복분자 열매(Rubus coreanus Miquel)의 항산화 활성 및 생리활성물질 (Antioxidant Activity and Effective Compounds of Black Raspberry (Rubus coreanus Miquel) Extracted by Different Solvents)

  • The black raspberry (Rubus coreanus Miquel) contains anthocyanin, tannins, gallotannin, gallic acid, ferulic acid and phenolics. It brightens the eyes and protects the liver and kidneys. It was effective for anti-aging. Thus, the purpose of this study was to inform the excellence of black raspberry and to screen antioxidant activity to ensure the possibility as a functional material. In this study, bioactive compounds in black raspberry were determined. Additionally, black raspberry was extracted by CM (chloroform:methanol, 2:1, v/v), 70% methanol and 70% ethanol, and were investigated and compared in vitro methods. Total phenol and flavonoid contents were measured to compare each different solvents. In addition, DPPH radical scavenging activity and ABTS radical scavenging activity, ferric reducing antioxidant power, reducing power were determined to measure the antioxidant activity. The results were the highest in 70% ethanol extracts, and the higher the concentration showed a significantly higher antioxidant capacity. When observed the relationship between the study, antioxidant activity of black raspberry was supposed to affect by the anthocyanin, phenol and flavonoid contents.

복분자 증편의 품질특성에 관한 연구 (A Study on Quality Characteristics of Jeungpyeon with Added Rubus coreanus Miquel)

  • This study sought to develop a healthier type of jeungpyeon, or Korean rice cake, by adding Korean black raspberry powder (Rubus coreanus). The moisture content, color, volume, symmetry, uniformity, texture, and sensory quality characteristics were evaluated in order to determine the optimal amount of raspberry powder to add to rice flour in order to get the best-tasting, healthiest food. We substituted 0.5%, 1.0%, 1.5% and 2.0% of rice flour with Rubus coreanus powder and mixed 3%, 6%, 9% and 12% Rubus coreanus concentrate with water to make dough. The result was stored for up to 48 hours at $20^{\circ}C$, and then evaluated. Jeungpyun with Rubus coreanus powder and Rubus coreanus concentrate had the right flavor, color, fermentation, and taste, compared to the other groups. Consequently, in overall acceptability, the highly rated Rubus coreanus powder-PW(1.5%) and Rubus coreanus concentrate - BA (6%) will be the most desirable.

  • 복분자 와인은 복분자 열매를 발효ㆍ숙성시켜 제조하기 때문에 그들 양자 간의 성분에는 차이가 있을 것으로 추측되었다. 그러나 복분자 와인의 음용률이 높음에도 불구하고 복분자 와인 중에 함유된 성분들에 관한 체계적인 연구는 거의 전무한 실정이다. 그래서 먼저 복분자 와인 중에 함유되어 있는 성분들의 chemical profile의 검토를 위해 항산화 활성에 초점을 맞추어 화합물의 단리ㆍ구조해석에 착수하였다. 복분자 와인 11 L(신선 복분자 열매 15.7 kg 상당량)의 농축액(928.3 g)을 용매분획한 후 얻어진 EtOAc층(20 g/56.2 g)을 대상으로 각종 column chromatography 및 HPLC를 행하여 5종의 항산화 활성 화합물을 단리하였다. 그들을 대상으로 $^1H$-NMR 분석 및 TMS 유도체화 후 GCMS 분석을 행한 결과, 4-hydroxybenzoic acid(1, 0.1 mg), 3,4-dihydroxybenzoic acid(2, 0.3 mg), 4-(2-hydroxyethyl)-phenol(3, 0.6 mg), pyrocatechol(4, 0.3 mg), 3,4,5-trihydroxybenzoic acid ethyl ester(5, 0.6 mg)로 동정하였다. 화합물 1과 2는 복분자 열매중에 존재함이 보고되어 있으나 화합물 3-5는 복분자 열매로부터 그 존재가 보고되어진 바 없으며, 이들 5종의 화합물 모두가 복분자 와인으로부터 동정되어진 것 또한 처음이다.

Freeze-Dried Powder of Rubus coreanus Miquel Ameliorates Isoproterenol-Induced Oxidative Stress and Tissue Damage in Rats

  • Rubus coreanus Miquel (bokbunja), Korean black raspberry, is known to possess various phytochemicals that exert antioxidative, anti-inflammatory, and anti-cancer effects. However, most studies on Rubus coreanus Miquel have been performed with the solvent extracts and/or a single component to demonstrate the efficacy, while studies evaluating the effect of the whole fructus of Rubus coreanus Miquel are limited. In this study, therefore, we employed the isoproterenol (IPN)-induced myocardial infarction model and investigated the effect of freeze-dried powder of Rubus coreanus Miquel (RCP) on oxidative stress and prevention of organ damage. Oral administration of RCP reduced the level of toxicity markers, alanine transaminase (ALT), aspartate transaminase (AST), and lactate dehydrogenase (LDH) without affecting body weight and diet intake. The oxidative stress marker glutathione (GSH) increased about 45% and malonaldehyde (MDA) decreased about 27% compared to the IPN group with RCP-H (3%) administration. By histological analysis, IPN induced significant myocardial damage in the heart and vascular injury in the liver, and RCP administration ameliorated the damages in a dose-dependent manner. Taken together, RCP activated the antioxidant system leading to prevention of damage to organs by IPN in rats, making it possible to expect beneficial efficacies by consuming the whole fructus of Rubus coreanus Miquel.

