파프리카 급여가 고콜레스테롤 식이 흰쥐의 혈청 지질과 $\alpha$-Tocopherol 농도 변화에 미치는 영향 (Effects of Paprika (Capsicum annuum L.) on Serum Lipid Profile and $\alpha$-Tocopherol Concentration in Rats Fed a High-Cholesterol Diet)

  • 본 연구는 추출물 형태에 따른 파프리카 첨가 식이가 고콜레스테롤 식이 흰쥐의 혈청 지질과 $\alpha$-tocopherol 농도에 미치는 영향을 알아보기 위해 Sprague-Dawley 수컷 25마리를 대조군(NC), 고콜레스테롤군(HC), 고콜레스테롤+동결건조 파프리카 분말 섭취군(HC-FDP), 고콜레스테롤+파프리카 물 추출물 섭취군(HC-WEP), 고콜레스테롤+파프리카 에탄올 추출물 섭취군(HC-EEP)으로 나누어 실험 식이를 6주간 공급하였다. 6주 후 HC군과 파프리카 급여군에서 체중 증가는 각각 85.2%와 65.5~68.88%로 HC군에서 체중이 유의적으로 증가하였다. 그리고 내장지방 무게는 HC군보다 파프리카 급여군에서 감소하였으며, 파프리카 급여군의 중성지질, 총 콜레스테롤, LDL-콜레스테롤과 atherogenic index 역시 HC군보다 유의적으로 감소하였다(p<0.05). 그리고 파프리카 급여군들 중에서는 HC-EEP 군이 혈청지질농도 감소 효과가 가장 큰 것으로 나타났다. 혈청 $\alpha$-tocopherol 농도는 HC-EEP에서 $30.5{\pm}0.6\;{\mu}mol/{\ell}$로 가장 높았고, 그 다음으로는 HC-WEP($30.0{\pm}0.8\;{\mu}mol/{\ell}$)>HC-FDP($29.1{\pm}0.5\;{\mu}mol/{\ell}$)>HC($27.7{\pm}0.3\;{\mu}mol/{\ell}$)이었다. 이는 고콜레스테롤 식이에 의한 산화적 스트레스를 감소시키기 위하여 혈청 $\alpha$-tocopherol이 소모되는 것을 파프리카 급여로 절약하는 효과를 나타낸 것으로 사료된다. 따라서 본 연구를 통해 파프리카는 혈중 지질 농도 저하 효과를 보였고, $\alpha$-tocopherol 절약 작용을 통해 체내 항산화제로서 활용 가능성을 확인할 수 있었다.

고지혈증 흰쥐에서 비배당체 이소플라본 고함유 대두분말의 혈청 지질 대사 영향과 항산화효과 (Effects of the Soybean Powder with Rich Aglycone Isoflavone on Lipid Metabolism and Antioxidative Activities in Hyperlipidemic Rats)

  • 비배당체 이소플라본 고함유 대두분말이 고지방식이를 섭취한 흰쥐의 지질대사와 항산화 효과에 미치는 영향을 조사하고자 식이성 고지혈 흰쥐를 정상군(NO), 고지방식이군(CO), 고지방식이에 비배당체 이소플라본 고함유 대두분말 10%, 20%(w/w) 첨가군(BFF10, BFF20)으로 나누어 7주간 사육한 결과는 다음과 같다. 비배당체 이소플라본 고함유 대두분말 섭취는 체중과 간 무게를 유의적으로 감소시켰으며, CO군에 비해 혈청의 총 콜레스테롤, LDL-콜레스테롤, 중성지질 농도가 유의적으로 감소하였다. 간조직 손상시 활성증가가 우려되는 alanine amino transferase와 aspartate amino transferase 수준도 유의적으로 감소하였다. Thiobarbituric acid reactive substance의 생성량은 비배당체 이소플라본 고함유 대두분말의 첨가량이 높을수록 현저하게 감소되었으며 superoxide dismutase 활성은 농도 의존적으로 증가시키는 것으로 나타났다.

