Experiment Study on Antioxidant Activites of Aqua-acupuncture of Cynanchum Wilfordii Hemsley (백화수오약침의 항산화작용에 관한 실험적 연구)

  • This study was performed to prove the antioxidant activities of Cynanchum Wilfordii Hemsley(白何首烏) by way of aqua- acupuncture methods. After 10% & 20% concentrations of liquid extract of Cynanchum Wilfordii Hemsley were acupunctured on Joksamri(足三里) of rats with acute and chronic experimental oxidation by AAPH(2, 2'-azobis(aminoidinopropane), hydrochoride), various kinds of experiments were measured. The results were obtained as follows: 1. In acute antioxidant experiment, the amount of TBARS indicated irregular increase and decrease to over 68% in experiment group in comparison with normal group. 2. In the serum test of acute antioxident experiment, uric acid and total protein and albumin were significantly increased in experiment group in comparison with control group. 3. In chronic antioxident experiment, the change of body weight was significantly decreased in 20% experimental groups in comparison with control group, and only liver significantly increased in 10% & 20% experiment group. 4. In chronic antioxident experiment, total protein significantly increased in 10%, 20% experimental groups and creatinine and BUN were significantly decreased in 10% experiment group. 5. In the chronic antioxident experiment, serum GOT, GPT activity didn't show no significantly change, LDH activity was significantly increase in 10% experiment group in comparison with control group. 6. In the chronic antioxident experiment, the tolerance test with enzyme activities of erythrocyte membrane was significantly decreased in 10% & 20% experimental groups. 7. In the chronic antioxident experiment, the change of TBA was significantly decreased in 10% & 20% experimental group. 8. In the chronic antioxident experiment, the amount of cytochrome p-450 was significantly increased in 10% & 20% experimental groups in comparison with control group. 9. In the chronic antioxident experiment, the amount of cytochrome b5 was significantly increase in 20% experimental group in comparison with control group. 10. In the chronic antioxident experiment, the change of activity of superoxide dismutase, and catalase were significantly increased in 20% experiment group in comparison with control group. From the above result, it was concluded considers the liquid extract of Cynanchum Wilfordii Hemsley(白何首烏) could be used as antiaging drugs, because it delayed the aging process and retarded senility.

Antioxident components from Aralia continentalis

  • The root of Aralia continentalis Kitagawa(Araliaceae) have been used as an analgesic and fever remedy, and for treatment of rheumatism in Chinese medicine, whereas the young leaves are used for salad. An antioxidant activity of the young leaves of A continentalis was determined by measuring lipid peroxide produced when a mouse liver homogenate was exposed to the air at 37$^{\circ}C$, using 2-thiobarbituric acid(TBA) and by evaluation the radical scavenging activity on 1,l-diphenyl-2 picrylhydrazyl (DPPH) radical. Bioassay guided fractionation of MeOH extract isolated six flavonoid compounds as active components from EtOAc fraction. Adenosine and two saponins were isolated from the weak active BuOH fraction. The antioxidant effect by DPPH radical scavenging activity showed that quercetin was the most active among these compounds. Hyperoside and kaempferol were also active, while 6"-O-acetyl astragalin, astragalin, trifolin, adenosine, oleanolic acid 28-O-glucosyl ester and salsoloside C methyl ester were almost inactive. All the compounds were identified by spectroscopic methods.

  • Antioxidant enzymes such as catalase (CAT), glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD) are known to inhibit oxidative reactions by incativating compounds responsible for the formation of ree radicals. SOD transforms superoxide radical into hydrogen peroxide which is precursor to active free radicals. CAT reduces hydrogen peroxide to water. GSH-Px reduces hydroperoxides to corresponding alcohols. Antioxidant enzyme activities of muscle are different by animal species age, stress and exercise, muscle type and part, conditions of post mortem, storage and processing which are related to oxidative deterioration I muscle foods as well as oxidative defence in living systems. Antioxidant enzyme systems are enhanced rather than weakened in aging skeletal muscle. Red muscle contains higher antioxidant enzyme activity than white muscle. The antioxidant enzyme activities of poultry are higher in leg than in breast, and those of beef are higher in redder and more unstable muscles. It is clear that the effectiveness of the antioxidant enzyme in muscle foods seems to be influenced by meat processing operations. Both GSH-Px and CAT are inactivated by heat processing NaCl also influence the efficiency of the antioxident enzymes since its presence diminishes their catalyitc activity.

