Analysis of Microflora Profile in Korean Traditional Nuruk

  • A variety of nuruk were collected from various provinces in Korea, and their microflora profiles were analyzed at the species level. A total of 42 nuruk samples were collected and when the viable cell numbers in these nuruk were enumerated, the average cell numbers of bacteria, fungi, yeast, and lactic acid bacteria from all nuruk were 7.21, 7.91, 3.49, and 4.88 log CFU/10 g, respectively. There were no significant differences in viable cell numbers of bacteria or fungi according to regions collected. Bacillus amyloliquefaciens and B. subtilis were the predominant bacterial strains in most samples. A significant portion, 13 out of 42 nuruk, contained foodborne pathogens such as B. cereus or Cronobacter sakazakii. There were various species of lactic acid bacteria such as Enterococcus faecium and Pediococcus pentosaceus in nuruk. It was unexpectedly found that only 13 among the 42 nuruk samples contained Aspergillus oryzae, the representative saccharifying fungi in makgeolli, whereas a fungi Lichtheimia corymbifera was widely distributed in nuruk. It was also found that Pichia jadinii was the predominant yeast strain in most nuruk, but the representative alcohol fermentation strain, Saccharomyces cerevisiae, was isolated from only 18 out of the 42 nuruk. These results suggested that a variety of species of fungi and yeast were distributed in nuruk and involved in the fermentation of makgeolli. In this study, a total of 64 bacterial species, 39 fugal species, and 15 yeast species were identified from nuruk. Among these strains, 37 bacterial species, 20 fungal species, and 8 yeast species were distributed less than 0.1%.

Multispecies probiotics alter fecal short-chain fatty acids and lactate levels in weaned pigs by modulating gut microbiota

  • Short-chain fatty acids (SCFAs) are metabolic products produced during the microbial fermentation of non-digestible fibers and play an important role in metabolic homeostasis and overall gut health. In this study, we investigated the effects of supplementation with multispecies probiotics (MSPs) containing Bacillus amyloliquefaciens, Limosilactobacillus reuteri, and Levilactobacillus brevis on the gut microbiota, and fecal SCFAs and lactate levels of weaned pigs. A total of 38 pigs weaned at 4 weeks of age were fed either a basal diet or a diet supplemented with MSPs for 6 weeks. MSP administration significantly increased the fecal concentrations of lactate (2.3-fold; p < 0.01), acetate (1.8-fold; p < 0.05), and formate (1.4-fold; p < 0.05). Moreover, MSP supplementation altered the gut microbiota of the pigs by significantly increasing the population of potentially beneficial bacteria such as Olsenella, Catonella, Catenibacterium, Acidaminococcus, and Ruminococcaceae. MSP supplementation also decreased the abundance of pathogenic bacteria such as Escherichia and Chlamydia. The modulation of the gut microbiota was observed to be strongly correlated with the changes in fecal SCFAs and lactate levels. Furthermore, we found changes in the functional pathways present within the gut, which supports our findings that MSP modulates the gut microbiota and SCFAs levels in pigs. The results support the potential use of MSPs to improve the gut health of animals by modulating SCFAs production.

Screening of Bacterial Antagonists to Develop an Effective Cocktail against Erwinia amylovora

  • Several types of chemical bactericides have been used to control fire blight. However, their excessive usage leads to environmental deterioration. Therefore, several researchers have analyzed antagonistic microorganisms as promising, effective, and safe biological control agents (BCAs). The primary aim of this study was to screen for potential antagonistic bacteria that suppress Erwinia amylovora. Among the 45 isolates studied, 5 strains showed the largest inhibition zone against E. amylovora. 16S rRNA gene sequencing identified them as Bacillus amyloliquefaciens (KPB 15), B. stratosphericus (KPB 21), B. altitudinis (KPB 25), B. safensis (KPB 31), and B. subtilis (KPB 39). KPB 25 and 31 reduced the lesion size of fire blight by 50% in immature apple fruits, and did not show antagonism against each other. Therefore, KPB 25 and 31 were selected to develop an antagonistic mixture against fire blight. Although the mixture with KPB 25 and 31 showed a slightly increased ability to reduce lesion size on immature fruits, they did not exhibit a synergistic effect in reducing E. amylovora population compared to each strain alone. Nevertheless, we have identified these two strains as useful and novel BCAs against fire blight with additional benefits safety and potential in developing a mixture without loss of their activity, owing to the absence of antagonism against each other.

