In our search for the natural cosmetic ingredients, we found that Euryale ferox seed extract exhibited the strong antioxidative activity. Five active compounds were isolated from the ethyl acetate extract through various chromatographic methods and their structures were determined by NMR and MS spectral analysis. These compounds were identified as fucosterol (1), 3-(4-hydroxy-3-methoxybenzyl)-4-[(7'R),5'-dihydroxy-3'-methoxybenzyl]tetrahydrofuran (2), resorcinol (3), pyrogallol (4) and 4-O-methylgallic acid (5).We evaluated the antioxidative, antielastase activities and melanogenesis inhibitory effects of these compounds. The $SC_{50}$ values of compounds 2 ~ 5 for free radical scavenging activity were $17.0\;{\sim}\;100.2\;{\mu}M$ and especially compounds 4 and 5 were 6-fold more effective than ferulic acid as a positive control. And compounds 2 ~ 4 inhibited human neutrophil elastase with $IC_{50}$ values of $18.8\;{\sim}\;78.2\;{\mu}M$ and compound 3 also inhibited melanin synthesis in B16F10 melanoma cells with an $IC_{50}$ value of $492.8\;{\mu}M$. These results suggest that Euryale ferox extract having a lot of various active ingredients may be useful as a natural multi-functioning agent.
Free amino acid and mineral analysis of beverages with Gugija (Lycii fructus) extracts performed to determine the antioxidative and anti-diabetes activities of the beverages. The four major free amino acids in the water- or ethanol-extracts of the Gugija beverages were asparagine (115.23, 51.95 mg%), methionine (20.02, 22.07 mg%), aspartic acid (19.65, 13.72 mg%) and taurine (18.64, 22.44 mg%). The mineral contents of the Gugija beverages with water- and ethanol-extracts were in the order K>Mg>Na>Ca>Zn>Fe. Antioxidant activity determined by DPPH and ABTS radical scavenging activity. There was no significant difference in DPPH and ABTS radical scavenging activities between the water-extract beverages and the ethanol-extract beverages, although more phenolics and flavonoids were found in the ethanol-extract beverage. Higher anti-${\alpha}$-glucosidase activity was observed in the ethanol-extract beverage compared to acabose, which was used as the control and is an inhibitor of ${\alpha}$-glucosidase, suggesting that the Gugija beverage with ethanol extract could be a potential hypoglycemic agent.

In this study, the antioxidative effect, cellular protective effect and inhibitory effect on tyrosinase of Cosmos bipinnatus extracts were investigated. The ethyl acetate fraction of Cosmos bipinnatus flower extract ($11.48\;{\mu}g$/mL) showed more excellent free radical (1,1-diphenyl-2-picrylhydrazyl radical, DPPH) scavenging activity (FSC50) than those of leaf and stem extracts ($17.45\;{\mu}g$/mL). Reactive oxygen species (ROS) scavenging activity (OSC50) of Cosmos bipinnatus extracts on ROS generated in $Fe^{3+}$-EDTA/H2O2 system were investigated using the luminol-dependent chemiluminescence assay. The ethyl acetate fraction of Cosmos bipinnatus flower extract ($0.56\;{\mu}g$/mL) showed 3 times more excellent ROS scavenging activity than L-ascorbic acid ($1.50\;{\mu}g$/mL). The protective effects of the ethyl acetate fractions of extracts from different parts of Cosmos bipinnatus on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The ethyl acetate fractions of leaf and stem extract and flower extracts suppressed photohemolysis in a concentration dependent manner ($10\sim50\;{\mu}g$/mL). The inhibitory effect of ethyl acetate fraction of Cosmos bipinnatus flower extract ($62.75\;{\mu}g$/mL) on tyrosinase was investigated to assess the whitening efficacy. The ethyl acetate fraction of Cosmos bipinnatus flower extract showed 3.5 times higher tyrosinase inhibitory effect than arbutin ($226.88\;{\mu}g$/mL) known as an effective whitening agent. These results indicate that fractions of Cosmos bipinnatus extracts can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging $^1O2$ and other ROS, and protect cellular membranes against ROS. Fractions of Cosmos bipinnatus extracts can be applicable to new functional cosmetics for antioxidant and whitening.

