We investigated the effects of newly synthesized 4-[(butylsulfinyl)methyl]-1,2-benzenediol (SMBD) on the prevention of atherosclerosis in hypercholesterolemic rabbits. SMBD exhibited stronger inhibition of $Cu^{2+}$-induced low-density lipoprotein oxidation than that of ascorbic acid or simvastatin. Three-month-old rabbits were fed an atherogenic diet containing 0.5% cholesterol and 10% coconut oil, while other two groups were given an atherogenic diet with intravenous injection of either simvastatin or SMBD (0.33 mg/kg/day) for 4 weeks. The concentrations of plasma cholesterol and thiobarbituric acid reactive substances were significantly decreased in SMBD groups, compared to the control group. Also, aortic lipid level in the SMBD group significantly lower than that in the control group. Furthermore, compared with the control group, the SMBD group significantly inhibited the increase of aortic intimal thickness by 36% via reducing of aortic reactive oxygen species and cyclooxygenase-2 protein levels. We conclude that raised antioxidant effect of SMBD results in significant prevention against hypercholesterolemia.
The wild edible mushroom, Lentinus lepideus has recently been cultivated for commercial use in Korea. While the mushroom has been widely used for nutritional and medicinal purposes, the possible anti-hyperlipidemic action is unclear. The effects of dietary L. lepideus on plasma and feces biochemical and on the liver histological status were investigated in hypercholesterolemic rats. Six-wk-old female Sprague-Dawley albino rats were divided into three groups of 10 rats each. Biochemical and histological examinations were performed. A diet containing 5% L. lepideus fruiting bodies reduced plasma total cholesterol, triglyceride, low-density lipoprotein, total lipid, phospholipids, and the ratio of low-density to high-density lipoprotein. Body weight was reduced. The diet did not adversely affect plasma biochemical and enzyme profiles. L. lepideus reduced significantly plasma ${\beta}$- and pre-${\beta}$-lipoprotein, while ${\alpha}$-lipoprotein content was increased. A histological study of hepatic cells by conventional hematoxylin-eosin and oil red O staining revealed normal findings for mushroom-fed hypercholesterolemic rats. The present study suggests that a diet supplemented with L. lepideus can provide health benefits by acting on the atherogenic lipid profile in hypercholesterolemic rats.

This study was performed to investigate the effect of dietary crab shell powder on lipid metabolism in diet induced hyperlippidemic rats. Sixteen male Sprague-Dawley rats were divided into two groups and fed 20% (w/w) carb shell powder supplemented diet for 4 weeks. Energy intake and weight gain were lower in experimental group than in control group. Concentrations of plasma triglyceride, total cholesterol, LDL-cholesterol, and atherogenic index (AI) were decreased and concentration of HDL-cholesterol was increased by crab shell powder supplementation. Concentrations of liver total cholesterol and triglyceride were significantly lower in experimental group. The crab shell decreased triglyceride and cholesterol concentration in plasma and liver, and increased bile acid excretion in feces. Therefore, the crab shell powder is assumed to exhibit function of chitin or chitosan that effects on lipid metabolism and cholesterol reabsorption.

The effects of chitin and chitosan on the contents of plasma and liver lipids and the fecal excretion of neutral and acidic steroids were investigated in rats fed with atherogenic diet. Dietary chitin did not affect plasma total cholesterol, LDL cholesterol and HDL cholesterol, while chitosan diet de creased total cholesterol and LDL cholesterol. Plasma content of triglyceride was reduced signif icantly by chitin diet but not by chitosan. Increases in fecal weight were observed in rats fed with either chitin or chitosan, and the increase was greater with chitosan than with chitin. Dietary chitosan resulted in an increase of fecal excretion of neutral steroids without any changes in the excretion of acidic sterols. Putting the results together, dietary chitosan reduces plasma cholesterol probably due to the reduction of cholesterol absorption. The decrease in plasma triglyceride level by dietary chitin requires further research to understand the mechinism.

