Objectives: Accumulation of cellular reactive oxygen species (ROS) leads to oxidative stress. Increased production of ROS, such as superoxide anion, or a deficiency in their clearance by antioxidant defences, mediates cellular pathology. Grewia Spp fruits are a source of bioactive compounds and have notable antioxidant activity. Although the antioxidant capacity of Grewia Spp has been studied, there is very limited evidence that links the antioxidant activities of Grewia bicolor and Grewia flava to the inhibition of free radical formation associated with damage in biological systems. Methods: This study evaluated the protective effects of Grewia bicolor and Grewia flava extracts against free radical-induced oxidative stress and the resulting cytotoxicity effect using HeLa cells. Antioxidant properties determined using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and total phenolic content (TPC) assays showed significantly higher (p < 0.05) antioxidant activity in Grewia flava (ethanol extract) than Grewia flava (water extract) and Grewia bicolor (ethanol and water extracts). Results: Using 3-(4,5-dimethylthiazol-2-yl)-2,5diphenyltetrazolium bromide or MTT assay, cytotoxicity results showed that extracts of Grewia bicolor and Grewia flava were less toxic to HeLa cells at tested concentrations compared to the untreated control. This confirmed the low toxicity of these edible fruits at the tested concentrations in HeLa cells. Furthermore, hydrogen peroxide (H2O2)-induced cell loss was effectively reduced by pre-incubating HeLa cells with Grewia bicolor and Grewia flava extracts, with Grewia flava (ethanol extract) revealing better protection. Conclusion: The effect was speculated to be associated with the higher antioxidant activity of Grewia flava (ethanol extract). Additional studies will warrant confirmation of the mechanism of action of such effects.
Hepatic antioxidant defense systems were examined in rats treated with tetrabromobisphenol-A (TBBPA), a brominated flame retardant, at the doses of 0, 250, 500 and 1,000 mg/kg for four weeks. Hepatic ratio of glutathione disulfide to glutathione (GSH) and levels of malondialdehyde, oxidative stress markers were not changed in rats treated with TBBPA. Hepatic expression of antioxidant enzymes including GSH peroxdiase-1 (GPX-1)/GSH reductase (GR), alpha-, mu- and pi-class glutathione-S-transferase (GST) and gamma-glutamylcysteine ligase catalytic subunit was determined using immunoblot analysis. Alpha-class GSTs, GPX-1 and GR levels were significantly decreased in rats treated with TBBPA at the dose of 500 or 1,000 mg/kg. These results show that TBBPA results in down-regulation of hepatic expression of antioxidant enzymes related with GSH, suggesting the liver in TBBPA-treated rats may be more sensitive to oxidants.

This study was carried out to investigate the antioxidant effect of Rhus verniciflua Stokes (RVS) extracts. The antioxidant activity of ethanol and water extract from RVS was examined on model systems and cooked beef, respectively. As concentration of RVS ethanol extract increased (1, 10, 100, and 1,000 ppm), the reductive activity of DPPH (2, 2-diphenyl-1-picrylhydrazyl) was significantly increased (14.79, 75.08, 82.02, and $83.97\%$ respectively) (p<0.05). The RVS ethanol extract (10 ppm) was showed higher antioxidant activity than control in liposome and meat homogenate (p<0.05). It had more antioxidative effect in 10 ppm RVS ethanol extract with 2 ppm $\alpha-tocopherol$ treatment The maximum antioxidant activity appeared at pH 6.0 in meat homogenate and at pH $5.0\~6.0$ in liposome. Cooked beef mixed with Rhus verniciflua Stokes water extract showed significantly lower TBARS value, POV during storage for 4 days at $4^{\circ}C$ (p<0.05). And the RVS water extract also showed strong antioxidant activity in cooked beef which accelerated NaCl-catalyzed oxidation. Therefore, the results suggest that RVS extract may be used in commercial meat products as natural antioxidant in the near future.