복분자(Rubus coreanus Miquel) 와인의 ethyl acetate 획분으로부터 저분자 휘발성 화합물들의 분리 및 구조해석 (Isolation and Identification of Low Molecular Volatile Compounds from Ethyl Acetate Layer of Korean Black Raspberry (Rubus coreanus Miq.) Wine)

  • 복분자 와인은 복분자 열매를 발효 숙성시켜 제조한 것으로, 소비자에게 널리 음용되고 있으나 그 원재료인 복분자 열매에 대한 연구에 비해 복분자 와인에 존재하는 성분들에 대한 체계적인 연구는 거의 전무한 실정이다. 최근 우리는 복분자주에 함유되어 있는 화합물들의 분자 수준에서의 연구를 통하여 4-hydroxybenzoic acid, 3,4-dihydroxybenzoic acid, 4-(2-hydroxyethyl)-phenol, pyrocatechol, ethyl gallate 등의 5종의 저분자 페놀성 화합물을 단리 구조해석하여 보고한 바 있다. 그 연속된 연구로써 본 논문에서는 복분자주에 존재하는 성분들에 대한 추가적인 분리 및 구조해석을 행하였다. 제조된 복분자 와인(11 L, 복분자 열매 15.7 kg)을 용매분획하여 얻어진 EtOAc층(56.2 g)의 일부(20 g)를 silica gel column chromatography와 ODS-HPLC로 정제하여 5종의 화합물을 단리하였다. 이 화합물들을 대상으로 MS 및 NMR 등의 기기분석을 행한 결과, ethyl succinate(1, 13.1 mg), vanillic acid(2, 2.6 mg), ethyl 3,4-dihydroxybenzoate(3, 13. 1 mg), furan-2-ol(4, 1.3 mg), 그리고 4-(4-hydroxyphenyl) butan-2(S)-ol(5, 1.1 mg)로 동정하였다. 이 화합물들 대부분은 발효식품에서 향기성분으로 동정된 바 있으며, 화합물 2는 복분자 열매에 존재함이 이미 보고되어 있으나, 화합물 1과 3-5는 복분자 열매 및 복분자와인으로부터 처음으로 동정되었다.

Fermentation properties of beer produced from Korean two-row barley or malt (Gwangmaek) supplemented with Korean red ginseng extracts and Bokbunja (Rubus coreanus Miquel) juice

  • This study involved the production of specialty lager beers supplemented with Korean red ginseng extracts or Bokbunja (Korean black raspberry, Rubus coreanus Miquel) juice. The effects of the Korean red ginseng extracts or Bokbunja juice on the specific gravity, pH, yeast viability, free amino nitrogen content, reducing sugar content, color, alcohol content, turbidity, and sensory evaluation were evaluated. The alcohol content of the beers containing the extracts or juice were within the standard alcohol amounts (3.63-4.0%, v/v). The pH values of the three samples containing Bokbunja juice were lower than that of the control values. The sensory evaluation showed that the addition of Bokbunja juice was superior to the ginseng extracts, and the optimal addition time was before or after the secondary fermentation. These data indicate that the flavor and odor of the Bokbunja juice are more persistent than that of the ginseng extracts.

완숙 복분자 부위별 지방산 조성과 여드름 원인균에 대한 항균 활성 (Comparison of Fatty Acids and Antibacterial Activity against Pathogen of Acne in Different Parts of Ripened Black Raspberry (Rubus coreanus Miquel))

  • 본 연구는 복분자의 활용도를 높이기 위한 자료를 제공하고자 동결 건조한 완숙 복분자를 종자와 종자를 분리한 과육, 그리고 분리과정을 거치지 않은 과실로 구분하여 지방산조성 분석과 각 부위별 methanol 추출물의 여드름 관련 균주에 대한 항균 활성, 그리고 polyphenol 함량 및 DPPH radical 소거능을 비교 분석하였다. 지방산 조성 분석 결과 종자의 총 지방 및 지방산 함량이 다른 부위에 비해 월등히 높은 수준임을 알 수 있었다. 아울러 종자의 주요 지방산으로 밝혀진 linoleic acid와 linolenic acid의 함량이 전체 지방산 조성의 81.262%로 대부분을 차지하였다. Polyphenol 함량 측정에서 123.64 mg/g으로 가장 높은 수준의 polyphenol 함량을 보였던 종자의 methanol 추출물이 DPPH radical 소거능에서도 다른 추출물에 비해 높은 활성을 나타내었다. 여드름 원인균에 대한 항균 활성에서 실험에 사용된 모든 균주에 대해서 종자를 분리한 과육의 methanol 추출물이 상대적으로 가장 높은 항균 활성을 나타내었으며, 종자 추출물은 상대적으로 낮은 활성을 보여주었다. 그러나 가장 높은 활성을 나타낸 과육 추출물의 항균 활성과 비교해서 종자 추출물의 항균 활성이 각 균주별로 약 76.96~89.08%에 해당함을 알 수 있었다. 결과적으로 완숙 복분자의 과육 부분뿐만 아니라 잔사나 폐기물로 버려질 수 있는 종자 부위의 활용도를 높일 수 있는 가능성을 확인하였다.