솔잎, 약쑥추출물을 첨가한 초고추장의 품질평가 (Quality Evaluation by the Addition of Pine Needle and Artemisia princeps Extracts in Vinegared Kochujang)

  • 솔잎, 약쑥추출물을 첨가하여 제조한 초고추장을 3주 동안 $25^{\circ}C$, $37^{\circ}C$에 저장하면서 각각 2일, 1일 간격으로 분석 후 초고추장의 품질을 검토하였다. 우선, 초고추장에서 우점균주가 Bacillus amyloliquefaciens ER282 라는 것을 분리${\cdot}$동정 하였고 이에 대하여 솔잎, 약쑥추출물의 1, 3, 5, 10% 농도로 항균력 실험을 한 결과 대부분의 추출물에서 효과를 나타내었다. 초고추장 저장중의 총균수 변화는 저장온도와 관계없이 $25^{\circ}C$$37^{\circ}C$ 모두 무첨가구에 비해 현저히 감소하였다. pH와 산도의 변화는 $25^{\circ}C$, $37^{\circ}C$에서 무첨가구와 저장 기간에 따른 뚜렷한 차이가 없었다. 당도는 $25^{\circ}C$에 저장한 무첨가구가 초기 $67^{\circ}$Brix, 21일째 $77^{\circ}$Brix를 주정 3% 첨가구는 초기 $63^{\circ}$Brix, 21일째 $74^{\circ}$Brix로 나타났으며 추출물 첨가구는 초기 $61{\sim}74^{\circ}$Brix, 21일째 $67{\sim}78^{\circ}$Brix의 변화를 보여 약간 증가하는 양상을 보였다. $37^{\circ}C$에 저장한 무첨가구는 초기 $67^{\circ}$Brix, 21일째 $83^{\circ}$Brix를 주정 3% 첨가구는 초기 $63^{\circ}$Brix, 21일째 $73^{\circ}$Brix를 보였으며 추출물 첨가구는 $61{\sim}74^{\circ}$Brix, 21일째 $73{\sim}85^{\circ}$Brix로 나타나 $25^{\circ}C$와 마찬가지로 증가하였다. 색도는 솔잎, 약쑥추출물 대부분이 첨가량이 증가함에 따라 명도값은 증가하였으나 적색과 황색값은 크게 감소하였고 무첨가구와 주정 3%첨가구도 같은 경향이었다. 그러나 $25^{\circ}C$에서 주정 1%와 약쑥추출물 1% 첨가구가 21일이 되어도 저장 초기값과 비슷한 값을 나타내었고, $37^{\circ}C$에서는 약쑥추출물 1%, 약쑥추출물 3% 첨가구가 저장 초기값과 큰 차이를 보이지 않았다. 관능검사 결과 추출물 첨가구가 무첨가구 및 주정 3% 첨가구보다 기호도가 높은 편이였으나 저장기간에 따라 전체적인 기호도는 감소하는 경향이었다. 그러나 저장온도에 관계없이 솔잎추출물 3% 첨가구가 전체적인 기호도면에서 유의적으로 높은 기호도를 나타내었다.

