After-infection Activity of Protective Fungicides against Apple White Rot

  • In a trial to select suitable fungicides for developing a spray program that can control apple white rot effectively, after-infection activities in some protective fungicides were detected. Six fungicides, mancozeb, propineb, benomyl, folpet, azoxystrobin and iminoctadine-triacetate, which had been extensively used in apple orchards, were sprayed on 12-year-old apple trees (cv. Fuji) at 15-day intervals from late May to late July. Disease incidences and infection frequencies of the fruit bagged just before and soon after each spray were examined. When the infection frequency or disease incidence of the fruit bagged after each spraying of fungicide was significantly lower than those of the fruit bagged before spraying, the fungicides appeared to confer after-infection activity. The six fungicides showed diverse activities on white rot: folpet showed after-infection activity on disease development, iminoctadine-triacetate showed after-infection activity on infection, azoxystrobin showed after-infection activity on disease development and infection, and mancozeb, propineb and benomyl showed no distinct activity. The activity of a fungicide became much higher when it was sprayed alternately with other fungicide rather than successive spraying of the same fungicide. Analysis of the properties of these protective fungicides could lead to the development of a highly effective spray program against white rot.

Potential for Augmentation of Fruit Quality by Foliar Application of Bacilli Spores on Apple Tree

  • Previous studies have addressed the management of phyllosphere pathogens by leaf and root-associated microbes. The present study evaluated the effect of the foliar application of three strains of Bacillus spp. on plant growth and fruit quality. The application of a bacilli spore preparation significantly improved leaf growth parameters such as leaf thickness and photosynthesis capacity, indicating that bacilli treatment directly promoted leaf growth. In addition, foliar treatment resulted in an improvement in the key indicators of fruit quality including water, glucose, and sucrose contents. The present results suggest that foliar spraying of beneficial bacilli is a potential treatment of wide application for the improvement of apple quality. Foliar application of bacilli preparation as effective plant growth-promoting rhizobacteria broadens the spectrum of their availability for orchard application.

Identification and characterization of S-RNase genes in apple rootstock and the diversity of S-RNases in Malus species

  • We isolated and confirmed two S-RNases, denoted as mpS1 and mpS2, from apple rootstock 'Marubakaido' (Malus prunifolia Borkh. Var. ringo Asami). These S-RNases contained and conserved five cysteine residues and two histidine residues, which are essential for RNase activity. The mpS1 showed high similarity to S5 (99.1%) of Malus spectabilis, whereas the mpS2 showed 99.5% nucleotide sequence similarity to S26 of (Malus ${\times}$ domestica) and 99.6% to S35 of (Malus sieversii) when compared with reported S-RNases. In amino acid sequences, the mpS1-RNase was almost similar to the S5-RNase of Malus spectabilis, and the mpS2-RNase was similar to the S35 of Malus sieversii, with only one bp being different from the S26-RNase of Malus ${\times}$ domestica. The 57 S-RNases of Malus species were renamed and rearranged containing the new S-RNases, as mprpS35 (mpS2) and mprpS57 (mpS1), for determining S-genotypes and identifying new alleles from apple species (Malus spp.).

First Report of Two Colletotrichum Species Associated with Bitter Rot on Apple Fruit in Korea - C. fructicola and C. siamense

  • Bitter rot caused by the fungal genus Colletotrichum is a well-known, common disease of apple and causes significant yield loss. In 2013, six fungal strains were isolated from Fuji apple fruits exhibiting symptoms of bitter rot from Andong, Korea. These strains were identified as Colletotrichum fructicola and C. siamense based on morphological characteristics and multilocus sequence analysis of the internal transcribed spacer rDNA, actin, calmodulin, chitin synthase, and glyceraldehyde-3-phosphate dehydrogenase Pathogenicity tests confirmed the involvement of C. fructicola and C. siamense in the development of disease symptoms on apple fruits. This is the first report of C. fructicola and C. siamense causing bitter rot on apple fruit in Korea.

