Antioxidative Effects of Diarylheptanoids from Alnus hirsute (물오리나무에서 분리된 Diarylheptanoid의 항산화작용)

  • Four diarylheptanoids oregonin, hirsutanonol, 1,7-bis- (3,4-dihydroxyphenyl)-heptane-5-O-$\beta$-D-xylopyanoside and 1,7-bis-(3,4-dihydroxyphenyl)-heptane-5-0-$\beta$-D-glucopyranoside isolated from Alnus hirsuta were evaluated for their antioxidative effects with 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and lipid peroxidation generation system mediated by addition of $H_2O$$_2$and/or Fe$^{2+}$ to rat liver homogenate. All these diarylheptanoids showed significant inhibitory activities against the DPPH radical and demonstrated strong activities in preventing the lipid peroxidation induced by $H_2O$$_2$.>.

  • Rhus verniciflua Stokes (RVS) is a widely used herbal plant with various biological properties. Our previous study using in vitro platelet aggregation in whole blood showed that ethyl acetate layer of RVS had strong anti-aggregatory activity. In this study. to investigate the anti-aggregatory activity and antioxidative effects of RVS ethyl acetate layer. the layer was subsequently fractionated by ODS columm chromatograph (50% MeOH). As a result. the fraction 3 was most inhibited the aggregation of platelet in rat whole blood induced by thrombin and all fraction of RVS was detected strong antioxidative effect. (omitted)

Antioxidative Effect of Phenolic Acids in Defatted Perilla Flour on Soybean Oil (탈지들깨박 중 페놀산의 대두유에 대한 항산화 효과(I))

  • Free phenolic acid, soluble phenolic acid ester and insoluble bound phenolic acid were extracted from defatted perilla flour. Their antioxidative effects were compared with those of BHA, AE and TBHQ for soybean oils by measuring acid and peroxide values at 60$^{\circ}C$ for 25 days. The patterns of these extracts were compared by using gas chromatography. Free phenolic acid and soluble phenolic acid ester extracts showed a higher antioxidative effects than BHA and AP. Among phenolic extracts, free phenolic acid showed the most effective antioxidant activity in soybean oil. Six types of free phenolic acid, 3 types of soluble phenolic acid ester, and 2 types of insoluble phenolic acid were found in the extract.

Antimicrobial and Antioxidative Effects of Roselle (Hibiscus sabdariffa L.) Flower Extract and Its Fractions on Skin Microorganisms and Oxidation

  • Antimicrobial and antioxidative effects of 14 different herbal flower extracts on skin microorganisms and oxidation were tested in this research. Herbal flower extracts were prepared with 70% ethanol. Among the herbal flower extracts, roselle (Hibiscus sabdariffa L.) flower extract showed the highest antimicrobial activity against Staphylococcus epidermidis as determined by a paper disc method. The seventy % ethanol extract of roselle flower was fractionated by sequential hexane, chloroform, ethyl acetate, n-butanol, and water fractionation. The growth of S. epidermidis, Streptomyces collinus, Streptomyces coeruleoprunus, Salmonella enteritidis, Vibrio parahaemolyticus, and Malassezia pachydermatis was most efficiently inhibited by ethyl acetate fraction of roselle flower extract as determined by a paper disc method and growth inhibition curves. In addition, the ethyl acetate fraction, water fraction and butanol fraction showed free radical scavenging and DNA cleavage inhibition activities. These results demonstrate that roselle flowers hold antimicrobial and antioxidative activities against skin microorganisms and oxidants.

Antioxidative Activity and Varietal Difference of Cyanidin 3-glucoside and Peonidin 3-glucoside Contents in Pigmented Rice

  • The content of anthocyanin, cyanidin 3-glucoside (C3G) and the peonidin 3-glucoside (P3G) from 591 rice cultivars and the antioxidative activities of MeOH extract from 8 rice cultivars were evaluated. Among them, C3G content of pigmented rice were ranged from 0 to 451.9mg in 100 g brown rice, while the P3G contents were in the range from 0 to 42.7mg in 100g brown rice. There was no correlation between C3G and P3G content. Total anthocyanin content ranged from 0 to 475.1mg in 100g brown rice. The antioxidative activity, the scavenging activity on DPPH (2,2-diphenyl-picryl-hydrazyl) radical of MeOH extracts from rice grain, were different according to cultivars. The activity of blockish purple pericarp rice cultivars was twice stronger than that of white pericarp cultivar. Especially, the antioxidative activity of Heugjinjubyeo was four times stronger than that of white pericarp cultivar. The scavenging effect on DPPH radical in rice extracts was related to the total anthocyanin contents of the extracts.

  • The rhizome of Alisma orientale Juzep (Alismataceae) has been used as a crude drug for diabetes, edema, inflammation and urinary disturbances in oriental medicine. Recent animal studies have shown that the extract of Alisma orientale rhizome (AOR) can potently lower high levels of serum lipids and improve insulin resistance, which are usually detected in patients and animals with non-alcoholic fatty liver disease. So, we studied the antioxidative effects of AOR extracts and fraction in vitro and their protective effects against acute hepatotoxicity induced by $CCl_4$ in vivo.. We then investigated the effects of each fraction on hepatotoxicity induced by tert-butyl hydroperoxide (t-BHP). DAOR (dichloromethane fraction of the Alisma orientale rhizome) scavenged free radicals and superoxide anions. DAOR protected against $CCl_4$ induced hepatotoxicity. DAOR had hepatoprotective and antioxidative effects against t-BHP-induced hepatotoxicity in HepG2 cells and in rats.

