The relationship between water deficit stress and monodehydroascorbate reductase (MDHAR) activity was determined in lettuce (Lactuca sativa L.) leaves under water stress condition imposed by with-holding water for 72 hrs. Relative water content determined in water deficit stressed lettuce leaves gradually reduced from 91.29% to 74.58%, and water content of medium drastically decreased 4.73% after quitting of irrigation. Hydrogen peroxide content in leaves subjected to water deficit stress rapidly increased, but soluble protein content rapidly decreased when those were compared to control plant. The relationship between relative water content and hydrogen peroxide content in stressed leaves positively correlated with $R^2$=0.8851, but soluble protein content reversely correlated with $R^2$=0.9826. Total chlorophyll content in stressed plant leaves was higher than that of control plant, and increased rapidly in early stage of treatment of both stressed and control plants. Carotenoid content was higher than that of control plant, and the ratio of carotenoid to total chlorophyll in stressed plant was higher as compared to control plant. As water deficit stress continued progressively, total ascorbate content in stressed plant leaves was a little higher than that of control plant. But dehydroascorbate (DHA) content within 6 hr of water deficit stress was higher than that of control plant, and then, content of control plant in 12 hr of stress treatment higher than that of stressed leaves. The activity of monodehydroascorbate reductase of cytosolic and chloroplastic tractions increased dramatically, and mRNA of MDHAR was highly detected by probing $^{32}P$-labeled single stranded MDHAR RNA of lettuce plant leaves subjected to water deficit stress. Relationship between MDHAR activity and relative water content and hydrogen peroxide highly correlated with $R^2$=0.9937 and 0.8645, respectively.
Tomato (Lycopersicon esculentum) seedlings exposed to various concentrations of $CdC1_2$ (0∼100 $\mu$M) in the nutrient solution for up to 9 days were analyzed with the seedling growth, $H_2O_2$ production, glutathione levels and activity changes of enzymes related to $H_2O_2$ removal. The growth of seedlings was inhibited with over 50 $\mu$M Cd, whereas the levels of $H_2O_2$ and glutathione were enhanced with Cd exposure level and time. Meanwhile, Cd exposure increased the activities of catalase (CAT) and glutathione reductase (GR) but decreased the activities of dehydroascorbate acid reductase (DHAR) and ascorbate peroxidase (APX) in both leaves and roots. These results suggest that the altered activities of antioxidant enzymes particularly involved in the $H_2O_2$ removal and the subsequent $H_2O$$_2$ accumulation could induce the Cd-induced phytotoxicity.
The effects of light on the activities of several antioxidative enzymes, catalase (CAT), superoxide dismutase(SOD), ascorbate oxidase(AO), and peroxidase(POD) were examined in the hairy root cultures of Phytolacca esculenta van Houtte induced by Agrobacterium tumefaciens $A_4$T. Activities of CAT, SOD, and AO were significantly decreased with incresing light intensity (500-2,000 lx). The activity of AO under high light condition (2,000 lx)was decreased by 92% compared to the dark condition. The activities of glutathoine peroxidase (GPO), ascorbate peroxidase (APO) and general POD were increased under lower light intensify below 500 lx. The activity of GPO under 2,000 lx was decreased by 85% compared to the dark condition. The activities of antioxidative enzymes were more decreased in blue light (400-500nm). The activities of antioxidative enzymes in blue light intensity were increased in lower light intensity below 30 lx, but decreased 21-70% under 200 lx. The activity of AO was decreased by 70% under 200 lx with increasing blue light intensity. Our results suggest that the activities of antioxidative enzymes in hairy roots might be inhibited by endogenous oxidants generated under the high blue light conditions.
Monodehydroascorbate reductase (MDHAR) is an important enzyme that plays a role in the detoxification of reactive oxygen species (ROS) by maintaining reduced pool of ascorbate through recycling the oxidized form of ascorbate. In this study, we isolated a PagMDHAR1 gene from Populus alba ${\times}$ P. glandulosa, and investigated its expression characteristics. The PagMDHAR1 cDNA encodes a putative 434 amino acids containing FAD- and NAD(P)H-binding domains. Southern blot analysis indicated that a single nuclear gene encodes this enzyme. Northern hybridization analysis revealed that PagMDHAR1 is highly expressed in both suspension cells and flower tissues, while its expression levels were enhanced by drought, salt, cold, wounding and ABA. Therefore, PagMDHAR1 might be expressed in response to abiotic stress through the ABA-mediated signaling pathway in this poplar species, suggesting that the PagMDHAR1 plays an important role in the defense mechanisms against oxidative stress.

We examined two Populus species (Populus euramericana and Populus alba ${\times}$ Populus glandulosa) trees grown under livestock waste leachate treatment in the field. We investigated their physiological properties (photosynthetic rate, chlorophyll contents, transpiration rate and intercellular $CO_2$ concentration) and antioxidant enzyme activities. Ascorbate peroxidase and glutathione reductase were analyzed. According to our measurements, P. euramericana at control site showed a lower total chlorophyll contents, photosynthetic rate, intercellular $CO_2$ concentration, transpiration and stomatal conductance than those of trees at treatment site. P. alba ${\times}$ P. glandulosa showed low stomatal conductance and low photosynthetic rate.
