Formalin, 37% formaldehyde, has been employed as a chemical for controlling ectoparasites and aquatic fungi responsible for infectious fish of diseases in hatcheries and culture facilities (Roberts, 1978; Schnick, 1991; Rach et al., 1997). Regarding the commercial usage of formaldehyde in the aquaculture industry, the U.S. Food and Drug Administration (FDA) approved three commercial products as parasiticides in a species-specific manner: Paracide-F and Formalin-F for bluegill, catfish, largemouth bass, salmon, and trout and Parasite-S for all finfish (FDA, 1998). Withdrawal time for these products was legally zero when used as permitted under the regulations. With the increased production of cultured fish in Korea, such as olive Hounder Paralichthys olivaceus and black rockfish Sebastes schlegeli, application of formalin to diseased fish has become more frequent. Moreover, there is still some concern about environmental exposures caused by effluents from fish culture facilities. The purpose of the present study was to evaluate residues in fish resulting from therapeutic usage of formalin in the aquaculture industry and to document the rate of disappearance of formaldehyde in seawater treated with formalin. We constructed a black rockfish (Sebastes schlegeli) leukocyte cDNA library and a total of 386 expressed sequence tag (EST) clones were generated. Gene annotation procedures and homology searches of the sequenced ESTs were locally done by BLASTX for amino acid similarity comparisons. Of the 386 EST clones, 199 different ESTs showed significant homology to previously described genes while 97 ESTs were unidentified, hypothetical, or unnamed proteins. Encoding 38 different sequences were identified as putative bio-defense genes or genes associated with immune response.
In this study, we evaluated the efficiency of chemical marking of black rockfish scales by immersion in oxytetracycline hydrochloride (OTC, 500 ppm), alizarin red S (AR, 250 ppm) and calcein (CARL, 250 ppm) diluted rearing water. Immersion treatment of chemicals had no effects on both mortality and growth of black rockfish. Marking sucess was 100% in all treatment durations (24, 48 and 72 hours) with three chemicals and marking quality was higher in 48 and 72 hours than 24 hours treatment. Marking retention rates at 24 weeks after treatment were 100% in OTC and CAL treated group, but marking quality was higher in CAL treated group (brilliant 92%, bright 8% and dim 0%) than in OTC treated group (brilliant 4%, bright 70% and dim 26%). AR treated group had lower marking retention rates and marking quality than OTC and CAL treated group. As a results, immersion treatment with OTC and CAL was effective in marking scales of black rockfish and practical in releasing program and other studies requires same rearing environment.
Vibrio harveyi, one of the major causal agent of vibriosis, affects a diverse range of marine vertebrates and invertebrates over a wide geographical area. The aims of this study were to investigate the characteristics of extracellular products (ECPs) from an isolate of the pathogenic non‐luminous V. harveyi and the luminous V. harveyi. And ECPs of V. harveyi were examined the pathogenicity to the black rockfish, Sebastes schlegeli, and histopathological traits of internal organs injected by ECPs. Four strains of V. harveyi cultures produced ECPs showing various enzymatic activites (caseinase, gelatinase, phospholipase, lipase, haemolysin). The ECPs showed strong cytotoxicity on macrophages of black rockfish, Sebastes schlegeli and olive flounder, Paralichthys olivaceus. Especially, the virulence of ECPs from the isolate of V. harveyi FR 2 was higher in the intraperitoneally injected black rockfish. Also, the ECPs of V. harveyi FR 2 caused the expansion of sinusoids in the liver, the activation of ellipsoid in the spleen and the sloughing of the epidermal cell in the intestine. It was suggested that the ECPs from V. harveyi play an important role in the pathogenicity process of the V. harveyi.
Seventeen fish species were counted in an underwater visual census, and the most common species, Sebastes inermis and Halichoeres poecilopterus, were released. More than 60% of the released S.inermis occurred near a fish farm 5 days after release, but they moved to a distant rocky area, and decreasing numbers were seen near the farm 90 days after release. In pots, a total of 722 individuals belonging to 43 species occurred, and released S.inermis dominated with 174 individuals recaptured during the study period. The other common fish species were Stephanolepis cirrhifer and Acanthopagrus schlegeli. The percentage of recapture was 0.23%, but declined to 0.12% at 90 days after release. The potential predators of released S.inermis included Lateolabrax japonicus and Sebastes schlegeli.
