This study was carried out to investigate the artificial cultivation of Pleurotus eryngii on the optimal medium vessel, periods of cultivation and the optimal method of pinheading for both yield and quality of fruiting body were also performed. The optimal composition of sawdust medium in polypropylene(PP) bottle was combination of sawdust(70%) and corncob(30%) but increased amount of corncob delayed the period of mycelial growth. The mycelial growth and the yield of fruiting body in the medium with beat pulp were worse than that without beat pulp. The optimal composition of nutrients for both yield and quality of fruit body tuned out to be a combination of rice bran(12%), wheat bran(12%) and cottonseed cake(6%). Additions of zeolite, shell lime and bean curd dregs were not effective in mycelial growth and yield of fruit body. When testing size of PP bottle for cultivation, the larger of bottle mouth is, the more pinheading number found, but the number of available fruit body is not significantly different. The culture in $1100\;ml-{\phi}75\;mm$ bottle is the best in the yield and quality of fruit body than those in $555\;ml-{\phi}50\;mm,\;850\;ml-{\phi}58\;mm,\;850\;ml-{\phi}65\;mm\;and\;1100\;ml-{\phi}65\;mm$ bottle. Using the PP bag for cultivation, a square shaped bag was better than a round shaped and black square shaped in mycelial growth and yield of fruit body. The most suitable period of incubation was 35 days after inoculation at $22{\pm}2^{\circ}C$. When the incubation periods was decreased less than 35 days, the pileus formation and yields were very bad but a pinheading condition looked similar, For an optimal pinheading, turning upside down was better than standing and covering.
BACKGROUND: Various culture media have been used for hydroponic cultures of horticultural plants under the smart greenhouses with natural and artificial light types. Management of the culture medium for the control of medium amounts and/or necessary components absorbed by plants during the cultivation period is performed with ICT (Information and Communication Technology) and/or IoT (Internet of Things) in a smart farm system. This study was conducted to develop the cloud-based data analysis system for effective management of culture medium applying to hydroponic culture and plant growth in smart greenhouses. METHODS AND RESULTS: Conventional inorganic Yamazaki and organic media derived from agricultural byproducts such as a immature fruit, leaf, or stem were used for hydroponic culture media. Component changes of the solutions according to the growth stage were monitored and plant growth was observed. Red and green lettuce seedlings (Lactuca sativa L.) which developed 2~3 true leaves were considered as plant materials. The seedlings were hydroponically grown in the smart greenhouse with fluorescent and light-emitting diodes (LEDs) lights of $150{\mu}mol/m^2/s$ light intensity for 35 days. Growth data of the seedlings were classified and stored to develop the relational database in the virtual machine which was generated from an open stack cloud system on the base of growth parameter. Relation of the plant growth and nutrient absorption pattern of 9 inorganic components inside the media during the cultivation period was investigated. The stored data associated with component changes and growth parameters were visualized on the web through the web framework and Node JS. CONCLUSION: Time-series changes of inorganic components in the culture media were observed. The increases of the unfolded leaves or fresh weight of the seedlings were mainly dependent on the macroelements such as a $NO_3-N$, and affected by the different inorganic and organic media. Though the data analysis system was developed, actual measurement data were offered by using the user smart device, and analysis and comparison of the data were visualized graphically in time series based on the cloud database. Agricultural management in data visualization and/or plant growth can be implemented by the data analysis system under whole agricultural sites regardless of various culture environmental changes.
