BACKGROUD: ${\alpha}$- and ${\beta}$- Endosulfan isomers of endosulfan, an endocrine disrupting chemical, are widely used cyclodiene organochlorine pesticide in worldwide, and it has widespread application in agriculture and can contaminate river-system as runoff from soil or aerial deposition METHOD AND RESULTS: In this study, an attempt was made to isolate an endosulfan degrading fungus from endosulfan-polluted agricultural soil. Through repetitive enrichment and successive subculture in media containing endosulfan and its metabolites as the sole carbon source, a fungus designated KEF-1 was isolated. Based on phylogenetic analysis, strain KEF-1 was assigned to the genus Eutypella. Also, the ITS (internal transcribed spacer) sequences of KEF-1 were submitted to GenBank under accession number EF581006. In potato dextrose broth containing 8 ${\mu}g$/mL endosulfan, strain KEF-1 completely degraded the endosulfanin 12 days. CONCLUSION: These results suggested that Eutypella sp. KEF-1 has potential as a biocatalyst for endosulfan bioremediation
Viruses have various applications in the biopharmaceutical industry. They are used in pesticide production, production of vaccines, gene transfers, cancer therapeutics, and more. The downstream processing of viruses is an essential step for their biological and pharmaceutical applications. Among the various processes, the purification of viruses is critical. Membrane chromatography plays a vital role in this process. While ion exchange membrane chromatography is a primarily used method, it has various limitations regarding size exclusion and insufficient purification. Also, it cannot be applied to the rapidly changing strains of viruses such as influenza. This review examines various improved methods of membrane chromatography or alternatives. It focuses on purification, viral recovery rates, and scalability of the methods.
Cheju island depends on a hydrogeologically vulnerable aquifer system as its principle source of drinking water. Most of golf courses are located in the area which is important for the ground water recharge, and pesticides are applied to golf courses often at relatively high rates. Therefore, turf pesticides in golf course should be applied without adversely impacting ground water. In this experiment, downward movement of pesticides was monitored in model greens of golf course, where different adsorbents were layered in 3-cm thickness at 35-cm depth, and effect of the adsorption layer on the leaching loss of pesticides was investigated. Major leachings were observed in the periods of heavy rain and very limited leaching was observed under artificial irrigation. Fenitrothion and triadimefon, which have relatively short persistence and high adsorption coefficient, were found in the leachate in low concentrations only at the first rainfall event, around 20 days after the pesticide application. However, diniconazole, which has a relatively long half-life (97 days), was detected in the leachate during the whole period of experiment and concentration was much higher than those of the other pesticides. Maximum leachate concentrations were 1.9, 10.3, and 84.5 ${\mu}l^{-1}$ for fenitrothion, triadimefon, and diniconazole, respectively. Therefore, in golf course green which allows rapid water percolation and has extremely low adsorption capacity, persistence in soil could be more important factor in determination of leaching potential of pesticides. Total quantity of pesticides leached from the model green was <0.2% for fenitrothion and triadimefon and 1.8% for diniconazole. Adsorption layers significantly reduced pesticide leaching, and active carbon and Orpar were more effective than zeolite. In the model green having adsorption layer of active carbon or Orpar, leaching loss of pesticides was reduced below 0.01% of the initial application.
Cypermethrin, a commonly used domestic and agricultural pyrethroid pesticide, is widely considered detrimental to the environment and to many organisms because of its residual property and toxicity. Cellulophaga lytica DAU203, isolated from coastal sediment, was chosen because it degrade cypermethrin. Cellulophaga lytica DAU203 effectively degraded cypermethrin, as the utilized carbon source and substrate, in a mineral salt medium. Effective factors, such as carbon source, nitrogen source, initial pH, and temperature, for cypermethtin biological degradation by Cellulophaga lytica DAU203 were analyzed by one factor at a time method. Temperature ($22{\sim}42^{\circ}C$), initial pH (5~9), and yeast extract concentration (0.1~2.5%[w/v]) were selected as the three most important factors. There were optimized at $33.4^{\circ}C$, pH 7.7, and 2.4%(w/v) by response surface methodology, respectively. The Box- Behnken design consisting of 46 experimental runs with three replicates was used to optimize the independent variables which significantly influenced the cypermethrin biological degradation. This model for cypermethrin degradation by Cellulophaga lytica DAU203 is highly significant (p<0.05). Under the optimized condition, Cellulophaga lytica DAU203 degraded approximately 83.7 % of the cypermethrin within 5 days. These results suggest that Cellulophaga lytica DAU203 may be useful for the biological degradation of cypermethrin in cypermethrin-contaminated environments.

