An Experimental Study on Some Effects of SOEUMIN-HYANGSAYANGYUI-TANG (소음인향사양위탕(少陰人香砂養胃湯)의 효능(效能)에 관(關)한 실험적(實驗的) 연구(硏究))

  • We have studied some effects of the SOEUMIN-HYANGSAYANGYUI-TANG (S.H.Y.) on the C.N.S. and on the intestines. Several empirical remarks, depending on the different treatments, are investigated throughout this study as follows; 1) The treatment, using atropine bromide, represses the effect of the S.H.Y. on the contractive force of intestines. 2) The treatment, using pheniramine maleate, seems not be able to repress the effect of the S.H.Y. on the contractive force of intestines. 3) The treatment, using cyproheptadine, seems not be able to repress the effect of the S.H.Y. on the contractive force of intestines. 4) The analgesic effect as well as the antipyretic effect are remarked while the acetic acid is applied. 5) The suppressive action on the convulsion, induced by strychine, is not observed. However the significant effect on the convulsion induced by picrotoxin is remarked. 6) The sedactive effect of the S.H.Y., using the Rotor rod test, is not obviously observed.

  • Scolopendra corpus has been used as anticonvulsants and antispasmodics in the oriental medicine. It was previously shown that water extract of Scolopendra corpus had an inhibitory action on ileum in mouse. To investigate the anticonvulsant and antispasmodic activity of Scolopendra corpus, pharmacological studies have been carried out with the water extract of Scolopendra subspinipes mutilans $L.\;K_{OCH}$, which is widely distributed in Korea. 1. The extract depressed convulsions induced by picrotoxin and strychnine. 2. Sedative and antipyretic analgesic action were observed. 3. In mouse and rabbit, tone of intestinal smooth muscle was suppressed with the treatment of the extract and intestinal contraction induced by $BaCl_2$ was also inhibited, suggesting that the extract has a papaverine-like effect. Whereas, in guinea pig, intestinal and tracheal smooth muscle were stimulated, and the effect was antagonized by pre-and after-treatment of diphenhydramine, suggesting that the extract has a histamine-like effect. 4.Flow rate was increased when hind-limb of Toad was perfused with saline containing the extract, but returned to normal within 10 min. Hypotensive effect was observed in rabbit and the effect was abolished by vagotomy.

A Study on Acupuncture of Spring-point(형혈); Effect on Body Temperature and Central Immune System (형용자침이 발열 흰쥐의 체온하강과 중추성 면역에 미치는 영향)

  • Objective : There are many reports that acupuncture has thermoregulatory effects on human and animals. To investigate the effect and mechanism of antipyretic action of acupuncture, we observed the body temperature and cytokine expressions in the hypothalamus of rats. Methods : Lipopolysaccharide (LPS, i.p., 2.5mg/kg) was injected to conscious rats (Sprague-Dawley, male, n=4l) to cause hyperthermia and simple needling (stainless steel, 0.25 mm o.d., 5 mm insertion for 10 sec with no manipulation) was performed bilaterally with the measurement of rectal temperature. Next, we sacrificed rats to remove brain and determined the level of mRNA for interleukin-6 (IL-6), $interleukin-1{\beta}{\;}(IL-1{\beta})$, interleukin-2 (IL-2) and $interferon-{\gamma}{\;}(IFN-{\gamma})$ in the hypothalamus by using reverse transcriptase-polymerase chain reaction (RT-PCR). Resul1s : Needling on forepaw (acupoint HT8) and needling on hindpaw (acupoint BL66 and acupoint LR2) significantly inhibited LPS-induced fever of rats (P<0.01, 10 min after treatment respectively), but same treatment on proximal tail (non-acupoint) did not cause any change on fever. The levels of IL-6 and $IL-1{\beta}$ mRNA in the hypothalamus was significantly enhanced by LPS-injection, while the level of IL-6 and $IL-1{\beta}$ mRNA was markedly reduced after treatment on BL66 (P<0.01). Treatment on forepaw reduced it slightly, but not significantly. Equivalent stimulation on proximal tail did not cause any changes. Conclusions : Our results indicate that acupuncture stimulation on various body parts has differential thermoregulatory effects on LPS-induced fever of rats with site-specificity. And, we suggest that its antipyretic action might be accompanied with the suppression of hypothalamic production of pro-inflammatory cytokine of immune system, IL-6 and $IL-1{\beta}$.

Effect of Solanum lyratum Extract on Dimethylniatrosamine-Induced Liver Damage in Rats (Dimethylnitrosamine에 의해 유도된 흰쥐 간독성에 대한 배풍등 추출물의 보호효과)

  • Solanum lyratum(Solanaceae) has been used as a traditional analgesic, antipyretic and hepatoprotective agents in Korea. In this study, we investigated the hepatoprotective effect of ethylacetate extract of Solanum lyratum (SL) on the dimethylnitrosamine (DMN)-induced liver damage in rats. Oral administration of SL (150, 300 mg/kg daily for 4 weeks) into the DMN-treated rats remarkably prevented the elevation of serum alanine transaminase, aspartate transaminase and alkaline phosphatase levels. SL also increased serum protein level and reduced the hepatic level of malondialdehyde in DMN-treated rats. Furthermore, DMN-induced elevation of hydroxyproline content was reduced by the treatment of SL. In conclusion, these results demonstrated that SL exhibited in vivo hepatoprotective effect against DMN-induced liver injury, and suggest that SL may be useful in the prevention of liver damage.