Antioxidant activity of 70% aqueous ethanolic extract of leaves of Justicia gendarussa (EJ) was evaluated. EJ was prepared by cold maceration method. The antioxidant potency of EJ was investigated employing various established in vitro systems, such as DPPH radical scavenging, nitric oxide (NO) scavenging, ${\beta}-carotene$ linoleic acid module system (${\beta}$ CLAMS), hydroxyl (OH) radical scavenging, anti lipid peroxidation. $IC_{50}$ values were determined in each experiment. Also, ferric ion reduction capacity of extracts in presence and absence of chelating agent (EDTA) and total antioxidant capacity were determined. Preliminary phytochemical investigation was carried out to know the nature of constituents present in the leaves and correlate it with antioxidant activity. Further total phenolic content was determined in EJ. $IC_{50}$ values of EJ were 123.09 ${\pm}$ 3.01, 643.0 ${\pm}$ 61.10, 132.3 ${\pm}$ 6.03, 68.5 ${\pm}$ 11.5 and 68.13 ${\pm}$ 1.38 ${\mu}g/mL$ in DPPH radical scavenging, NO scavenging, ${\beta}$ CLAMS, OH radical scavenging and anti lipid peroxidation activity respectively. In total antioxidant capacity assay, ascorbic acid equivalent value was found to be 205.56 ${\pm}$ 4.69 ${\mu}g/mg$ of extract. Total phenolic content was found to be 43.76 ${\pm}$ 4.27 ${\mu}g$ equivalent of gallic acid per mg of extract. Phytochemical investigation reveals the presence of flavonoids. The results indicate that EJ possess antioxidant activity and flavonoids are responsible for this activity.
Background: Explosive puffing can induce changes in the chemical, nutritional, and sensory quality of red ginseng. The antioxidant properties of ethanolic extracts of red ginseng and puffed red ginseng were determined in bulk oil and oil-in-water (O/W) emulsions. Methods: Bulk oils were heated at $60^{\circ}C$ and $100^{\circ}C$ and O/W emulsions were treated under riboflavin photosensitization. In vitro antioxidant assays, including 2,2-diphenyl-1-picrylhudrazyl, 2,2'-azinobis-3-ethyl-benzothiazoline-6-sulfonic acid, ferric reducing antioxidant power, total phenolic content, and total flavonoid content, were also performed. Results: The total ginsenoside contents of ethanolic extract from red ginseng and puffed red ginseng were 42.33 mg/g and 49.22 mg/g, respectively. All results from above in vitro antioxidant assays revealed that extracts of puffed red ginseng had significantly higher antioxidant capacities than those of red ginseng (p < 0.05). Generally, extracts of puffed red and red ginseng had high antioxidant properties in riboflavin photosensitized O/W emulsions. However, in bulk oil systems, extracts of puffed red and red ginseng inhibited or accelerated rates of lipid oxidation, depending on treatment temperature and the type of assay used. Conclusion: Although ethanolic extracts of puffed red ginseng showed stronger antioxidant capacities than those of red ginseng when in vitro assays were used, more pro-oxidant properties were observed in bulk oils and O/W emulsions.

Chlorophytum borivilianum Baker (Antharicaceae) commonly referred as 'Safed Musli' has been widely used in the Indian traditional systems of medicine to treat various diseases like rheumatism apart from having immunomodulating property and is used as general tonic. It is also known as 'Ayurvedic viagra' for its aphordisiac properties. C. borivilianum was screened for the first time to determine its antioxidant activity, isolation of the sapogenins and standardization of the isolated sapogenin fraction using HPTLC. Potent antioxidant activity of ethanolic extract was found by their ability to scavenge DPPH (84.51%), hydroxyl radical (48.95 %), ferryl bi-pyridyl complex (84.53%) along with the inhibition of lipid peroxidation (67.17%) at $100\;{\mu}g/ml$ concentration. The ethanolic extract also exhibited significant inhibition of superoxide anion radical generated by photochemiluminescence. Thus, the potent antioxidant activity validates the innumerable therapeutic claims of the plant in the traditional system especially its use as a Rasayana drug.