This study was carried out to investigate the residual characteristics of flubendiamide in kale to establish pre-harvest residue limits (PHRL) and the removal efficiency according to the washing solvent and method. Field tests were conducted at two different greenhouses, field 1 (Anseong-si, Gyeonggi-do) and field 2 (Incheon-si, Gyeonggi-do). According to the safe use guidelines kale was sprayed with flubendiamide twice every 10 days and harvested 0 (after 2 h), 1, 2, 3, 5, 7 and 10 days after the final application. The biological half-live of flubendiamide in kale was calculated based on dissipation curves of the pesticide in samples analyzed by liquid chromatography coupled with tandem mass spectrometry. In the analysis, method limits of quantitation (MLOQ) were 0.01 mg/kg, and recoveries performed with two different fortification levels of 10 MLOQ and maximum residue limit (0.7 mg/kg) were 104.2±3.6 and 101.9±10.2%, respectively. The dissipation rate constant of flubendiamide in kales were 0.2437 at field 1 and 0.1981 at field 2. PHRL calculation equations obtained using the dissipation constants estimated as follows: if the residual concentration of flubendiamide in kale on 10 days before harvest is less than 8.0 mg/kg, the residual concentration on the harvest would be under MRL. The removal of flubendiamide from kale was the greatest when it was washed with vinegar (39.8%), followed by baking soda (31.7%), calcium powder (30.2%), neutral detergent (27.2%), and tap water (15.9%). The results of this study would be useful for both farmers and consumers to produce or consume safe agricultural products.
This study was carried out to survey the residual characteristic of chlorpyrifos and estimate their residues in squash before harvest. The pesticide was sprayed onto the crop at the recommended and its double rates 10 days before the prearranged harvest and sampling was done at 0, 2, 3, 5, 6, 7 and 10 days after spraying. The amounts of the chlorpyrifos residue in the crop was analyzed by chromatographic method. Limit of detection (LOD) of chlorpyrifos was 0.005 mg/kg and its recovery ranged from 95.21 to 102.69%. The initial concentration of chlorpyrifos sprayed with recommended dose exceeded its MRL of 0.1 mg/kg but its concentration was less than its MRL 10 days after application. However its concentration in case of the double dose was over its MRL both immediately and 10 days after application. Biological half-lives of chlorpyrifos sprayed onto squash was 2.5 and 2.9 days at the recommended and double doses, respectively. Ten days later, the residual concentration of chlorpyrifos in squash was decreased substantially. The concentration of chlorpyrifos was estimated in squash at the given day using its regression equations. The estimated concentration of chlorpyrifos in case of application with recommended dose was below its MRL at 10 days after application but its concentration in case of application with double dose was over its MRL at 10 days of the prearranged harvest. The rate of the estimated daily intake (EDI) of chlorpyrifos to its acceptable daily intake (ADI) was 282% right after application but it decreased to less than 18% at 10 days of the prearranged harvest.
This study established hazards which may cause risk to human at farm during cultivation stage of paprika. Samples of plants (paprika, leaf, stem), cultivation environments (water, soil), personal hygiene (hand, glove, clothes), work utensils (carpet, basket, box) and airborne bacteria were collected from three paprika farms (A, B, C) located in Western Gyeongnam, Korea. The collected samples were assessed for biological (sanitary indications and major foodborne pathogens), chemical (heavy metals, pesticide residues) and physical hazards. In biological hazards, total bacteria and coliform were detected at the levels of 1.9~6.6 and 0.0~4.610g CFU/g, leaf, mL, hand or 100 $cm^2$, while Escherichia coli was not detected in all samples. In major pathogens, only Bacillus cereus were detected at levels of ${\leq}$ 1.5 log CFU/g, mL, hand or 100 $cm^2$, while Staphylococuus aureus, Listeria monocytogenes, E. coli O157 and Salmonella spp. were not detected in all samples. Heavy metal and pesticide residue as chemical hazards were detected at levels below the regulation limit, physical hazard factors, such as insects, pieces of metal and glasses, were also found in paprika farms. Proper management is needed to prevent biological hazards due to cross-contamination while physical and chemical hazards were appropriate GAP criteria.

