Resistome Study in Aquatic Environments

  • Since the first discovery of antibiotics, introduction of new antibiotics has been coupled with the occurrence of antibiotic resistant bacteria (ARB) and antibiotic resistance genes (ARGs). Rapid dissemination of ARB and ARGs in the aquatic environments has become a global concern. ARB and ARGs have been already disseminated in the aquatic environments via various routes. Main hosts of most of ARGs were found to belong to Gammaproteobacteria class, including clinically important potential pathogens. Transmission of ARGs also occurs by horizontal gene transfer (HGT) mechanisms between bacterial strains in the aquatic environments, resulting in ubiquity of ARGs. Thus, a few of ARGs and MGEs (e.g., strA, sul1, int1) have been suggested as indicators for global comparability of contamination level in the aquatic environments. With ARB and ARGs contamination, the occurrence of critical pathogens has been globally issued due to their widespread in the aquatic environments. Thus, active surveillance systems have been launched worldwide. In this review, we described advancement of methodologies for ARGs detection, and occurrence of ARB and ARGs and their dissemination in the aquatic environments. Even though numerous studies have been conducted for ARB and ARGs, there is still no clear strategy to tackle antibiotic resistance (AR) in the aquatic environments. At least, for consistent surveillance, a strict framework should be established for further research in the aquatic environments.

수종의 역충전 재료의 치근단 밀폐력 비교 (COMPARISON OF THE SEALING ABILITY OF VARIOUS RETROGRADE FILLING MAIERIALS)

  • This study was performed to evaluate the sealing ability of various retrograde filling materials by using bacterial penetration and dye penetration test. One hundred and forty extracted human teeth with single, straight canals and mature apiece were collected and used for this study. All canals were instrumented using an engine driven Ni-Ti file (ProFile). After removing 3mm from the apex of tooth, a standardized 3mm root end cavity was prepared using an ultrasonic instrument. The 70 teeth were randomly divided into 7 groups : 6 groups for retrograde filling using Super-EBA, ZOE, Chelon-Silver, IRM, ZPC and amalgam. The 7th group was used as a negative control. Nail varnish was applied to all external root surfaces to the level of the reseated root ends to prevent lateral microleakages. The specimens were then sterilized in an ethylene oxide sterilizer for 24 hours. 2 mm of the reseated root was immersed in a culture chamber containing a Tripticase Soy Broth with a phenol red indicator. The coronal access of each specimen was inoculated every 72 hours with suspension of Proteus vulgaris. The culture media were observed every 24hours for color change indicating bacterial contamination. The specimens were observed for 4weeks. The remaining 70 teeth were submitted to a dye penetration test. The canals of all teeth were first sealed with AH26 and obturated using an Obtura II system. Root resection, root end preparation and retrograde filling was performed as above. All specimens were suspended in 2% methylene blue dye for 72 hours before being ion gitudinally split. The degree of dye penetration was then measured using a stereomicroscope at 10 magnification and evaluated. The results were as floows : 1. In the bacterial penetration, the degree of leakage was the lowest in the Super-EBA, followed by, in ascending order, ZOE, Chelon-Silver IRM and ZPC. The amalgam showed highest bacterial leakage of all(p<0.01). 2. In the dye penetration, the degree of microleakage was the lowest in the Chelon-Silver and Super-EBA, followed by, in ascending order, IRM, ZPC. The ZOE and amalgam showed the highest microleakage of all (p<0.05). These results suggested that the eugenol based cement, Super-EBA, have excellent sealing ability as a retrograde filling material.

Ultraviolet 및 건조 처리에 의한 마스크에 오염된 미생물 살균 효과 (Bactericidal Effect of Ultraviolet and Dry Treatment on Bacterial Contaminants in Facial Masks)

  • Due to the pandemic caused by COVID-19, the demand for face masks is soaring and has often caused a shortage. The aim of this study was to evaluate the effect of ultraviolet (UV) and drying treatments on microbial contaminants in facial masks. To conduct this study, standard procedures were designed to develop samples contaminated by the control bacteria Escherichia coli, Staphylococcus aureus, and Pseudomonas aeruginosa. The contamination level of the standard samples was approximately 6.30 × 106 CFU/ml, and the UV light treatment was performed 1, 3, 5, and 7 times. To evaluate the effect of the UV and drying treatments, the masks were first treated with UV 1, 2, and 3 times, followed by the drying process. As a result, the mask contaminated with E. coli and P. aeruginosa showed a bacterial rate of approximately 99.9% after 1 UV irradiation, and in the case of the S. aureus-contaminated mask, it exhibited a bactericidal rate of approximately 99.9% after 7 UV irradiations. However, when the drying process was included after UV irradiation, all the samples contaminated with E. coli, S. aureus, and P. aeruginosa showed a bactericidal rate of 99.9% or more. The results of this study suggest that UV and drying treatments can effectively reduce the bacterial contaminants in facial masks. In addition, these results provide fundamental data and appropriate sterilization methods for reusing masks.