Biodegradable barrier membrane has been demonstrated to have guided bone regeneration capacity on the animal study. The purpose of this study is to evaluate the effects of cultured calvarial cell inoculated on the biodegradable barrier membrane for the regeneration of the artificial bone defect. In this experiment 35 Sprague-Dawley male rats(mean BW 150gm) were used. 30 rats were divided into 3 groups. In group I, defects were covered periosteum without membrane. In group II, defects were repaired using biodegradable barrier membrane. In group III, the defects were repaired using biodegradable barrier membrane seeded with cultured calvarial cell. Every surgical procedure were performed under the general anesthesia by using with intravenous injection of Pentobarbital sodium(30mg/Kg). After anesthesia, 5 rats were sacrificed by decapitation to obtain the calvaria for bone cell culture. Calvarial cells were cultured with Dulbecco's Modified Essential Medium contained with 10% Fetal Bovine Serum under the conventional conditions. The number of cell inoculated on the membrane were $1{\times}10^6$ Cells/ml. The membrane were inserted on the artificial bone defect after 3 days of culture. A single 3-mm diameter full-thickness artificial calvarial defect was made in each animal by using with bone trephine drill. After the every surgical intervention of animal, all of the animals were sacrificed at 1, 2, 3 weeks after surgery by using of perfusion technique. For obtaining histological section, tissues were fixed in 2.5% Glutaraldehyde (0.1M cacodylate buffer, pH 7.2) and Karnovsky's fixative solution, and decalcified with 0.1M disodium ethylene diaminetetraacetate for 3 weeks. Tissue embeding was performed in paraffin and cut parallel to the surface of calvaria. Section in 7${\mu}m$ thickness of tissue was done and stained with Hematoxylin-Eosin. All the specimens were observed under the light microscopy. The following results were obtained. 1 . During the whole period of experiment, fibrous connective tissue was revealed at 1week after surgery which meant rapid soft tissue recovery. The healing rate of defected area into new bone formation of the test group was observed more rapid tendency than other two groups. 2 . The sequence of healing rate of bone defected area was as follows ; test group, positive control, negative control group. 3 . During the experiment, an osteoclastic cell around preexisted bone was not found. New bone formation was originated from the periphery of the remaing bone wall, and gradually extended into central portion of the bone defect. 4 . The biodegradable barrier membrane was observed favorable biocompatibility during this experimental period without any other noticeable foreign body reaction. And mineralization in the newly formed osteoid tissue revealed relatively more rapid than other group since early stage of the healing process. Conclusively, the cultured bone cell inoculated onto the biodegradable barrier membrane may have an important role of regeneration of artificial bone defects of alveolar bone. This study thus demonstrates a tissue-engineering the approach to the repair of bone defects, which may have clinical applications in clinical fields of the dentistry including periodontics.

We aimed to evaluate the effectiveness of the Giggenbach bottle method and develop the related pretreatment and analytical methods using artificial fumarolic gases. The artificial fumarolic gases were generated by mixing $CO_2$, CO, $H_2S$, $SO_2$, $H_2$, and $CH_4$ gas streams with a $N_2$ stream sparged through an acidic medium containing HCl and HF, with their compositions varied by adjusting the gas flow rates. The resultant fumarolic gases were collected into an evacuated bottle partially filled with a NaOH absorption solution. While non-condensible gases such as CO, $H_2S$, and $CH_4$ accumulated in the headspace of the bottle, acidic components including $CO_2$, $SO_2$, HCl, and HF that were dissolved into the alkaline solution. Like other acidic components, $H_2S$ also dissolved into the solution, but it reacted with dissolved $Cd^{2+}$ to precipitate as CdS when $Cd(CH_3COO)_2$ was added. The non-condensible gases were analyzed on a gas chromatography. Then, CdS precipitates were separated from the alkaline solution by filtration, and they were pretreated with $H_2O_2$ to oxidize CdS-bound sulfide into sulfate. In addition, a portion of the solution was also pretreated with $H_2O_2$ to oxidize sulfite to sulfate. Following the pretreatment, the resultant samples were analyzed for $SO_4^{2-}$, $Cl^-$ and $F^-$ on an ion chromatography. In the meanwhile, dissolved $CO_2$ was analyzed on a total organic carbon-inorganic carbon analyzer without such pretreatment. According to our experimental results, the measured concentrations of the fumarolic gases were shown to be proportional to the gas flow rates, indicating that the Giggenbach bottle method is adequate for monitoring volcanic gas. The pretreatment and analytical methods employed in this study may also enhance the accuracy and reproducibility of the Giggenbach bottle method.

Lactobacilli have been generally recognized as an important lactic acid bacteria present in the normal human intestinal flora. Fifty two Lactobacillus isolates were recovered from the feces of healthy Koreans whose age ranged from thirteen days after birth to 37 years. Among the isolates above, 17 isolates were tentatively identified as strains of Lactobacillus acidophilus and 3 isolates as strains of Lactobacillus casei. For their characterization, these twenty isolates were subjected to the experiments for the resistance to the artificial gastric juice, pH2.5 and bile salt. Interestingly, 3 strains survived pH 2.5 after 3 hour incubation in the artificial gastric juice with more than 75% of survival rate. L. acidophilus a-4 had the highest survival rate of 100%. Four strains including L. acidophilus a-3 grew gradually in MRS broth in the presence of the artificial bile salt. Cholesterol assimilation was also tested for the 20 isolates. The result showed that cholesterol concentration of the medium was reduced by 10 Lactobacillus isolates with more than 60%, as compared to the control. L. acidophilus a-2 had the highest reduction rate of 77%. Judging from these data obtained in vitro, some isolates ware likely to reach the lower small intestine after consumption without a significant loss of viability, suggesting that they had the potential to be developed as a probiotic culture which might lower the cholesterol level in human.
