Nutritional Compositions of Varieties of Foxtail millet and Proso millet Developed in Korea (국내 육성 조와 기장 품종의 영양성분 특성)

  • Foxtail millet (Setaria italica L.) and proso millet (Panicum miliaceum L.) were not the major cereal crops in Korea. Although, they are attracting attention due to their nutritional excellence. This study focused on nutritional composition of Korean foxtail millet and proso millet varieties. The protein content of foxtail millet is lower than that of proso millet, and the fat content tends to be higher. Especially, Daname (foxtail millet) and Geumsilchal (proso millet) showed the highest protein content. Also, among these, Chohwangme (foxtail millet) and Manhongchal (proso millet) were the highest fat content, respectively. In the essential amino acids content results, these crops were rich in leucine, phenylalanine and valine. Also, lipids were composed of palmitic acid (C16:0), stearic acid (C18:0), oleic acid (C18:1), linoleic acid (C18:2), linolenic acid (C18:3), arachidic acid (C20:0), gadoleic acid (C20:1), and behenic acid (C22:0). Especially, linoleic acid content was the highest.

A study on the thermally-stimulated displacement current(TSDC) of the PAAS Langmuir-B1odgett(LB) films (PAAS LB 박막의 열자격 변위 전류에 관한 연구)

  • This paper describes a thennally stimulated displacement current(TSDC) of polyamic acid alkylamine salts(PAAS) Langmuir-Blodgett(LB) films, which is a precursor of polyimide(PI). The TSDC measurements of PAAS LB film were performed from room temperature to about $250^{\circ}C$ and the temperature was increased at a rate of $0.2^{\circ}C/s$. This show that this is TSDC peaks at about $70^{\circ}C$ in the arachidic acid LB films, and at about $70^{\circ}C$ and $160^{\circ}C$ in the PAAS LB films. Results of this measurements indicate that one small peak at $70^{\circ}C$ is resulted from a softening of the alkyl group and the large peak at $160^{\circ}C$ is possibly due to dipole moment of C-O group in the PAAS molecule. We have calculated the vertical component of dipole moment of the P AAS LB film out of the TSDC curves. It shows that the dipole moment of PAAS LB film is about -40mD at $70^{\circ}C$ and about 200mD at $160^{\circ}C$ in the first measurement of TSDC. In the second measurement of TSDC of PAAS LB film after cooling down to room temperature, the TSDC peaks are almost disappeared.

  • We investigate characteristics of J-aggregation as take advantage of LB technic. In order to confirm the applications possible for the molecular electronic device, the morphological properties of merocyanine dye were investigated by AFM. ${\pi}-A$ curves investigated the surface pressure of the LB film from a liquid to a solid state ranged between 90 and 100 mN/m. We observed aggregation and it's characteristics by using visible reflection spectroscopy. We have observed morphology of merocyanine dye on gold surface by STM. focuses on results obtained in mercocyanide dye of J-aggregation. When LB films of merocyanine dye are mixed with arachidic acid, J-aggregate formation is exhibited. J-aggregate formation has been serving as typical systems in revealing the physical and structural aspects of nano-sized molecular aggregates constructed as muiltilayers.

A Sensitive Fluorescent Detection Method for Prostaglandins by High Performance Liquid Chromatography (고속액체크로마토그라피에 의한 프로스타글란딘류의 고감도 형광 분석법)

  • The Prostaglandins were derivatized rapidly with monodansyl cadaverine as a fluorophore in mild conditions. The carboxylic moiety of prostaglandins was activated with diethyl phosphorocyanidate and successively coupled with fluorophore in dimethylformamide at room temperature. The labeling yield was reached about 95% at 15 min using arachidic acid $(C_{20:0})$ as a test sample. This derivative showed constant fluorescent intensity at $4^{\circ}C$ for 180 days. The derivatives of prostaglandins were shown high solvent selectivity with tetrahydrofuran in reversed-phase column. therefore, these derivatives could be successfully separated on YMC pack A-212(S-5 120A C8) column in tetrahydrofuran-based eluents. The detection limits of these derivatives was ca. 500 fmol and determination limits was ca. 5 pmol as injected amount in fluorescent detection $({\lambda}ex.\;340\;nm,\;{\lambda}em.\;520\;nm)$. In this method, the ranges of recovery and coefficient of variation were $93.6{\sim}102.7%$ and $4.3{\sim}5.8%$, respectively.

