Effect of Kyungohkgo(瓊玉膏) on Antioxidant Capacity in D-galactose Induced Aging Rats (경옥고(瓊玉膏)가 노화유발 흰쥐의 항산화능에 미치는 영향)

  • Objectives : In order to examine the antioxidant activities of Kyungohkgo(瓊玉膏), the study was done through measurement of parameters such as Thiobarbituric acid reactive substance(TBARS), Superoxide dismutase(SOD), Catalase(CAT), Glutathione peroxidase(GSH-px), Plasma total lipid, Plasma total triglyceride, Plasma total cholesterol, HDL-cholesterol concentrations in rat erythrocytes and plasma. Methods : Sprague-Dawley rats divided into 3 groups, Normal group(l2 weeks old), pathologically induced group(injected D-galatose 50mg/kg, 1time/day for 6 weeks, CONTROL) and Kyungohkgo(瓊玉膏) administered group(D-galactose 50mg/kg and Kyungohkgo extracts 1125.0mg/kg 1time/day for 6 weeks, KOG). Rats were sacrificed and TBARS, SOD, CAT, Plasma total lipid, Plasma triglyceride, Plasma total cholesterol, Plasma HDL-cholesterol concentrations and GSH-px were measured in rat erythrocytes and plasma. Results : Plasma TBARS concentrations of KOG group were significantly lower than those of control. Red blood cell(RBC) SOD activities of KOG group was increased(F=3.619, p=0.052, ANOVA test), and RBC catalase activities of all experimental group were not significantly different. RBC GSH-px activities of KOG group was increased(F=6.844, p=0.008, ANOVA test). The changes of Plasma triglyceride was not significantly different. Plasma total lipid of KOG group showed significant decrease compared to the control group(F=19.337, p=0.0001, ANOVA test). Plasma total cholesterol and HDL-cholesterol concentrations of all experimental groups were not significantly different. Conclusions : According to the above results, it is considered that Kyungohkgo(瓊玉膏) is effective in inhibiting lipid peroxidation and increasing antioxidative enzyme activities in D-galactose induced aging rat.

  • This study was conducted to investigate the protox activity, the PPIX accumulation and the activity of antioxidative enzymes of the oxyfluorfen-tolerant and -susceptible rice cultivars by oxyfluorfen and oxytluorfen-similar herbicides treatment. When treated with acifluorfen, bifenox or oxadiazon, the oxyfluorfen-tolerant rice cultivars showed less decreased in fresh weight than the susceptible rice cultivars. The inhibition of protox activity was in the order of acifluorfen > oxyfluorfen > bifenox > oxadiazon, and the PPIX accumulation was in the sequence of oxadiazon > acifluorfen > oxyfluorfen > bifenox. The inhibition of protox activity and the PPIX accumulation by the herbicide was greater in the susceptible rice cultivars than in the tolerant rice cultivars. The effect inhibiting the decrease of chlorophyll content resulting from the treatment of GC, tetrapyrrole biosynthesis inhibitor, was in the order of oxyfluorfen > acifluorfen > bifenox > oxadiazon, and the tolerant rice cultivar had more than the susceptible rice cultivar. In the treatment of DPE and oxadiazon, the activities of MDAR, POX and GR was higher in the tolerant rice cultivar than in the susceptible rice cultivar, and in the case of POX isozyme the activities of main D and E bands increased in the tolerant rice cultivars at tested herbicides but they didn't in the susceptible rice cultivar.

Studies on Antioxidant Activity and Inhibition of Nitric Oxide Synthesis from Codium fragile (청각추출물의 항산화 및 일산화질소 합성 저해 연구)

  • This study was carried out to investigate the biological effects from Codium fragile. Methanol extract of Codium fragile increased two times at 2500 ㎍/ml the growth of Lactobacillus plantarum that associated with probiotic properties of lactic acid bacteria of Kimchi. Ethyl acetate extract of Codium fragile inhibited the cellulase activity up to approximately 60% at $2500\;{\mu}g/ml$. Methanol extract of Codium fragile was fractionated into several subfractions and their antioxidant activities were measured by using DPPH radical scavenging and SOD-like activity. Especially the antioxidative activity of ethyl acetate fraction was shown higher than that of other fractions and its fraction showed higher contents of total phenolic compounds, indicating the positive relationship between DPPH radical scavenging effect and total polyphenol content. Stimulation of the macrophages RAW264.7 cells with lipopolysaccharide (LPS) resulted in increased production of nitric oxide (NO) in the medium. However, the methanol extract of Codium fragile showed marked inhibition of NO synthesis in a dose-dependent manner. This result suggest that Codium fragile plays significant role for activation of immune system in the pathogenesis of inflammatory diseases.

