Screening of Crude Drugs for Antioxidative Activity (생약추출물의 항산화 활성검색)

  • Based on DPPH radical scavenging activity and lipid peroxidation inhibitory activity, the MeOH extracts of 139 crude drugs were screened in order to search for antioxidants. Among tested samples, the extracts from the seed of Nelumbo nucifera, the fruit of Terminalia chebula var. gangetia, the root of Salvia miltiorhiza, the fruit of Ziziphus jujuba var. innermis, the root bark of Paeonia moutan, the fruit of Rubus coreanus, the fruit of Zanthoxylum schinifolium, the lignum of Caesalpinia sappan, the leaf of Pinus densiflora, the rhizome of Alpinia officinarum, the fruit of Syzygium aromaticum, the ramulus and uncus of Uncaria rhynchophylla, the root bark of Lycium chinense, and the fruit of Alpinia katsumadai showed a relatively strong antioxidative activity. Furthermore, the BuOH fraction from the extract of N. nucifera showed a potent activity in each assay. The isolation of bioactive compounds has been carried out and will be reported in the next paper.

Anti-hyperlipidemic Effect of Vitex doniana Ethanol Extract in Poloxamer Induced Hyperlipidemia

  • To determine the effect of Vitex doniana (leaves stem and root bark) ethanolic extracts on lipid profiles of Poloxamer 407 (P407) induced hyperlipidemic and normal rats. Fifty four mixed sex rats weighing 100-200g were divided into nine groups comprising six animals per group. At the end of the 21 day, the animals were sacrificed and blood sample were collected for determination of serum levels of: Total cholesterol (TC), Triacylglycerides (TAG), High-density lipoprotein cholesterol (HDL-c) and Low-density lipoprotein cholesterol (LDL-c). The studies showed that all induced treated groups significantly (P<0.05) lower serum levels of TC, TAG, LDL-c and significantly (P<0.05) increased HDL-c when compared to the P407 induced hyperlipidemic control. The normal treated groups showed no significant (P>0.05) difference in the serum levels of TC, TAG, LDL and HDL when compared to the normal control group. Calculation of atherogenic risk predictor indices of the induced treated groups showed that all the extracts significantly (P<0.05) lowered the LDL-c/HDL-c, log (TAG/HDL-c) and significantly (P<0.05) increased HDL-c/TC ratio when compared to the P407 induced hyperlipidemic control group. The atherogenic risk predictor indices of normal treated groups showed no significant difference (P>0.05) in LDL-c/HDL-c, Log (TAG/HDL-c) and HDL-c/TC ratio when compared to the normal control group. The study demonstrates the phytotherapeutic effect of Vitex doniana (leaves, stem and root bark) ethanolic extract in poloxamer 407 induced hyperlipidemia.

Biological Activity Investigation, and Phenol Compounds Isolation from Barks of Prunus Sargentii R. (산벚나무 수피추출물의 생리활성 및 페놀성 화합물 분리)

  • Prunus sargentii of Rosaceae familiy, has been reported to have radical scavenging activity and anti-inflammatory effect. On these facts, this research was conducted to evaluate pharmaceutical activities of the bark extracts P. sargentii. Free radical scavenging activities of fraction (Fr) -5${\sim}$10 isolated from P. sargentii was higher than 80% respectively at 10ppm. The superoxide dismutase (SOD)-like activity of Fr-5, 9 were about 97, 84%, respectively at 1,000 ppm. Xanthine oxidase inhibitory effect of Fr-9, 10 were about 75, 78%, respectively at 1,000 ppm. The tyrosinase inhibitory effect related to skin-whitening was 72, 68% in Fr-2, 9 isolated from Prunus sargentii R. at 1,000 ppm. Hyaluronidase inhibition activity related to the anti-inflammation effect was 98% for Fr-8 at 500 ppm. Isolation of the methanol soluble fraction from P. sargentii yielded two major phenol compounds, (-)-epicatechin and taxifolin. The structure of the compound was certainly determined by chemical analyses, as well as NMR and Mass spectroscopy. The present study was carried out in a search for new cosmetic material from the bark from P. sargentii. and, (-)-epicatechin and taxifolin were isolated as active principles. So P. sargentii R. methanolic extracts may be used for the cosmetic material.