Lipid composition of Korean safflower seeds (한국산 재래종 잇꽃 종실의 지질 성분)

  • The lipid compositions, physical and chemical characteristics of crude oil in Korean safflow, Carthamus tinctorius L., seeds were determined and proxmate composition of it were also analyzed. The proximate composition showed moisture 7.2%, carbohydrate 34.4%, crude protein 18.2%, crude fat 34.8% and ash 5.35. Specific gravity, refractive index, smoking point, melting point and titer of the crude oil were $0.922{\sim}0.927,\;1.468{\sim}1.469,\;210^{\circ}C,\;-15{\sim}-18^{\circ}C\;and\;15{\sim}18$, respectively. Iodine value, saponification value, acid value, esterification value, unsapoifiable matter and saturated fatty acid content of the crude oil were $140{\sim}152,\;186{\sim}192,\;2.6{\sim}3.5,\;179{\sim}186,\;0.8{\sim}1.3%\;and\;6.6{\sim}7.2%$, respectively. Fatty acid compositions were quantitatively determined by G.L.C. to give $70{\sim}78%$ linoleic acid, $10{\sim}23%$ oleic acid and $5{\sim}8%$ palmitic acid as main components, stearic, linolenic and myristic acid were presented in small quantities.

Comparison of Functional Constituents and Biological Activity of the Seed Extracts from Two Mulberry Fruits

  • The seeds from two mulberry fruits [Morus alba (MA) and Cudrania tricuspidata (CT)] were examined for their oil content, and fatty acid, phytosterol and tocopherol compositions and contents. Moreover, polyphenolic compounds and biological activity of the two defatted seed residue extracts were also evaluated. Oil contents of MA and CT seeds were 29.36% and 16.69%, respectively, while MeOH extracts of the defatted MA and CT seed residues were 5.10% and 6.22%, respectively. The two seed oils were composed of 81.4 and 74.37% linoleic, 5.75 and 11.39% oleic, 8.40 and 10.18% palmitic acid, and 3.52 and 3.0% stearic acids, and two other minor fatty acids, such as linolenic and arachidic acids. MA seed had higher contents of phytosterols (507.59 mg/100 g of oil), tocopherols (99.64 mg/100 g of oil), and total flavonoid (106.50 mg/100 g of seed) than CT seed, whereas CT seed had higher levels of total polyphenol than MA seed. The MeOH extract of MA seed residue showed higher antioxidant, anti-diabetic, and anti-melanogenic activity than that of CT seed residue. trans-Resveratrol (9.62 mg/100 g), quercetin (54.83 mg/100 g), and 4-prenylmoracin (48.70 mg/100 g), were found to be the main polyphenolic components in the MeOH extract of MA seed residue. These results indicate that MA seeds are good sources of essential dietary phytochemicals with antioxidant, anti-diabetic and anti-melanogenic activity.

Rapid Gas Chromatographic Screening of Vegetable Oils for Free Fatty Acids (기체크로마토그래피법에 의한 식물성 유지내 유리 지방산의 신속한 스크리닝)

  • A rapid gas chromatographic profiling method for the simultaneous analysis of free fatty and other acids was applied to vegetable oils. Oil samples were dissolved in dichloromethane and the free acids were extracted with saturated $NaHCO_3$ solution. The aqueous extract was acidified and then loaded onto the Chromosorb P column for the extraction. The acids were eluted with diethyl ether selectively from Chromosorb P column and were treated with triethylamine to prevent the losses of volatile acids. Several long chain fatty acids were detected from soybean oil, rice-bran oil, sesame oil and perilla oil. Various organic acids including odd number fatty acids were detected in crude oil, especially sesame oil. Arachidic acid from perilla oil and vanillic acid from sesame oil, which were not reported before were detected. The content ratio of free linoleic acid to oleic acid was $1.02{\sim}1.18$, which was similar to the reported data. When the GC profile of organic acids were simplified to their corresponding retention index spectra of bar graphical forms, they presented characteristic pattern of each vegetable oil that can be quickly recognized.