Changes in the Antioxidative and Antigenotoxic Effects After the Cooking Process of Sulgidduk Containing Pine Needle Juice (솔잎착즙액을 첨가한 설기떡의 가열조리에 따른 항산화 및 항유전독성 효능 검증)

  • Recently, two formulas of sulgidduk added to pine needle juice (PNJ) with various physiological activities were developed for metabolic syndrome patients in our lab. According to previous studies, cooking may alter antioxidant properties by initiating destruction, release or transformation of antioxidant compounds contained in food. Therefore, the aim of this study was to compare the antioxidant activities and antigenotixic effects of sulgidduk with/without PNJ and to note changes in these activities after cooking. The ingredients of sulgidduk was added on the basis of 100% rice flour as follows: conventional sulgidduk (S): 1.5% salt, 30.0% sugar; PNJ added to sulgidduk A (PS-A): 1.4% salt, 30.0% sugar, and 1.0% PNJ; PNJ added to sulgidduk B (PS-B): 1.5% salt, 21.4% sugar, and 1.4% PNJ. Ethanol and water extracts of sulgidduk were analyzed for the total phenolic content (TPC), DPPH radical scavenging activity (DPPH RSA), total radical-trapping antioxidant potential (TRAP), oxygen radical absorbance capacity (ORAC), and antigenotoxic effect by comet assay. The ethanol extracts PS-A and PS-B showed higher TPC and antioxidant activities (DPPH RSA, TRAP, and ORAC) than did the S ethanol extract before cooking. The more PNJ was added, the higher TPC and anitoxidant activities were observed in sulgidduk (PS-A$200{\mu}M$ of $H_2O_2$. Taken together, this study suggests that sulgidduk added to 1.44% of pine needle juice may be a good option antioxidant and antigenotoxic source.

Studies on the Chemical Constituents Biological Activities of Mulberry Extracts (오디(Mulberry) 추출물의 성분분석 및 생리활성에 관한 연구)

  • The freezed Mulberry (10 kg) was extracted with 80% EtOH, concentrated, and fractionated with a series of n-hexane, ethyl acetate, and water on a separatory funnel. A portion of ethyl acetate soluble (22 g) was chromatographed on a Sephadex LH-20 column using a series of aqueous methanol as eluents. The isolated compounds were identified by cellulose TLC, $^1H-$,$^{13}C$-NMR, FAB, and EI-MS. Quercetin-3-O-${\beta}$-D-glucopyranoside (Compound I), protocatechuic acid (Compound II), p-hydroxybenzoic acid (Compound III) were isolated from the ethyl acetate soluble fraction. In antioxidative activities of the fractionated extractives using DPPH radical scavenging test, EtOAc and water soluble fractions indicated better than BHT as contro and in in vitro tests using MTT assay, there was no cytotoxicity. Also, tyrosinase inhibition and anticancer activities were not so good, but there may be a potential as a cosmetic raw material because the cell extension effect was excellent.

  • To investigate the effect of large patch infection on oxidative stress induction, growth, $H_2O_2$ concentration and lipid peroxidation were compared between pathogen-infected and healthy (control) zoysiagrass. The sampling for leaves and roots were carried out every 2 days for a period of 6 days. Pathogen-infection increased root mortality by 30% compared to control. Dry mass was not significantly affected by pathogen-infection until day 4, but significant decreases in both leaves (-14%) and roots (-20%) were observed at day 6. The $H_2O_2$ concentration in pathogen-infected leaves rapidly increased within the first 2 days(+28%) and then slightly decreased. The increase of $H_2O_2$ in pathogen-infected roots was distinct, showing 1.7-fold higher level than control at day 6. The extent of lipid peroxidation caused by pathogen-infection continuously increased for the first 4 days. This was then stagnated until day 6. In roots, the significant increase of lipid peroxidation was observed only at day 2. These results indicate that large patch-infection induces oxidative stress, and that the oxidative stress responsive pattern was plant organ specific.

