 검정콩 껍질에서 분리한 주요 안토시아닌은 glycopyranose를 함유하고 있는 배당체이다. 천식은 호염기성 세포(basophils)와 비만세포(mast cells)를 포함한 다양한 면역세포와 관련된 알레르기 관련 질병이다. 호산구(eosinophils), 호염기성 세포, 비만세포는 탈과립화에 의한 천식-특이적 보조 2(T-helper 2) 사이토카인 분비 그리고 계속해서 일어나는 증폭과 같은 염증을 일으키는 매개체의 증가를 통해 알레르기 천식(allergic asthma)에서 중요한 역할을 한다. 흰쥐 호염기성 백혈병(rat basophilic leukemia) RBL-2H3 세포는 알레르기 반응을 측정하기 위해 가장 일반적으로 사용되는 in vitro 모델이다. 본 연구에서는 검정콩 껍질 안토시아닌이 항원으로 자극한 RBL-2H3 세포에서 탈 과립 그리고 Th2 사이토카인 생산에 미치는 효과를 조사하였다. 세포 탈과립은 ${\beta}$-hexosaminidase의 방출을 검출함으로써 평가하였다. IgE-항원 복합체로 자극한 RBL-2H3 세포내 ${\beta}$-hexosaminidase 방출과 Th2 사이토카인 생산이 무처리군의 그것과 비교하여 더 높았다. 안토시아닌은 RBL-2H3 세포의 IgE-항원 복합체 유도 탈과립을 현저하게 억제시켰고 RBL-2H3 세포내 IgE-항원 복합체-매개체 interleukin(IL)-4, IL-13 그리고 TNF-${\alpha}$ 생산을 저해하였다. 이러한 결과는 검정콩 껍질 유래 안토시아닌이 알레르기 반응(allergic reaction)을 현저히 저해하는 효과가 있음을 보였고, 이들 안토시아닌이 향후 알레르기 천식을 억제하거나 개선하는데 유용한 물질로 사용될 가능성이 있을 것으로 판단되었다.

Changes of Phenol Compounds according to Storing Years in Soybean

  • The objective of this study was to determine the role of storing years with the variation of total phenol and individual phenolic compounds in soybean (Glycine max L.) seeds. The total phenol content varied from 0.36 to 0.42% over four years, with the highest value (0.42%) found at storage for two and three years. Among the nine soybean varieties examined, Daweonkong had the highest total average phenol content (0.58%). The total content of 11 phenolics varied from 730.0 to 1812.8 $\mu\textrm{g}\;\textrm{g}^{-1}$ over storage for four years, and the highest concentration (1812.8 $\mu\textrm{g}\;\textrm{g}^{-1}$) was found at storage for two years. Myeongjunamulkong (1465.4 $\mu\textrm{g}\;\textrm{g}^{-1}$) had the highest mean content among the nine soybean varieties. The total content of 11 phenolic compounds measured in this study occupied from 20.96 to 47.73% of the total phenol contents. The highest total phenol contents were in seeds with black coats (5279.4 $\mu\textrm{g}\;\textrm{g}^{-1}$), while the highest concentration of individual phenolic compounds were in seeds with green coats (1419.5 $\mu\textrm{g}\;\textrm{g}^{-1}$). Our study suggests that it may be feasible to improve soybean varieties with high functional substances such as phenolic compounds.

남부지역 논에서 파종 시기별 검정콩의 종실 생산량 및 품질 특성 (Influence of Sowing Date on Seed Yield and Quality of Black Soybean (Glycine max (L.) Merrill cv. C heongja-3ho) in the Southern Paddy Field)

  • 본 연구는 남부지역 논에서 파종 시기를 달리하여 검정콩(청자3호)을 재배하였을 때 생육 및 종실 생산성 등을 조사하여 최근의 기후변화와 소비 트랜드, 그리고 논을 이용한 2모작 콩 재배의 확대 등에 대처하기 위하여 수행되었다. 남부지역에서 검정콩 파종 시기는 지상부 생육, 종실 수량, 유용 성분 등과 유의미한 상관을 보였다. 5월 파종 시에는 영양생장이 활발하여 지상부가 무성해지나 생식생장이 부진하여 수확량이 크게 감소하였다. 반면에 6월과 7월 파종에서는 생식생장이 활발하여 종실 수량이 200 kg/10a 이상으로 많고, 비립 꼬투리와 종자 표면의 열피 발생 또한 감소하였다. 그리고, 파종 시기가 늦어질수록 이소플라본 함량이 크게 증가하였다. 따라서 검정콩 파종은 6월 상순부터 7월 상순까지가 적절한 것으로 보인다. 그렇지만, 2모작 작부체계 시 동계작물의 파종 시기와 겹치는 것을 피하기 위해서는 남부지역에서 검정콩의 파종은 6월 하순이 최적시기로 보인다.

Anthocyanin Extracts from Black Soybean (Glycine max L.) Protect Human Glial Cells Against Oxygen-Glucose Deprivation by Promoting Autophagy

  • Anthocyanins have received growing attention as dietary antioxidants for the prevention of oxidative damage. Astrocytes, which are specialized glial cells, exert numerous essential, complex functions in both healthy and diseased central nervous system (CNS) through a process known as reactive astrogilosis. Therefore, the maintenance of glial cell viability may be important because of its role as a key modulator of neuropathological events. The aim of this study was to investigate the effect of anthocyanin on the survival of glial cells exposed to oxidative stress. Our results demonstrated that anthocyanin extracts from black soybean increased survival of U87 glioma cells in a dose dependent manner upon oxygen-glucose deprivation (OGD), accompanied by decrease levels of reactive oxygen species (ROS). While treatment cells with anthocyanin extracts or OGD stress individually activated autophagy induction, the effect was signifi cantly augmented by pretreatment cells with anthocyanin extracts prior to OGD. The contribution of autophagy induction to the protective effects of anthocyanin was verifi ed by the observation that silencing the Atg5 expression, an essential regulator of autophagy induction, reversed the cytoprotective effect of anthocyanin extracts against OGD stress. Treatment of U87 cells with rapamycin, an autophagy inducer, increased cell survival upon OGD stress comparable to anthocyanin, indicating that autophagy functions as a survival mechanism against oxidative stress-induced cytotoxicity in glial cells. Our results, therefore, provide a rationale for the use of anthocyanin as a preventive agent for brain dysfunction caused by oxidative damage, such as a stroke.