The purpose of the study was to find an effect of medium chain triglyceride (MCT)diet on the serum lipids and lipoprotein of Sprague-Dawley rats when the were fed with cholesterol. All experiment-groups were fed with different diet such as soybean oil, shortening,MCT and MCT mixed with oil respectively. After fed to each group, the levels of serum lipids and lipoprotein among fed rats were examined and the data from the experiment were compared with a control group. The results obtained from the study are as follows : (1) The average boby weight gain in MCT diet and soybean oil diet groups were lighter than that of the control group, while the food efficiency ratio in each experimental diet group was almost same as than in the control group. (2) Total cholesterol levels in serum of all experimental diet groups were lower than that of the control group except the level in shortening diet group. Especially, the levels of total cholesterol in MCT diet and soybean oil diet groups were significantly lower than that of the control group. (3) VLDL, LDL-cholesterol levels of all experimental groups were almost same except the case of the MCT diet and soybean oil diet group which was significantly low. (4) The ratio of VLDL, LDL-cholesterol to HDL-cholesterol, so called, atherogenic index in serum of MCT and soybean oil diet groups were significantly lower than that of the control group. (5) The ratio of total cholesterol to phospholipids in serum of MCT diet group was decreased, because total cholesterol levels were decreased more than phospolipid levels. (6) The ratio of the serum VLDL, LDL to HDL in MCT diet group. in MCT and soybean oil mixed diet group, and in soybean diet group was decreased. In other wards, it means that the decrease of the serum VLDL, LDL in each group and the increase of the serum HDL.

The purpose of this study was to investigate the effects of dietary xylooligosaccharide on lipid levels of serum in rats fed high cholesterol diet. Male Sprage-Dawley rats weiging 100$\pm$10g were randomly assigned to groups of two normal(N, N+10X) and four high cholesterol diets which contained 1%(w/w) cholesterol. High cholesterol diet groups were classified to xylooligosaccharide free diet (C group), 5% xylooligosaccharide diet(C+5X group), 10% xylooligosaccharide diet(C+10X group) and 15% xylooligosaccharide diet(C+15X group) according to the level of dietary xylooligosaccharide supplementation. These experimental diets were fed ad libidum for 4 weeks. The weight gain of high cholesterol diet group were significantly increased more than that of normal group, but those of 10% and 15% dietary xylooligosaccharide groups were significantly decreased more than that of high cholesterol diet(C)group. The higher content of xylooligosaccharide, the more food intake was increased. The food efficiencies of 10%, 15% cholesterol diet groups were lower than that of high cholesterol diet(C)group. The levels of serum triglyceride(TG) and total cholestoral were significantly high in cholesterol diet groups compared with normal diet group but were decreased in groups fed 5% and 10% dietary xylooligosaccharide. Especially, the lowest level showed in group fed high cholesterol diet containing 10% xylooligosacchride. High cholesterol diet group containing 10% xylooligosaccharide increased HDL-cholesterol level and then decreased LDL-cholesterol level and atherogenic index compared with other groups. The level of TBARS(thiobarbituric acid reactive substances) in serum was decreased in rat group fed xylooligosaccharide in high cholesterol diet. The higher content of xylooligosacchride, the more gastrointestinal transit time was decreased. The results indicate that dietary xylooligosaccharide can improve status of TG and total cholesterol and repress lipid peroxidation in serum lipid at hypercholesterolemia induced by high cholesterol diet.
Effects of dietary supplementation of Pleurotus eryngii(PE) to a diet containig the cholesterol on lipid levels in the liver and serum, and enzyme activities of rats were studied by feeding 8-weeks old male rats with the four diets for 4 weeks, respectively: normal diet, control diet (10% fat + 0.5% cholesterol), 3% and 5% PE diets (control diet + 3% and 5% PE dry powder). The body weight gains and food efficiency ratios of the 3% and 5% PE diet groups were significantly lower than those of the control group. The liver and epididymal fat pad weights, and hepatic triglyceride Levels of the 5% PE diet group were more significantly decreased than those of the control group. The concentrations in serum triglyceride, total cholesterol, LDL+VLDL-cholesterol, and atherogenic index ratios were significantly lower in the 3% or 5% PE groups compared to those of the control group. Ratio of HDL-cholesterol to total-cholesterol was significantly higher in the 3% and 5% PE groups compared to that of the control group. The fecal total lipid excretion of the 5% PE diet group was more significantly decreased than that of the control group. There was no significant difference in the hepatic cholesterol, and the serum HDL-cholesterol concentrations, and the GOT, GPT and ${\gamma}-GTP$ activities among the experimental groups. These results showed that the Pleurotus eryngii powder feeding decreased the total cholesterol, LDL-cholesterol and atherogenic index, and increased the ratio of HDL-cholesterol to total-cholesterol in serum of the rats.