Effect of Whey Protein Isolate and Lactobacillus spp. Cell Extracts on Intracellular Antioxidative Activities in Human Prostate Epitherial Cells (유청단백질 및 Lactobacillus spp. 추출물이 전립선 세포 내 항산화 활성에 미치는 영향)

  • Bovine whey protein are rich in cysteine, which is the rate limiting amino acid for synthesis of antioxidant glutathione(GSH). Some strains of Lactobacillus caseihas been reported to contain high level of GSH in cell extracts. The objective ofthis study was to determine whether enzymatically hydrolyzed whey protein isolate(WPI) and cell extract of Lb. casei HY2782 could increase intracellular GSH concentrations and protect against oxidant induced cell death in human prostate epithelial cell line (designated as RWPE1, and PC3MMM2 cells). Treatment of RWPE1 cellsandPC3MMM2 cells with hydrolyzed WPI (500g/ml) significantly increased GSH by28.2% and38.4% respectively. Compared with control cells receiving no hydrolyzed WPI(P<0.05). hydrolyzed WPI and Lb casei HY2782 cell extracts significantly protected RWPE1 and PC3MMM2 cellsfrom oxidant induced cell death compared with controls receiving no WPI. DNA damage associated with oxidant treatment was demonstrated by single cell gel (SCG) electrophoresis.

  • N-acetyl-L-cysteine (NAC) having a thiol, a precursor for glutathione (GSH), is known as one of the antioxidants. NAC used as a radioprotector against ionizing radiation (IR)-induced injury and damage. The aim of this study was to evaluate the radioprotective effects of NAC against IR-induced cell damage in Saccharomyces cerevisiae and the antioxidative effect of NAC on transcriptional level of yeast antioxidant enzyme genes such as superoxide dismutase (SOD) and catalase. In the present study, yeast cells were pretreated with various concentrations of NAC and/or irradiated with various doses of gamma rays. The cell viability was measured by counting the cell forming unit (CFU). The quantitative real-time PCR was performed for analysis of gene expression of SOD and catalase. The viability of irradiated cells was not improved by pretreatment with NAC. Ionizing radiation with 100 Gy highly induced the gene expression of antioxidant enzymes. In the irradiated group with NAC pretreatment, the gene expression of SOD and catalase was gradually reduced with the increased concentrations of NAC. These results indicate that NAC can act as a useful antioxidant to scavenge reactive oxygen species in vivo, but does not protect cells against IR-induced cell death in S. cerevisiae.

Changes in 4'-O-methylpyridoxine (ginkgotoxin) and antioxidant activity in ginkgo biloba seeds in different cooking conditions (조리조건에 따른 은행알의 4'-O-methylpyridoxine (ginkgotoxin) 함량 및 항산화 활성 변화)

  • The purpose of this study was to examine the best cooking condition to decrease 4'-O-methylpyridonxine (MPN, ginkgotoxin) and increase the antioxidative effect. We also examined the change in color of ginkgo biloba seeds after different cooking methods and times. MPN content was decreased with an increase in the cooking time. For the reduction of MPN content, the most efficient cooking method was pan-frying. In particular, MPN content was largely reduced after 8 min of pan-frying. 1,1-Diphenyl-2-picrylhydrazyl radical scavenging activity and total flavonoid content were increased after 11 min of pan-frying and this level was maintained until 14 min. The total phenolic compound content was maintained for up to 30 min by steaming and 11 min by pan-frying. Therefore, the optimum condition was established by pan-frying from 8 to 11 min.

DNA Breakage by Salvianolic acid B in the Presence of Cu (II) (구리이온(II)이 존재할 때 Salvianolic acid B에 의한 DNA 절단)

  • Salvianolic acid B, which is a compound in the Salvia miltiorrhiza, has diverse biological activities, In particular, the antioxidative effects were reported to be involved in the protection of hepatocytes, neurons, and various cell types. On the other hand, some phenolic compounds, such as ferulic acid, which is regarded as an antioxidant, plays a pro-oxidative role in the specific transitional metal environment, which could explain the anticancer effect. This study examined the pro-oxidative effects of salvianolic acid B in the presence of $Cu^{2+}$. Treatment with both salvianolic acid B and $Cu^{2+}$ induced the transition of supercoiled DNA to the open circular or linear form but not in the sole salvianolic acid B or $Cu^{2+}$ treatments. Salvianolic acid B reduced the $Cu^{2+}$ to $Cu^+$ using neocuproine, a $Cu^+$ specific chelator. In addition, catalase, an enzyme that breaks down the $H_2O_2$ to water and molecular oxygen, inhibited the DNA breakage. $H_2O_2$, a reactive oxygen species, has detrimental effects on biological molecules, particularly DNA. Overall, the reduction of $Cu^{2+}$ by salvianolic acid B could lead to the production of $H_2O_2$ followed by DNA breakage. These results suggest that the pro-oxidative effects could be the one of the anti-cancer mechanisms of salvianolic acid B, which remains to be explained.