Effects of Organic fertilizer Application on Growth and Medicinal ingredients of Platycodon grandiflorum Radix (유기질 비료시용에 따른 도라지의 생육 및 약용성분에 미치는 효과)

  • This study was conducted to elucidate the effect of organic fertilizer on growth characteristics, saponin contents and antioxident activity of Platycodin grandiflorum Ridix roots for organic farming. As basal fertilizers, chemical fertilizer, mixed organic fertilizer, bacterial cultures and fermented oil cake and decomposed manure were treated based on 3 kg/10a level before transplanting Platycodin grandiflorum Ridix. In root length, when chemical fertilizer plot and mixed organic fertilizer plot were treated, root length was recorded the highest scores (25.3 and 24.0 cm) Root width was recorded the highest score (26.6 cm) in chemical fertilizer plot. The number of fine-roots was the highest in mixed organic fertilizer plot and chemical fertilizer plot (20.0 and 17.0), respectively. Fresh weight, which affects directly yield, was the highest in organic fertilizer plot (55.7 g/plant). The content of Platycodin D was shown to 327.4~373.8 mg/100 g, the highest values were observed in organic fertilizer plot. The total polyphenol and flavonoid contents were recorded the highest scores (15.5 and 15.3 mg/g, respectively) in organic fertilizer plot. In this study we confirmed that application of organic fertilizer was effective to increase yield and pharmacological effect through increase of the number of fine-roots with high saponin contents.

Effects of Hot Water Soluble Extract from Angelicae Gigantis Radix on the Lipid Metabolism and Antioxident Defense System in rats fed Hypercholesterol Diet (당귀 열수추출물이 고콜레스테롤 식이를 공급한 흰쥐의 지질대사와 황사화계에 미치는 영향)

  • To investigate the effects of hot water soluble extract from Angelicae Gigantis Radix on lipid metabolism and antioxidant defense system in rats fed hypercholesterol diet, Sprague-Dawley male rats were fed either hypercholesterol diet with water or hot water soluble extract from Angelicae Gigantis Radix. Hypercholesterol diet contained 18 % beeftallow and 5 % cholesterol. After 4 weeks of experimental periods serum and liver obtained. The level of serum lipid and the activities of GOT and GPT was measured. The level of lipid and TBARS and the activity of GSH-Px was measured also. The results obtained were: 1) Food intake in the experimental group supplied hot water soluble extract from Angelicae Gigantis Radix was low significantly. 2) In the experimental group supplied hot water soluble extract from Angelicae Gigantis Radix of serum total lipid and total cholesterol was significantly low. 3) Serum GOT activity was significantly low in the experimental group supplied hotwater soluble extract from Angelicae Gigantis Radix. There was no difference of serum GPT activity between control and experimental group supplied hot water soluble extract from angelical Gigantis Radix. 4) There was no difference of lipid composition, TBARS level and GSH-px activity in liver between control and experimental group. The result of this study indicates that the hot water soluble extract from Angelicae Gigantis Radix was effective in decreasing the food intake of the experimental animals in the hypercholesterol diet and also effective in decreasing the levels of serum total lipid, total cholesterol and GOT activity. However it was not effective to change the level of lipid, TBARS and GSH-Px activity of liver.

Optimization of Cookies Prepared with Mori cortex radicis Powder (상백피 분말 첨가 쿠키의 제조조건 최적화)

  • The purpose of this study was to investigate the antioxidant effects of Mori cortex radicis powder and to determine the optimal composite recipe by testing different amount of Mori cortex radicis powder and sugar in cookies prepared with Mori cortex radicis powder. In regard to its antioxident effects, Mori cortex radicis powder had a total phenolic content and DPPH free radical scavenging activity of 149.56 mg GAE/g and $137.77{\mu}g/mL$, respectively. The response surface methodology was used to obtain ten experimental points (including two replicates for Mori cortex radicis powder and sugar) and Mori cortex radicis cookie formulation was optimized using rheology. The results of the sensory evaluation produced significant values for color (p<0.05), texture (p<0.05), sweetness (p<0.01) and overall quality (p<0.05), and the results of instrumental analysis showed significant values in sweetness (p<0.001), redness (p<0.01) and spread ratio (p<0.5). As a result, the optimum formulations obtained by numerical and graphical methods were found to be 16.84 g of Mori cortex radicis powder and 64.42 g of sugar.

Effect of Dry Powders, Ethanol Extracts and Juices of Radish and Onion on Lipid Metabolism and Antioxidative Capacity in Rats (무, 양파의 시료제조 방법에 따른 흰쥐의 지방대사와 항산화능에 관한 연구)

