Mechanism of amyloidogenesis: nucleation-dependent fibrillation versus double-concerted fibrillation

  • Amyloidogenesis defines a condition in which a soluble and innocuous protein turns to insoluble protein aggregates known as amyloid fibrils. This protein suprastructure derived via chemically specific molecular self-assembly process has been commonly observed in various neurodegenerative disorders such as Alzheimer's, Parkinson's, and Prion diseases. Although the major culprit for the cellular degeneration in the diseases remains unsettled, amyloidogenesis is considered to be etiologically involved. Recent recognition of fibrillar polymorphism observed mostly from in vitro amyloidogeneses may indicate that multiple mechanisms for the amyloid fibril formation would be operated. Nucleation-dependent fibrillation is the prevalent model for assessing the self-assembly process. Following thermodynamically unfavorable seed formation, monomeric polypeptides bind to the seeds by exerting structural adjustments to the template, which leads to accelerated amyloid fibril formation. In this review, we propose another in vitro model of amyloidogenesis named double-concerted fibrillation. Here, two consecutive assembly processes of monomers and subsequent oligomeric species are responsible for the amyloid fibril formation of $\alpha$-synuclein, a pathological component of Parkinson's disease, following structural rearrangement within the oligomers which then act as a growing unit for the fibrillation.

  • Product Lifecycle Management (PLM) is an integrated business approach to manage the creation and distribution of product information throughout the product development process. From the product perspective, PLM encompasses a holistic approach to product development and product information management. It supports the integrated product information in conjunction with the efficient product structures and BOM (Bill Of Material), user interfaces, proper functions, design processes and enterprise integration. Therefore, PLM should not only satisfy required functions as an enterprise software but also offer a systematic method for the efficient application from the initial stage of its development. Recently, many shipyards have been considering the PLM as a strategic solution to get the efficient management of product information such as 3-D models, BOM, drawings, documents, and the other product data. Though many studies on PLM are performed, most of them are performed in a function-based approach adequate for mass productive assembly industries. It could not help having limitations on applying the proper PLM system to the shipbuilding business since the requirements of shipbuilding PLM are too diverse and huge to design the architecture. This study presents the PLM framework which effectively reflects the diverse requirements of shipbuilding PLM. In order to get the macroscopic architecture of shipbuilding PLM, authors suggest the four-tier architecture model which considers the various requirements collected from shipyards. Entities of ship design data are modeled BOM in terms of product structure and hierarchical class diagram. Applicable functions of shipbuilding PLM are also investigated by analysis of issues of ship design. Finally, by reflecting the design process of shipbuilding, To-Be ship design procedure cooperated with the suggested PLM framework has been summarized.

  • The Purpose of this paper is to automate the representation of manufacturing line using Petri net model. In the manufacturing cell, the line can be represented using workflow which is composed of Bill of Material (BOM) and Bill of Processes (BOP). BOP shows the precedence of processes and the relationship between assembly and disassembly. As a modeling scheme, generalized stochastic Petri net is adopted. For a problem domain with flexible manufacturing cell, Petri net model is made and behavioral properties are analyzed.

INTERBUSs를 이용한 모델 플랜트의 구동 소프트웨어의 개발 (Development of an Operating Software for a Model Plant using INTERBUSs)

  • This study develops and operating software for a model plant. The model plant consists of an AS/RS system, two machining centers, an assembly line, and supplementary material handling equipments. Devices of each component are connected with the IBS RT24 DIO 16/16-T I/O module. Each I/O module communicates digital signals with the INTERBUS controller board via SUB-D 9 Connector cable. This study is a previous stage for developing an educational CIM software. Petri Nets is used for modelling the storage/retrieval of the AS/RS system, the flow of workpieces and the assembly line for parts. The operating software is coded with Microsoft Visual $C^{++}$ 5.0 and Interbus Library which is a software driver for the controller board. The operating software can be run on a MS Windows 95. Microsoft Access is used for the implementation of databases for BOM, AS/RS, and parts.

Development of Information System for Product Redesign: Evaluation of Assembly Process and Characteristics of Product Functions

  • Since product design strongly depends on the experience and ability of the designers, and a long lead time is required for the product design stage, introducing a support system related to this experience and ability is an effective technique to reduce the lead time. In this paper, an information system is developed to support the development of engineering mechanisms with the evaluation of the assembly process on the basis of the abstract functions required for new products. The developed system is based on a database system involving the following data structure: (1) a hierarchical structure related to information about the product functions, (2) the relationship between the parts and engineering functions and/or mechanisms of products, and (3) the relationship between the parts and manufacturing characteristics. The system stores the relationships between the product functions, structure of parts, and assembly characteristics. This information can then be interactively retrieved using the data structure described in (1), (2), and (3). A procedure for designing new products is proposed that involves using the information about existing products. This paper presents the characteristics of the proposed procedure and the developed information system. In addition, a case study of the redesign of a simple structured robot by using the proposed procedure is discussed.

