50세 이하 급성기 대상포진의 한양방 복합치료 치험 2례 (A Case Report of the Korean-Western Medicine Combination Treatment of Two Patients with Acute Herpes Zoster under 50 Years of Age)

  • Objectives : The purpose of this study is to report the effect of Korean-Western medicine combination treatment administered to two patients with acute herpes zoster. Methods : During hospitalization, the two patients were treated with Hyeonggaeyeongyotang-gagam, Hwangryunhaedok-tang wet dressing, pharmacopuncture, acupuncture, and other western medications including antiviral and analgesic agents. Photographs of skin lesions, numeric rating scale(NRS), and visual analog scale(VAS) were used to assess the improvement of symptoms. Results : At the time of discharge, both patients showed remarkable improvement in skin lesions, NRS for pain, and VAS for pruritus and insomnia. Conclusions : The results demonstrated that Korean-Western medicine combination treatment may be effective for treating herpes zoster.

잔나비걸상 수용성성분의 항-Varicella Zoster Virus 작용과 Interferon과의 병용효과 (Anti-Varicella Zoster Virus Activity of Water Soluble Components of Elfvingia applanata Alone and in Combinations with Interferons)

  • 잔나비걸상 Elfvingia applananta 자실체의 수용성성분 EA의 varicella zoster virus(Oka strain; VZV/oka)에 대한 항바이러스효과를 plaque reduction assay에 따라 실험한 결과 EA는 용량의존적으로 plaque 형성을 억제하였으며 $EC_{50}$$464.14\;{\mu}g/ml$이었다. EA와 단백질성 항바이러스제인 interferon(IFN) alpha 및 IFN gamma와의 병용시험 결과, 단독처리시와 병용처리시에 m(slope) value가 서로 유사한 값을 보였으므로 이들은 상호 배타적인 약물(mutually exclusive drug)임을 알 수 있었고, IFN alpha와의 병용시 combination index(CI)는 f(a)가 0.50에서 0.90인 유효농도 범위내에서 $0.83{\sim}1.09$의 값을 나타내어 부분적 상승 또는 상가효과를 보였으며, IFN gamma와의 병용시에는 $1.20{\sim}1.24$를 나타내어 길항효과를 보였으므로, IFN alpha와의 병용이 IFN gamma와의 병용보다 더 우수한 병용효과를 나타내었다.

Morin Hydrate Inhibits Influenza Virus entry into Host Cells and Has Anti-inflammatory Effect in Influenza-infected Mice

  • Influenza virus is the major cause of seasonal and pandemic flu. Currently, oseltamivir, a potent and selective inhibitor of neuraminidase of influenza A and B viruses, is the drug of choice for treating patients with influenza virus infection. However, recent emergence of oseltamivir-resistant influenza viruses has limited its efficacy. Morin hydrate (3,5,7,2',4'-pentahydroxyflavone) is a flavonoid isolated from Morus alba L. It has antioxidant, anti-inflammatory, neuroprotective, and anticancer effects partly by the inhibition of the NF-κB signaling pathway. However, its effects on influenza virus have not been studied. We evaluated the antiviral activity of morin hydrate against influenza A/Puerto Rico/8/1934 (A/PR/8; H1N1) and oseltamivir-resistant A/PR/8 influenza viruses in vitro. To determine its mode of action, we carried out time course experiments, and time of addition, hemolysis inhibition, and hemagglutination assays. The effects of the co-administration of morin hydrate and oseltamivir were assessed using the murine model of A/PR/8 infection. We found that morin hydrate reduced hemagglutination by A/PR/8 in vitro. It alleviated the symptoms of A/PR/8-infection, and reduced the levels of pro-inflammatory cytokines and chemokines, such as TNF-α and CCL2, in infected mice. Co-administration of morin hydrate and oseltamivir phosphate reduced the virus titers and attenuated pulmonary inflammation. Our results suggest that morin hydrate exhibits antiviral activity by inhibiting the entry of the virus.