Diversity in Lipid Contents and Fatty Acid Composition of Soybean Seeds Cultivated in Korea (국내 유통 콩의 지방함량 및 지방산 조성변이)

  • The 117 soybeans seeds were collected from the nine provinces of Korea, and protein and lipid contents, and fatty acid composition levels were evaluated to investigate their relationship. The 100-seed weights of the black soybeans were varied $27.7{\sim}33.1g$, while the 100-seed weight of yellow soybeans were varied $24.6{\sim}36.6g$. Protein and lipid content of the 117 soybean seeds was 38.3% and 17.8%, respectively Protein contents of the 59 black soybean seeds (38.6%) were significantly higher than those of the 58 yellow soybean seeds (37.9%). However, lipid contents of the black soybean seeds (17.6%) were lower than those of the yellow soybean seeds (18.1%). Linoleic and oleic acid composition levels of the 117 soybean seeds were 53.75% and 22.08%. Unsaturated fatty acid levels of soybean seeds showed a statistically significant variability among the nine provinces of Korea, however, the differences were not found in the linoleic (18:2) and oleic acid (18:1) levels. Therefore, it was considered that the significant variability of unsaturated fatty acid were mainly due to the variations of linolenic acid (18:3) level. The composition levels of linoleic, oleic, palmitic (16:0), and linolenic acid in the yellow soybean seeds were 53.43%, 22.73%, 12.23%, and 8.24%, while those of the black soybean seeds were 54.13%, 21.48%, 12.47%, and 8.31%. Obtained results suggested that fatty acid composition levels were varied and possibly influence by the phenotype of seed coat colors. Oleic acid, mono-saturated fatty acid, showed the most remarkable variability between yellow and black soybean seeds, and the composition levels were higher in the yellow soybean seeds. Relationship between unsaturated fatty acid levels and 100-seed weights in the yellow soybean seeds showed a negative correlation (r=-0.513, P<0.01), but no relationship $(r=0.154^{ns})$ was observed in the black soybean seeds.

  • The lipids from the seeds of Pinus koraiensis mostly composed of triacylglycerols (TGs), in which linoleic acid (46.2 mol%) and oleic acid (25.6 mol%) are present as main components in the fatty acid composition. Surprisingly, they also have unusual fatty acids with ${\Delta}^5$-double bond systems such as ${\Delta}^{5.9.12}-C_{18:3}$ (16.0 mol%), ${\Delta}^{5.9}-C_{18:2}$ (2.3 mol%) and ${\Delta}^{5.11.14}-C_{20:3}$ (0.8 mol%). Saturated fatty acids of palmitic, stearic and arachidic acid were present in less than 8.0 mol%. TG was resolved into 17 fractions by reverse-phase HPLC according to so-called partition number (PN) suggested by Plattner, in which TG molecules with ${\Delta}^{5}$-NMDB acyl chains eluted later than did those with ${\Delta}^{9}$-MDB acyl radicals. $Ag^+$-HPLC separated the TG into 14 fractions more clearly than did those with ${\Delta}^{9}$-MDB acyl radicals. $Ag^+$-HPLC separated the TG into 14 fractions more clearly than did reverse-phase HPLC, and the complexity of ${\Delta}^{5.9.12}-C_{18:3}$ moiety with silver ion impregnated in the column bed was in the level between ${\Delta}^{9.12.15}-C_{18:3}$ ($C_{18:3{\omega}3}$) and $C_{18:2{\omega}6}$ (${\Delta}^{9.12}-C_{18:2}$). In the $Ag^+$-HPLC, it was found that the molecular species having a given-numbered double bonds widely spreaded in the acyl chains eluted earlier than those concentrated in one acyl chain. The main molecular species are $(C_{18:2{\omega}6})_2/{\Delta}^{5.9.12}-C_{18:3}$ (14.8 mol%), $C_{18:1{\omega}9}/C_{18:2{\omega}6})_2$ (12.8 mol%) and $C_{18:1{\omega}9}/C_{18:2{\omega}6}/{\Delta}^{5.9.12}-C_{18:3}$ (10.9 mol%).

Chemical Comparison of Germinated- and Ungerminated-Safflower(Carthamus tinctorius) Seeds (홍화(Carthamus tinctorius L.)씨와 발아홍화씨의 화학성분 비교)

  • This study was to investigate the chemical compositions of germinated (GSS)- and ungerminated (UGSS)-safflower (Carthamus tinctorius) seeds. GSS had higher amount of sugar and crude fiber than UGSS, but less amounts of protein and lipid. Levels of $\alpha$-tocopherol and essential amino acids of GSS were higher than those of UGSS, although there are no difference in fatty acid composition between GSS and UGSS. Among the nine phenolic compounds detected, five phenolic compounds, except for two lignans and two flavonoids, were found in both GSS and UGSS. Four serotonin derivatives accounted for about 80 per cent of total phenolic compounds, and levels of five phenolic compounds decreased slightly with germination. These results suggest that germination may enhance the functionality of safflower seed by increasing nutritional compositions and by decreasing phenolic compounds with bitter taste and cathartic effects.