  • Recently it has been reported that vitamin A and retinol binding proteins (RBPs) in blood and urine were changed in the condition of diabetes mellitus or hyperlipidemia. Fruits and vegetables are recommended to consume for the people suffered from these chronic degenerative diseases. The main components of fruits and vegetables are dietary fibers, for example cellulose and pectin, of which function to affect the absorption and excretion of dietary fat and fat-soluble substances. This study was conducted to investigate the effect of dietary fibers on RBPs mRNA expression in liver, small intestine and serum of rat fed high fat diet during 4 weeks. Sprague-Dawley rats, weighing 121g on average, were divided into four groups; (Control; $17\%$ fat & cellulose supplement diet, HF0: $25\%$ fat & fiber free diet, B:.Uc: $25\%$ fat & cellulose supplement diet and HF0: $25\%$ fat & pectin supplement diet) . The rats fed high fat diet groups (HF0, HFC, HFP) tended to consume the food less than the control group, but FER of HF0 groups was significantly higher than the control (p < 0.05) . The weight of adrenal gland in high fat diet groups (HF0, HFC, HFP) was significantly less than the control. Total lipid in feces daily excreted and in liver did not show any significant differences among the groups. Total cholesterol in HFP group was significantly different from that of HFC group. Serum total cholesterol and triglyceride in other group tended to lower than other groups and HDL cholesterol higher. Consequently, AI (atherogenic index) was the lowest in HFP group. Vit A contents in feces daily excreted tended to lower in high fat diet groups (HF0, HFP) compared to the control group. That content in adrenal gland was the lowest in HF0 group, but not in liver. In HFP group were down-regulated cRBPI mRNA in liver and cRBPII mRNA in small intestine and up-regulated RBP and transthyretin expression in serum compared to the other groups. In conclusion, dietary fibers, especially pectin, in high fat diet might down-regulate the expression of CRBP I, CRBP II mRNA in liver and small intestine, but increase the secretion of RBP into serum and therefore inhance the bioavailability of Vit A through the body. Feeding of Dehulled-micronized Faba Bean (Vicia faba var. minor) as Substitute for Soybean Meal in Guinea Fowl Broilers: Effect on Productive Performance and Meat Quality

  • The present study aimed to assess the effect of dietary substitution of soybean meal (SBM) with dehulled-micronized faba bean (Vicia faba var. minor) in guinea fowl broilers on their growth traits, carcass quality, and meat fatty acids composition. In this trial, 120 day-old guinea fowl keets were randomly assigned to two treatments which were fed from hatch to 12 weeks of age. Birds were fed two wheat middlings-based diets comprising of a control treatment which contained SBM (78.3 g/kg) and a test diet containing dehulled-micronized faba bean (130 g/kg) as the main protein source. Substituting SBM with faba bean had no adverse effect on growth traits, dressing percentage, or breast and thigh muscles relative weight of the guinea fowls. Conversely, a decrease (p<0.05) of abdominal fat was found in guinea fowls fed the faba bean-diet. Breast muscle of birds fed faba bean had higher $L^*$ score (p<0.05) and water-holding capacity (p<0.05) than the SBM control diet. Meat from guinea fowls fed faba bean had less total lipids (p<0.05) and cholesterol (p<0.01), and higher concentrations of phospholipids (p<0.01). Feeding faba bean increased polyunsaturated fatty acid concentrations in breast meat and decreased the saturated fatty acid levels. Moreover, dietary faba bean improved the atherogenic and thrombogenic indexes in guinea fowl breast meat. Results indicated that substitution of SBM with faba bean meal in guinea fowl diet can improve carcass qualitative traits, enhancing also meat lipid profile without negatively affecting growth performance.

FIB 햄스터에서 양파즙과 나린진 추출 혼합물의 항고지혈증 효과 -투과전자현미경적 관찰- (Effects of Onion and Naringin Extract Mixed Products on Antihyperlipidemic Levels of Bio FIB Hamster)

  • 본 연구는 고지혈을 일으킨 햄스터에게 나린진과 양파즙을 투여하여 고지혈을 낮추는 효과를 조사하고자 하였다. 고지혈 햄스터는 basal diet에 10% 코코넛오일과 0.05% 콜레스테롤로 고지혈을 일으켰다. 실험군은 5군의 그룹으로 나뉘어 40일간 실험식이를 각각 제공하였는데 즉 정상군, 대조군, 50%의 양파즙과 50%의 물을 혼합한군(OEN), 100% 양파즙군(OES), 100% 양파즙과 1% 나린진을 혼합한군(OESN)이다. 나린진을 처리한 그룹에서는 혈청 총 콜레스테롤, 중성지질 레벨은 현저하게 감소되었고 HDL 콜레스테롤은 증가하였다. 한편 간조직은 대조군과 비교하여 미토콘드리아와 핵의 형태가 현저하게 달랐다. 결론적으로 나린진을 함유한 양파즙은 간에서 콜레스테롤을 감소시켰다. 양파즙과 나린진추출물의 혼합은 콜레스테롤을 감소시키는 시너지 효과를 나타내었다.

