The performance of immobilized fungal cells on celite beads for the production of gibberrelic acid was investigated in flasks and 7-L stirred-tank reactor. Repeated incubations of immobilized fungal cells increased cell concentrations and volumetric productivity. The maximum volumetric productivity obtained in the immobilized-cell culture was 3-fold greater than that in suspended-cell culture. The concentration of cotton seed flour (CSF), among the various nutrients supplied, most significantly influenced productivity and operational stability. Notably, insoluble components in CSF were found to be essential for production. CSF at 6 g/L with 60 g/L glucose was found to be optimal for gibberellic acid production and stable operation by preventing excessive cell growth.
A $\beta$-glucosidase from the algal lytic bacterium Sinorhizobium kostiense AFK-13, grown in complex media containing cellobiose, was purified to homogeneity by successive ammonium sulfate precipitation, and anion-exchange and gel-filtration chromatographies. The enzyme was shown to be a monomeric protein with an apparent molecular mass of 52 kDa and isoelectric point of approximately 5.4. It was optimally active at pH 6.0 and $40^{\circ}C$ and possessed a specific activity of 260.4 U/mg of protein against $4-nitrophenyl-\beta-D-glucopyranoside$(pNPG). A temperature-stability analysis demonstrated that the enzyme was unstable at $50^{\circ}C$ and above. The enzyme did not require divalent cations for activity, and its activity was significantly suppressed by $Hg^{+2}\;and\;Ag^+$, whereas sodium dodecyl sulfate(SDS) and Triton X-100 moderately inhibited the enzyme to under 70% of its initial activity. In an algal lytic activity analysis, the growth of cyanobacteria, such as Anabaena flos-aquae, A. cylindrica, A. macrospora, Oscillatoria sancta, and Microcystis aeruginosa, was strongly inhibited by a treatment of 20 ppm/disc or 30 ppm/disc concentration of the enzyme.
This study was conducted to improve biological degradation efficiency of toluene as a model volatile organic compound (VOC) using yeast Candida tropicalis and to suggest an effective method for bioreactor operation. The yeast strain was immobilized with polyethylene glycol (PEG), alginate, and powdered activated carbon (PAC). The yeast-immobilized polymer media were used as fluidized materials in an airlift bioreactor. Polymer media without PAC were also made and operated in another airlift bioreactor. The two bioreactors showed toluene removal efficiencies ranging 80-96% at loading rates of $10-35 g/m^3-hr$, and the bioreactor containing the polymer media with PAC achieved higher removal efficiency. Protein contents in the liquid phase showed that the bioreactor using the yeast-immobilized polymer media with PAC had a higher rate of microbial growth initially than that without PAC. In addition, the microbial growth rate inside of the polymer media with PAC was five times higher than that without PAC. Consequently, the polymer media containing the yeast strain and PAC could enhance removal efficiencies for VOCs, and the immobilization method improve microbial activity and stability for a long-term operation of biological systems.
An adventitious root formation protocol from Smilax china L. was established for in vitro production of dioscin, a steroidal saponin having various bioactivities such as anticancer, antifungal, antiviral, and antiobesity. Optimal medium for root initiation from leaf explant was MS medium containing $30\;g{\cdot}L^{-1}$ of sucrose supplemented with $1.0\;mg{\cdot}L^{-1}$ kinetin + $2.0\;mg{\cdot}L^{-1}$ NAA. The induction of adventitious roots from in vitro initiated root segments was most favorable to MS liquid medium with $0.1\;mg{\cdot}L^{-1}$ kinetin + $2.0\;mg{\cdot}L^{-1}$ NAA. Among the 20 different adventitious roots originated from different plants, strain No. 10 was selected based on production ability of dioscin, and its stability through the successive suspension culture. The maximum growth stage of adventitious roots was noticed at 5 weeks after subculture while that of dioscin production in the adventitious root was at 7 weeks after subculture in suspension culture system. These results provide that suspension culture of adventitious roots of Smilax china L. have a potential for in vitro mass production of dioscin.
Recently, various researches on materials and equipment have been actively conducted to overcome the limitations of conventional output methods due to the increase of diversity of 3D printing materials and to adopt an output method suitable for the characteristics of each material. As the range applicable to outputable materials is expanded, manufacturing of medical devices applied to patients is in a more rapid growth trend than other fields. In this study, we investigated the suitable materials for fabricating 3D printer using photocurable resin. As a result, one suitable material was selected through biological safety experiment and thermal stability experiment. Next, to optimize the output of the selected materials, we have developed a system that optimizes the equipment according to the characteristics of the material. The results of this study enabled the implementation of personalized medical implants that could not be made from 3D printer dependent materials, thereby overcoming the limitations of existing 3D printer output conditions and dedicated materials.