Differential Display PCR을 이용한 사과 자가적과성 연관 유전자 탐색 (Identifying Genes Related with Self-thinning Characteristics in Apple by Differential Display PCR)

  • 사과의 경우 한 과총에 5개의 꽃이 피는데 그 중 한 가운데의 중심화가 먼저 개화하여 과일로 발달하고 주위의 측과 4개는 스스로 낙과되는 현상을 자가적과성이라고 한다. 적과는 인위적으로 과실의 숫자를 줄여 잎 수와 과실 수의 균형을 맞추는 작업으로 과실의 크기를 증가시키고, 수세, 수형을 유지시켜 안정적인 생산에 도움을 준다. 노동력 절감을 위해 인간에게 유용하게 사용될 수 있는 특성이 자가적과성인데, 자가적과성 품종의 사과에서 측과는 만개 후 30일 이내에 떨어지고 중심과만 남아서 성숙하게 된다. 51개의 사과 품종으로부터 비자가적과성 그룹 20종, 6월 생리적 낙과 그룹 16종, 자가적과성 그룹 15종을 분류하였다. 대표적인 자가적과성 품종인 Aori #9 로부터 중심과와 측과에서 다르게 발현이 되는 유전자들을 DD-PCR 방법으로 확인하였다. 중심과에서 30개의 clones 과 측과에서 24개의 clones을 선발하여 염기서열을 분석하였다. 주로 측과에서 발현되는 유전자들은 pathogenesis, senescence, temperature stress, protein degradation, fruit browning, sorbitol metabolism에 관여하는 유전자들과 높은 상동성을 나타내고, 중심과에서 발현되는 유전자들의 염기서열을 분석해 보면 anthocyanin의 up-regulation이나 flavonol 생합성, ethylene 생합성에 관여하는 유전자들이 분포한다. Cytochrome P450 유전자의 발현양상을 보기 위해 Real time PCR 분석을 한 결과 중심과보다 측과에서 발현량이 높게 나타났다. 중심과와 측과에서 다르게 발현되는 유전자들의 실제 발현양상을 분석하기 위해 Real time PCR을 이용해서 상대정량을 분석할 계획이며 분자수준에서의 자가적과를 조절하는 기작에 대한 앞으로의 연구는 생력 재배가 가능한 품종 육성의 육종 소재 개발에 활용될 수 있을 것이다.

Morphological Differences between Larvae of the Oriental Fruit Moth (Grapholita molesta Busck) and the Peach Fruit Moth (Carposina sasakii Matsumura) in Korea

  • The oriental fruit moth (Grapholita molesta Busck) and the peach fruit moth (Carposina sasakii Matsumura) are the most severe insect pests affecting apple orchards in Korea. To prevent an outbreak of these two species and to control these agricultural insect pests, it is important to identify them accurately. However, it is hard to classify them when they were in the larval stage since they tunnel into the apple fruit. In this study, surface structures of the two species of larvae were observed using stereo microscope and scanning electron microscope. Distinct differences between the two species of larvae were found. The prothorax spiracles of oriental fruit moth larvae were approximately twice as large as those of peach fruit moth larvae. The arrangements of subventral setae, located around the proleg, were different between oriental fruit moth and peach fruit moth larvae. Furthermore, subdorsal setae of oriental fruit moth were located next to the spiracle on the 8th abdominal segment, while that of peach fruit moth was located above the spiracle. The identification of the two species of larvae observed in this study was confirmed using polymerase chain reaction-restriction fragment length polymorphism method. Surface structural differences are intrinsic characteristics for each species of larvae and can easily be identified using stereo microscope. These specificities will be helpful where a large number of field-collected larvae need to be identified routinely in pest control research.

사과 경도의 비파괴측정을 위한 검량식 개발 및 정확도 향상을 위한 연구 (Development of Calibration Model for Firmness Evaluation of Apple Fruit using Near-infrared Reflectance Spectroscopy)

  • Using Fuji apple fruits cultivated in Kyungpook prefecture, the calibration model for firmness evaluation of fruits by near infrared(NIR) reflectance spectroscopy was developed, and the various influence factors such as instrument variety, measuring method, sample group, apple peel and selection of firmness point were investigated. Spectra of sample were recorded in wavelength range of 1100∼2500nm using NIR spectrometer (InfraAlyzer 500), and data were analyzed by stepwise multiple linear regression of IDAS program. The accuracy of calibration model was the highest when using sample group with wide range, and the firmness mean values obtained in graph by texture analyser(TA) were used as standard data. Chemometrics models were developed using a calibration set of 324 samples and an independent validation set of 216 samples to evaluate the predictive ability of the models. The correlation coefficients and standard error of prediction were 0.84 and 0.094kg, respectively. Using developed calibration model, it was possible to monitor the firmness change of fruits during storage frequently. Time, which was reached to firmness high value in graph by TA, is possible to use as new parameter for freshness of fruit surface during storage.