Investigation of the groundwater contamination around landfill where slaughtered animals were buried

  • This study was designed to investigate if there were groundwater contamination in 17 landfill where slaughtered animals were buried during the crisis of 2002 foot-and-mouth-disease (FMD) outbreaks in Gyeonggi province. From March to August 2005 groundwater was collected once a month from 17 sites, and examined with potential for hydrogen (pH), colour, turbidity, lead (Pb), arsenic (As), mercury (Hg), cadmium (Cd), copper (Cu), zinc (Zn) , iron (Fe), manganese (Mn) , aluminium (Al), nitrate-nitrogen $(NO_3-N)$, ammonia-nitrogen $(NH_3-N)$, microbial pathogen and Escherichia spp. In the examination of $NH_3-N$ which of the mean concentration was from not-detected (ND) to 0.05 mg/l. The range of $NH_3-N$ level was $0.3-24.1mg/{\ell}$. However, groundwater from four sites was to go beyond the drinking water quality standard (DWQS), i.e., the mean concentration of those were $15.5mg/{\ell}\;(site\;1),\;20.7mg/{\ell}\;(site\;9),\;24.1mg/{\ell}\;(site\;13)\;and\;10.6mg/{\ell}\;(site\;17)$. In the investigation of pH, colour and turbidity, all of the pH were below of DWQS (pH 5.8-6.6), but one site in color test and four sites in turbidity test were over the standard level. Among 9 metal ions examined, Mn was in excess of DWQS, and its concentration was $2.4mg/{\ell}$. Pb, Cd, Hg and As were not traced. The contents of Cu, Zn, Fe and Al were $ND-0.22mg/{\ell},\;0.01-0.05mg/{\ell},\;ND-0.05mg/{\ell}\;and\;0.03-0.16mg/{\ell}$, respectively. Escherichiae spp were not identified, but bacterial colonies were detected at 3 groundwater including 2 sites over the DWQS at the level of $491CFU/m{\ell}\;(site\;4)\;and\;217CFU/m{\ell}\;(site\;15)$.

Activity of Essential Oils Against Bacillus subtilis Spores

  • Alternative methods for controlling bacterial endospore contamination are desired in a range of industries and applications. Attention has recently turned to natural products, such as essential oils, which have sporicidal activity. In this study, a selection of essential oils was investigated to identify those with activity against Bacillus subtilis spores. Spores were exposed to 13 essential oils, and surviving spores were enumerated. Cardamom, tea tree, and juniper leaf oils were the most effective, reducing the number of viable spores by 3 logs at concentrations above 1%. Sporicidal activity was enhanced at high temperatures ($60^{\circ}C$) or longer exposure times (up to 1 week). Gas chromatography-mass spectrometry analysis identified the components of the active essential oils. However, none of the major oil components exhibited equivalent activity to the whole oils. The fact that oil components, either alone or in combination, did not show the same level of sporicidal activity as the complete oils suggested that minor components may be involved, or that these act synergistically with major components. Scanning electron microscopy was used to examine spores after exposure to essential oils and suggested that leakage of spore contents was the likely mode of sporicidal action. Our data have shown that essential oils exert sporicidal activity and may be useful in applications where bacterial spore reduction is desired.

자외선을 이용한 Film Cassette의 소독 효과 (Disinfection Effect of Film Cassettes by Ultraviolet Irradiation)

  • 본 연구에서는 진단방사선과에서 사용하는 필름카세트에 자외선을 조사하여 소독효과를 알아보기 위해 필름카세트의 자외선 조사 전 후의 동정과 응고효소검사 (Coagulase test)와 선량을 측정하였다. 필름카세트에 자외선을 1분, 2분, 3분 간격으로 조사 후에 세균을 동정하여 자외선이 세균을 불활성화 하는데 자외선 조사시간은 2분 이상이 필요하다. 자외선 조사시의 선량을 30초 간격으로 측정하였다. 자외선 조사시의 30초에서 선량은 $1565 {\mu}W{\cdot}s/cm^2$ 이었다 자외선선량은 시간에 선량이 정비례하였다. 이를 통하여 병원감염의 예방과 병원감염관리의 활성화를 위한 기초자료를 마련하고, 병원감염을 보다 효과적으로 예방하기 위한 자외선 소독효과를 알고자하는데 목적이 있다.