Inhibitory Effect of Hizikia fusiformis Solvent-Partitioned Fractions on Invasion and MMP Activity of HT1080 Human Fibrosarcoma Cells

  • Matrix metalloproteinases (MMPs) are endopeptidases that take significant roles in extracellular matrix degradation and therefore linked to several complications such as metastasis of cancer progression, oxidative stress, and hepatic fibrosis. Hizikia fusiformis, a brown algae, was reported to possess bioactivities, including but not limited to, antiviral, antimicrobial, and anti-inflammatory partly due to bioactive polysaccharide contents. In this study, the potential of H. fusiformis against cancer cell invasion was evaluated through the MMP inhibitory effect in HT1080 fibrosarcoma cells in vitro. H. fusiformis crude extract was fractionated with organic solvents, $H_2O$, n-BuOH, 85% aqueous MeOH, and n-hexane (n-Hex). The non-toxicity of the fractions was confirmed by MTT assay. All fractions inhibited the enzymatic activities of MMP-2 and MMP-9 according to the gelatin zymography assay. Cell migration was also significantly inhibited by the n-Hex fraction. In addition, both gene and protein expressions of MMP-2 and -9, and tissue inhibitor of MMPs (TIMPs) were evaluated by reverse transcription-polymerase chain reaction and Western blotting, respectively. The fractions suppressed the mRNA and protein levels of MMP-2, MMP-9 while elevating the TIMP-1 and TIMP-2, with the $H_2O$ fraction being the least effective while n-Hex fraction the most. Collectively, the n-Hex fraction from brown algae H. fusiformis could be a potential inhibitor of MMPs, suggesting the presence of various derivatives of polysaccharides in high amounts.

위염 및 위궤양에 미치는 Propolis 추출물의 영향 (Effects of Propolis Extract on Antigastritic and Antiulcer)

  • 벌집으로부터 채취한 수지상의 물질인 propolis는 다양한 생화학적 성분을 함유하며 기원전 300년부터 사용되어 온 전통약물이다. 최근 항균, 항 바이러스, 항진균, 국소마취, 면역 활성, 항염, 항산화 작용등의 생물학적 활성이 보고된 바 있다. 본 연구에서는 위염과 위궤양에 미치는 propolis추출물과 분획물의 약조학적 효과를 평가하고자 에탄올 추출물을 hexane, toluene, ethyl acetate로 분획하여 항위염 및 항위궤양 활성을 조사하였다. Propolis 에탄을 추출물은 염산·에탄을 위손상, Shay의 위액 분비 실험에서 용량 의존적인 보호효과를 나타내었고 hexnae과 toluene 분획물은 염산·에탄을 위손상, aspirin에 의한 위손상, Shay의 위액 분비 실험에서 위손상과 위액분비를 감소시켰다. propolis 추출물과 분획물의 위염 및 위궤양에 대한 보호효과는 H+/K+ATPase activity의 억제에 의한 위액분비의 감소에 의한 것으로 생각된다.

Ribavirin의 소아만성 B형 간염에 대한 치료효과 (Therapeutic Trial of Ribavirin on Chronic Hepatitis B in Children)

  • 소아만성B형간염 환아 24명에게 항virus제인 ribavirin을 15mg/kg/day로 14일간 경구투여한 군과 투여하지 않은 대조군으로 나누어 12개월간 혈청 HBsAg과 HBeAg을 추적 검사한 결과 두군 모두에서 HBsAg과 HBeAg의 감소를 볼 수 없었으므로 ribavirin의 소아만성B형간염에 대한 치료효과는 없는것으로 보여진다. 또한 ribavirin 치료에 따른 특별한 부작용도 볼 수 없었다.

Antimicrobial Activity of Korean Propolis Extracts on Oral Pathogenic Microorganisms

  • Propolis has been used as a natural remedy in folk medicine worldwide. The antibacterial, antiviral, antifungal, and antiprotozoal aspects of its antimicrobial properties have been widely investigated. However, few studies focused on its applications in dentistry. Many dental diseases are related to various microorganisms in the oral cavity. In this study, we assessed the antimicrobial activity of Korean propolis extract, collected from 6 different regions, on oral pathogenic microorganisms. The propolis samples, collected from 6 different regions (P1: Uijeongbu, P2: Ansan, P3: Hongcheon, P4: Iksan, P5: Gwangju, and P6: Sangju), were dissolved in ethanol at two different concentrations (10 and 50 mg/ml). Three oral bacteria (Streptococcus mutans, Staphylococcus aureus, and Enterococcus faecalis) and one fungus (Candida albicans) were activated in general broth for 24 hours. Microorganisms were diluted and spread onto agar plates, onto which sterilized 6 mm filter papers with or without each propolis sample were placed. After 24 hours of incubation, clear zones of inhibition were observed. All tests were performed in triplicate. The propolis samples showed significant antibacterial and antifungal activity on oral pathogenic microorganisms; in addition, low-concentration groups showed outstanding antimicrobial efficacy on the 4 different microorganisms. Among the samples, P6 had significantly higher antibacterial activity than that of the others against three different bacteria. In particular, a high concentration of P6 showed a significant antifungal effect. In conclusion, we confirmed that Korean propolis has an inhibitory effect on oral pathogenic bacteria and fungi. Therefore, we suggest the possibility of developing oral medicine and oral care products based on Korean propolis.