오이 검은별무늬병 방제에 효과적인 유기농업자재 선발 (Screening for Effective Organic Farming Materials for the Control of Cucumber Scab Caused by Cladosporium cucumerinum)

  • 본 연구는 Cladosporium cucumerinum PT1 (KACC 48094)에 의해 발생하는 오이 검은별무늬병에 대하여 유기농업자재의 방제효과를 평가하기 위해 수행되었다. 43품목의 유기농업 자재를 대상으로 실내에서 항균활성 검정과 온실에서 오이 유묘에 포자현탁액 접종을 통한 유묘검정을 실시하였다. 13개 품목의 유기농업자재가 균사생장을 억제하였고, 이 중 9개 품목이 미생물 제제였다. 오이 유묘를 이용한 검정에서 6품목의 유기농업자재가 90% 이상의 우수한 방제효과를 나타냈다. 그 중에서 Bacillus amyloliquefaciens M27이 유효성분인 미생물 제제의 억제효과가 가장 뛰어났다. 그 다음으로 9개의 유기농업자재를 선발하여 예방적 및 치료적 효과를 알아보기 위해 적용하였는데, 예방적으로 처리했을 때 9개 자재 모두 병 발생을 상당히 억제하였다. 특히, 보르도액 I과 III은 방제가가 각각 96.7%, 73.3%로 훌륭한 예방효과를 나타냈다. 이 중 백리향+고삼 추출물이 함유된 자재에서만 방제가 가 50%로 다소 낮았지만 치료적 처리의 효과가 나타났다. 이 연구를 통해 시설 내 온화하고 습한 조건에서 잘 발생하는 오이 검은별무늬병은 유기농업자재로 방제될 수 있고 예방적 처리가 더욱 중요하고 효과적임이 확인되었다.

Bacillus amyloliquefaciens로 제조한 콩 발효물의 발효시간에 따른 품질 변화 (Changes in Quality of according to Fermentation Time of Fermented Soybean Produced Made with Bacillus amyloliquefaciens)

  • The purpose of this study was to evaluate the appearance, physicochemical, physical, and fermentation properties of the fermented soybean produced by manufacturing with inoculation the different types of microbial strains. The strains were inoculated by the NSI (natural strains inoculation), and the SSI (selective strain inoculation) were treatments. The appearance showed differences in color, viscous substance, and hardness depending on strains inoculation and fermentation duration. The pH, and total acidity were 6.40~7.26%, and 0.10~0.39% respectively with differences depending on the samples. The moisture content as the fermentation duration increased, the NSI (56.03~57.66%) decreased and the SSI (56.71~58.63%) increased. The physical characteristics of the hardness increased as the fermentation duration increased for the NSI and the SSI decreased. The color values for the L, a, and b values were 47.64~58.56, 7.15~9.08, and 12.41~17.30, respectively. The α-amylase and protease activities of the SSI were the highest among all treatments. The total viable cell counts of the fermented soybean products by strains were 5.02 to 9.77 log CFU/g, and SSI (fermentation, 48 hours) was the highest. The amino-type nitrogen contents of all samples were 301.62~746.97 mg% and the SSI showed the highest content. The amino acid had the highest glutamic acid content.

초고압처리가 생막걸리의 저장기간 연장에 미치는 영향 (Effect of High Hydrostatic Pressure Treatment on Extension of Shelf Life of Draft Makgeolli)

  • 생막걸리의 유통기간은 연장하고자 UV조사와 초고압처리를 하였다. UV조사는 막걸리내 미생물을 감소시키지 못하였으나 초고압처리는 급격하게 감소시켰다. 막걸리의 생균수는 2.2-5.8 × 107 CFU/mL이었으나 300MPa로 1분간 처리하였을 때 1.4-10 × 103 CFU/mL로 감소하였고 400MPa 처리에 의해서는 4-68 CFU/mL, 500MPa 처리에 의해서는 40CFU/mL이하로 감소하였다. 효모는 400MPa 압력에서 사멸하였고 Bacillus amyloliquefaciens와 Rummeliibacillus stabekisii 500MPa 압력에서도 생존하였다. 400과 500MPa의 압력으로 처리한 막걸리는 상온(25℃)에서 30일간 저장하여도 균체수가 증가하지 않았고 알코올, 산도, 아미노산도의 변화도 억제되었다. 그러나 30일 이상 상온에서 저장하면 막걸리의 산도와 아미노산도가 증가하면서 음용이 어려웠다. 저온(4℃)저장에서는 저장 70일까지 막걸리의 품질변화를 억제시켜 신선한 막걸리와 유사한 맛을 가지고 있었다.