  • This study was performed to investigate the effects of dry powders, ethanol extracts and juices of radish and onion on lipid metabolism, lipid peroxidation and antioxidative enzyme activity in rats. Forty-nine male Sprague-Dawley rats weighing 157$\pm$6g were blocked into seven groups according to body weight and raised for four weeks with diets containing 5%(w/w) dry powders of two different vegetables consumed frequently by Korean-radish(Raphanus sativus L.) and onion(Allium cepa L.), ethanol extracts and juices from equal amount of each dry powder. All the powders, ethanol extracts and juices of radish and onion decreased total lipids, triglycerides and total cholesterol concentrations in plasma and liver. Above all, onion ethanol extract decreased them most remarkably. It was thought that organosulfur compounds and flavonoids extracted from onion by ethanol inhibited biosynthesis and absorption of lipid and promoted degradation of lipid. Radish powder also decreased them by increasing fecal excretions of total lipids, triglycerides and total cholesterol most effectively. Catalase and glutathine peroxidase(GSH-px) activities in red blood cell(RBC) were most remarkably increased by radish powder and onion powder respectively. Superoxide dismutase(SOD), catalase and GSH-px activities in liver were most remarkably increased by onion ethanol extract, radish powder and onion ethanol extract respectively. Xanthine oxidase(XOD) activities in liver were most effectively decreased by ethanol extracts of radish and onion. Thiobarbituric acid reactive substance (TBARS) levels in plasma and liver of experimental groups were significantly lower than those of controls. Above all, onion powder decreased them most effectively. It was thought that vitamin E and high flavonoids in onion powder inhibited lipid peroxidation, promoting liver and RBC SOD, catalase and GSH-px activities and inhibiting XOD activities effectively. Flavonoids in onion ethanol extract inhibited lipid peroxidation, promoting three antioxident enzyme activities and inhibiting XOD activities most remarkably. Also flavonoids and high vitamin C in radish powder inhibited lipid peroxidation, promoting liver and RBC catalase most remarkably and inhibiting XOD activities. In conclusion, radish and onion were effective in lowering lipid levels and inhibiting of lipid peroxidation in animal tissue. From these data, radish and onion can be recommended in the treatment and prevention of diseases such as cardiovascular disease and cancer and in delaying aging. As ethanol from onion were most effective in lowering lipid level and promoting three antioxident enzymes, and inhibited lipid peroxidation as did we should try to utilize onion skin which is discarded though reported to have abundant flavonoids. Extraction and Biological Activities of Essential Oil from Thuja occidental Leaves (서양측백나무 잎으로부터 식물정유 추출 및 생리활성)

  • Essential oil of Thuja occidental leaves was extracted by steam distillation method, and chemical composition of essential oil was analyzed by GC-MS. The essential oil was tested to antimicrorbiaI activity against 28 test microorganisms and antioxident assay through DPPH free radical scaveging test and insecticidal activity by mosquito insecticide assay. The major constituents in the essential oil of T. occidental were monoterpene as d-isothujone, ${\alpha}-thujone$, camphor, L-fenchone, and hornyl acetate. The essential oil have broad antimicrobiaI activity, which showed strong antimicrobiaI activity for Streptococcus pyogenes as bacteria, Alternaria alternata, Aspergillus niger, Aspergillus oryzae, Saccharomyces cerevisiae, and Streptococcus mutans as fungi. The essential oil also showed DPPH free radical scavenging ability at concentration of $36\;{\mu}{\ell}/m{\ell}$. In spraying and fumigation tests, the essential oil had strong insecticidal activity against mosquito.

Enhanced Antioxident Effect by over Expression of Tomato β-carotene Hydroxylase Gene (ChyB) Using Agrobacterium-infiltration in Tobacco Plant (Agro-infiltration을 이용한 토마토 β-carotene hydroxylase 유전자(ChyB) 과발현 및 담배식물체의 항산화 효과 증진)

  • Several reports indicated that astaxanthin and zeaxanthin have more active anticancer activity than pro-vitamin A carotenes. ${\beta}$-carotene hydroxylase is a key enzyme to synthesize zeaxanthin and astaxanthin in carotenoids biosynthesis pathway. We isolated the ChyB gene encoding ${\beta}$-carotene hydroxylase from tomato leaves. The ChyB gene (1.5Kbp) fragment was cloned into the binary vector and designated to pIG121-ChyB-tom. Agrobacterium-mediated infiltration was used for transient assay in Nicotiana benthamiana. Leaf samples were collected 0, 1, 2, 3 days after infiltration (DAI). RT-PCR result showed that the expression of ${\beta}$-carotene hydroxylase transcripts was not detected in control (0DAI), but its expression was detected after 1 DPI and increased later on. When the activity of ${\beta}$-carotene hydroxylase was measured, the 1,1-diphenyl-pricryl hydrazyl (DPPH) radical scavenging activity (27%) at 2 DAI was significantly higher than that (21%) at 0 DAI. These results indicated that anti-oxidant activity dramatically increased at 2 DAI in tobacco leaves was due to over expression of tomato ${\beta}$-carotene hydroxylase. These results can be the foundation to develop tomato cultivars with high oxy-carotenoids content using the ChyB gene transformation.

Antioxidant and Synergist Effect of Extract Isolated from Commercial Green, Oolong and Black Tea (시판 녹차, 홍차, 오룡차의 항산화 및 상승효과)

  • The antioxidative effect of tea extracts from green tea, oolong tea and black tea was investigated. The extraction yields of crude catechin were green tea 5.4%, oolong tea 3.5%, and black tea 2.5%. Green tea, oolong tea and black tea of crude catechin showed antioxident activity at concentrations of 200ppm, and the oolong tea showed highest antioxidant effect. The browning intensities(at 420 nm) of green, oolong and black tea were 0.140, 0.582 and 0.915, respectively and green tea showed highest hydrogen donating ability(HDA) and than followed by oolong or black tea. Also, when the crude catechin(100 ppm) was used in combination with ${\alpha}$-tocopherol(50 ppm) and ascorbyl palmitate(50 ppm), the crude cathechin showed very strong synergistic effect, comparable to that of crude catechin(200 ppm).