에스트로겐에 의한 암쥐의 심장조직의 COX-2, Mapkinases 및 관련된 Apoptotic Markers의 발현의 변화에 관한 연구 (Changes in the Expression Pattern of Cyclooxygenase-2, Mapkinases and Related Apoptotic Markers by Different Levels of Estrogen Supplementation in Mature or Ovariectomized Female Rat Heart)

  • The effects of different concentrations of estrogen supplementation to mature female rats or estrogen supplementation to ovariectomized rats on cyclooxygenase-2 (COX-2) expression, PGE$_2$ production and mapkinases expression were investigated in experimentally induced atherogenic rats with feeding a high fat. high cholesterol diet. In the first experiment using 48-week old mature rats, the supplementation of three different levels of estrogen was compared to the basal diet. The high concentration of estrogen supplementation induced the marked up-regulation of COX-2 protein and the increase in plasma PGE$_2$ production and this seems to be followed by the up-regulation of p38 among mapkinases. The regulation of bax showed in a reverse trend of COX-2 in heart tissues of mature female rats. In the second ex-perimental system, female Sprague-Dawley rats were bilaterally ovariectomized; sham-operated animals were used as controls. Three weeks later, the animals were supplied with basal diet to sham-operated control group and ovariectomized control group, and estrogen supplemented diet to ovariectomized group for an eight-week experimental period. In a group supplemented with a medium dose of estrogen, COX-2 expression was up-regulated. This up-regulation was accompanied by the elevated expression of pERK1/2. Bax was increased in estrogen-fed animals indicating bax might be involved in estrogen feeding state in ovariectomized rats. Further investigations on the relationship between COX-2 and biological activities such as vasodilation by estrogen are required in in vivo system of female rats at the various physiological states.

고지방 식이로 유도된 비만 흰쥐에 인진(茵蔯), 지실(枳實), 산사가 미치는 영향 (The Effects of Artemisiae Capillaris, Ponciri Fructus and Cartaegi Fructus in Obese Rats Induced by High Fat Diet)

  • Objectives : This study was designed in order to determine the effects of Artemisiae Capillaris, Ponciri Fructus and Cartaegi Fructus so as to care the obesity and obesity-related diabetes. Methods : After induction of obesity by high fat-diet in rats, the each extracts of three medicinal stuffs were administrated with continuous high fat-diet for 4 weeks. And the changes of lipid metabolism, liver function and blood levels of glucose and insulin were examined. Results : Blood concentrations of triglyceride, free fatty acid and LDL-cholestrol were increased in obese group compared with standard dieted group. By treatment of Ponciri Fructus or Cartaegi Fructus with high fat-diet, their concentrations of glyceride and free fatty acid were significantly decreased. Blood level of LDL -cholesterol was decreased in all stuffs treated groups, however atherogenic index was significantly decreased in treatment of Cartaegi Fructus with high fat-dieted group. Conclusion : High fat-dieted with treatment of Ponciri Fructus or Cartaegi Fructus, fatty degeneration was remarkably diminished.

퀘르세틴 첨가가 고지방 및 고콜레스테롤 식이 랫드의 혈청 성분 농도의 변화에 미치는 영향 (Effect of Quercetin Supplement on Major Biochemical Parameters in Sera of Rats Fed High Fat and High Cholesterol Diet)