Dried samples of Ligustrum japonicum were extracted twice: with methylene chloride and with methanol (MeOH), respectively. The combined crude extracts were successively fractionated into n-hexane, 85% aqueous methanol (85% aq.MeOH), n-butanol (n-BuOH), and water fractions by liquid-liquid partition. Antioxidant activities of crude extract and its solvent fractions were evaluated by measuring authentic $ONOO^-$, and $ONOO^-$ generated from 3-morpholinsydnonimine (SIN-1) as well as degree of occurrence of intracellular ROS in HT 1080 cells, and genomic DNA oxidation. The 85% aq.MeOH and n-BuOH fractions exhibited the good antioxidant activity. Further purification of the 85% aq.MeOH fracition led to the isolation of Oleanolic acid (1), Maslinic acid (2), and Ursolic acid (3). All compounds showed the significant antioxidant effects in all assay systems.

Ligularia fischeri (LF) has been used to treat jaundice, scarlet-fever, rheumatoidal arthritis, and hepatic diseases. This study was carried out to determine the antioxidant activity of the extract from the leaves of LF by using various established in vitro systems with ${\alpha}$-tocopherol as positive control. The results showed that the ethanol extract of LF (0.05-0.5 mg/mL) displayed a strong free radical scavenging activity against 1,1-diphenyl-2-picrylhydrazyl, ${O_2}^{{\cdot}-}$, and $H_2O_2$ radicals. It was observed that LF extract (1 mg/mL) significantly decreased the levels of malondialdehyde (MDA) to 69 and 89% of control, measured by ferric thiocyanate (FTC) and thiobarbituric acid (TBA) test, respectively. LF extract (0.5-2 mg/mL) was also found to inhibit significantly the amount of malondialdehyde formed from liver homogenate to 69-56% of control. Similarly, in the high $Fe^{2+}/ascorbate$ induction system LF extract (1-2 mg/mL) inhibited carbonyl formation measured by 2,4-dinitrophenylhydrazine reaction to 89-79% of control. Like antioxidant activity, the reducing power of LF extract was excellent at concentration of 0.5-2 mg/mL. These results indicate that LF could be used as a potential source of natural antioxidant.
This study was undertaken to determine free radical scavenging capacity and oxidative DNA damage protecting activity of methanol extract of red tea stem. The extract was subjected to assess their antioxidant potential using various in vitro systems such as $DPPH^{\bullet}$, $ABTS^{{\bullet}+}$, super oxide and nitric oxide free radicals and it exhibited $IC_{50}$ values of $68.88{\pm}1.1$, $12.08{\pm}0.65$, $404.38{\pm}1.6$, $93.6{\pm}2.7{\mu}g/mL$ respectively. Red tea extract also showed ferric reducing ability (FRAP) with 2606.85 mmol Fe (II)/g of extract. Furthermore, Methanol extract of red tea stem showed significant DNA damage protecting activity in concentration dependent manner against $H_2O_2+UV$ induced photolysis on pUC19 plasmid DNA. Results of this study showed that the methanol extract of Red Tea stem has strong antioxidant potential along oxidative DNA damage protecting capacity that would be the significant sources of natural antioxidants, which might be helpful in preventing the progress of various oxidative stress generated diseases. Further study is necessary for isolation and characterization of the active antioxidants, which may serve as a potential source of natural antioxidant.

Oxidative stress is prerequisite for the development of atherosclerosis. Apart from the traditional risk factors that contribute to this devastating condition, in the past few decades, much attention has been focused on plasma total homocysteine mainly because of its strong association with coronary artery disease. It has been suggested that homocysteine induces oxidative stress and hence the present work was undertaken to assess the total homocysteine status and plasma total antioxidant capacity in the acute myocardial infarction (AMI) patients among Tamil population. The study subjects included only the Tamil population. Blood samples were collected from 100 AMI patients and 100 controls. Plasma was separated and the total antioxidant status was assessed as a measure of ferric reducing power of antioxidants using spectrophotometric method. Plasma total homocysteine concentrations were assessed by automated chemiluminescence method. While Total antioxidant status was significantly decreased, the plasma homocysteine concentrations were elevated in AMI patients compared to the controls. However, there was no correlation between the homocysteine levels and total antioxidant status. The findings of this study may have therapeutic implications, including food sources rich in antioxidants for all AMI patients to minimize the effect of free radicals formed during oxidative stress among Tamil population.