This study investigated the antioxidative activity and whitening efficacy of Crataegus pinnatifida Bunge fruit 70% ethanol extract (CFE). The total polyphenol contents of CFE was 61.31 mg/g, and the total flavonoid contents was 25.42 mg/g. The electron donating ability of CFE at a concentration of $1,000{\mu}g/mL$ was 85.80%. The ABTS radical scavenging activity, and reducing power of CFE at a concentration of $1,000{\mu}g/mL$ were 17.3% and 0.31, relatively. The maximum permissible levels of CFE in melanoma cells were $100{\mu}g/mL$. CFE at $50{\mu}g/mL$ reduced melanin contents by 8.5%. CFE at $1,000{\mu}g/mL$ reduced intracellular tyrosinase activity by 46.83%. Collectively, the results of this study suggest that CFE effectively inhibited tyrosinase activity, whereas melanin synthesis was weak. These results suggest that Crataegus pinnatifida Bunge fruit could be used as a whitening agent and antioxidant resources for functional foodstuffs, cosmetics, and beauty industrial materials.
This study examined the neurotoxicity induced by lead acetate (LA) on cultured C6 glioma cells and the protective effects of Euphorbiae humifusae L. (EH) extract against LA-induced cytotoxicity. In this study, LA exhibited neurotoxicity in a dose-dependent manner compared to the control, and was determined to be highly-toxic according to the toxic criteria. The $XTT_{50}$ value of LA was calculated at a concentration of $38.6{\mu}M$ after C6 glioma cells were incubated for 72 hours in the media containing $30{\sim}50{\mu}M$ of LA, respectively. In addition, LA-induced neurotoxicity was suggested to correlate with the level of oxidative stress because vitamin E, an antioxidant, increased the cell viability damaged by LA-induced cytotoxicity. The EH extract effectively prevented cell injury from LA-induced cytotoxicity via its antioxidative effects, such as inhibitory ability of lipid peroxidation, superoxide dismutase-like activity and 1,1-diphenyl-2-picrylhydrazyl-radical scavenging activity. These antioxidative effects may result in components, such as polyphenol or flavonoids including gallic acid or quercetin. In conclusion, natural resources, such as EH extracts, may be a useful putative agent for the treatment of diseases associated with oxidative stress, such as lead neurotoxicity.
This study examined the neurotoxicity of aluminum sulfate (AS), an environmental pollutant, and the protective effect of Phrymaleptostachya var. asiatica HARA (PLVAH) extract on the neurotoxicity induced by AS in the cultured C6 glioma cells. For this study, the cell viability and antioxidative effects, such as electron donating (ED) activity, lipid peroxidation (LP) activity, and superoxide anion-radical (SAR) scavenging activity, were analyzed. AS decreased the cell viability significantly in a dose-dependent manner and the $XTT_{50}$ value was measured at $120.0{\mu}M$ of AS. The neurotoxicity of AS was determined to be mid-toxic by Borenfreund and Puerner's toxic criteria. In addition, the catalase (CAT), antioxidant enzyme remarkably increased the cell viability injured by AS-induced neurotoxicity in these cultures. Regarding the protective effect of the PLVAH extract on AS-induced neurotoxicity, PLVAH extract significantly increased the ED ability, and the inhibitory ability of the LP and SAR scavenging ability. These findings suggest that oxidative stress is involved in the cytotoxicity of AS, and the PLVAH extract effectively protected against AS-induced neurotoxicity by its antioxidative effects. Natural resources, such as the PLVAH extract may be a putative therapeutic agent for the treatment of the toxicity induced by heavy metallic compounds, such as AS correlated with the oxidative stress.
The protective effect of Agrimonia pilosa var. (AP) extract on potassium dichromate ($K_2Cr_2O_7$)-induced cytotoxicity in cultured NIH3T3 fibroblasts, was examined by performing an XTT assay for the cell viability and antioxidative effects, such as lactate dehydrogenase (LDH) activity and superoxide anion-radical (SAR) scavenging activity. In this study, $K_2Cr_2O_7$ decreased the cell viability significantly in a dose-dependent manner, and the $XTT_{50}$ value was determined to be $37.5{\mu}M$, which was highly-toxic according to the Borenfreund and Puerner' toxic criteria. The antioxidant, butylated hydroxytoluene (BHT), increased remarkably the cell viability damaged by $K_2Cr_2O_7$-induced cytotoxicity in these cultures. With regard to the protective effect of the AP extract on $K_2Cr_2O_7$-induced cytotoxicity, AP extract produced a significant increase in cell viability and antioxidative effects as the inhibitory ability LDH and SAR scavenging ability. These findings suggest that oxidative stress is involved in the cytotoxicity of $K_2Cr_2O_7$, and the AP extract effectively protected the cells from $K_2Cr_2O_7$-induced cytotoxicity by antioxidative effects. These results suggest that natural resources, such as AP extract, may be a putative therapeutic agent for the diminution or treatment of cytotoxicity induced by heavy metallic bases, such as $K_2Cr_2O_7$ correlated with oxidative stress.
