Scavenging Effects of Free Radicals and Inhibitory Effects of Lipid Peroxidation of Bupleury Radix Aqua-Acupuncture Solution in Vitro (시호 약침제제의 자유기 소거능 및 지질과산화 억제효능에 관한 연구)

  • Bupleury radix has been used for the treatment of fever, liver disease, inflammation in traditional medicine. The present study was carried out to evaluate the antioxidant effects of Bupleury radix aqua-acupuncture solution (BRAS) in vitro. Oxygen derived free radicals produced in the course of normal aerobic life, such as superoxide anion radical($O_2^-$ ), hydroxyl radicaI( OH), hydrogen peroxide($H_2O_2$) and singlet oxygen($^1O_2$) can attack polyunsaturated fatty acid in cell membranes, enzymes, other cell compounds, and give rise to lipid peroxidation, DNA damage, lipofuscin accumulation, structure alteration of cell membrane and cell death. In this study, antioxidant effects of BRAS on lipid peroxidation were determined according to the method of TBA. BRAS inhibited markedly peroxidation of linoleic acid during the autoxidation, and also inhibited lipid peroxidation induced by hydroxyl radical derived from $H_2O_2-Fe^{2+}$ in rat liver homogenate. And BRAS showed 30% scavenging effect on DPPH radical, also exhibited a 30% inhibitory effect on superoxide generation from xanthine-xanthine oxidase system. In addition, BRAS protected the cell death induced by tert-butyl hydroperoxide(t-BHP) and significantly increased cell viability in the normal rat liver cell(Ac2F).

Studies on the Lipid Components of Red Pepper Seed oil (고추씨 기름의 지질성분에 관한 연구)

  • This studies were conducted to find out the possibility of utilizing red pepper seed as resources of food fats, the research method was designed to make a comparison between crude and refined oil, and the results of the studies are as follows : The red pepper seed contained 28% of crude fat and 21% of crude protein. The main fatty acids of red pepper seed oil were linoleic acid(72.10~72.31%), palmitic acid(12.81~13.28%) and oleic acid(9.47~10.48%). The linolenic acid content was so small that is will not influence the autoxidation of the red pepper seed oil. The major triglyceride type of crude and refined oil of red pepper seeds were C52 and C54. The other types were found in a small quantity. The sterol composition of crude oil was $\beta$-sitosterol, campasterol, stigmasteral and brassicasterol,in the quantity order. after refining, brassicasterol was not detected, and the content was decreased by one six and one eight. The toropherol composition of crude and refined oil, tocopherol analog was composed of three kinds $\alpha$-, ${\gamma}$-, $\delta$-, but no $\beta$-form. the quantity of ${\gamma}$-, $\alpha$- and $\delta$-tocopherol were 162.91, 83.72, 43.98mg% respectively. The Quantity of and capsaicin in crude oil was 1,296 ppm, and it was reduced consicerably by refining and removed completely after the process of redeodorization.

  • Doenjang(fermented soybean paste) was prepared by the series of processes including soaking, cooking, first fermentation(3 days at 30$\pm$2$^{\circ}C$) for the preparation of meju(soybean koji) after inoclulation of Aspergillus oryzae, and further fermentation (60 days at 30$\pm$2$^{\circ}C$) for the ripening after addition of salt 13% to meju. the crude phenolics extracted from defatted soybean and doenjang were fractionated onto the neutral phenolics(isofavonoids) and acidic phenolics(phenolic acids), respectively. Composition and antioxidative characteristics of phenolic fractions were determined. The neutral phenolic fractions contained genistin, genistein and daidzein; on the other hand, acidic phenolic fractions had syringic acid and seven other components. The content of genistin in doenjang dramatically decreased at the early stage of fermentation, whereas the content of genistein rather increased. In addition, the content of syringic acid of acidic phenolic fractions were increased during fermentation. These changes in individual phenolic components affected the antioxidative activity of neutral phenolics or acidic phenolics. antioxidative activity of phenolic compounds were evaluated during soybean fermentation. The antioxidative and free radical scavenging activity of neutral phenolic fractions and acidic phenolic fractions on linoleic acid autoxidation were also investigated.

Free Radical Scavenging and Antioxidative Activities by Ethanol Extract from Capsella bursa-pastoris (냉이(Capsella bursa-pastoris) 에탄올 추출물의 유리라디칼 소거 및 항산화 활성)

  • Screening was performed on edible natural sources to examine superoxide radical scavenging activities by using the method of superoxide dismutase assay. Among 47 kinds of samples, the extract of Capsella bursa-pastoris showed a potent superoxide radical scavenging activity of 11.6 unit / mg solid and was selected for further studies. In order to select optimal extraction solvent system, Cappella bursa-pastoris was extracted with various solvents and the electron donating abilities and inhibitory effects on lipid peroxidation of linoleic acid were measured. Among them, the ethanol extract of Capsella bursa-pastoris possessed the highest level of activities. The ethanol extract of Cappella bursa-pastoris was found to have an inhibitory effect on autoxidation of soybean oil at 5$0^{\circ}C$ from peroxide value, conjugated diene value and refractive index. In the soybean oil containing 0.2% of ethanol extract, induction period was increased 2 times in comparison with control.

Antioxidative Components of Pueraria Root (칡뿌리의 항산화 성분)

  • As a search for natural antioxidants, antioxidative fractions in pueraria root were extracted and identified using column chromatography, thin layer chromatography and high performance liquid chromatography. Components which have most effective antioxidative activities were futher identified by IR and GC/MS. The strongest antioxidative component of pueraria root methanol extract was identified as puerarin. Puerarin obtained from pueraria root was practically effective as antioxidant at the level of 100 ppm. Antioxidative activity of the puerarin was higher in linoleic acid-water system than in a linoleic acid substrate. Puerarin, daidzin and daidzein contents in pueraria root juice were 0.39%, 0.45% and 0,03%, respectively.