This study was performed to select potentially available biological control agent from soil bacteria for prevention of ginseng damping off. More than five hundred strains were isolated from ginseng rhizosphere soil. By testing antifungal activity, we have selected three soil bacteria strains and their ability to produce antibiotics and lytic enzymes such as cellulase, protease and pectate lyase was examined. Also, the presence of genes for biosynthesis of lipopeptide such as fengycin, bacillomycin D, surfactin, iturin A, and zwittermicin A was investigated in selected strains. All three strains produced cellulase, protease, and xylanase. Moreover, these strains had gene for biosynthesis of bacillomycin D, surfactin, and iturin A. ES1 and ES3 strains were identified Bacillus methylotrophucus and ES2 was confirmed Bacillus amyloliquefaciens using phylogenetic analysis on the basis of 16S rRNA gene sequences. In field test, control value of ES1, ES2 and ES3 treatment was 32.4%, 46.8% and 36.7%, respectively. This results indicate that antagonistic microbes with high ability of antifungal and lytic enzyme activity can be used as a useful biological control agent to control ginseng damping off.
Physical, chemical and biological hazards of strawberry farms at the cultivation stage were analyzed to establish the GAP(Good Agricultural Practice) system. Samples were collected from the plants, cultivation environments(water, soil and air), and personal hygiene (hand, glove, and clothes) of three strawberry farms(A, B, and C) and were tested to analyze physical, chemical (heavy metals and pesticide residues), and biological(sanitary indications and foodborne pathogens) hazards. Physical hazards such as insects and pieces of metal and glass were found in the strawberry farms and can be potential bow for strawberry products. Heavy metal and pesticide residue as chemical hazards were detected at levels lower than the regulation limit. In case of biological hazards, total bacteria and coliform were detected at the levels of 1.6~7.3 and 1.3~5.6 log CFU/g, leaf, mL, hand or $100cm^2$. However, Escherichia coli was not detected in all samples. Bacillus cereus and Staphylococuus aureus were detected at levels of ${\leq}$ 1.1~6.1 log CFU and 4.7~5.4 log CFU/g, mL, hand or $100cm^2$, whereas Listeria monocytogenes, E. coli O157 and Salmonella spp. were not detected in all samples. This study demonstrates that various harzards were in strawberry farms at the growing stage. Therefore proper management such as GAP is needed to prevent the occurrence of food poisoning associated with the hazards revealed in this study.
The construction procedures and artificial turf maintenance program on golf course definitely influence on the distortion of its environment. Soil microbial communities in soil profile were affected directly by those practises on turf areas. In Jeju island, the environmental impact assessment has been required to apply the first quality class granular activated carbon(GAC), which has a high absorbent character to agricultural chemicals, on the soil profiles of golf green system to reduce the pesticide leaching to ground water. This research was carried out to analyze the changes of microbial communities and chemical properties on soil profiles where GAC had been applied at the construction stage at two golf courses in Jeju. The changes of soil microbial population and chemical properties associated with construction methods of soil profile and agrochemical management program were analyzed by monthly at the surface and sub-soil profiles during April through October, 2007. The total numbers of bacteria and fungi, soil moisture content, soil physio-chemical properties were measured on greens and fairways of the both golf courses with different GAC treatment on the green and fairway soil profiles. The results showed that GAC had positive effects on the water holding capacity, pH and EC, however, it did not improved the holding capacity of available nutrients ${NO_3}^-,{NH_4}^+$, and phosphorus by its sorption phenomenon. In microbial count test, the total numbers of bacteria and fungi showed a great variation during sampling dates. That may directly relate to the agrochemical application, however, the ratio of total bacterial number versus total fungus number showed a constant value on a sub-soil of 15~30cm depth. Thus, the construction method of GAC in soil profile, and application of fertilizer and pesticide, both impacted on the changes of microbial population. It's means that the construction method of soil profile and turf management using agro-materials might greatly affect on the turfgrass culture and the environment of golf course.
