EFFECTS OF ASCORBIC ACID (VITAMIN C) SUPPLEMENTATION IN LAYER AND BROILER DIETS IN THE TROPICS

  • An experiment was conducted to study the effects of Ascorbic Acid (AA) supplementations in the layer and broiler diets kept in the natural hot humid tropical climate ($20-35^{\circ}C$). The layers and the broilers were fed on normal commercial diet as control while supplementation of 400 and 600 mg/kg Ascorbic Acid made up the experimental diets. The results showed that AA supplementation in the layers significantly reduced egg weight and increased Haugh unit values of the eggs, but produced no significant effects on feed intake, body weight, egg production, respiratory rate and body temperature. The shell thickness was slightly improved, though not significantly, with AA supplementation. AA supplementation in broilers improved body weight gain and FCR and reduced the effect of heat stress as shown by lower body temperature and respiratory rates.

  • Encapsulation of L-ascorbic acid(AA) into BGsome was attempted to improve its stability. BGsome is a bio-compatible vesicular system prepared by dispersion of hydrated liquid crystalline phase formed through hydration of 1,3-butylene glycol(BG)-dissolved lecithin with an aqueous solution containing hydrophilic component. The characteristics of AA encapsulated BGsome, such as droplet size, surface charge, and solution appearance, was investigated. The concentration of AA solution had considerable effect on droplet size and surface charge of BGsome. Several tens nanometer droplet made by sonication treatment did not showed any change of size with storage time. Stability of AA was improved by encapsulation into BGsome, which was verified through DPPH test and HPLC assay.

Synergistic Effect of Methanol Extract from Kalopanax pictus and Ascorbic Acid on Antioxidant, Anticancer and Immunomodulatory Activities (음나무(Kalopanax pictus) 추출물과 비타민 C의 항산화, 항암 및 면역활성 상승효과)

  • The 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid (ABTS), nitric oxide (NO) scavenging activities and ferric- reducing/antioxidant power (FRAP) assay against extracts of Kalopanax pictus (KP) were measured. Radical scavenging and antioxidant activities were increased depend on the concentration and the effects were enhanced by ascorbic acid (AA). KP extracts and AA had a good anti-proliferating activity against HepG2 cells by MTT assay and induces cells apoptosis, which was demonstrated by flow cytometric analysis. KP extracts and AA caused the arrest of cell-cycle progression at either G0-G1-phase or G2/M-phase, which might be depending upon the KP extracts concentration. In addition, KP extracts and AA are effective in enhancing immunity and nitric oxide production by RAW 264.7 macrophages cells. KP extracts and AA inhibited tumor cell growth and exerted antioxidant effects as compared to controls. These results demonstrate that simultaneous AA and KP extracts treatment could be useful in preventing the oxidative damage and anti-proliferating HepG2 cells, and are effective in enhancing immunomodulatory and antioxidant activity.

Effect of Ascorbic Acid on the Gravitropic Response of Primary Roots in Maize (옥수수 일차뿌리에서 Ascorbic acid가 굴중성 반응에 미치는 효과)

  • Ascorbic acid (AA) is a multifunctional metabolite in plants that is essential for plant development and growth. We examined the effect of AA, an antioxidant, on the gravitropic response of primary roots in maize. The application of $10^{-3}$ M AA to the elongation zone did not affect the gravitropic response and slightly inhibited the root growth. However, treatment with both $10^{-5}$ M and $10^{-3}$ M AA at the root tip increased the gravitropic response and inhibited root growth. Differences in indole-3- acetic acid (IAA) activity between the upper and lower hemispheres of the root resulted in differential elongation along the horizontal root. Roots are extremely sensitive to IAA, and increasing the amount of IAA in the lower hemisphere of the root inhibited elongation. Therefore, we examined the effect of IAA in the presence of AA. The inhibitory effect of AA on the gravitropic response was greater in combination with IAA. To understand the role of AA in the regulation of root growth and the gravitropic response, we measured ethylene production in the presence of AA in the primary roots of maize. AA stimulated ethylene production via the activation of the 1-aminocyclopropane-1-carboxylic acid (ACC) oxidase gene, which regulates the conversion of ACC to ethylene. These results suggest that AA alters the gravitropic response of maize roots through modification of the action of ethylene.