Antifungal Activity of the Extracts of Zanthoxylum Schinifolium Sieb. et Zucc. against Dermatophytes (산초나무 추출물의 피부사상균에 대한 항균활성과 그 성분)

  • The antifungal activity of methanol extracts against dermatophytes was the highest at root-bark methanol extract, and the highest inhibitory effect was revealed in petroleum ether fraction of root-bark methanol extract. Compound I and compound II with significant antifungal activity were isolated from the fractions by silica gel column chromatography. As a result of the instrumental analyses, compound I and compound II were already known alkaloids. Compound I was identified as 4-methoxyfuro[2,3-6]quinoline (dictamnine ; $C_{12}H_9NO_2$) and compound II was identified as 4-methoxy-lmethyl-2(1H)-quinolinone ($C_{11}H_{11}NO_2$). The MIC of compound I against T. mentagrophytes and T. rubrum was $40{\mu}g/m\ell$ and the MIC of compound II against the same fungi was $800{\mu}g/m\ell$.

Antioxidant and Antimicrobial Activities of The Extracts from Native Camellia japonica in Korea (국내 자생 동백나무(Camellia japonica L.) 추출물의 항산화 및 항미생물 활성)

  • This research has been undertaken to increase availiability of native Camellia japonica leaf and flower in Korea as a edible-medicinal resource. Chemical compositions, antioxidant and antimicrobial activities in different parts of camellia were investigated. Crude protein contained the highest in young leaves(14.22%) but less than 10% in different parts. The contents of crude fat and crude ash were 60.48% in seeds and 5.16% in mature leaves, respectively. Methanol extract of young leaf, flower, and flower bud in camellia showed strong antioxidant activity compared with different parts. Also, antioxidant activity of these was higher than that of BHT, but weaker than that of VtC. Antioxidant activity of various parts in camellia were in order of young leaf &gt; flower bud &gt; flower &gt; mature leaf &gt; stem &gt; bark. Extracts of mature leaf and bark had a remarkable antibacterial activity(0 CFU/$m\ell$) on Bacillus subtilis. The young leaf extract displayed effective growth inhibition against B. subtilis, Candida albicans, and Trichosporon beigelii.

Alterations of Antioxidant Status and Mitochondrial Succinate Dehydrogenase Activity in the Liver of Wistar Strain Albino Rats Treated with by Ethanol Extracts of Annona senegalensis Pers (Annonaceae) Stem Bark

  • Numerous ethnomedicinal uses have been attributed to different parts of Annona senegalensis (ASE), including its uses as food and food additives. The present study investigated toxicological and antioxidant effects of 28 days administration of ethanol extracts of ASE stem bark to Wistar strain albino rats. Acute toxicity test was done to determine lethal dose in Wistar rats while sub-acute toxicity test was conducted on rats divided into four groups (A - control, B - 50 mg/kg, C - 100 mg/kg, D - 150 mg/kg, respectively and treated for 28 days. Oxidative stress markers in liver and kidney as well as hepatic succinate dehydrogenase activity in the mitochondrial and post mitochondrial fractions (PMF) were evaluated. The $LD_{50}$ value of ASE was > 2,000 mg/kg. White blood cell counts gradually increased, but red blood cell counts and haematocrits level decreased significantly (p < 0.05) by about 50%. Liver enzymes in the serum and mitochondrial succinate dehydrogenase activity increased significantly (p < 0.05). Superoxide dismutase and catalase activities also increased in liver mitochondria and PMF while malondialdehyde (MDA) and reduced glutathione levels increased only in the PMF. Furthermore, only MDA levels increased significantly in the kidney after 28 days extract administration. Histopathological examination showed hepatic necrosis and no obvious signs of nephrotoxicity. Anona senegalensis is relatively safe, but prolonged ingestion could induce oxidative stress and impair ATP synthesis through the modulation of the activity of mitochondrial succinate dehydrogenase.

