Antioxidative Effects of Purple Sweet Potato Extracts (자색고구마 추출물의 항산화 효과)

  • The colored sweet potato, particularly purple sweet potato, has been well known to contain anthocyanins abundantly. This study was conducted to examine the antioxidant properties of purple sweet potato. The chopped purple sweet potato was extracted 2 times with water or acetone for 18 hours at $28^{\circ}C$. The antioxidative potential of each solvent extract was assessed by DPPH free radical scavenging activity assay, FRAP assay, and total phenolic contents. The results showed that both extracts had not only high DPPH free radical scavenging activity but had high level of total phenolic compounds. Furthermore, both solvent extracts were found to have antioxidative effects in human colon cancer cells (HCT 116, HT 29) in DCFDA assay. The notable antioxidant activity of purple sweet potato suggests its significant health benefit and deserves further study to develop into functional food ingredient.

Antilipid Peroxidative Effect of Houttuynia cordata (즙채의 과산화지질 생성 저해효과)

  • Houttuynia cordata (Saururaceae) has pungent smell and taste. It has been regarded as detoxicant, antipyretic, antiinflammantory and diuretic agents. In order to evaluate antioxidative and antilipidperoxidative efficacies, its fractions ($H_2O$, 20% MeOH, 40% MeOH, 60% MeOH, 170% MeOH) were measured by DPPH method and TBARS assay on rat liver homogenate. It was revealed that 60% MeOH fractions had potential antioxidative activity and inhibited lipid peroxidation significantly: In active fractions, we isolated rutin, hyperin, quercitrin and quercetin.

Antioxidative Stress and Antimutagenic Effects of Lentinus edodes Ethanol Extracts (표고버섯 에탄올 추출물의 산화적 스트레스 억제 효과와 항돌연변이 효과)

  • The antioxidative effect and antimutagenic capacity in ethanol extracts of Lentinus edodes were studied for suggestion of prevention and dietetic treatment of chronic diseases and development of antioxidative and antimutagenic functional food by employing biological and biochemical assay. The $IC_{50}$ of MDA with BSA conjugation reaction, lipid peroxidation and scavenging effect on DPPH radical in ethanol extracts of Lentinus edodes showed 74.58 mg/assay, 5.747 mg/assay and 0.939 mg/assay respectively. So, the most effective antioxidative capacity in ethanol extracts of Lentinus edodes was the scavenging effect on DPPH radical, among the method used this study. The indirect and direct antimutagenic effects of ethanol extracts of Lentinus edodes were examined by Ames test using Salmonella typimurium TA98 and TA100. The inhibition rates on indirect mutagenicity mediated by 2-anthramine showed 91.67% in the Salmonella typimurium TA98 and 96.60% in the Salmonella typimurium TA100. The inhibitory effect on direct mutagenicity mediated by sodium azide in Salmonella typimurium TA100 was 22.83%. and mediated by 2-nitrofluorene in Salmonella typimurium TA98 was 5.34%. This data indicates that ethanol extracts of Lentinus edodes have more effective effects on indirect mutagenicity than direct mutagenicity. From this result, it believed to have a possible antioxidative and antimutagenic capacities, and taken for the candidate of prevention and dietetic treatment of chronic diseases and development of antioxidative and antimutagenic functional food.

  • Acanthopanax species have traditionally been used as a tonic, a sedative as well as in the treatment of rheumatism, hypertension and diabetes. In the present study, oxidative stress was induced in Vero cells by incubating the cells with glucose and the cell viability was measured by MTT assay. The concentration of glucose which 50% of cell viability was 125 mM $(IC_{50})$ and the cell viability was increased to $87.6{\pm}8.8%$ by treatment of the extracts of Acanthopanax divaricatus var. albeofructus. The antioxidative activity of water extract of different parts of the Acanthopanax plant was investigated by DPPH (1,1-diphenyl-2-picrylhydrazyl) assay, xylenol orange assay, TBARS (thiobarbituric acid reactive substances) assay and enzyme (superoxide anion and catalase) assay. Each extract (leaves, root, stem and fruits) of the plant showed free radical and $H_2O_2$ scavenging activity. The extract also inhibited lipid peroxidation and recovered enzyme (superoxide anion dismutase and catalase) activity in Vero cells treated with glucose.

Antioxidative Activity of Crackers Made with a Guava(Psidium guajava Linn.) Leaf Extract Harvested in Korea (한국산 구아바 잎 추출물을 첨가한 크래커의 항산화활성)

  • The objectives of this study were to investigate the antioxidative activity of crackers made with a guava(Psidium guajava Linn.) leaf extract harvested in Korea. Guava leaf extraction using boiling water showed significantly higher antioxidative activities than extracting using 70% ethanol based on the higher total phenolic contents, FRAP, and ABTS assays(p<0.05). The crackers containing 1% guava leaf extract, and 0.075% BHT were stored at $63^{\circ}C$ for 7 days for the Schaal oven test, and the oxidative stability(AV, POV), antioxidative activity(DPPH, FRAP, ABTS assay), and sensory evaluation were compared. The crackers containing 1% guava leaf extract were found to have a higher oxidative stability than the control due to a lower acid value and peroxide value after 7 days of storage. The antioxidative activities of the crackers containing 1% guava leaf extract was the highest after 7 days as determined in the DPPH and ABTS assay, and was lower than crackers containing 0.075% BHT after 4 days as assessed by the FRAP assay. In the sensory evaluation, the crackers containing the 1% guava leaf extract had the highest scores in terms of taste, texture, and overall palatability than others at increasing storage time. As a result, the addition of 1% guava leaf extract harvested in Korea increased the antioxidative effect as well as the sensory acceptability of crackers.