Growth Performance and Antibody Response of Broiler Chicks Fed Yeast Derived β-Glucan and Single-strain Probiotics

  • A study was conducted to evaluate the effects of dietary yeast derived ${\beta}$-glucan and single-strain probiotics on the growth performance and antibody response in broiler chicks. Six hundred and thirty 1-d-old male broiler chicks were divided into seven groups, placed into three pens per group (30 birds per pen) and fed one of seven non-medicated corn-SBM based experimental diets containing 0.025, 0.05 or 0.1% Saccharomyces cerevisiae ${\beta}$-glucan and 0.05, 0.1 or 0.2% Bacillus amyloliquefaciens (BA-pro, $1.3{\times}10^9/g$) or devoid of them for 5 wk. The body weight gains in groups fed diets containing 0.025 or 0.1% ${\beta}$-glucan, 0.1% or 0.2% BA-pro were significantly higher (p<0.05) than the control over 1-35 d. Feed conversion rates of groups fed ${\beta}$-glucan and BA-pro tended to be improved compared to the control group. There were no significant differences in the relative weights of liver, abdominal fat and breast muscle. No significant differences were observed in the activities of serum enzymes and concentrations of various cholesterol fractions. The antibody titers against Newcastle disease or infectious bronchitis virus in the chicks fed diets containing ${\beta}$-glucan and BA-pro were significantly higher (p<0.05) than in the control. The concentrations of cecal lactic acid bacteria in all groups fed BA-pro were significantly increased (p<0.05) compared to the control. These results indicated that dietary yeast derived ${\beta}$-glucan and BA-pro exerted growth-promoting and immune-enhancing effects in broiler chickens. In addition, BA-pro added to the diets modulated the profiles of cecal microflora, reflecting a potential to be beneficial microorganisms in chickens.

Effects of dietary supplementation with Taiwanese tea byproducts and probiotics on growth performance, lipid metabolism, and the immune response in red feather native chickens

  • Objective: This study compared the catechin composition of different tea byproducts and investigated the effects of dietary supplementation with green tea byproducts on the accumulation of abdominal fat, the modulation of lipid metabolism, and the inflammatory response in red feather native chickens. Methods: Bioactive compounds were detected, and in vitro anti-obesity capacity analyzed via 3T3-L1 preadipocytes. In animal experiments, 320 one-day-old red feather native chickens were divided into 4 treatment groups: control, basal diet supplemented with 0.5% Jinxuan byproduct (JBP), basal diet supplemented with 1% JBP, or basal diet supplemented with 5×106 colony-forming unit (CFU)/kg Bacillus amyloliquefaciens+5×106 CFU/kg Saccharomyces cerevisiae (BA+SC). Growth performance, serum characteristics, carcass characteristics, and the mRNA expression of selected genes were measured. Results: This study compared several cultivars of tea, but Jinxuan showed the highest levels of the anti-obesity compound epigallocatechin gallate. 3T3-L1 preadipocytes treated with Jinxuan extract significantly reduced lipid accumulation. There were no significant differences in growth performance, serum characteristics, or carcass characteristics among the groups. However, in the 0.5% JBP group, mRNA expression of fatty acid synthase (FAS) and acetyl-CoA carboxylase (ACC) were significantly decreased. In the 1% JBP group, FAS, ACC and peroxisome proliferator-activated receptor γ levels were significantly decreased. Moreover, inflammation-related mRNA expression levels were decreased by the addition of JBP. Conclusion: JBP contained abundant catechins and related bioactive compounds, which reduced lipid accumulation in 3T3-L1 preadipocytes, however there was no significant reduction in abdominal fat. This may be due to a lack of active anti-obesity compounds or because the major changes in fat metabolism were not in the abdomen. Nonetheless, lipogenesis-related and inflammation-related mRNA expression were reduced in the 1% JBP group. In addition, dietary supplementation with tea byproducts could reduce the massive amount of byproducts created during tea production and modulate lipid metabolism and the inflammatory response in chickens.

