Comparison of Inhibitory Effects of 17-AAG Nanoparticles and Free 17-AAG on HSP90 Gene Expression in Breast Cancer

  • Background: HSP90 may be overexpressed in cancer cells which are greatly dependent on Hsp90 function. Geldanamycin derivative 17 allylamino-17-demethoxygeldanamycin (17-AAG) inhibits the function and expression of HSP90. 17-AAG has poor water-solubility which is a potential problem for clinical practice. In this study for improving the stability and solubility of molecules in drug delivery systems we used a ${\beta}$-cyclodextrin-17AAG complex. Materials and Methods: To assess cytotoxic effects of ${\beta}$-cyclodextrin-17AAG complexes and free 17AAG, colorimetric cell viability (MTT) assays were performed. Cells were treated with equal concentrations of ${\beta}$-cyclodextrin- 17AAG complex and free 17AAG and Hsp90 gene expression levels in the two groups was compared by real-time PCR. Results: MTT assay confirmed that ${\beta}$-cyclodextrin- 17AAG complex enhanced 17AAG cytotoxicity and drug delivery in T47D breast cancer cells. The level of Hsp90 gene expression in cells treated with ${\beta}$-cyclodextrin- 17AAG complex was lower than that of cells treated with free 17AAG (P=0.001). Conclusions: The results demonstrated that ${\beta}$-cyclodextrin- 17AAG complexes are more effective than free 17AAG in down-regulating HSP90 expression due to enhanced ${\beta}$-cyclodextrin-17AAG uptake by cells. Therefore, ${\beta}$-cyclodextrin could be superior carrier for this kind of hydrophobic agent.

  • We consider a subdiagonal bilinear model and give sufficient conditions for the associated Markov chain defined by Pham (1985) to be uniformly ergodic and then obtain the $\beta$-mixing property for the given process. To derive the desired properties, we employ the results of generalized random coefficient autoregressive models generated by a matrix-valued polynomial function and vector-valued polynomial function.

  • A sequential procedure for estimating a linear function of two normal means which satisfies the two requirements, i.e. one is a condition of coverage probability, the other is a condition of $\beta$-protection, is proposed when the variances are unknown and not necessarily equal. We give asymptotic behaviors of the proposed stopping time.

  • In this paper we investigate a generalization of the Hyers-Ulam-Rassias stability for a functional equation of the form $f(\varphi(X))\;=\;\phi(X)f(X)$, where X lie in n-variables. As a consequence, we obtain a stability result in the sense of Hyers, Ulam, Rassias, and Gavruta for many other equations such as the gamma, beta, Schroder, iterative, and G-function type's equations.

  • PALMIWON is composed of 8 oriental herbs which has been known to show some pharmacological effects in kidney, blood vessels and immune systems, and used for the treatment of kid ney disease, hypertension, nervous disease and diabetic mellitus in the Orient for a long time. Based on our previous report that PALMIWON showed different effects on immune cells and ${\beta}$-cells, the immunoreactivity of ICSA (Islet Cell Surface Antibody) with ${\beta}$-cell (RINm5F) and the cell proliferation and function of interleukin-1${\beta}$ damaged ${\beta}$-cells in the presence of PALMIWON were examined. It was observed that PALMIWON significantly inhibited the immunoreactivity of ICSA with ${\beta}$-cell, and markedly increased cell proliferation and insulin release of interleukin-1${\beta}$ damaged ${\beta}$-cells.

Momordica charantia Protects against Cytokine-induced Apoptosis in Pancreatic $\beta$-Cells

  • The unripe fruit of Momordica charantia (MC) has been shown to possess antidiabetic activity. However, the mechanism of its antidiabetic action has not been fully understood. In this study, the effects of the aqueous ethanolic extract of MC (AEE-MC) were evaluated on the apoptosis in pancreatic $\beta$-cells treated with a combination of the cytokines, interleukin (IL)-$1{\beta}$, tumor necrosis factor (TNF)-$\alpha$, and interferon (IFN)-$\gamma$. In MIN6N8 cells, the inhibitory effect of AEE-MC was significantly observed at 2 to 50 ${\mu}g/mL$: a 26.2 to 55.6% decrease of cytoplasmic DNA fragments quantified by an immunoassay. The molecular mechanisms, by which AEE-MC inhibited $\beta$-cell apoptosis, appeared to involve the inhibition on the expression of p21, Bax, and Bad, the up-regulation of Bcl-2 and Bcl-$X_L$, and the inhibition on the cleavage of caspase-9, -7, and -3 and poly (ADP-ribose) polymerase. This study suggests that MC may inhibit cytokine-induced apoptosis in $\beta$-cells and, thus, may contribute via this action to the antidiabetic influence in diabetes.

Impact of Cognitive Function and Cancer Coping on Quality of Life among Women with Post-chemotherapy Breast Cancer (항암화학요법을 받은 유방암 여성의 인지기능과 암 대처가 삶의 질에 미치는 영향)

  • Purpose: This study was done to identify effects of cognitive function and cancer coping on quality of life among women with breast cancer treated with antineoplastic agents. Methods: The study was correlational research and participants were 145 women with breast cancer who had received antineoplastic agents. Data were collected from October to November, 2015 via online replies. Cognitive function was measured with the Functional Assessment of Cancer Therapy-Cognitive Function Version-3 (FACT-Cog), cancer coping, with the Korean Cancer Coping Questionnaire (K-CCQ), and quality of life with the Functional Assessment of Cancer Therapy-Breast Version-4 (FACT-B). Data were analyzed using descriptive statistics, t-test, ANOVA, $Scheff{\acute{e}}$ test, ANCOVA, Bonferroni test, partial correlation coefficient, and hierarchical multiple regression with SPSS 21. Results: Cognitive functions, total individual coping, and interpersonal coping explained 42% of quality of life. Cognitive function (${\beta}=.35$, p<.001) was the best predictor of quality of life, followed by total individual coping (${\beta}=.34$, p<.001), and interpersonal coping (${\beta}=.26$, p<.001). Conclusion: Results indicate that cognitive function and cancer coping are meaningful factors for quality of life among breast cancer survivors. Therefore when developing intervention programs for these women, content on cognitive function and coping skills as well as coping resources should be included.

INEQUALITIES FOR THE NON-TANGENTIAL DERIVATIVE AT THE BOUNDARY FOR HOLOMORPHIC FUNCTION

  • In this paper, we present some inequalities for the non-tangential derivative of f(z). For the function $f(z)=z+b_{p+1}z^{p+1}+b_{p+2}z^{p+2}+{\cdots}$ defined in the unit disc, with ${\Re}\(\frac{f^{\prime}(z)}{{\lambda}f{\prime}(z)+1-{\lambda}}\)$ > ${\beta}$, $0{\leq}{\beta}$ < 1, $0{\leq}{\lambda}$ < 1, we estimate a module of a second non-tangential derivative of f(z) function at the boundary point ${\xi}$, by taking into account their first nonzero two Maclaurin coefficients. The sharpness of these estimates is also proved.