시중 판매 후추의 오염도 및 회분식 광펄스 처리에 의한 살균 효과 (Contamination level of commercialized pepper and sterilization effect by intense pulsed light in batch system)

  • 시중에서 판매되고 있는 후추 29종을 수거하여 오염도를 조사한 결과, 전통시장에서 구입한 후추는 일반세균 $10^6-10^7$, Bacillus $10^4-10^5$, 효모와 곰팡이 $10^2$ 이하의 오염도를 보였고, 유기농 제품으로 판매되는 후추의 경우에는 일반세균 $10^4$, Bacillus $10^2-10^3$, 효모는 $10^1$ 이하의 오염도를 나타내었다. 대형마트에서 판매되고 있는 후추는 낮은 오염도를 나타내었으나 일부에서는 일반세균이 $10^3$ 정도의 오염도를 보였다. 전압 1,000 V, 펄스수 5 pps, 시료와 램프사이의 거리 4 cm의 조건에서 10분간 광펄스 처리하였을 경우 통후추는 1.45-1.55 log, 흑후추분말과 백후추분말은 0.8-0.85 log의 사멸율을 나타내었다. 살균 용기의 차이에 따른 살균율에는 유의적인 차이를 나타내지 않았다. 광펄스에 의한 후추의 살균율은 오존처리나 감압방전프라즈마 살균 등 다른 비가열 살균방법에 비해서는 높은 사멸율을 보여 산업적 적용 가능성을 보였으나, 추가적인 연구를 통해 살균 효과를 높일 수 있는 방법이 제시되어야 할 것으로 판단된다.

병원근무자 유니폼에 의한 병원 내 감염에 대한 체계적 문헌고찰 (The Association of Health Care Workers' Uniforms and Health Care-associated Infection: Systematic Review)

  • Purpose: To identify an associations between health care workers' uniforms and health care-associated infection. Methods: Electronic databases, including Ovid-Medline, the Cochrane Library, CINAHL, EMBASE, KMbase, and KoreaMed, were searched. The search terms included doctor, nurse, health care worker/staff/assistant, clothing, (white) gown, uniform, (neck)tie, and attire. Only papers published in English and Korean were included. Results: 16 studies were selected from 1,900 references screened. All of the studies were non-comparative studies except for one. Four were conducted with doctors, six with nurses, one with health care workers including physiotherapists and one for medical staff plus visitors in a neonatal intensive care unit. Doctors more frequently changed their uniforms than neckties; therefore, the degree of contamination was more serious in neckties. The cuff zone was more likely to be heavily contaminated than other areas of long-sleeve gowns. Coats become contaminated quickly once worn, and colony counts reached a similar level within the first few hours after wearing them. Wearing a plastic apron or protective clothing did not prevent the bacterial contamination of nurses' uniforms, and the best way to decrease the contamination was changing to newly laundered uniforms before starting every duty. Conclusion: Healthcare workers' uniforms are a potential source of health care-associated infection although there was no robust evidence. The government must establish standards for laundering of uniforms or a requirement for institutions to provide a laundering service for healthcare workers' uniforms.

유수의 세균오염도 조사 (A Study on Bacterial Contamination of Rainfall)

  • During April and May, 1973 rain water samples were collected five different times in nine selected areas of Seoul city. The pH factor and bacteria counts were measured. The following results were obtained. 1. The lowest pH was 6.18 at Moon Rae Dong and the highest was 6.30 at Jong Am Dong. This difference is not significant at the 0. 05 level. 2. The pH factor did not vary significantly with the time of sample collection. 3. Dae Bang Dong had the highest Total Plate Count of $1.9\times 10^3/ml$, Bo Moon Dong and An Kook Dong had the lowest Total Plate Count at $4.1\times 10^2/ml$. 4. The total plate count varied greatly according to the time of sample collection. This result can be used to remove are pollutants. 5. The Total plate Count is nearly inversely proportional to the length of time during which the sample is collected.

닭고기의 오염미생물 제거를 위한 감마선의 이용 (Irradiation of Chicken for the Improvement of Hygiene)

  • Gamma irradiation(1~10 kGy) was applied to chicken for the evaluation of their microbiological safety. In 3kGy-irradiated sample, the initial level of microbial contamination in chicken was 1.8$\times$10$^4$CFU/g (Coliforms), 2.2$\times$10$^3$CFU/g(Salmonella) and 2.7$\times$10$^3$CFU/g(Listeria), All kinds of contaminated microorganism were sterilized by 7kGy-irradiation. Also, irradiation followed by freeze storage at the same time was very effective in inhibiting bacterial growth. The pH was slightly increased during storage period and titratable acidity decreased. However, no significant changes in pH and acidity were observed by gamma irradiation.