수용성 β-1,3-glucans의 면역 활성 효능에 대한 연구 (Immune Stimulating Efficacy of Soluble β-1,3-glucans)

  • Background: $\beta$-1,3-glucans are well known to enhance the immune reactions, resulting in antitumor, antibacterial, antiviral, anticoagulatory and wound healing activities. $\beta$-1, 3-glucans have various activities depending on molecular weight, degree of branching, conformation, water-solubility and intermolecular association. However, the $\beta$-1,3-glucan linked backbone structure is essential and $\beta$-D-glucopyranosyl units are required for immuno-potentiating activities. Result: In this study, we tested the immunophamacological activities of soluble $\beta$-1,3-glucans and confirmed the following activities: (1) $IFN-{\gamma}$ production in PBMCs in the presence or the absence of PHA, LPS, or IL-18; (2) induction of various cytokines in the spleen and thymus; (3) adjuvant effect on the antibody production; (4) nitrogen oxide synthesis in macrophages; (5) the cytotoxic and antitumor effects on cell lines and ICR mice. Conclusion: These results strongly suggested that $\beta$-1,3-glucans possessed various immuno-pharmacological activities.

Antitumor Effect of Soluble ${\beta}$-1,3-Glucan from Agrobacterium sp. R259 KCTC 1019

  • [ ${\beta}$ ]-1,3-Glucans enhance immune reactions such as antitumor, antibacterial, antiviral, anticoagulatory, and wound healing activities. ${\beta}$-1,3-Glucans have various functions depending on the molecular weight, degree of branching, conformation, water solubility, and intermolecular association. The molecular weight of the soluble glucan was about 15,000 as determined by a high-performance size exclusion chromatography. From the infrared (IR) and $^{13}C$ NMR analytical data, the purified soluble glucan was found to exclusively consist of ${\beta}$-D-glucopyranose with 1,3 linkage. We tested the immunestimulating activities of the soluble ${\beta}$-1,3-glucan extracted from Agrobacterium sp. R259 KCTC 1019 and confirmed the following activities. IFN-$_{\gamma}$ and each cytokines were induced in the spleens and thymus of mice treated with soluble ${\beta}$-1,3-glucan. Adjuvant effect was observed on antibody production. Nitric oxide was synthesized in monocytic cell lines treated with ${\beta}$-1,3-glucan. The cytotoxic and antitumor effects were observed on various cancer cell lines and ICR mice. These results strongly suggested that this soluble ${\beta}$-1,3-glucan could be a good candidate for an immune-modulating agent.

제브라피쉬 interferon regulatory factor 10의 주사에 따른 면역 유전자 발현과 VHSV에 대한 방어 효과 (Immune gene expression and protection effect against VHSV by injection of interferon regulatory factor 10 in zebrafish (Danio rerio))

  • Interferon regulatory factors (IRFs) are a family of transcription factors essential to the control of antiviral immune response, cell growth, differentiation and apoptosis. IRF10 of zebrafish (Danio rerio) was negative regulation of the interferonΦ1 and 3 response in vitro. In this study, we analyze the induction of in vivo immune response activation from the IRF10 gene of zebrafish and the protective effect against VHSV. As the results, the group inoculated with IRF10 expression vectors, there was no expression of IFNΦ1, suggestion that IRF10 may function as a negative regulator of IRF3, which binds to the IFNΦ1 promoter. And other types of interferon genes (IFNΦ2-4) are thought to have been activated, inducing to the expression of pro-inflammatory cytokine and Mx genes. As the results of challenge test performed at 14 days after inoculation of the expression vectors, the maximum survival rate [50% (1㎍ DNA) and 42.5% (10㎍ DNA)] for IRF10 group were recorded. Meanwhile, the survival rates of pcDNA3.1 and PBS as the control groups were 10% and 15%, respectively. This study suggests that the possibility that activation of IRF10 molecule could be exploited as a VHS control method.