Agricultural reservoirs are an important water resource nationwide and vulnerable to abnormal climate effects such as drought caused by climate change. Therefore, it is required enhanced management for appropriate operation. Although water-level tracking is necessary through continuous monitoring, it is challenging to measure and observe on-site due to practical problems. This study presents an objective comparison between multiple AI models for water-body extraction using radar images that have the advantages of wide coverage, and frequent revisit time. The proposed methods in this study used Sentinel-1 Synthetic Aperture Radar (SAR) images, and unlike common methods of water extraction based on optical images, they are suitable for long-term monitoring because they are less affected by the weather conditions. We built four AI models such as Support Vector Machine (SVM), Random Forest (RF), Artificial Neural Network (ANN), and Automated Machine Learning (AutoML) using drone images, sentinel-1 SAR and DSM data. There are total of 22 reservoirs of less than 1 million tons for the study, including small and medium-sized reservoirs with an effective storage capacity of less than 300,000 tons. 45 images from 22 reservoirs were used for model training and verification, and the results show that the AutoML model was 0.01 to 0.03 better in the water Intersection over Union (IoU) than the other three models, with Accuracy=0.92 and mIoU=0.81 in a test. As the result, AutoML performed as well as the classical machine learning methods and it is expected that the applicability of the water-body extraction technique by AutoML to monitor reservoirs automatically.

This study was carried out to obtain morphological and physiological characteristics of ear mushrooms for an artificial cultivation. Eighteen strains were cultivated with bag culture and classified into mainly five groups such as brown, black, white, purple and others group. The highest yield was shown in 43007 strain as 98.3 g/bag and 43009, 43016, 43025 and 44035 strains were more than 60 g/bag. Among collected strains, 43007, 43009 and 43035 were selected in this study as superior strains. Three selected strains were investigated for optimal mycelial growth conditions. MCM and GPYM media were selected for the favorable culture medium. The carbon sources of 43007, 43009 and 43035 on mycelial growth were maltose, fructose and glucose, respectively and peptone was selected as a nitrogen source. Highest mycelial growth was observed when the C/N ratio was 10 for 43007 and 20 for 43009 and 43035.

This study was executed in an attempt to investigate a artificial requisites of fruitbody occurrence. Environmental requirements on habitat for fruitbody occurrence of collected cultures resulted in leading to $13-16^{\circ}C$ and 75% relative humidity, and requiring silt loam of soil texture which had more nutritional substances than a dry field. Optimal temperature was $25^{\circ}C$, medium PDB, and pH 5.0 in cultural conditions. Mannose required of 5% in ASI 59002, 59003, 59004, but 3% in ASI 59001 was selected as optimum carbon source. The substrates stimulating sclerotium formation were cotton waste, or cotton waste + oak sawdust (mixture ratio of 8:2), which had 20% additive of wheat barn respectively. Sclerotium was formed well in the substrate adjusted chemical properties by applying 2% of calcium sulfate. Sclerotium formation was the most effective in the treatment of peat moss + oak sawdust (mixture ratio of 5:5) + 30% of wheat barn.
We applied the seismic method to investigate the site characteristics around the Circular Pond (top diameter 7.3 m, bottom diameter 3 m, and depth 4.78 m) at the Ssangsujung Park within the Gongsansung in Gongju. Previous excavations for the cultural assets beneath the Ssangsujung Park disclosed the assumed site of the Palace of the Beakje Dynasty and the Circular Pond containing the Bakje relics. We demonstrated that the seismic prospecting can be applicable to delineate the underground structure around the cultural properties by the three kinds of seismic approaches: walk-away test, conventional refraction method, and equal-distance refraction survey. The last method which is designed by this work ran detect the I-W variations of seismic velocity in the subsurface medium across the Circular Pond on the basis of the difference of the P-wave arrival times between the 1-m-spacing 24 geophones and the corresponding 24 shots parallel with the geophone profile. From the combined results, prominent three-layer velocity structure is observed around the Circular Pond. The bottom layer is interpreted as the basement rock which is exposed near the Ssangsujung whereas the upper layer with relatively lower velocities is interpreted to be the artificial covering. The basement depth beneath the Circular Pond is deeper than the norhern area. The western basement of Circular Pond has the thicker weaker layer compared with the eastern part. Thus, the middle layer could be constructed as the artificial foundation during the Beakje Dynasty. Consequently, the Kong-sansung Circular Pond is possibly built upwardly rather than digging.