  • In the course of screening for antioxidant compounds by measuring the DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging effect and superoxide quenching activity, methanol extract of Cyperus difformis (Cyperaceae) was found to show potent antioxidant activities. Subsequent activity-guided fractionation of the methanolic extract led to the isolation of three phenolic compounds, rosmarinic acid (1), luteolin (2) and caffeic acid (3). Their structures were elucidated by spectroscopic studies. Three compounds showed the significant antioxidative effects on DPPH and nitric oxide radical scavenging effects, riboflavin and xanthin originated superoxide quenching activities. Among them, compound 3 showed the most significant antioxidative effects on those four antioxidant tests. Compound 3 showed better antioxidative activities than vitamin C.

Effect of Dried Powder and Juice of Aster Scaber on Lipid Metabolism and Antioxidative Capacity in Rats (참취의 건분 및 녹즙이 흰쥐의 지방대사와 항산화능에 미치는 영향)

  • This study was performed to investigate the effects of dried powder and juice of Aster scaber on lipid metabolism and antioxidative capacity in rats. Twenty one male Sprague-Dawley rats weighing 170.9$\pm$15.1g were blocked into 3 groups according to body weight and raised for 4 weeks with diets containing 5%(w/w) dried powder and juice from the same amount of Aster scaber. Food intake, body weight gain, food efficiency ratio and organ weights were not different among all the experimental groups. On lipid metabolism, both dried powder and juice supplementation decreased plasma total lipid, triglyceride(TG), total cholesterol and liver cholesterol levels by increasing fecal lipid excretions. Plasma thiobarbituric acid reactive substance(TBARS) concentrations were significantly decreased in dried powder and juice groups, whereas liver TBARS concentrations were not affected by experimental diets. Red blood cell(RBC) superoxide dismutase(SOD) activities were significantly increased by dried powder and juice supplementation, while activities of other antioxidative enzymes in RBC and liver were decreased or showed no changes. In conclusion, both Aster scaber dried powder and juice had the effects on lowering lipid levels by increasing fecal lipid excretions, and inhibiting of plasma lipid peroxidation in animals and the effects of juice were higher than that of dried powder. Effects of Korean Green Tea, Oolong Tea and Black Tea Beverage on the Antioxidative Detoxification in Rat Poisoned with Cadmium (한국산 녹차, 우롱차 및 홍차가 카드뮴에 중독된 흰쥐 간조직의 항산화적 해독작용에 미치는 영향)

  • In order to investigate the effect of Korean green tea, oolong tea and black tea beverage on the antioxidative detoxification in cadmium(Cd) poisoned rat liver, male Sprague-Dawley rat weighing 143$\pm$3.2g were divided into control and experimental groups. The experimental groups were fed standard diet containing 40ppm Cd and were given distilled water(CD), 5% black tea(BT), oolong tea(OT) and green tea(GT), respectively. Tea beverages were extracted from 5G dry leaves of teas in 100ml hot distilled water by the treatment at 85$^{\circ}C$ for 3 min. Liver xanthine oxidase(XOD) activity was increased by the administration of Cd except GT group. Liver superoxide dismutase(SOD), glutathione peroxidase(GSH-px), glutathione S-transferase(GST) activities were decreased by te administration of Cd but did not decreased by the administration of green tea(in GT group). Vitamin E and reduced glutathione contents were significantly decreased in Cd administered groups. Liver lipid peroxide value in Cd administered groups were increased compared to control group, but was not increased in GT group. Serum glutamic oxaloacetic transaminase(GOT) activities in CD, OT, BT groups were higher than control, but that in GT group was similar to control group. Serum glutamic pyruvic transaminase(GPT) activity was not significantly different among various groups. It was concluded that green tea might alleviate peroxidative damage in Cd-administered rat liver by reinforcing antioxidative detoxification system.

  • Phytochemical examination of seeds of Astragalus sinicus has led to the isolation and characterization of kaempferol $3-O-{\beta}-D-apiofuranosyl-(1{\rightarrow}2)-{\alpha}-L-rhamnopyranosyl-(1{\rightarrow}6)-{\beta}-D-glucopyranoside$ (1), quercetin $3-O-{\beta}-D-xylopyranosyl-(1{\rightarrow}2)-{\beta}-D-glucopyranoside$ (2), ampelopsin (3), ampelopsin $3'-O-{\beta}-D-xylopyranoside$ (4), ampelopsin $3'-O-{\beta}-Dxylopyranoside$ (5), myricetin (6), myricetin $3'-O-{\beta}-D-glucopyranoside$ (7), myricetin $3'-O-{\beta}-D-xylopyranoside$ (8). Antioxidative activity of these compounds was determined by measuring the radical scavenging effect on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals. Flavanonols(3,4, and 5) showed potent antioxidative activity.
