Global warming has accelerated in recent decades due to the continuous consumption of petroleum-based fuels. Cyanobacteria-derived biofuels are a promising carbon-neutral alternative to fossil fuels that may help achieve a cleaner environment. Here, we propose an effective strategy based on the large-scale cultivation of a newly isolated cyanobacterial strain to produce phycobiliprotein and biodiesel, thus demonstrating the potential commercial applicability of the isolated microalgal strain. A native cyanobacterium was isolated from Goryeong, Korea, and identified as Pseudanabaena mucicola GO0704 through 16s RNA analysis. The potential exploitation of P. mucicola GO0704 was explored by analyzing several parameters for mixotrophic culture, and optimal growth was achieved through the addition of sodium acetate (1 g/l) to the BG-11 medium. Next, the cultures were scaled up to a stirred-tank bioreactor in mixotrophic conditions to maximize the productivity of biomass and metabolites. The biomass, phycobiliprotein, and fatty acids concentrations in sodium acetate-treated cells were enhanced, and the highest biodiesel productivity (8.1 mg/l/d) was achieved at 96 h. Finally, the properties of the fuel derived from P. mucicola GO0704 were estimated with converted biodiesels according to the composition of fatty acids. Most of the characteristics of the final product, except for the cloud point, were compliant with international biodiesel standards [ASTM 6761 (US) and EN 14214 (Europe)].
Chlorella vulgaris는 인간과 동물에서 주요한 식품의 원료로 이용되는 수 조류이다. 최근 클로렐라는 어류 양식, 생물연료 및 영양물질과 치료용 단백질의 생산을 위한 이용 가능성과 관련하여 많은 관심을 받고 있다. 최근 본 연구실에서는 빠른 성장, 배양의 용이성, 암 조건에서의 배양성을 특징으로 하는 새로운 C. vulgaris PKVL7422균주를 분리하였으나, 이 균주의 질소 이용에 관련된 유전자에 대하여는 보고된 바가 없다. 본 연구에서는 cDNA 말단의 신속 증폭과 genome walking을 이용하여 C. vulgaris PKVL7422의 질산환원 효소(nitrate reductase, NR)를 동정하였다. C. vulgaris PKVL7422 NR 유전자는 약 8 kb이며, 18개의 인트론과 877개의 아미노산을 암호화 하는 19개의 엑손으로 되어 있다. 기존의 알려진 NR 단백질과의 비교 분석 결과 C. vulgaris PKVL7422 NR 단백질은 식물의 NR에 존재하는 5개의 도메인과 다수의 비변이성 아미노산 잔기를 가지고 있는 것으로 확인되었으며, 이러한 결과는 조류 NR 유전자의 분자생물학적 구조에 대한 새로운 정보를 제공한다.

The development of a yeast strain capable of fermenting mixed sugars efficiently is crucial for producing biofuels and value-added materials from cellulosic biomass. Previously, a mutant Pichia stipitis YN14 strain capable of co-fermenting xylose and cellobiose was developed through evolutionary engineering of the wild-type P. stipitis CBS6054 strain, which was incapable of co-fermenting xylose and cellobiose. In this study, through genomic and transcriptomic analyses, we sought to investigate the reasons for the improved sugar metabolic performance of the mutant YN14 strain in comparison with the parental CBS6054 strain. Unfortunately, comparative whole-genome sequencing (WGS) showed no mutation in any of the genes involved in the cellobiose metabolism between the two strains. However, comparative RNA sequencing (RNA-seq) revealed that the YN14 strain had 101.2 times and 5.9 times higher expression levels of HXT2.3 and BGL2 genes involved in cellobiose metabolism, and 6.9 times and 75.9 times lower expression levels of COX17 and SOD2.2 genes involved in respiration, respectively, compared with the CBS6054 strain. This may explain how the YN14 strain enhanced cellobiose metabolic performance and shifted the direction of cellobiose metabolic flux from respiration to fermentation in the presence of cellobiose compared with the CBS6054 strain.

The surge in the oil prices, increasing global population, climate change, and waste management problems are the major issues which have led to the development of biofuels from lignocellulosic wastes. Cellulosic or second generation (2G) bioethanol is produced from lignocellulosic biomass via pretreatment, hydrolysis, and fermentation. Pretreatment of lignocellulose is of considerable interest due to its influence on the technical, economic and environmental sustainability of cellulosic ethanol production. In this study, furniture waste sawdust was subjected to alkaline peroxide (H2O2) for the production of reducing sugars. Sawdust was pretreated at different concentrations from 1-3% H2O2 (v/v) loadings at a pH of 11.5 for a residence time of 15-240 min at 50, 75 and 90 ℃. Optimum pretreatment conditions, such as time of reaction, operating temperature, and concentration of H2O2, were varied and evaluated on the basis of the amount of total reducing sugars produced. It was found that the changes in the amount of lignin directly affected the yield of reducing sugars. A maximum of 50% reduction in the lignin composition was obtained, which yielded a maximum of 75.3% total reducing sugars yield and 3.76 g/L of glucose. At optimum pretreatment conditions of 2% H2O2 loading at 75 ℃ for 150 min, 3.46 g/L glucose concentration with a 69.26% total reducing sugars yield was obtained after 48 hr. of the hydrolysis process. Pretreatment resulted in lowering of crystallinity and distortion of the sawdust after the pretreatment, which was further confirmed by XRD and SEM results.