Bactericidal/permeability-increasing protein (BPI) and lipopolysaccharide-binding protein (LBP) are important components of the mammalian innate defence system against Gram-negative infections. The BPI/LBP cDNA was identified from the black rockfish ConA/PMA or LPS stimulated leukocyte cDNA library. The full-length BR-BPI/LBP cDNA was 2118 bp long and contained an open reading frame (ORF) of 1422 bp that encoded 473 amino-acid residues. The 5' UTR had a length of 57 bp, and the 3' UTR 639 bp. The molecular weight and theoretical isoelectric point (pI) values were calculated 51.4 kDa and 9.72, respectively. Compared with other known BPI or BPI/LBP peptide sequences, the most conserved regions of the black rockfish BPI/LBP peptide were found to be the BPI1 N-terminal, BPI2 C-terminal domains and a LPS binding domain. Phylogenetic analysis based on the deduced amino acid sequence revealed a homologous relationship between the BPI/LBP sequence of black rockfish and that of other teleosts. The black rockfish BPI/LBP gene was predominantly expressed in the PBLs, head kidney, trunk kidney and spleen. The expression of the black rockfish BPI/LBP molecule was induced in the peripheral blood leukocytes (PBLs) from 1 to 24 h following LPS stimulation, with a peak at 12 h post-stimulation.
조피볼락(Sebastes schlegeli) 소형어와 중형어를 대상으로 MS-222와 lidocaine-HCl 마취농도에 따른 마취 및 회복시간과 혈액생리학적 반응, 생존율 등을 조사하여 마취효과를 비교하였다. 마취농도에 따른 마취시간은 MS-222와 lidocaule-HCl 모두에서 마취농도가 증가함에 따라 마취시간이 유의하게 감소하였다. 어체크기에 따른 마취시간은 소형어(평균전장 18.2$\pm$1.0cm, 평균체중 100.6$\pm$21.1 g)보다는 중형어(평균전장 14.2$\pm$0.9 cm, 평균체중 51.3$\pm$10.5 g)에서 길었다. 그러나 회복시간은 MS-222와 lidocaine-HCl 모두에서 농도가 증가함에 따라 회복시간이 길어졌다. 회복시간은 소형어보다는 중형어에서 더욱 짧았다. 마취농도에 바른 cortisol 농도는 MS-222 300 ppm에서 마취 6시간후 96.1$\pm$12.1 ng/ml로 가장 높은 값을 나타내었으나, 실험종료시까지 모든 실험구에서 실험 개시전 보다 높은 수준을 보였다. Lidocaine-HCl도 모든 실험구에서 실험 종료시까지 실험개시전보다 높은 값을 나타냈다. Glucose 농도는 소형어의 경우 MS-222 마취 1시간 후 $30.3{\pm}15.4{\~}69.5{\pm}23.2 mg/dl$로 다소 높았으나 마취 6시간 후에는 유의한 차이를 보이지 않았다. 중형어는 소형어 보다 다소 높은 농도를 나타내었으나 12시간 후에는 실험 개시전 수준으로 회복되었다. Lidocaine-HCl 경우는 MS-222와 비슷한 경향을 보였다. Na+ 농도는 모든 실험구에서 실험개시전과 차이를 나타내지 않았다. An epizootic causing mortality among cultured marine finfishes occurred in 1999 in the province of Kyoungsang, Korea. The disease was characterized by the presence of enterocele, abdominal swelling, and gastroenteritis. The causative bacteria were isolated from olive flounder (Paralichthys olivaceus), black rockfish (Sebastes schlegeli), turbot (Scophthalmus maximus) and the rearing water. These bacteria showed swarming activity on agar plates and yellowish or greenish colonies on thiosulfate-citrate-bile salts-sucrose (TCBS) agar plates, but no luminescence. The pathogen was identified as Vibrio harveyi based on morphological and biochemical characteristics and the sequence of l6S rDNA. The lethal doses (LD$_{50}$) of olive flounder and black rockfish were estimated to be $1.24\times10^6-1.36\times10^8$ and $3.24\times10^5-5.8\times10^7$ CFU/fish respectively following intraperitoneal injection.
