Spontaneous pyometra induced by Staphylococcus aureus in a Maltese (황색포도상구균 감염에 의한 어린 말티스의 자궁축농증 증례)

  • An 8-months-old young Maltese bitch was presented for investigation of anorexia and occasional vomiting. The body weight was 2 kg. Abdominal sonography demonstrated abnormal enlargement of uterus. The dog had been clinically and pathologically examined. Also, ovariohystectomy was performed. Staphylococcus aureus was identified in the pus-like materials of the uterine lumen. This case was confirmed as pyometra induced by Staphylococcus aureus. Gross examination of the dissected uterus revealed enlarged uterine horn, which had yellowish or brownish pus-like materials in the lumen. Also, there was several segments formation between the enlarged areas of left and right uterine horns. Histopathologically, endometritis and necrosis were present in uterine wall. The bitch made a complete recovery following an ovariohystectomy. This case was spontaneous pyometra induced by Staphylococcus species. On our knowledge, this is a very rare report of pyometra occurrence in young dog and Staphylococcus species-induced pyometra. This study may provide a valuable data on the study of canine pyometra.

Paternity test in dogs by microsatellite allele analysis (Microsatellite 대립유전자 분석을 통한 개에서의 친자감별)

  • Microsatellite allele analysis has been used for individual identification and paternity test. In the present study, the biological father of three puppies was determined by using microsatellite allele amplification analysis. The mother bitch of the litter was a Poongsan dog. The three stud dogs that could have inseminated the bitch, by being in the same residence, were a white Poosan dog, a mixed breed, and a white Jindo dog. DNA was obtained from all the relevant dogs by buccal swabbing. Four loci of tetranucleotide repeat microsatellite were PCR-amplified, and analyzed by polyacrylamide gel electrophoresis and silver staining. The results of genotyping unambigously assigned the Poongsan dog as the biological father. There was no evidence of superfecundation. Therefore, the present study demonstrated the usefulness of microsatellite allele analysis as a simple, efficient method of paternity test in dogs.

Experimental Production of Canine Pyometra by Inoculation with Escherichia coli into the Uterus (대양균 자궁내 접종에 의한 개의 자궁축농증 발증시험)

  • An attempt to reproduce pyometra by means of experimental uterine infection was conducted with the 10 mongrel hitches of the anestrus, proestrus, metestrus, and postpartum uterine involution stages. Organism used was hemoltic E. coli. origially isolated from uterine pus of a bitch that had the canine pyometra. One ml of broth culture containing $2{\times}10^6$ CFU was inoculated into the uterine horn with the uterine cervix ligated. Aか bitches were ovariohysterectomized at the 7th or 14th day after inoculation and the uterus was provided for macroscopic and microscopic observations. An experimental production of pyometra was successful in 7(70%) of 10 mongrel bitches, and the incidence of pyometra was seen 100% in the stage of anestrus, 100% in the stage of metestrus, 50.0% in the stage of pyomestrus, and 33.359 in the stage of uterine involution. Histologically, these uteri were characterized by the presence of cystic. endomeetrial hyperplasia(CEH) and acute endometritis superimposed on the cystic hyperplasia. And the pyometra with CEH could be seen 66.7% in metestrus 50% in anestrus, 33.3% in involution and in 0% in proestrus stage. Blood progesterone concentrations were showed as a normal range of the each stage of sexual cycle and any relationships between the progesterone concentrations and the incidence of pyometra were not observed.

Comparison of Plasma Progesterone Concentrations between Spontaneous Delivery Bitches and Fetal Death Bitches (정상 분만견과 태아사 발생견에서의 혈장 Progesterone 농도 비교)

  • This study investigated potential relationship between fetal deaths and plasma progesterone concentrations of bitches. Serial ultrasonographic examinations were performed on small-pet 23 dogs from gestation day (GD) 15 through parturition. The dogs were 3 non-pregnant bitches, 9 spontaneous delivery bitches, 6 partial early embryonic death bitches, 2 whole early embryonic death bitches, and 3 aborted bitches. The late pregnancy (GD 51-54) appeared in 2 of the 3 aborted bitches and the hypoluteoidism appeared in 1 of the 3 aborted bitches. The plasma progesterone concentrations of partial early embryonic death bitches (n=6) showed no significant difference when compared with the spontaneous delivery bitches. We observed that plasma progesterone concentrations were dramatic decrease before the onset of embryonic death in whole early embryonic death bitches that plasma progesterone concentrations of aborted bitches at late pregnancy were significantly decreased when compared with those of spontaneous delivery bitches. The plasma progesterone concentrations of the hypoluteoidism bitch were lower than those of spontaneous delivery bitches. At the hypoluteoidism bitch, fetuses were resorbed in early pregnancy and aborted in late pregnancy. On the basis of the results, the diagnosis of partial early embryonic death could not be confirmed without ultrasonographic examination. The partial early embryonic death was considered a spontaneous phenomenon and uncorrelated with plasma progesterone concentration. However, aborted bitches and whole early embryonic death bitches were related to plasma progesterone concentrations and that of bitches gradually decreased before fetal death. These findings suggest that administration of progesterone may be a useful preventing agent against fetal death.