This experiment was conducted to find out the effects of ABA and IAA on activities of antioxidant enzymes, antioxidant content, and growth of tobacco under exposure to ozone. The exposure to ozone in tobacco plant significantly decreased plant height, but it did not show any difference in vegetative characteristics except plant height of IAA $10^{-3}$M treated. Total chlorophyll content of NC 82 was dramatically decreased with increase in days after ozone treatment. However, reduction of chlorophyll was minimized when plant growth regulators were treated before ozone exposure. Three days treament of ozone in tobacco increased ascorbic acid of oxidised form, while slightly decreased `in reduced ascorbic acid by IAA treatment. But seven days of ozone treatment showed increase in ascorbic acid and decrease in dehydroascorbic acid. Ozone treatment did not show any difference in glutathione content and glutathione reductase activity when plant growth regulators were treated. Activities of superoxide dismutase(SOD), ascorbate peroxidase(AP) and guaiacol peroxidase(GP) were increased by the exposure to ozone for three days. However, there were no difference in activities of SOD, AP and GP due to exposure to ozone for seven days. These reactions may be interpreted as protective responses to prevent or alleviate the damage of tobacco plant by ozone exposure.
Maintaining redox balance is one of the crucial requirements for a cell to endure stress from the outside. Dehydroascorbate reductase (DHAR; EC 1.8.5.1) plays an important role in the ascorbate-glutathione cycle; one of the major ROS scavenging systems in most known biological systems. A cDNA clone of the DHAR gene from Oryza sativa (OsDHAR) was isolated and overexpressed in Escherichia coli BL21 (DE3) strain from the pET-28a(+) expression vector. The OsDHAR transformed E. coli cells showed significantly higher DHAR activity and a lower level of ROS than the E. coli cells transformed by an empty pET-28a(+) vector. Also, the DHAR-overexpressing E. coli strain was more tolerant to oxidant- and heavy metal-mediated stress conditions than the control E. coli strain. The results suggest that the overexpressed rice DHAR gene effectively functions in a prokaryotic system and provide protection to various oxidative stresses.
Methyl jasmonate (MeJA) is a signal molecule in the activation of defense responses in plants. In this study, we isolated 15 MeJA-inducible genes by subtractive hybridization. These genes encode two myrosinase-binding proteins, five lipase-like proteins, a polygalacturonase inhibitor, a putative chlorophyll-associated protein, a terpene synthase, a dehydroascorbate reductase, an ascorbate oxidase, a cysteine protease, an O-methyltransferase, and an epithiospecifier protein. Northern analysis showed that most of the Chinese cabbage genes are barely expressed in healthy leaves, but are strongly induced by MeJA treatment. We also examined whether these MeJA-inducible genes were activated by ethethon, BTH, and Pseudomonas syringae pv. tomato (Pst), a nonhost pathogen of Chinese cabbage. The results showed that none of the MeJA-inducible genes was strongly induced by ethephon or by BTH. The genes encoding lipase-like proteins and a myrosinase-binding protein were weakly induced by Pst. Other MeJA-inducible genes were not activated at all by the pathogen.
This study investigated concentrations of the several air pollutants and compared antioxidative enzyme activities on Platanus occidentalis because this tree species is one of the widespread street trees in Korea. This species has been emerging the ambient air pollutants during its growing periods. The purpose of this study was to identify the relationship between air pollution on the tree species and antioxidant enzyme activities on the trees. $O_3$, $NO_2$, CO and $SO_2$ concentrations of several cities in Korea were compared for last decades. Among the air pollutants, $O_3$ and $NO_2$ concentrations in six big cities in Korea showed similar increasing trends during this period. In contrast, $SO_2$ and CO concentrations in the same cities dramatically decreased between 1994 and 2005. Platanus occidentalis trees were controlled to investigate, ascorbate peroxidase (APX) and glutathione reductase (GR) activity. Ozone exposure generally increased APX and GR activities of tree seedlings. It is a typical compensatory strategy of stressed trees.
The environmental pollutions were a serious problem in Korea recently. So many researcher have studied the effect of environmental pollution on plants and agro-ecosystem, but the basic mechanisms of environmental stresses were various. One of the important mechanisms was oxidative stress caused by active toxic oxygen. The toxic oxygen was generated by several stresses, abnormal temperature, many xenobiotics, air pollutants, water stress, fugal toxin, etc. In the species of toxic oxygen which is primary inducer of oxidative stresses, superoxide, hydrogen peroxide, hydroxyl radical and singlet oxygen were representative species. The scavenging systems were divided into two groups. One was nonenzymatic system and the other enzymatic system. Antioxidants such as glutathione, ascorbic acid, and carotenoid, have the primary function in defense mechanisms. Enzymatic system divided into two groups; First, direct interaction with toxic oxygen(eg. superoxide dismutase). Second, participation in redox reaction to maintain the active antioxidant levels(eg. glutathione reductase, ascorbate peroxidase, etc.).