Measurement of Antioxidant Activity of Anserine, Taurine, and L-Histidine in vitro and Content of Anserine, Taurine, and L-Histidine in Mature and Juvenile Rainbow Trout (Onchorhynchus mykiss) Muscle

  • The content of anserine, taurine, and L-histidine was measured by HPNC in the muscle of mature(670~690g) and juvenile(80~120g) rainbow trout fatmed in Chungsun, Korea. The concentration of anserine and taurine was higher in mature rainbow trout than in juvenile, but that of L-histidine was lower in mature than in juvenile. When measured with the chemiluminescence(CL) assay, anserine and taurine showed very powerful antioxidative activity above physiological concentration rainbow trout. Taurine still showed antioxidative activity below physiological concentration, while anserine showed prooxidative activity below that. L-Histidine was prooxidative dose-dependently. In TBA method, while taurine showed very week antioxidative effect, anserine appeared very powerful antioxidant and L-histidine prooxidant at physiological concentration. There was no synergism between anserine and taurine and anserine inhibited prooxidative effect of L-histidine.

Functional Activities of Ethanol Extracts from Flammulina velutipes (팽이버섯 에탄올 추출물의 기능적 특성)

  • This study was conducted to evaluate the antioxidative effect and antimutagenic capacity of ethanol extracts of Flammulina velutipes by employing biological and biochemical assays. The $IC_{50}$ of MDA with BSA conjugation reaction, lipid peroxidation and scavenging effect on DPPH radical in ethanol extracts of Flammulina velutipes was found to be 28.39 mg/assay, 9.33 mg/assay and 144.61 mg/assay respectively. Therefore, the most effective antioxidative capacity of ethanol extracts of Flammulina velutipes was $Fe^+$-induced linoleate peroxidation, among the method used this study. The indirect and direct antimutagenic effects of the ethanol extracts of Flammulina velutipes were examined by the Ames test using Salmonella typimurium TA98 and TA100. The inhibition rates on indirect mutagenicity mediated by 2-anthramine and on direct mutagenicity mediated by sodium azide in Salmonella typimurium TA100 and mediated by 2-nitrofluorene in Salmonella typimurium TA98 were 0%, respectively. These findings indicate that ethanol extracts of Flammulina velutipes have no effects on indirect and direct mutagenicity. Based on these results, it believed that the ethanol extracts of Flammulina velutipes has antioxidative capacities, and is a the candidate for the prevention and dietetic treatment of chronic diseases and the development of antioxidative functional food.

Study on Antioxidant and Cytotoxic Activities in Ethanol Extract from Prunus mume (오매 에탄올 추출물의 항산화 활성 및 항암 활성 연구)

  • Prunus mume is well known contain many functional materials that play beneficial roles in the human body. Studies have found that many organic acids and polyphenol compounds exist in Prunus mume. In this study, content of total polyphenols and flavonoids, antioxidative activity and cytotoxicity of ethanol extract from Prunus mume were measured Contents of total polyphenolic and total flavonoid compounds in ethanol extract from Prunus mume were 16.92 mg/g and 59.95 mg/g, respectively. The $IC_{50}$ of ethanol extract from Prunus mume were $237.72 {\mu}g$/assay and $239.58 {\mu}g$/assay by DPPH and ABTS radical cation scavenging test, respectively. Additionally, ferric ion reducing antioxidant power (FRAP) value of ethanol extract from Prunus mume was 0.94 mM ($FeSO_4$ eq.) by $800 {\mu}g$/assay. Cytotoxicity of Prunus mume against five kinds of cancer cell lines increased as the extract concentration increased Especially, cytotoxicity of the ethanol extract against A-549 (lung cancer line) was higher than that against any other cancer cell line by both MTT and SRB assay. These results show that ethanol extract of Prunus mume has considerably high antioxidative and cytotoxic activities.

Antioxidative Constituents from Lycopus lucidus

  • Three phenolic compounds, rosmarinic acid (1), methyl rosmarinate (2), ethyl rosmarinate (3), and two flavonoids, luteolin (4), luteolin-7-O-$\beta$-D-glucuronide methyl ester (5) were isolated from the aerial part of Lycopus lucidus (Labiatae). Their structures were determined by chemical and spectral analysis. Compounds 1-5 exhibited potent antioxidative activity on the NBT superoxide scavenging assay. The $IC_{50}$ values for compounds 1-5 were 2.59, 1.42, 0.78, 2.83, and 3.05 $\mu\textrm{g}$/mL respectively. In addition, five compounds were isolated from this plant for the first time.

Comparative Study of the Antioxidative Potential of Common Natural Flavonoids and Isoflavonoids (천연물인 플라보노이드와 이소플라보노이드의 항산화 효과 비교연구)

  • The half maximal inhibitory concentration ($IC_{50}$) values and trolox equivalent antioxidant capacity (TEAC) values were calculated by a 2,2'-diphenylpicrylhydrazyl (DPPH) assay and a 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid ($ABTS^{+{\cdot}}$) assay, in order to determine the antioxidative activities of the compounds. On the basis of the DPPH assay, quercetin had the strongest antioxidative potential of the flavonoids, followed in order by fisetin, 7,8-dihydroxyflavone, morin and kaempferol. Quercetin, fisetin and 7,8-dihydroxyflavone had higher antioxidant potentials than butyl hydroxyl anisole. Quercetin had the highest TEAC value amongst the flavonoids and isoflavonoids, followed in order by 3-hydroxyflavone, fisetin, 7,8-dihydroxyflavone and morin. Comparatively, isoflavonoids were found to have significantly weaker antioxidative potential than the flavonoids.