3차원 CAD라이브러리를 이용한 프레스 금형 부품의 설계 (A Design of Press Die Components by Use of 3D CAD Library)

  • Using standard components of a press die is recognized as a way for a cost reduction and a short lead time. It also provides a way for a quick maintenance of a die under repair. It is expected to contribute to integration of CAD/CAM system for manufacturing dies in the future. This paper presents a 3D CAD library which is constructed using the standard components and is used for designing a press die. This 3D CAD library is generated by a database system made of Microsoft Access for standard components and by CATIA V5 R10 API for geometric features. The library is implemented using Visual Basic 6.0 utility of CATIA API function in the Windows NT environment. It creates a 3D model of the standard components of press die easily when a die designer inputs numerical values of geometric features and the BOM of the completely assembled parts. It also generates automatically the assembly drawing of die set by using variables for standard values of die parts. Therefore users can save the cost of time to design the press die components, and even a beginner can use this program with ease. The test results of the 3D CAD library for designing shearing and bending dies verify its usefulness and feasibility.

  • 본 연구에서는 STEP 표준을 고려하지 않고 설계·개발된 기존의 PDM 시스템이 CAD 시스템에서 생성된 STEP 포맷의 제품 데이터를 활용할 수 있도록 하기 위한 데이터 변환 방법을 제시하고자 한다. 이를 위하여 AP203 응용 프로토콜의 분석과 함께 스키마 매핑을 위한 방법과 절차를 제시하였다 그리고, 이를 바탕으로 AP203 스키마와 ‘중소기업형 PDM 시스템'의 DB 스키마간에 매핑을 설계하였다. 또한, CAD 시스템에서 생성된 제품의 Assembly 구조를 PDM 시스템의 BOM 구조로 변환하기 위한 알고리즘을 함께 제시하였으며, 제품의 product structure를 중심으로 데이터 변환을 위한 STEP-to-DB 변환기 프로토타입을 개발하였다. 그리고, CAD 시스템에서 생성된 제품 데이터에 대한 STEP 스키마 구조와 AP203 응용 프로토콜 스키마와의 구조적 차이를 비교·분석함으로서 CAD 시스템에서 생성된 STEP 포맷의 제품 데이터 구조에 대해 엔지니어링 데이터로서의 구조를 갖도록 하기 위한 기초 자료를 제공하였다.

  • Nowadays, most automotives companies are making use of laser welding in car body assembly shop. But even though laser welding is better than resistance spot welding in many points, its application has been limited to special technology for manufacturing. The paper introduces in the field of remote welding system(RWS) to improve the process efficiency of laser welding. Positioning time of RWS between welding stitches are dramatically reduced to zero. It is a kind of solutions to generalize laser welding in mass production.

  • Presented in this paper is the development of an product cost estimation system at the product design stage. The efficient cost estimation function at the design stage is essential for the cost reduction activities through the entire product life cycle. For this purpose, it is necessary to establish a systematic working procedure, and to develop information system for managing a great deal of production and product-related data required for the cost estimation. The developed system has the capability of estimating a cost of assembly type products as well as unit-item type products. As proposed system is based on the variant approach, it can be used easily at an early design stage without the need for detail design information. Also, this system is integrated with legacy PDM (Product Data Management) and ERP (Enterprise Resource Planning) system for fast. accurate and easy product cost estimation. The estimated cost includes material cost, overhead cost as well as labor cost.

20, 40 wt% Pt/C 촉매를 사용한 MEA제조에서 나피온의 최적비 (Optimum Ratio between Nafion and 20, 40 wt% Pt/C Catalysts for MEAs)

  • Pt/C 촉매 (20, 40 wt% Pt/C)를 사용하여 고분자 전해질 연료전지의 MEA를 제조하고 각각의 촉매에서 최적의 나피온 이오노머 함량을 알아보았다. 나피온 함량에 따른 MEA의 전기화학적인 성능변화는 단위전지 성능평가, electrochemical impedance spectroscopy (EIS), cyclic voltammetry(CV)을 통해서 분석하였다. 나피온의 함량에 따라 전지의 활성화 분극, 옴 저항, 물질전달 저항 등의 변화가 나타났다. 이는 전극의 촉매층 내에서 발생되는 전기/이온 전도도 사이의 'trade-off'와 물질전달(물 배출과 반응가스 확산)에 의한 것이며, 대부분 활성화 분극과 물질전달 저항의 변화로 나타났다. 20 wt% Pt/C와 40 wt% Pt/C 촉매에서 최적의 나피온 함량은 각각 35 wt%와 20 wt%로 나타났다. 이는 Pt 중량비에 따른 Pt 입자간의 거리 및 촉매의 비표면적의 차이 때문에 나타난 결과이며 서로 다른 나피온 함량에서 최적의 삼상계면이 형성되는 것으로 판단된다.