Antioxidant Properties and Physicochemical Attributes of Meat from Berkshire Finishing Pigs Supplemented with Rubus coreanus By-Product

  • A 60-d feeding trial was conducted to evaluate the effects of diets supplemented with two concentrations (0% and 0.3%) of black raspberry (Rubus coreanus Miquel) fruit by-product (RCFB) on the physicochemical characteristics, oxidative stability, antioxidant capacity, antioxidant enzyme activity, and fatty acid profile of M. longissimus dorsi (LL) porcine muscle from Berkshire finishing pigs meat. Results revealed that regardless of the sex, diets supplemented with 0.3% RCFB reduced (p<0.05) the thiobarbituric acid reactive substances (TBARS) expressed as malonaldehyde (MDA) content effectively. A higher antioxidant capacity [2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity] was found (p<0.05) in response to feeding supplemented with 0.3% RCBF for male or female pigs. Moreover, 0.3% RCFB dietary feed increased (p<0.05) the glutathione peroxidase enzyme activities (GPX1) in blood plasma for male or female pigs. However, no influences were observed (p>0.05) on meat color, WHC, shear force, and fatty acid contents while fed diet supplemented with 0% or 0.3% RCFB for male or female pigs. Overall, this study suggests that a diet supplemented with 0.3% RCFB may beneficially affect owing to better oxidative stability, higher antioxidant capacity, and antioxidant enzyme activity (blood plasma) in pigs which could be a promising natural antioxidant without affecting meat quality traits.

Anti-Inflammatory and Anti-Superbacterial Activity of Polyphenols Isolated from Black Raspberry

  • The fruit of the black raspberry (Rubus coreanus Miquel) has been employed in traditional medicine, and recent studies have demonstrated its measureable biological activities. However, the root of the black raspberry has not been studied. Therefore, in this study, we evaluated the anti-inflammatory and antibacterial properties of the root and unripe fruit polyphenols of the black raspberry. Both polyphenols proved to have anti-inflammatory activity as evidenced by the decreased nitric oxide (NO), cytokines (IL-$1{\beta}$, IL-6, and IL-10) and prostaglandin E2 ($PGE_2$) levels in lipopolysaccharide (LPS)-stimulated RAW 264.7 murine macrophages. However, root polyphenols showed stronger anti-inflammatory activity than fruit polyphenols. LPS-induced mRNA and protein expressions of inducible NO synthase (iNOS) and cyclooxygenase (COX)-2 levels were also decreased, confirming the anti-inflammatory activity. Root polyphenols showed lethal activity against methicillin-resistant Staphy-lococcus aureus (MRSA), carbapenem-resistant Acinetobacter baumannii (CRAB), and Bacillus anthracis. In contrast, the black raspberry fruit did not demonstrate these properties. These data provide the first demonstration that black raspberry root has potential anti-inflammatory and anti-superbacterial properties that can be exploited as alternatives for use in the food and cosmetic industries and/or as pharmaceuticals.

복분자 추출물의 Sprague-Dawley rat를 이용한 단회 경구 투여 독성시험 (Single Oral Dose Toxicity Study of Black Raspberry Extract in Sprague-Dawley Rats)

  • Objective : This study was performed to evaluate the toxicity after a single oral administration of black raspberry extract to male and female Sprague-Dawley (SD) rats and to determine the approximate lethal dose (ALD). Methods : We previously showed that the black raspberry extract repressed the simvastatin-mediated expression of Proprotein convertase subtilisin/kexin type 9 (PCSK9) and improved Low-Density Lipoprotein cholesterol (LDL-C) uptake by hepatocytes through the induction of the Low-Density Lipoprotein Receptor expression in hepatocytes. The groups consisted of black raspberry extract groups, as an oral dose of 2,000 mg/kg and a control group. 5 weeks SD rats were randomly assigned to 4 groups of 5 rats. Each male and female SD rats were administered orally once. For 14 days after the administration, mortality, clinical signs, changes in body weight, and necropsy findings were observed according to the "Standard for Toxicity Study of Pharmaceuticals" of Korea Food and Drug Administration (KFDA) guideline and "Acute Oral Toxicity- Fixed Dose Procedure" of OECD Test Guideline. Results : There were no cases of mortality in the group administered with 2,000 mg/kg of male and female, and no abnormalities in body weight change and clinical signs. Also, no gross abnormalities were observed at the autopsy. Conclusions : As a result of a single oral administration of the black raspberry extract to SD rats, the ALD was determined to exceed 2,000 mg/kg for both male and female SD rats.