푸르푸랄(furfural)은 리그노셀룰로오스 바이오매스(lignocellulose biomass)의 헤미셀룰로오스(hemicellulose) 성분 중 하나인 자일로스(xylose)로부터 생산되는 플랫폼 화학물질이다. 푸르푸랄은 페놀류 화합물이나 바이오 연료 등의 중요한 원료로 사용될 수 있다. 본 연구에서는 푸르푸랄 생산공정에서 일반적으로 사용되는 산 촉매인 황산(sulfuric acid)과 친환경적 촉매인 포름산(formic acid) 두 가지 촉매를 이용하여 회분식 반응 시스템(batch system)에서 자일로스로부터 푸르푸랄을 생산하기 위한 조건을 비교 및 최적화하였다. 자일로스의 초기 농도(10 g/L~100 g/L), 반응 온도(140~200 ℃), 황산 촉매(1~3 wt%), 포름산 촉매(5~10 wt%), 반응 시간에 따라 자일로스로부터 푸르푸랄 수율에 미치는 영향을 조사하였다. 촉매 종류에 따른 최적 조건은 다음과 같았다. 황산 촉매의 경우, 3 wt%의 촉매농도, 50 g/L의 초기 자일로스 농도, 180 ℃의 온도 10분의 반응시간에서 최대 58.97%의 푸르푸랄 수율을 얻었다. 포름산 촉매의 겨우, 5 wt%의 촉매농도, 50 g/L의 초기 자일로스 농도, 180 ℃의 온도, 150분 반응 시간에서 65.32%의 푸르푸랄 수율을 확보하였다.
Carbon dioxide (CO2) is the most abundant component of greenhouse gases (GHGs) and directly creates environmental issues such as global warming and climate change. Carbon capture and storage have been proposed mainly to solve the problem of increasing CO2 concentration in the atmosphere; however, more emphasis has recently been placed on its use. Among the many methods of using CO2, one of the key environmentally friendly technologies involves biologically converting CO2 into other organic substances such as biofuels, chemicals, and biomass via various metabolic pathways. Although an efficient biocatalyst for industrial applications has not yet been developed, biological CO2 conversion is the needed direction. To this end, this review briefly summarizes seven known natural CO2 fixation pathways according to carbon number and describes recent studies in which natural CO2 assimilation systems have been applied to heterogeneous in vivo and in vitro systems. In addition, studies on the production of methanol through the reduction of CO2 are introduced. The importance of redox cofactors, which are often overlooked in the CO2 assimilation reaction by enzymes, is presented; methods for their recycling are proposed. Although more research is needed, biological CO2 conversion will play an important role in reducing GHG emissions and producing useful substances in terms of resource cycling.
Glucosylation is a well-known approach to improve the solubility, pharmacological, and biological properties of flavonoids, making flavonoid glucosides a target for large-scale biosynthesis. However, the low yield of products coupled with the requirement of expensive UDP-sugars limits the application of enzymatic systems for large-scale. C. glutamicum is a Gram-positive and generally regarded as safe (GRAS) bacteria frequently employed for the large-scale production of amino acids and biofuels. Due to the versatility of its cell factory system and its non-endotoxin producing properties, it has become an attractive system for the industrial-scale biosynthesis of alternate products. Here, we explored the cell factory of C. glutamicum for efficient glucosylation of flavonoids using apigenin as a model flavonoid, with the heterologous expression of a promiscuous glycosyltransferase, YdhE from Bacillus licheniformis and the endogenous overexpression of C. glutamicum genes galU1 encoding UDP-glucose pyrophosphorylase and pgm encoding phosphoglucomutase involved in the synthesis of UDP-glucose to create a C. glutamicum cell factory system capable of efficiently glucosylation apigenin with a high yield of glucosides production. Consequently, the production of various apigenin glucosides was controlled under different temperatures yielding almost 4.2 mM of APG1(apigenin-4'-O-β-glucoside) at 25℃, and 0.6 mM of APG2 (apigenin-7-O-β-glucoside), 1.7 mM of APG3 (apigenin-4',7-O-β-diglucoside) and 2.1 mM of APG4 (apigenin- 4',5-O-β-diglucoside) after 40 h of incubation with the supplementation of 5 mM of apigenin and 37℃. The cost-effective developed system could be used to modify a wide range of plant secondary metabolites with increased pharmacokinetic activities on a large scale without the use of expensive UDP-sugars.