Extracuterine Pregnancy in a Bitch (개의 자궁외 임신 증례)

  • A 3-year-old yorkshire terrier was examined because of the green-like vaginal discharge. Abdominal ultrasonographs revealed dead fetuses in the abdominal cavity. Surgical exploration revealed fetuses located in the region of the uterine wall, the omentum, the ileum, and the upper right side of the liver respectively. Fetal structures were removed and an ovariohysterectomy was performed. The abdominal cavity was flushed with warm saline and then the incision closed. These fetuses were at different stages of development. It was suggested that the development of the fetuses apparently continued in extrauterine pregnancy until the blood supply became inadquate for further growth.

  • Lameness of front limbs or hind limbs was occurred in German Shepherd dogs firm one brood bitch between 5 and 6 months of age. Physic81 examinations revealed pain associated with deep palpation of the affected area(u1nar, humerus, tibia, etc). But, no significant result was found for the clinical test including complete blood test and so oa Radiographic examination showed an increased radiopacity in the region of the nutrient foremen. Panosteitis was diagnosed on the basis of breed age, clinical signs and radiographic signs.

  • In dogs, correct diagnosis of estrus is important and the exact time of ovulation can be determined by variouse methods. Vaginal cytology has commonly used in conjunction with the physical examination, clinical history, vaginoscopy, and hormonal assays to determine the stage of the reproductive cycle. This study was therefore investigated the effectiveness of direct ovulation detector designed by changes of electrical resistance in vaginal mucus following different estrus cycles with several methods; vaginal cytology, concentration of plasma estrogen and progesterone, and direct examination by laparotomy. A total of 12 bitches was selected for the study and observed estrus signs. The bitches were evaluated clinical sign (vulvar swelling and bleeding), cytological examination (keratocyte and RBC), electrical resistance, plasma estrogen and progesterone concentration for estrus assessment. Accuracy of ovulation detection by vaginal cytology was significantly (p<0.05) lower than those by electrical resistance and plasma progesterone concentration, based on the confirmation by laparotomy. Vaginal smear is not confidential method compared to detection of electrical resistance and plasma progesterone concentration at ovulation. Although the value of electrical resistance was varied at the same points of estrus in individuals, ovulation was occurred at the first day which shown the peak of electrical resistance and mating time was third day after peak. In conclusion, ovulation detector designed by changes of electrical resistance is an effective and economic instrument for predicting estrus and ovulation in bitches.

Changes of Estradiol, Progesterone and Vaginal Epithelial Cells of a Pseudopregnant Captive Coyote (Canis latrans) during Breeding Season

  • Most captive canids and felids at Zoos in advanced countries have been examined enough to apply artificial reproductive techniques to them. We investigated reproductive hormones and vaginal epithelial cells of a 6-year-old, female coyote, hoping these data could eventually be extended to artificial insemination with frozen-thawed conspecific semen at Seoul Zoo. As a relative of pet dogs, coyote exhibited a similar appearance with only minor differences. In vaginal smear, an increase in the number of superficial cells suggests that the bitch has reached a state close to estrus. A sudden decrease of estradiol and increase of progesterone is considered as a preovulatory event. Vaginal epithelial cells and hormones might be useful for determining the optimal time of artificial insemination in coyotes' breeding.

Comparison of Appearance Rate of Vaginal Cytology According to Estrous Cycle in Small Pet Bitches (소형 애완견에서 발정주기별 질세포 출현율의 비교)

  • The aim of this study was to compare the appearance rate of vaginal cytology during estrous cycle in small pet bitches. A characteristic features of vaginal cytology during the estrous cycle were the high proportion of superficial cell in proestrus, anuclear cell in estrus, small intermediate cell in diestrus, and small intermediate cell in anestrus, respectively. There were no statistically significant differences of appearance rate of vaginal cytology among small pet bitches in the each phase of estrous cycle. These results indicated that the vaginal cytology was useful method for estimating estrous cycle and optimal breeding time in small pet bitches.