This study describes the anti-inflammatory, anti-oxidant, and melanogenesis inhibition activities of methanol extract and various organic solvent fractions of Arecae Pericarpium. We examined the inhibition of lipopolysaccharide (LPS)-induced nitric oxide (NO) production in RAW 264.7 cells, 1,1-diphenyl-2-picrylhydrazine (DPPH) scavenging activity, mushroom tyrosinase inhibition activity and melanin contents. The study showed that, among all tested fractions, methylene chloride fraction showed the strongest inhibition of LPS-induced NO production in RAW 264.7 cells ($IC_{50}$ value $8.89{\mu}g/mL$) and DPPH radical scavenging activity ($EC_{50}$ value $21.39{\mu}g/mL$). Methylene chloride and ethyl acetate fractions similarly inhibited mushroom tyrosinase activity. Methanol extract exhibited strongest reduction of melanin content in B16F10 melanoma cells. Based on the bioactivity assay results, methylene chloride and ethyl acetate fractions were further separated. Eight phenolic compounds were isolated, which are dimeric syringol (1), catechol (2), 4-hydroxybenzaldehyde (3), vanillin (4), 4-hydroxyacetophenone (5), apocynin (6), protocatechuic acid (7) and 4-hydroxybenzoic acid (8). Among the isolated compounds tested, catechol showed the strongest inhibition of LPS-induced NO production in RAW 264.7 cells. Catechol also showed the concentration-dependent NF-${\kappa}B$ inhibition activity. Arecae Pericarpium might have potentials to be developed as anti-inflammatory agent or dermatological product for skin-whitening agent.
The unicellular green alga Haematococcus pluvialis has recently attracted great inter-est due to its large amounts of ketocarotenoid astaxanthin, 3,3'-dihydroxy-${\beta}$,${\beta}$-carotene-4,4'-dione, widely used commercially as a source of pigment for aquaculture. In the life cycle of H. pluvialis, astaxanthin biosynthesis is associated with a remarkable morphological change from green motile vegetative cells into red immotile cyst cells as the resting stage. In recent years we have studied this morphological process from two aspects: defining conditions governing astaxanthin biosynthesis and questioning the possible function of astaxanthin in protecting algal cells against environmental stress. Astaxanthin accumulation in cysts was induced by a variety of environmental conditions of oxidative stress caused by reactive oxygen species, intense light, drought, high salinity, and high temperature. In the adaptation to stress, abscisic acid induced by reactive oxygen species, would function as a hormone in algal morphogenesis from veget ative to cyst cells. Furthermore, measurements of both in vitro and in vivo antioxidative activities of astaxanthin clearly demonstrated that tolerance to excessive reactive oxygen species is greater in astaxanthin-rich cysts than in astaxanthin-poor cysts or astaxanthin-less vegetative cells. Therefore, reactive oxygen species are involved in the regulation of both algal morph O-genesis and carotenogenesis, and the accumulated astaxanthin in cysts can function as a protective agent against oxidative stress damage. In this study, the physiological roles of astaxanthin in stress response and cell protection are reviewed.
This study was carried out to investigate the antibacterial activity against pathogens of acne and the anti-inflammatory effect of 75% ethanol extract and its fractions from the leaves of Prunus sargentii. In the antibacterial activity by the disc diffusion assay, the extract showed the highest effect against Propionibacterium acnes, Staphylococcus aureus and Staphylococcus epidermidis in 5 mg/disc. However, the ethyl acetate fraction showed the highest antibacterial activity in 1 mg/disc. On the other hand, the hexane and chloroform fraction showed strong nitric oxide (NO) production inhibitory effect in lipopolysaccharide (LPS)-stimulated Raw 264.7 cell. In the cell viability of Raw 264.7 by MTT assay, the extract and all fractions were exhibited normal viabilities as nontoxic result. Consequently, the extract from the leaves of P. sargentii and its ethyl acetate fraction could be applicable to functional materials for antibacterial activity related fields. Moreover, the hexane and chloroform fraction could be applicable to candidate materials as anti-inflammatory agent.