The present study examined the lipid lowering and antioxidant activity of Paecilomyces tenuipes. Eight week-old male Sprague-Dawley rats were fed one of the three diets, a reference diet without cholesterol addition (NC), a high fat (17 g/ 100 g)-high cholesterol (1 g/100 g) diet (HC) and a HC diet supplemented with 3% P. tenuipes (PT) for 30 days. Total lipid and total cholesterol were reduced significantly by 33 and 37%, respectively, in the PT diet group compared with controls. A similar reduction was found for low density lipoprotein (LDL) cholesterol levels, while plasma high density lipoprotein (HDL) cholesterol and triglyceride (TG) concentrations were not significantly different among groups. Hepatic total lipid and total cholesterol levels, but not hepatic TG levels, were significantly decreased in the PT group compared to the HC group. The administration of P. tenuipes increased the plasma total antioxidant potential and decreased the levels of lipid peroxidation. These results suggest that P. tenuipes exerts significant health benefits through the modulation of physiological functions including a variety of atherogenic lipid profiles and antioxidants in hypercholesterolemia.

Objective & Methods : The present study was to investigate the effect of Moutan Cortex Radicis Herbal-acupuncture (MCR-HA) at GB34 (Yangleungchean) on hyperlipidemia and arteriosclerosis induced by high fat diet in rats. Results : 1. Moutan Cortex Radicis herbal-acupuncture infusion solution (MCR-HAS) increased DPPH radical scavenging activity and HMG-CoA reductase inhibition rate in rat liver cells. 2. MCR-HA at GB34 decreased serum total cholesterol level, and increased serum phospholipid/total cholesterol in rats with high fat diet. 3. MCR-HA at GB34 decreased atherogenic index in rats with high fat diet. 4. MCR-HA at GB34 increased hepatic catalase activity in rats with high fat diet. Conclusion : These results suggested that Moutan Cortex Radicis herbal-acupuncture at GB34 has therapeutic effects on hyperlipidemia and related complications in rats with high fat diet.
Objective: The antiplatelet agent aspirin has been widely used for treating atherosclerosis in western medicine, and its efficacy has been proven in cardiac and extracardiac vascular diseases. On the other hand, Hyeolbuchukeo-tang has been widely used for treating blood stasis syndrome in traditional medicine. Therefore we investigated whether Hyeolbuchukeo-tang could have a synergic effect along with aspirin. Methods & Materials: Male $ApoE^{(-/-)}$ mice were randomly divided into three different experimental groups: a non-treated group(Control group), an aspirin-treated group(AP group), and an aspirin with Hyeolbuchukeo-tang-treated group(APH group). The control group was fed only an atherogenic diet, the AP group an atherogenic diet plus Aspirin 5 mg/kg, and the APH group an atherogenic diet plus Aspirin 5 mg/kg with Hyeolbuchukeo-tang 100 mg/kg. We investigated plasma lipid with liver function test, and performed the histological investigation of liver and abdominal aorta. Results: 1. We investigated photomicrographic changes of liver and abdominal aorta tissue. They showed that histological injury of aorta and lipid accumulations of the liver were lower in the AP and APH groups than in the control group. 2. In the APH group, plasma triglyceride levels were significantly lower than those in the control and AP groups. 3. There were no differences in aspartate aminotransferase and alanine aminotransferase levels among the control, AP and APH groups. Conclusion: The above results show that a combined treatment of Hyeolbuchukeo-tang and aspirin has a somewhat synergic effect in terms of inhibiting vessel injury and decreasing lipid deposits on liver cells without liver toxicity.