  • This presentation reviewed the dietary vitamin C requirements in main marine finfish culture in Korea. In experiment I, an II-week feeding trial was conducted to study the effects of the different levels of dietary vitamin C on growth, tissue ascorbic acid concentrations and histopathological changes in parrot fish. Casein and gelatin based diets were formulated to contain 0, 60, 120, 240, 480 and 2000mg L-ascorbic acid (AA)kg diets on Ah equivalent basis in the form of L-ascorbyl-2-monophosphate (AMP), 60 and 240 mg AA/kg diet as L-ascorbic acid. Broken line analysis of weight gain indicated that the dietary vitamin C requirement of parrot fish is l18$\pm$12 mg AA/kg diet in the form of L-ascorbyl-2-monophosphate for maximum growth. In experiment II, a 12-week experiment was conducted to compare L-ascorbyl-2-monophosphate-Ca (AMP-Ca) with L-ascorbyl-2-monophosphate-Na/Ca (AMP-Na/Ca) for supplying the dietary vitamin C for juvenile Korean rockfish Sebastes schlegeli. Fish were fed one of 11 semi-purified diets containing equivalent of 0, 50, 100, 200, 400, and 800 mg ascorbic acid (AA)kg diet in the form of AMP-Ca or AMP-Na/Ca for 12 weeks. Broken line analysis of weight gain indicated that the dietary vitamin C requirement of Korean rockfish is 100 mg AA/kg diet in the form of AMP-Na/Ca, and 117 nag AA/kg diet in the form of AMP-Ca. In experiment III, a 12-week experiment was conducted to study the effects of different dietary levels of vitamin C, L-ascorbyl-2-polyphosphate (ASPP), on growth and tissue vitamin C concentrations in juvenile olive flounder. Fish were fed one of six semi-purified diets containing an equivalent of 0, 25, 50, 75, 150, or 1500 mg ascorbic acid (AA) kg 1 diet in the form of ASPP for 12 weeks. Based on broken line analyses for WG and PER, the optimum dietary levels of vitamin C were 91 and 93 mg AA/kg diet, respectively.

Oxidation of Ascorbic Acid by Crosslinked Poly(4-vinyl pyridine)-Cu(II) Complexes 2. Effect of Crosslinker (가교 폴리(4-비닐피리딘)-구리(II) 착물에 의한 Ascorbic Acid의 산화반응 2. 가교제의 영향)

  • Various crosslinked poly(4-vinylpyridines) (CHP4VP) having different degrees of crosslinking were synthesized by radical copolymerization of 4-vinylpyridine with if N,N' -1, 6-hexamethylenebisacrylamide, and CHP4VP- Cu(II) complexes were prepared by the method of adsorption equilibrium. The catalytic activity of the complexes for the oxidation of ascorbic acid (AA) was investigated. The oxidation of AA by these complexes showed a kinetic behavior of the Michaelis-Menten type. The catalytic activity of CHP4VP-Cu(I ) catalytic system was increased with increasing the degree of crosslinking of CHP4VP, and its activity was scarcely decreased even after repeated use. However, the tendency of the catalytic activity of CHP4VP-Cu(II) complexes was decreased for the oxidation of AA when compared with that of the previously reported catalytic system containing crosslinked poly(4-vinylpyridine) prepared using N,N'-methylenebisacrylamide as a crosslinker. These results indicate that the degree of crosslinking of CHP4VP and the hydrophobicity of the crosslinker play an important role in the catalytic system of the oxidation of AA.

  • Aflatoxin $B_1$ ($AFB_1$) was reacted with ascorbic acid (AA) alone, with AA plue cysteine and with AA plus cupric ion for 5 days (at $37^{\circ}C$ and pH 5), and the mutagenicity of the reaction products was tested with Salmonella typhimurium TA 100. About 10% of AFBl induced mutagenesis was reduced when $AFB_1$ reacted with AA. This decreasing effect was more severe when $AFB_1$ reacted with AA plus cysteine. The mutagenicity of $AFB_1$ when reacted with AA plus cupric ion was almost completely inhibited, however, eupric ion itself was shown to enhance the mutagenicity of $AFB_1$. Therefore, $AFB_1$ may be degraded in the presence of AA under the given reaction condition and the reaction products was observed to have nonmutagenic effects on the bacterial mutagenecity trials.