Mycelial Yield of Pleurotus ostreatus Using Thinned Apple, Pear, and Peach on Submerged Culture

  • The effect of thinned fruits, apple, pear and peach, on the mycelial growth of mushrooms was investigated. The growth of mycelia with the addition of thinned fruit was clearly better than that in the control for all the tested mushrooms. The growth rate of Pleurotus ostreatus was faster than any other mushroom. The optimal concentrations of thinned apple, pear, and peach in a solid culture were 1.0%, 1.0%, and 3%, respectively, while in a liquid culture the optimal concentrations were 5,0%, 3.0%, and 5.0%, respectively. When Pleurotus ostreatus was incubated in a 20-L pilot scale fermenter with 10 L of a liquid medium containing 3% thinned fruit at 25$^{\circ}C$ and 6 vvm for 10 days, the mass-production of mycelia was 74.2 g/10 L (apple), 96.2 g/10 L (pear), and 86.3 g/10 L (peach). The mycelial yield of Pleurotus ostreatus in a medium containing thinned fruit was 2 ∼ 3 times higher than that in the control.

Macrophoma sp.에 의한 사과 부패의 생화학적특성 II. 감염과일의 페놀함량 (Biochemical Characteristics of Apple Rot Caused by Macrophoma sp. II. Phenolic Compound Content in Infected Fruits)

  • 사과 부패병균(Macrophoma sp.)에 감염된 과일에서 총페놀, flavonol, anthocyanin 함량의 병화를 사과의 여러 발육시기에서 조사하였다. 사과가 성숙됨에 따라 과육, 과피의 총페놀함량은 급격히 감소하였다. 부패병균에 감염된 과육에서 건전한 과육보다 다소 총페놀함량이 낮았으며 품종밀러보다 더 감수성인 후지의 감염된 과육에서 이의 감소가 뚜렷하였다. 건전한 과피에서 보다 감염된 과피에서 높은 페놀함량을 보였으며 밀러에서 이의 증가가 현저하였다. 부패된 사과에서 flavonol 함량이 건전 사과보다 높았으며 7월 10일에 Macrophoma에 완전 저항성이었던 사과의 감염된 과피에 flavonol이 크게 축적되었다. anthocyanin 생성도 감염된 과피에 상당히 증가되었으며 품종 밀러에서 뚜렷했다. 이들 결과에서 미루어 사과의 페놀대사변동은 사과 부패병진전과 관계가 있을지 모른다.

Effects of Prohexadione-Ca, Ethephon, and Water Stress on Growth and Productivity of 'Golden Delicious'/M.9 Apple

  • Prohexadione-calcium (Pro-Ca), ethephon and transient water stress were evaluated in a factorial design, as potential inhibitors of early-season shoot growth of high density orchard management of apple (Malus domestica Borkh.) trees. In the experiment, water stress was imposed to one-half of the 7-year-old 'Golden Delicious'/M.9 apple trees in each of 5 blocks, by stopping irrigation for 3 weeks between 35 and 56 days after full bloom (AFB). Within each whole unit, the following Pro-Ca and ethephon treatments were randomly allocated at $2{\times}2$ factorial: a) 0 or 250 $mg{\cdot}L^{-1}$ a.i. Pro-Ca applied at 28 days AFB and b) 0 or 300 $mg{\cdot}L^{-1}$ a.i. ethephon applied twice (35 and 71 days AFB). All trees were hand thinned to king flowers prior to treatments. Vegetative shoot growth was markedly reduced by Pro-Ca, with its effect being obvious within 14 days after application, while ethephon and water stress treatments were less effective. Pro-Ca had no effect on fruit set and yield but slightly increased fruit size. Ethephon substantially reduced the fruit size and yield but had no effect on fruit set. Water stress reduced fruit set, fruit size and yield. With regard to fruit quality, Pro-Ca did not influence fruit shape, flesh firmness and soluble solids contents (SSC) but slightly reduced titratable acidity. Ethephon had no effect on fruit shape but increased firmness, SSC and acidity, while water stress did not influence these fruit quality attributes. Dry weight of dormant spur buds was reduced by both Pro-Ca and water stress, while increased by ethephon. The larger dormant buds led to the larger spur flowers at the tight cluster stage the following spring. Return flowering was promoted only by ethephon, especially on previous season's shoots. There were no significant interactions between Pro-Ca and ethephon or water stress on most variables observed in this study.