발효콩 유지의 3T3-L1 지방전구세포와 고지방식이를 급여한 C57BL/6J 생쥐에 대한 항비만 효과 (Anti-Obesity Effects of Fermented Soybean Oils in 3T3-L1 Pre-Adipocytes and High Fat Diet-Fed C57BL/6J Mice)

  • 본 연구에서는 발효콩 유지가 비만 억제 및 예방, 3T3-L1 지방전구세포의 성장 및 분화 억제 효과를 관찰하기 위해 실험을 실시하였다. 3T3-L1 지방전구세포에 발효콩 유지를 처리하여 세포생존율을 측정한 결과, NF군 $100{\mu}g/mL$에서 3T3-L1 지방전구세포의 생존율을 유의적으로 감소시켜 세포독성이 나타났다. 분화를 유도한 다음 세포 내 triglyceride 함량을 측정한 결과, 비발효콩 유지(NF) 처리군보다 발효콩 유지처리군에서 triglyceride 함량 저해 효과가 높게 나타났으며, 특히 BS, LBA, BLO군 순으로 높게 나타났다. 또한, BS군, LBA군, BLO군 순으로 지방세포분화 관련 유전자인 $PPAR{\gamma}$의 mRNA 발현을 농도 의존적으로 감소시켰으며, $PPAR{\gamma}$ 유전자와 상관관계에 있는 $C/EBP{\alpha}$ 유전자도 농도 의존적으로 감소시켰다. 그리고 adiponectin 유전자의 발현은 농도 의존적으로 증가시켰다. 고지방식이로 비만을 유도한 C57BL/6J 생쥐를 이용하여 항비만 활성을 관찰한 결과, 총 체중증가량은 대조군에 비해 4주째에 감소하는 경향이 나타났다. 혈액 내 지질농도를 측정한 결과 중성지방, 총콜레스테롤, LDL-콜레스테롤 함량은 LBA, BLO군에서 유의적으로 낮게 나타났으며(P<0.05), HDL-콜레스테롤 함량은 대조군보다 증가하였으나 유의적인 차이는 나타나지 않았다. 동맥경화 지수인 AI(atherogenic index)는 대조군보다 감소시키는 경향을 보였다. 비만관련 호르몬인 adiponectin의 농도는 SO, BS, LBA, BLO군에서 유의적으로 증가하였고(P<0.05), insulin 농도는 유의적으로 감소하였다(P<0.05). Leptin은 대조군보다 감소하였으나 유의적인 차이는 나타나지 않았다. 따라서 이러한 결과를 종합하여 볼 때 발효콩 유지가 3T3-L1 지방전구세포의 mRNA 단계에서부터 지방분화를 억제하여 항비만 활성을 가지고 있는 것으로 추정되고, 고지방식이로 비만이 유도된 C57BL/6J 생쥐에서 총체중을 감소시키고, 혈액 내 지질농도인 triglyceride와 총콜레스테롤, LDL-콜레스테롤 및 HDL-콜레스테롤과 비만관련 호르몬인 adiponectin, insulin, leptin을 조절하여 항비만 활성을 가지고 있는 것으로 확인되었다. 이 결과로 발효콩 유지가 항비만 생리활성을 나타내는 새로운 식품 소재로서 이를 활용한 기능성 식품 개발의 가능성이 있다고 생각된다.