  • This study was performed to investigate the changes of the serological parameters of the rats fed a high fat and high cholesterol diets with or without quercetin supplement for five weeks. Thirty Sprague-Dawley male rats ($152.1{\pm}17.0\;g$ of body weight) were randomly divided into three groups with 10 rats in each group. Rats in the control(C) group were fed the high fat and high cholesterol diet containing 15% lard, 2% cholesterol and 0.5% sodium cholate (w/w). Rats in two treatment groups were fed the same diet supplemented with 0.25% quercetin (Q-0.25) or 0.5% quercetin (Q-0.5) on the weight to weight basis, respectively. The final body weight, gain of body weight, the amount of feed intake and the feed efficiency of rats in between control and treatment groups were not significantly different. Serum glucose, total protein, albumin, globulin and albumin/globulin (A/G) ratio of rats showed no significant differences between control and treatment groups. Serum total cholesterol (TC), low density lipoprotein-cholesterol (LDL-C) and atherogenic index(AI) of rats in both Q-0.25 and Q-0.5 groups were significantly lower than in C group (p < 0.05). However serum high density lipoprotein-cholesterol (HDL-C) was significantly higher in both Q-0.25 and Q-0.5 groups than in C group (p < 0.05). The levels of triglyceride in sera of rats showed no significant differences between control and treatment groups. The values of AST and ALT in sera of rats showed no significant differences between control and treatment groups. Therefore the supplementation of quercetin to high fat and high cholesterol diet in rats was effective in reducing the levels of serum lipids to cause cardiovascular diseases and in elevating the level of HDL-C to protect cardiovascular diseases.

Effect of Phellinus baumii -Biotransformed Soybean Powder on Lipid Metabolism in Rats

  • In this study, we evaluated the hypolipidemic and antioxidative effects of biotransformed soybean powder (BTS; Phellinus baumii-fermented soybean) on lipid metabolism in rats. Sprague-Dawley (SD) male rats were divided into basal diet group (BA), high fat diet group (HF), high fat diet containing 10% BTS group (10 BTS), and high fat diet containing 20% BTS group (20 BTS). Changes in the content of various isoflavones, including daidzein and genistein, within the soybean after fermentation to BTS were investigated. The levels of daidzein and genistein were $149.28{\mu}g/g$ and $364.31{\mu}g/g$, respectively. After six weeks experimental period, Food efficiency ratio in the 10 and 20 BTS group was significantly lower than the HF group (P<0.05). Total serum levels of cholesterol, triglycerides, low-density lipoprotein cholesterol, and atherogenic index ratio in the 10 or 20 BTS group were significantly lower than the HF group. The levels of alanine aminotransferase, aspartate aminotransferase and thiobarbituric acid reactive substance were significantly lower in the groups that received 10% and 20% BTS than the HF. The activities of SOD and CAT were significantly higher in the 10 and 20 BTS group than the HF group. The activity of XO in the 10 and 20 BTS group was significantly lower than in the HF group by 20% and 23%, respectively. In conclusion, these data suggest that BTS is an effective agent in improving lipid metabolism and antioxidant enzyme system.

Effects of Portulaca oleracea Powder on the Lipid Levels of Rats Fed a Hypercholesterolemia Inducing Diet

  • This study was initiated to investigate the effects of lyophilized purslane (Portulaca oleracea) powder (5% and 10%) on lipid levels in rats fed a hypercholesterolemia-inducing diet (1% cholesterol). During the four week study, there were no significant differences in either weight change or the food efficiency ratio between the group fed the hypercholesterol diet alone (HC) and the groups fed the purslane powder (HCPO-I and HCPO-II, 5% and 10% purslane, respectively). In serum, the levels of total lipid, total cholesterol and LDL-C decreased significantly for the group fed the 10% purslane powder (HCPO-II) in comparison with the group fed the 5% purslane powder (HCPO-I). The atherogenic index (AI) was reduced by about 51% for the group fed the 10% purslane powder (1.47) in comparison with the HC group (3.03). The activities of GOT, GPT, ALP and LDH decreased significantly for the groups fed the purslane powder in comparison with the HC group. Regarding liver tissue, the levels of total lipid, total cholesterol and triglyceride decreased significantly for the purslane powder-fed rats compared to the HC group. The fecal lipid profiles increased significantly as the amount of purslane powder was increased. Compared to the HC group, the fecal total cholesterol and triglyceride levels were higher in the group fed the 10% purslane powder by about 2.8 times and 2.3 times, respectively. For the serum and liver tissue, the content of lipid peroxide decreased significantly in the groups fed purslane powder compared to the HC group. The data from this experiment show an increase in the lipid levels discharged in feces, suggesting that the supplementation of purslane powder to a hypercholesterolemia-inducing diet reduces lipid levels.