Mucolytic Effects of Various Parts of FRUCTUS BENINCASAE Extracts in the Rat Trachea (백동과(白冬瓜)와 청동과(靑冬瓜)의 부위별(部位別) 추출물(抽出物)에 의(依)한 거담효과(祛痰效果)의 비교연구(比較硏究))

  • The in vivo effects of Fructus Benincasae (FB-Baekdongkwa(B) and FB- on the expectoration (decrease in sputum viscoelasticity) by their sorts and using rats (Sp. D. male, $150{\sim}160g$). FB was divided by seed, flesh, and bark, extracted by 95% ethanol for 3 hr. The extracts were given to rats administration and the following results were obtained: 1. When FB Recens-C extract was administered at the concentration of 300 mg/kg b.w., mucus secretion effect in the trachea was desirably stimulated. 2. The secretion of phenol red was increased in the FB-treated tracheas in the order of Semen Benincasae(SB)-B $(153{\pm}8\;%)$, FB-B $(149{\pm}10\;%)$, and FBR-C $(117{\pm}26\;%)$. In general, the effect of FB-B extract on phenol red secretion was stronger than that of FB-C. 3. When tracheobronchial lavage fluid was analyzed, the mucus secretion was relatively high $(111{\pm}14\;%)$ in FB-B compared with other extracts. 4. Microscopic analysis after direct treatment of the FB extracts to the rat tracheal tissue showed that all the FB extracts possessed no effects for the activity of the ciliary movement. 5. Glycoprotein content secreted by the seed extract of FB-B was increased compared with the control group, which represents the highest secretion effect of mucus. From the above results. we could conclude that the seed of SB-B possesses better activity for mucus secretion from trachea than the extracts of any other parts. Therefore, it is expected that the seed of SB-B may be available for the purpose of expectorant activity in the prescription of traditional medicine.

Isolation of Lipoxygenase Inhibitor from Indonesian Herb

  • A total of 20 extracts derived from different plant family commonly used in Indonesian traditional inflammation medicine were screened for their inhibitory effect on soybean lipoxygenase (SBL) and hyaluronidase (HAse) activity. Three methanol extracts, the bark of Cinnamomum burmanni (CB), the leaves of Piper betel (PB), and fruit of Barringtonia acutangula (BA) were found to have high inhibitory effects, whereas the methanol extract of the leaves of Mimusops elengi (ME) have medium inhibitory effect. The IC50 of CB, PB, BA and ME were found to be 21.7, 16.9, 39.1 and 62.8 g/$m\ell$, respectively. Among the tested extracts, only CB inhibited HAse (IC50 = 27g/$m\ell$). CB was successively fractionated with n-hexane, ethyl acetate, butanol and water. The EtOAc fraction having the strongest activity was fractionated and some compounds were isolated and purified by a preparative HPLC(Develosil ODS-HG-5 column). Coumarin and 2-hydroxy cinnamaldehyde. were identified through the analyses of UV-Vis absorption 1H-NMR, 13C-NMR and FAB+-MS spectra.

Anti-inflammatory activity of methanol extract isolated from stem bark of Albizia julibrissin

  • Albizia julibrissin (AJ) has been used widely as a traditional medicine. In macrophages nitric oxide (NO) is released as an inflammatory mediator and has been proposed to be an important modulator of many pathophysiological conditions including inflammation and carcinogenesis. In this study we have examined the NO inhibition effect of 85% methanol extracts of AJ in mouse macrophage. Lipopolysaccharide (LPS) has been reported to induce production of NO. Extracts of AJ (1, 10, $100{\mu}g/ml$) suppressed nitric oxide production in LPS-stimulated ($100{\mu}g/ml$) mouse (C57BL/6) macrophages and analyzed by ELISA. In addition, it also attenuated the expression of inflammatory products like Interleukin-6 (IL-6), cyclooxygenase-2 (COX-2), and inducible NOS (iNOS) as assessed by immunoblotting with specific antibodies. These results suggest that 85% methanol extracts of AJ would be useful in inflammatory diseases.

Screening of Thrombin Inhibitors from Medicinal and Wild Plants (약용 및 야생식물로부터 트롬빈 저해물질의 탐색)

  • Inhibitory activities of 264 methanol extracts, which were prepared from different parts of 210 kinds of wild and medicinal plants, against human thrombin were evaluated. Based on the anti-coagulation activity determined by thrombin time and activated partial thromboplastin time, the 14 extracts were screened. The fibrinolytic activity, heat stability and inhibition of other proteolytic digestive enzymes, such as pepsin, papain, trypsin and chymotrypsin, of the 14 extracts were further determined, and Ginko biloba (herba), Ephedra sinica (radix), Reynoutria elliptica (herba), Amomum tsao-ko Crevost (fructus), and Magnolia officinalis Rehd. et Wils (bark) were finally selected as possible plant sources for anti-thrombosis agent. These results suggested that medicinal and wild plants could be the potential source of thrombin inhibitor.