지구 온난화, 석유 고갈, 환경 오염에 대한 방안으로 수송부문에서 국제적으로 바이오연료에 관한 연구가 활발하게 이루어지고 있다. 그 중 바이오디젤은 이산화탄소 감소 효과와 인체에 무해하며 세탄가가 높아 석유디젤을 대체할 수 있는 장점을 가지고 있다. 현재 국내 바이오디젤 수요는 지속적으로 증가하고 있으나 원료부족으로 인해 수입의존도가 커지고 있는 상황이다. 이러한 문제를 해결하기 위해 본 연구는 현재 사용되지 않는 음폐유(약 33 % 유리지방산 함유)를 Amberlyst-15촉매를 이용한 에스테르화 반응을 통해 바이오디젤 원료로서 활용가능성을 확인 하였다. 다양한 반응 조건의 영향을 조사하기 위한 실험을 수행한 결과 반응온도 383 K에서 97.62 %의 전환율을 얻었으며, 반응속도는 353 K에서 373 K로 증가 할 때 최대 1.99 배까지 상승하였다. 또한 동역학적 결과를 이용하여 29.75 kJ/mol의 활성화 에너지를 확인하여 선행연구에서 연구된 타 고체촉매에 비해 에스테르화반응에 Amberlyst-15 더 적합함을 확인하였다. 그리고 메탄올 몰 비가 증가함에 따라 최대 91.43 %의 반응 전환율을 확인하였고, 촉매량 영향의 경우 0 wt%에서 20 wt%까지 증가시킨 결과 반응 전환율이 43.78 %에서 94.62 %까지, 초기 반응 속도는 1.1~1.4 배로 상승하는 것을 확인하였다. 교반속도의 경우 100~900 rpm의 조건에 따라 실험을 수행하였으나 반응 전환율에는 큰 영향을 주지 않음을 확인하였고 반응 시간에 따른 영향의 경우 240 분 까지 산가 감소를 보이다가 300 분이 지나면서부터 산가가 상승하는 결과를 가져왔다. 그리고 위 실험들을 통해 도출된 최적 조건을 적용하여 음폐유 에스테르화 반응에 적용하였고 그 결과 반응시간 60 분에서 음폐유와 모사 폐유지간의 13 %의 반응 전환율 차이를 보였으나 최종 240 분 반응 전환율은 모사 폐유지 98.12 %, 음폐유는 97.62 %로 거의 유사한 결과를 얻었다.
목질계 바이오매스는 바이오 연료 및 바이오 화학제품 생산을 위한 재생 가능 자원이다. 푸르푸랄(furfural, FF)은 목질계 바이오매스의 헤미셀룰로스로부터 화학적 촉매전환으로 유도되는 주요한 플랫폼 케미칼이다. 테트라히드로푸르푸릴 알코올(Tetrahydrofurfuryl alcohol, THFA)은 FF의 유도체로 열적 화학적 안정성을 지닌 친환경 용매로 이용 가능하다. FF를 THFA로 전환하는 실험적 연구가 다수 존재함에도 불구하고, FF로부터 THFA의 대량생산에 관한 경제적 실현가능성에 관한 연구는 거의 수행되지 않았다. 개발된 전환기술의 상용화 단계에서 기술적 병목점 확인과 스케일업 문제의 해결을 위한 정보를 얻기 위해 실증플랜트 규모의 연구가 필요하다. 본 연구에서는 FF의 THFA로의 화학적 촉매전환에 대해 공정 시뮬레이션 및 기술경제성 평가가 수행되며, 3가지 단계(통합 공정 디자인, 열 통합, 경제성 평가)를 거친다. 실험연구 결과를 기반으로 전환공정과 분리공정을 포함하는 실증플랜트 규모의 통합공정이 설계된다. FF 처리량은 일일 255톤이며, FF로부터 THFA로의 수율은 63.2~67.9 mol%이다. 통합공정에 대해 열 통합을 수행하여 가열요구량을 최초 대비 14.4~16.4% 감소시킬 수 있었다. 최종적으로 경제성 평가를 통해 전체 공정의 주요 비용원을 분석하고 THFA의 최소판매가격을 결정하였다. 개발된 공정에서 생산되는 THFA의 최소판매가격은 1톤당 2,120~2,340 달러로, 현재 THFA의 시장 가격에 근접한다.