  • A fast and simple method for synthesis of CuxO-ZnO/PPy/RGO nanocomposite by electrochemical manner have been reported in this paper. For testing the utility of this nanocomposite we modified a GCE with the nanocomposite to yield a sensor for simultaneous determination of four analytes namely ascorbic acid (AA), dopamine (DA), uric acid (UA), and folic acid (FA). Cyclic voltammetry (CV) and Differential pulse voltammetry (DPV) selected for the study. The modified electrode cause to enhance electron transfer rate so overcome to overlapping their peaks and consequently having the ability to the simultaneous determination of AA, DA, UA, and FA. To synthesis confirmation of the nanocomposite, Field emission scanning electron microscopy (FE-SEM), Raman spectroscopy, and electrochemical impedance spectroscopy (EIS) were applied. The linearity ranges were 0.07-485 μM, 0.05-430 μM, 0.02-250 μM and 0.022-180 μM for AA, DA, UA, and FA respectively and the detection limits were 22 nM, 10 nM, 5 nM and 6 nM for AA, DA, UA, and FA respectively Also, the obtained electrode can be used for the determination of the AA, DA, UA, and FA in human blood, and human urine real samples.

Production of 2-O-\alpha-D- Glucopyranosl L-Ascorbic Acid by Cyclodextrin Glucanotransferase from Paenibacillus sp. JB-13 (Paenibacillus sp. JB-13의 Cyclodextrin glucanotransferase에 의한 2-O-\alpha-D- Glucopyranosl L-Ascorbic acid 생산)

  • Paenibacillus sp. JB-13 producing the cyclodextrin glucan-otransferase(CGTase) [EC 2.4.1.19] that glucosylated ascorbic acid(AA) at the C-2 position was isolated form soil and the optimal conditions for the production of 2-O-$\alpha$-D- Glucopyranosl L-Ascorbic acid(AA-2G) with CGTase were investigated. CGTase produced AA-2G efficiently using dextrin as a substrate and AA as an aceptor. Several AA-2-oilgosaccharides(AA-2Gs) were also produced in this reaction mixture, and these were efficiently hydro-lyzed to AA-2G and glucose by the treatment with glucoamylase. The optimal temperature for AA-2G production was $37^{\circ}C$ and the optimal pH was around 6.5. CGTase also utilized $\alpha$-,$\beta$-,${\gamma}$-CDs, soluble starch, com statch, dia-static solution from rice and diastatic solution from malt as substrate, but not glucose. The reaction mixture for the maximal production of AA-2G was following; 15% total substrate concentration, 2,500 units/ml of CGTase and a mixing ration of 3:2(g of AA: g of dextrin). Under this condition, 56 mM of AA-2G ,which corresponded to 12.4% yield based on AA. was produced after incubation for 44 hrs at $37^{\circ}C$ and pH 6.5.

Protective Effect of Artificially Enhanced Level of L-Ascorbic Acid against Water Deficit-Induced Oxidative Stress in Rice Seedlings

  • Effects of the enhanced level of L-ascorbic acid (AA) on the water deficit-induced oxidative damage were studied in rice (Oryza sativa L.) seedlings. The seedlings sprayed with 20 to 80 mM L-galactono-${\gamma}$-lactone (GL), a putative precursor of AA, showed 2 to 5-fold higher levels of AA compared with controls. Pretreatment of the seedlings with GL prior to water stress imposition caused virtually no effect on dehydration of tissues during water deficit but substantially mitigated oxidative injury, as accessed by 2-thiobarbituric acid-reactive substances, ${\alpha}$-tocopherol, chlorophylls and ${\beta}$-carotene. Proline accumulation during water stress was also significantly lowered in the treated seedlings. In a complementary experiment, AA retarded photodegradation of ${\alpha}$-tocopherol in isolated thylakoids far more efficiently than glutathione. GL in itself did not show any noticeable reactivity toward ${\alpha}$-tocopheroxyl radical. The results demonstrate the antioxidative function of AA in rice seedlings encountering water-limited environments, suggesting a critical role of AA as a defense against oxidative stress in plants.