The objectives of this study were to evaluate the anti-aging effects and investigate the effect of simulated gastrointestinal (GI) digestion on the anti-aging properties and intestinal permeation of the potential fish collagen hydrolysates (FCH). Therefore, procollagen synthesis, matrix metalloproteinase-1 (MMP-1) production, and Caco-2 cell permeability were analyzed before and after in vitro digestion for FCHs, low-molecular weight fractions (<1 kDa), and high molecular weight fractions (>1 kDa). After being subjected to GI digestion, the level of MMP-1 inhibition was maintained, although the procollagen production was significantly (>20%) lower with all samples. Also, the digested FCHs and their <1 kDa fraction yielded 9.1 and 13.8% increased peptide transport, respectively, compared to undigested samples. Based on the effective intestinal permeation and high digestive enzyme stability, the <1 kDa fraction of FCHs is a potential bioactive material suitable for anti-aging applications in the food and cosmetics industries.
Phytases are enzymes that can hydrolyze phytate and its salts into inositol and phosphoric acid, and have been utilized to increase the availability of nutrients in animal feed and mitigate environmental pollution. However, the enzymes' low thermostability has limited their application during the feed palletization process. In this study, a combination of B-value calculation and protein surface engineering was applied to rationally evolve the heat stability of Escherichia coli phytase. After systematic alignment and mining for homologs of the original phytase from the histidine acid phosphatase family, the two models 1DKL and 1DKQ were chosen and used to identify the B-values and spatial distribution of key amino acid residues. Consequently, thirteen potential amino acid mutation sites were obtained and categorized into six domains to construct mutant libraries. After five rounds of iterative mutation screening, the thermophilic phytase mutant P56214 was finally yielded. Compared with the wild-type, the residual enzyme activity of the mutant increased from 20% to 75% after incubation at $90^{\circ}C$ for 5 min. Compared with traditional methods, the rational engineering approach used in this study reduces the screening workload and provides a reference for future applications of phytases as green catalysts.
Objectives In regard to the social competition hypothesis, depression is viewed as an involuntary defeat strategy. A previous study has demonstrated that adaptation in microenvironments can result in a wide range of behavioural patterns including defense activation disorders. Using a simulation model with evolutionary ecological agents, we explore how the fitness of various defence activation traits has changed over time in different environments with high and low social mobility. Methods The Evolutionary Ecological Model of Defence Activation Disorder, which is based on the Marginal Value Theorem, was used to examine changes in relative fitness for individuals with defensive activation disorders after adjusting for social mobility. Results Our study examined the effects of social mobility on fitness by varying the d-values, a measure of depression in the model. With a decline in social mobility, the level of fitness of individuals with high levels of defense activation decreased. We gained insight into the evolutionary influence of varying levels of social mobility on individuals' degrees of depression. In the context of a highly stratified society, the results support a mismatch hypothesis which states that high levels of defence are detrimental. Conclusions Despite the fact that niche specialization in habitats composed of multiple microenvironments can result in diverse levels of defensive activation being evolutionary strategies for stability, decreased social mobility may lead to a decrease in fitness of individuals with highly activated defence modules. There may be a reason behind the epidemic of depression in modern society.

Biological nutrient removal is gaining increasing attention in wastewater treatment plants; however, it is adversely affected by low temperatures. This study examined temperature effects on nutrient removal and morphological stability of the granular and denitrifying phosphorus accumulating organisms (PAO and DPAO, respectively) using sequencing batch reactors (SBRs) at 5, 10, and 20 ℃. Lab-scale SBRs were continuously operated using anaerobic-anoxic and anaerobic-oxic cycles to develop the PAO and DPAO granules for 230 d. Sludge granulation in the two SBRs was observed after approximately 200 d. The average removal efficiency of soluble chemical oxygen demand (SCOD) and PO43--P remained >90% throughout, even when the temperature dropped to 5 ℃. The average removal efficiency of NO3--N remained >80% consistently in DPAO SBR. However, nitrification drastically decreased at 10 ℃. Hence, the removal efficiency of NH4+-N was decreased from 99.1% to 54.5% in PAO SBR. Owing to the increased oxygen penetration depth at low temperatures, the influence on nitrification rates was limited. The granule in DPAO and PAO SBR was observed to be unstable and disintegrated at 10 ℃. In conclusion, morphological characteristics showed that changed conversion rates at low temperatures in aerobic granular sludge altered both nutrient removal efficiencies and granule formation.