  • Chunggisan has been widely used in treatment of urticaria which is based on. Oriental Medical literatures. These studies were attempted experimental effects of chunggisan on the analgesia, sedation, antipyretic, anti-inflammatory and isolated illeum of the experimental animals. The result of the studies were as follows: 1. The inhibitory effect was recognized on the Analgesic study by the Acetic acid Method in mice. 2. The spontaneous momentum of mice was restricted and the duration of Hypnosis induced by pentobarbital-sodium in mice was prolonged. 3. Anti-inflammatory effect was recognized significantly on the rat hind paws edema induced by dextran. 4. The inhibitory effect of leukocyte emigration was recognized by the CMC airpouch method of rats. 5. The inhibitory effect was noted on capillary permeability induced by histamin in mice. 6. Spontaneous mobilities in isolated ileum of mice were significantly suppressed, and contraction by acetylcholine, barium chloride and histamine were inhibited. According to the above results, it is expected that chunggisan can be widely used in treatment of urticaria.

  • In order to investigate experimentally the clinical effect of Soyangin-Hyongbangdojoksan (少陽人荊防導赤散) that was prescribed to cure the Bisuhanpyohanbyong (脾受寒表寒病) of Soyangin (少陽人). The author experimented various activities of dried extract from hyongbangdojoksan (Sample-I) and mixed extract of each dried extract of hyongbangdojoksan (Sample-II) by the methods prescribed in the experimental parts. The results summarized as follows. 1. In the acetic acid method experiment, analgesic effect was noted in sample-I & Sample-II. 2. Anti-inflammatory effect on the edema induced by carrageenin was noted in Sample-I. 3. Antipyretic effect is not noted in Sample-I and Sample-II. 4. On urinary volume change and blood electrolyte clearance, the result is not significant, on the contrary the urine electrolyte discharge is effective in Sample-I and Sample-II. According to the above results, the effects based on oriental medical references approximate to the actual experimental results.

  • In order to investigate experimentally the clinical effects of Soeuminsohaphyangwon that was prescribed to cure the Wisunanrihanbyung of Soeum-In, the author experimented various activities of mixed extract from the Soeuminsohaphyangwon by the method prescribed in the experimental part. The results of the studies were summerized as follows : 1. Suppressive action was not shown on the convulsion induced by strychnine but significant effect was noted on the convulsion induced by picrotoxin and caffeine. 2. In acetic acid method experiment analgesic effect was noted. 3. A prolongation of anesthetic time by Pentobarbital sodium and antipyretic effect was observed. 4. Relaxing action was noted on the ileums of mice, also same effect was recognized on contraction of the ileums due to acetylcholine, barium chloride and histamine 2 HCl. 5. The expansion of blood vessels by relaxation of smooth muscle and fall of blood pressure were noted. According to the above results, effects based on oriental medical references were approximate to the actual experimental results.

  • In order to investigate the pharmacological action of combined preparation of crude drugs, 'Whangryonhaedok-Tang' were studied. Pharmacological studies of the water extract, methanol insoluble fraction and methanol soluble fraction obtained from 'Whangryonhaedok-Tang' were conducted. The results of this investigation were summarized as follows; Central convulsions induced by strychnine, picrotoxine and caffeine were depressed in mice. Analgesic effect was observed in mice and rats. Antipyretic effect was observed in rats and rabbits. Sedative effect was observed.

  • This study has been carried out to investigate the effects of Ukgansan(UGS) extract on anti-convulsive, antipyretic, analgesic, sedative and GABAergic system of experimental animals. The results of this study were as follows : 1. UGS extract prolonged significantly the beginning time to convulsion and death induced by strychnine. 2. UGS extract prolonged significantly the time to death induced by electrical shock of ECT unit.(3 sec, 200 F, 25 mA) 3. On the experiment of hypothermic effects of UGS extract on the rectal temperature of mouse, UGS extract decreased significantly the rectal temperature of mouse 4. On the experiment of antipyretic effects of UGS extract on the febrile induced by the subcutaneous injection of $150\;{\mu}g/kg$ endotoxin in mouse, UGS extract decreased significantly the rectal temperature of mouse. 5. On the experiment of analgesic effects of UGS extract on the writhing syndrome induced by intraperitoneal injection 0.7% acetic acid 1 ml/100g in mouse, the writhing syndrome induced by acetic acid was reduced significantly by administration of UGS extract. 6. On the experiment of effects of UGS extract on spontaneous motor activity measured by wheel cage method in mice, the spontaneous motor activity was reduced significantly by administration of UGS extract. 7. On the experiment of effects of UGS extract on the activity of GABA-transaminase(GABA-T) in mouse brains after 21 days of oral administration of UGS extract. the activity of GABA-T was reduced significantly by administration of UGS extract. 8. On the experiment of effects of UGS extract on the activity concentration of GABA in mouse brain after 21 days of oral administration of UGS extract, the activity concentration of GABA was reduced significantly by administration of UGS extract. 9 On the experiment of effect of UGS water extract on the activity of GAD in mouse brain after 21 days of oral administration of UGS extract, the activity of GAD was reduced significantly by administration of UGS extract. According to the these results, Ukgansan extracts reveal the effects on the anti-convulsive, antipyretic, analgesic, sedative and GABAergic system.