  • Antioxidant effect of reconstituted liposome was studied for application to food and cosmetic industry. Vitamin-C/alginate alginate gel-entrapped DHA-PC liposome (AVDL) was prepared and studied with respect to morphology (TEM), loading efficiency (TLC) and antioxidant effect (TBARS assay). AVDL has spherical structure and 80 ∼ 250 ㎚ size. As DHA content increased, DHA and PC loading efficiency and antioxidant effect was increased. At addition of 0.2 % vitamin C in AVDL, DHA auto-oxidation was minimized and sustained about 3 days.

  • DMHF (2.5-dimethyl-4-hydroxy-3[2H]-furanone), an antioxidative compound from the reaction of L-cysteine/D-glucose scavenged efficiently 1,1-diphenyl-2-picryl hydrazyl free radicals. It exhibited an inhibitory effect on the autoxidation of linolenic acid, and the protective effect against UV cytotoxicity in cultured human fibroblast. In addition, DMHF appeared to prevent the cellular melanogenesis in the cultured murine melanoma cells more effectively than kojic acid, a well known inhibitor of melanogenesis, while the former was not so effective as the latter for the inhibition of the tyrosinase. Considering that cellular melanogenesis is a metabolic process triggered by oxidative stress, it ovas tentatively deduced that the antioxidative property of DMHF might afford the effect against cellular pigmentation by alleviating the causative stress. In toxicological tests such as irritation and sensitization, this compound turned out to be safe. The results of this study suggest that DMHF may be a novel inhibitor of melanogenesis, and that night be useful for application in cosmetics.

Antioxidative Activity and Nitrite Scavenging Ability of Methanol Extract from Ricinus communis (아주까리 메탄을 추출물의 항산화 효과 및 아질산염 소거작용)

  • Efficacy of antioxidative activity and nitrite scavenging ability of methanol extract from Ricinus communis. was investigated. Electron-donating ability of extract at RC$_{50}$ was 114.02 ${\mu}g$/mL. After addition of 0.46 mg/mL extract, autoxidation of pyrogallol decreased to 32.99% by superoxide dismutase-like activity. In antioxidative activity of extract against linoleic acid during incubation times of 24, 48, 96 hours at 40$^{\circ}C$, lipid peroxidation values significantly decreased by 85.50%, 87.77%, 90.95% with addition of 0.2 mg/mL, respectively. Total phenolic content was determined as gallic acid equivalents (GAE) and values revealed 83.98 ${\pm}$ 5.66 GAE ${\mu}g$/mg of extract. Nitrite scavenging ability showed the most remarkable effect at pH 1.2, decreasing to 47.24% by addition of 0.2 mg/mL. These results suggest that methanol extract from Ricinus communis. can be used as bioactive and functional material.

Antioxidant and ACE Inhibitory Activities of Soybean Hydrolysates: Effect of Enzyme and Degree of Hydrolysis

  • Native soy protein isolate (SPI) was hydrolyzed with 4 different proteolytic enzymes, including bromelain, papain, Neutrase, and Flavourzyme. SPI hydrolysates with the degree of hydrolysis (DH) in range of 6 to 15% were prepared by each enzyme. The angiotensin 1 converting enzyme (ACE) inhibitory and the antioxidant activities of the SPI hydrolysates, such as superoxide dismutase-like activity and inhibition of the linoleic acid autoxidation, were evaluated. Overall, as the DH increased, all evaluated bioactivities of the SPI hydrolysates significantly increased. The significantly highest ACE inhibitory and antioxidant activities were found in hydrolysates made with papain and bromelain, respectively. SPI hydrolysates by Flavourzyme showed the significantly lowest activity in all tested bioactivities. The results suggested that ACE inhibitory and antioxidant activities of SPI hydrolysates were determined by the DH and by the enzyme used.

Antioxidative Activity of Acylated Anthocyanin Isolated from Fruit and Vegetables

  • The antioxidative activity of seven different acylated anthocyanin pigments isolated from grape, sweet potato, eggplant and red cabbage was evaluated bvy using linoleic acid autoxidation and rat liver microsomal systems. The acylated anthocyanins were isolated and purified by Amberlite SAD-7, ODS and Sephadex LH-20 column omatography, and preparative HPLC. Most of acylated anthocyanisns exhibited antioxidative activity as strong as $\alpha$-tocoperol, and especially peonidin3-O--(6-O-trans-caffeyl)-2-O-(6-O-trans-feuloylglucopyranosyl)-$\beta$-D-glucopyranosyl-5-O-$\beta$-D-glucopyranoside from purple sweet potato showed the strongest activity, comparable to BHA (not significant, p<0.05) in the linoleic acid system. Meanwhile, two acylated anthocyanins from the pericarps of grape and eggplant inhibited considerably the MDA formation from rat liver microsomal lipid peroxidation induced by FeSO$_4$/$H_2O$$_2$. In particular, malvidin 3-O-(6-O-p-coumaroyl)-$\beta$-D-glucopyranosde from grape pericarps showed the strongest antioxidant activity, comparable to $\alpha$-tocopherol (not significant, p<0.05). These results suggest that the acylated anthocyanins from fruits and vegetables can be used as potential dietary ntioxidants and natural colorants.
